Effect of Organic vs. Inorganic Selenium Supplementation on Expression of Selected Defensin Genes in Somatic Milk Cells of Dairy Cows

The aim of the study was to evaluate the impact of organic selenium supplementation （selenium yeast） vs. inorganic selenium （selenium selenite） on the expression of selected defensin genes in milk somatic cells （MSC） of dairy cows. Sixteen polish Holstein-Friesian cows in second parity, with similar milk yield and milk composition were randomly divided into two equal groups. At the beginning of experiment, the animals were in the middle of their lactation curve （150, standard deviation （SD） = 26 d） and this experiment lasted 90 d. The basic diet, for both groups, consisted of corn silage, wilted grass silage and concentrates. The diet of control group was supplemented with commercial mineral and vitamin mixture with inorganic selenium and the for the experimental group daily selenium need was covered by the addition of Se-yeast （6 mg Se/cow/d）. Milk samples were collected three times during experiment （before experiment; on day 55; after the end the experiment）. Total RNA was isolated from milk somatic cells and the levels of transcripts of bovine β1-defensin （DEFB1）, β4-defensin （BNBD4）, β5-defensin （BNBDS）, β10-defensin （BNBD10） and lingual antimicrobial peptide （LAP） were measured with real time PCR, using glyceraldehyde 3-phosphate dehydrogenase （GAPDH） as a reference gene. The expressions of almost all studied genes were influenced by environmental conditions （very low and negative temperature）. However, BNBD4 （P 〈 0.05） and LAP （P ≤ 0.001 ） genes were influenced by selenium supplementation but in opposite ways, depending on the form of Se. These findings support that selenium is an important factor affecting the mRNA level in MSC, but the effect of the form of selenium might depend on genes.

The effect of the dietary inclusion of pea seeds of colored-flowered and white-flowered varieties on gastrointestinal function in turkeys

This study investigated the effects of dietary replacement of soybean meal(SBM)with graded levels of pea seeds(PS)on the gastrointestinal function of *** of 2 pea varieties,a colored-flowered variety and a white-flowered variety(CFP and WFP,respectively)were fed to 56-d-old birds for 8 wk.A total of 539 female Hybrid turkeys were allocated to 7 groups,each group consisted of 7 pens with 11 birds per *** experiment had a 2-factorial design,with 3 dietary inclusion levels of PS(100,200 and 300 g/kg)and 2 pea varieties(CFP and WFP).The control group(diets without PS)was compared with CFP and WFP treatments by simple contrast *** comparison with CFP seeds,WFP seeds contained 7-fold less tannins(0.67 vs.4.66 g/kg)and less non-starch polysaccharides(NSP,117.8 vs.132.7 g/kg),but more trypsin inhibitors(1.34 vs.0.98 g/kg)and starch(489 vs.455 g/kg).A rise in the PS content of diets from 100 to 200 and 300 g/kg increased the weight of the small intestine(P=0.031)and the dry matter(DM)content of intestinal digesta(P=0.001),but it had no effect on the pH of *** the highest PS content differentiated the concentrations of short-chain fatty acids(SCFAs)in the small intestinal digesta(WFP>CFP,P=0.008),whereas PS did not cause any changes in the morphological parameters of the small intestinal *** dietary inclusion of PS had no influence on the levels of acetate,butyrate,putrefactive SCFAs or total SCFAs in the cecal *** from increasing the activities of b-glucosidase(P=0.017)and b-galactosidase(P=0.025),pea varieties did not affect the activities of the analyzed cecal microbial ***,CFP seeds decreased the DM content(P=0.041)and increased the pH of cecal digesta,compared with WFP seeds(P=0.013).The results of this study,pointing to a few differences in the functional parameters of the small intestine and cecum,indicate that tannins are not a factor differentiating the suitability of CFP and WFP seeds in the nutrition of f

AIM:To assess the mucosa-associated bacterial microflora and mucus layer in adolescents with inflammatory bowel disease(IBD) .METHODS:Sixty-one adolescents(mean age 15 years,SD ± 4.13) were included in the *** biopsies from inflamed and non-inflamed mucosa of IBD patients and from controls with functional abdominal pain were cultured under aerobic and anaerobic *** number of microbes belonging to the same group was calculated per weight of collected *** mucus thickness in frozen samples was measured under a fluorescent ***:The ratios of different bacterial groups in inflamed and non-inflamed mucosa of IBD patients and controls were specific for particular *** *** predominant in the inflamed mucosa of Crohn's disease(CD) patients(80% of all bacteria) ,and Lactobacillus *** predominant in ulcerative colitis patients(90%) .The differences were statistically significant(P = 0.01-0.001) .Lower number of bifidobacteria was observed in the whole IBD group.A relation was also found between clinical and endoscopic severity and decreased numbers of Lactobacillus and,to a lesser extent,of Streptococcus in biopsies from CD *** mucus layer in the inflamed sites was significantly thinner as compared to controls(P = 0.0033) and to non-inflamed areas in IBD patients(P = 0.031) .CONCLUSION:The significantly thinner mucosa of IBD patients showed a predominance of some aerobes specific for particular diseases,their numbers decreased in relation to higher clinical and endoscopic activity of the disease.

Single crystal of Eu3þ-doped cadmium molybdate(Cd0.9268M0.0244Eu0.0488MoO_(4),where M denotes cationic vacancies)has been successfully grown by the Czochralski method in air and under 1 MPa.X-ray diffraction analysis indicates that as-grown single crystal exhibits tetragonal scheelite-type structure(a=b=5.16188(14)Å;c=11.2080(5)Å;space group I41/a).Eu^(3+) þions do not show long-range order and they are randomly distributed in CdMoO_(4) framework substituting Cd^(2+ )þ*** diffuse reflectance measurements revealed very close optical band gap(Eg)values,i.e.-1.74 eV along[100]and[001]crystallographic directions that are twice smaller than Eg of microcrystalline pure CdMoO_(4) as well as powder Eu3þ-doped single *** and electrical studies of Eu3þ-doped cadmium molybdate single crystal showed a paramagnetic and n-type semiconducting behaviour with the metal-insulator transition above 350 K along both crystallographic *** results analysis using the Cole-Cole fit function revealed that the dipole relaxation process has different time scale depending on the crystallographic direction and exhibits Arrhenius temperature dependence for both studied *** fact is accompanied by the colossal dielectric permittivity withεr>8,*** above results are considered in the framework of narrow europium multiplets of energy comparable to thermal energy.

During the acquisition of electroencephalographic(EEG) signals, data may be missing or corrupted by noise and artifacts. To reconstruct the incomplete data, EEG signals are firstly converted into a three-order tensor(multi-dimensional data) of shape time × channel × trial. Then, the missing data can be efficiently recovered by applying a tensor completion method(TCM).However, there is not a unique way to organize channels and trials in a tensor, and different numbers of channels are available depending on the EEG setting used, which may affect the quality of the tensor completion results. The main goal of this paper is to evaluate the robustness of EEG completion methods with several designed parameters such as the ordering of channels and trials, the number of channels, and the amount of missing data. In this work, the results of completing missing data by several TCMs were compared. To emulate different scenarios of missing data, three different patterns of missing data were ***, the amount of missing data on completion effects was analyzed, including the time lengths of missing data and the number of channels or trials affected by missing data. Secondly, the numerical stability of the completion methods was analyzed by shuffling the indices along channels or trials in the EEG data tensor. Finally, the way that the number of electrodes of EEG tensors influences completion effects was assessed by changing the number of channels. Among all the applied TCMs, the simultaneous tensor decomposition and completion(STDC) method achieves the best performance in providing stable results when the amount of missing data or the electrode number of EEG tensors is changed. In other words, STDC proves to be an excellent choice of TCM, since permutations of trials or channels have almost no influence on the complete results. Towards an optimum saturable absorber for the multi-gigahertz harmonic mode locking of fiber lasers

Ultra-high-pulse-repetition-rate lasers are essential for a number of applications, including, e.g., optical communication and ablation-cooled material processing. Despite several techniques to generate pulses with gigahertzrange repetition rate, incorporating mainly short-length resonators, more widespread applications are still limited by the lack of a robust, simple, and cost-effective solution. Here, we report for the first time, to the best of our knowledge, fully passive harmonic mode locking in an all-polarization-maintaining(PM) fiber laser. The design guarantees a fixed polarization state and stable operation, where the cavity harmonic number is controlled by the pump power only. Self-starting operation is provided by the antimony telluride(Sb2Te3) thin-film saturable absorber(SA), which facilitates multiple pulse operation. The SA acts by means of low modulation depth, low saturation fluence, and an inverse slope in the saturable absorption curve. The optimum features of the SA and limiting factors for high-repetition-rate pulse generation in this regime of operation are discussed. As a result, 2.2 ps pulses with 3 GHz repetition rate are generated at 1560 nm wavelength. The study reports a new approach towards an optimal SA for multi-gigahertz pulse generation in practical, all-PM fiber lasers.
