Glial fibrillary acidic protein levels are associated with global histone H4 acetylation after spinal cord injury in rats

Emerging evidence has suggested global histone H4 acetylation status plays an important role in neural plasticity. For instance, the imbalance of this epigenetic marker has been hypothesized as a key factor for the development and progression of several neurological diseases. Likewise, astrocytic reactivity-a wellknown process that markedly influences the tissue remodeling after a central nervous system injury-is crucial for tissue remodeling after spinal cord injury（SCI）. However, the linkage between the above-mentioned mechanisms after SCI remains poorly understood. We sought to investigate the relation between both glial fibrillary acidic protein（GFAP） and S100 calcium-binding protein B（S100B）（astrocytic reactivity classical markers） and global histone H4 acetylation levels. Sixty-one male Wistar rats（aged ~3 months） were divided into the following groups： sham; 6 hours post-SCI; 24 hours post-SCI; 48 hours post-SCI; 72 hours post-SCI; and 7 days post-SCI. The results suggested that GFAP, but not S100B was associated with global histone H4 acetylation levels. Moreover, global histone H4 acetylation levels exhibited a complex pattern after SCI, encompassing at least three clearly defined phases（first phase： no changes in the 6, 24 and 48 hours post-SCI groups; second phase： increased levels in the 72 hours post-SCI group; and a third phase： return to levels similar to control in the 7 days post-SCI group）. Overall, these findings suggest global H4 acetylation levels exhibit distinct patterns of expression during the first week post-SCI, which may be associated with GFAP levels in the perilesional tissue. Current data encourage studies using H4 acetylation as a possible biomarker for tissue remodeling after spinal cord injury.

Contextual Fear Extinction and Re-Extinction in Carioca High-and Low-Conditioned Freezing Rats

We recently reported two novel breeding lines of rats known as Carioca High- and Low-conditioned Freezing (CHF and CLF), based on defensive freezing responses to contextual cues previously associated with electric footshock. The present study used animals of the 8th generation of our selective breeding program to investigate both contextual fear extinction and re-extinction. The results consistently showed that CHF animals froze more than CLF animals. Long extinction training was able to extinguish phenotypic differences between lines, but the divergence was restored after just one fear reacquisition training session. These differences disappeared again during re-extinction training. The possible neural mechanisms involved in these two types of learning are discussed.

We review recent studies of cavity switching induced by the optical injection of free carriers in micropillar cavities containing quantum dots.Using the quantum dots as a broadband internal light source and a streak camera as detector,we track the resonance frequencies for a large set of modes with picosecond time resolution.We report a record-fast switch-on time constant(1.5 ps)and observe major transient modifications of the modal structure of the micropillar on the 10 ps time scale:mode crossings are induced by a focused symmetric injection of free carriers,while a lifting of several mode degeneracies is observed when off-axis injection breaks the rotational symmetry of the micropillar.We show theoretically and experimentally that cavity switching can be used to tailor the dynamic properties of the coupled QD-cavity system.We report the generation of ultrashort spontaneous emission pulses(as short as 6 ps duration)by a collection of frequency-selected QDs in a switched pillar microcavity.These pulses display a very small coherence length,attractive for ultrafast speckle-free imaging.Moreover,the control of QD-mode coupling on the 10 ps time scale establishes cavity switching as an appealing resource for quantum photonics.

BACKGROUND Obesity is a global health problem that is continuing to increase in the young population.In Brazil,the frequency of obesity in 2018 was 19.8%.Several comorbidities are directly associated with obesity,such as non-alcoholic fatty liver disease(NAFLD),which is considered the most common liver disorder in Western countries and affects up to 46%of adults.Bariatric surgery is effective in treating obesity and can improve NAFLD;however,the effect of bariatric surgery on body composition,phase angle(PA),and improving NAFLD needs to be further studied.AIM To analyze the PA in the postoperative period of bariatric surgery and to correlate it with changes in body composition and liver disease.METHODS This study is a retrospective cohort study of the analysis of the medical records of patients undergoing bariatric surgery in a reference center of a teaching hospital in Porto Alegre over a 2-year period.Patients older than 18 years whose record contained all information relevant to the study were included.The data analyzed were body composition and PA through electrical bioimpedance and NAFLD through liver biopsy in the pre-and postoperative period.The level of significance adopted for the statistical analyses was 5%.RESULTS We evaluated 379 patients with preoperative data.Regarding PA,169 patients were analyzed,and 33 patients had liver biopsy pre-and postoperatively with NAFLD information.In total,79.4%were female,with a mean age of 39.1±10.6 years.The average body mass index(BMI)was 45.9±7.5 kg/m².The PA showed a mean of 5.8±0.62°in the preoperative period and a significant reduction in the postoperative period.A postoperative reduction in body composition data(skeletal muscle mass,fat percentage,fat mass,body cell mass,BMI and visceral fat area)was shown as well.Regarding liver disease,all patients presented a reduction in the degrees and stages of liver disease in the postoperative period,and some had no degree of liver disease at all.CONCLUSION PA decreased after bariatric surgery,with a direct correlation

determination of the Antiretroviral Drug Zidovudine in Diluted Alkaline Electrolyte by Adsorptive Stripping Voltammetry at the Mercury Film Electrode

This paper describes a stripping method for the determination of zidovudine at the submicromolar concentration levels. This method is based on the controlled adsorptive accumulation of zidovudine at the thin-film mercury electrode, followed by a linear-sweep stripping voltammetry measurement of the surface species. Optimal experimental conditions include a NaOH solution of 2.0 × 10–3 mol●L–1 (sup-porting electrolyte), an accumulation potential of –0.30 V and a scan rate of 100 mV?s–1. The response of zidovudine is linear over the concentration range 0.01 - 0.08 ppm. After an accumulation time of 5 minutes, the detection limit was found to be 0.67 ppb (2.5 × 10–9 mol●L–1). More convenient methods to measure zidovudine concentration in the presence of the didanosine, acyclovir, nevirapine, lamivudine, and efavirenz, were also investigated. The presence of zidovudine together with ATP or ssDNA demonstrates the utility of this method.

Biogas in Brazil： A Governmental Agenda

Environmental issues have become an important aspect in the energy sector and also in governmental policies all over the world. Renewable energy plays a decisive role in a cleaner energy production with less environmental impacts. In this new world scenario, several countries create incentive programs aiming to increase the renewable energy share in their energy balance. Biogas is an interesting example of a smart and rational use of organic waste material. Nevertheless, international experience shows that its production relays mainly on government incentives. This paper investigates what the Brazilian government role is to introduce and promote the biogas industry in the country. We estimate the production of agricultural waste energy using biogas derived from cattle and swine waste and analyze the Brazilian legal and regulatory framework for renewable energy, focusing on biogas production, transport and sale. The results point to an unexplored potential of the use of cattle and swine waste for biogas production in Brazil. However, this potential can only become a reality if some legal and regulatory issues are solved. Brazilian government agenda has to include not only filling the legal and regulatory blanks but also creating incentives for the use of biogas.

Is the Distribution of Microorganisms and Peritonitis Affected by Seasonality in Peritoneal Dialysis?

Introduction: Peritonitis continues to be the main complication for patients on peritoneal dialysis (PD). Objective: To determine the frequency of peritonitis according to the disease-causing microorganism and its distribution throughout the year, linking to seasonality. Methods: A retrospective study conducted in the Dialysis Unit of the Hospital São Lucas, PUCRS (HSL-PUCRS). Patients undergoing PD between January 1984 and September 2013 were included. descriptive statistics were used and Fisher’s exact test with Monte Carlo simulation for comparison between the categorical variables. Results: Of 415 evaluated patients, 66% had at least one episode of peritonitis with an incidence rate of 0.68 episode/year. There were 601 peritonitis episodes in total. The most common microorganism was coagulase-negative Staphylococcus (26.6%, n = 160), followed by Staphylococcus aureus (16.3%, n = 98), with 16.3% of the sample being negative culture. Most episodes occurred in the months of January (10.3%, n = 62) and May (10.1%, n = 61), while June had the lowest occurrence (5.2%, n = 31). The objectives of this study were to assess the safety and efficacy of intralesionally administered diclofenac in the treatment of cutaneous neurofibromas in patients with NF1. This was a proof-of-concept, prospective, safety and efficacy study of the effect of intralesionally administered diclofenac 25 mg/ml given once a week to 3 target cutaneous neurofibromas for 4 consecutive weeks. Overall, there was no significant change in neurofibroma size. During the study, some treated lesions developed signs of necrosis and fell off after a few weeks, but none of the control neurofibromas fell off. There were no significant changes in patient’s vital signs. A few adverse events occurred, mostly at the injection sites. During the study, some neurofibromas developed necrosis after the diclofenac injections and eventually detached from the patient. Overall, diclofenac was well tolerated, suggesting minimal systemic exposure, which required confirmation and further studies, including bioavailability analysis.

Analysis of the Mechanical Properties of the Sintered Composite FeCuC in Two Different Atmospheres

This study aimed to analyze the mechanical properties of the compound FeCuC when compacted at varying pressures and sintered in two different types of furnaces. Besides the different models of furnace, the working atmospheres were varied: one is being composed with argon gas and another constituted with a balancing nitrogen and hydrogen. Atmospheres vary with the amount of production and the type of equipment used. The compound generated is used in the manufacture of rings for mechanical seals and is currently manufactured by the sintering process in passing furnace. The sintering was performed in a static furnace with argon atmosphere and compared with the same compound sintered in passage furnace with hydrogen and nitrogen atmosphere. The analysis of the properties of the tested material was performed with the aid of metallography using a scanning electron microscope, which verified the particle size distribution, chemical elements and pores present. Brinell hardness and Vickers micro hardness tests were also used to analyze the properties of this material after completion of the two processes. Thus, the research carried out has shown that variations may occur in the mechanical properties when processed in different furnace types and different sintering atmospheres.
