The rapid detection of glyphosate resistance in goosegrass（Eleusine indica） will enhance our ability to respond to new resistant populations of this major weed. Chlorophyll fluorescence（Fluo） and P700（reaction center chlorophyll of photosystem I） absorbance were analyzed in one biotype of goosegrass that is resistant to glyphosate and in another that remains sensitive to the herbicide. Both biotypes were treated with a foliar spray of glyphosate. Differences in photosystem II maximum quantum yield（Fv/Fm）, effective photochemical quantum yield（Y（II））, and non-photochemical quenching（NPQ） between the biotypes increased over time. Values for Fv/Fm and Y（II） differed between the two biotypes 24 h after treatment（HAT）. Differentiated activities and energy dissipation processes of photosystem II（PSII） and energy dissipation processes of photosystem I（PSI） were manifested in the two biotypes 24 HAT with 20 mmol L–1 glyphosate. Differentiated energy dissipation processes of PSI were still apparent 24 HAT with 200 mmol L–1 glyphosate. These results indicate that the Fluo parameters related to PSII activity and energy dissipation and the P700 parameters related to energy dissipation are suitable indicators that enable rapid detection of glyphosate resistance in goosegrass.

Glyphosate has been used worldwide for nearly 40 years,and 30 types of resistant weeds have been *** is mass-produced and widely used in China,but few studies and reports on glyphosate-resistant weeds and resistance mechanisms *** studies found a goosegrass species with high glyphosate resistance from orchards in South China and its glyphosate resistant mechanism was described in this *** cDNAof 5-enolpyruvylshikimate-3-phosphate synthase（EPSPS,EC 2.***.1.19）,the target enzyme of glyphosate,was cloned from the glyphosate-resistant and-susceptible goosegrass,respectively,and referred as EPSPS-R and *** Pro106 residue was known to be involved in the glyphosate resistance in most goosegrass ***,sequence analysis did not find the mutation at the Pro106 residue in the R biotype EPSPS amino acid *** residue 133 and 382 was mutated in the R biotype EPSPS amino acid sequence instead,but it did not affect the EPSPS-S and EPSPS-R genes sensitivities to ***-PCR and Western blot analyses suggested that EPSPS mRNA and protein are mainly present in the shoot tissues both in the R and S goosegrass *** EPSPS-R rapidly responds to the glyphosate in R-biotype goosegrass and the induced expression was detected at 12 h post glyphosate *** mRNA and protein expression of EPSPS-R increased constantly as the increasing concentration of ***,the expression of the EPSPS-S was not induced significantly by glyphosate in the S goosegrass *** of real-time PCR results showed that the copy number of the EPSPS in R-biotype goosegrass was 4.7 times higher than that in the S goosegrass *** the results implied that EPSPS gene amplification might mainly caused the glyphosate resistance of a goosegrass population collected from orchards in South China.

The insecticide chlorantraniliprole exhibits good efifcacy and plays an important role in controlling the diamondback moth, Plutella xylostella Linnaeus. However, resistance to chlorantraniliprole has been observed recently in some ifeld populations. At present study, diamondback moths with resistance to chlorantraniliprole （resistant ratio （RR） was 82.18） for biochemical assays were selected. The assays were performed to determine potential resistance mechanisms. The results showed that the selected resistant moths （GDLZ-R） and susceptible moth could be synergized by known metabolic inhibitors such as piperonyl butoxide （PBO）, triphenyl phosphate （TPP） and diethyl-maleate （DEM） at different levels （1.68-5.50-fold and 2.20-2.89-fold, respectively）, and DEM showed the maximum synergism in both strains. In enzymes assays, a high level of glutathione-S-transferase （GST） was observed in the resistant moth, in contrast, moths that are susceptible to the insecticide had only 1/3 the GST activity of the resistant moths. The analysis of short-term exposure of chlorantraniliprole on biochemical response in the resistant strain also showed that GST activity was signiifcantly elevated after exposure to a sub-lethal concentration of chlorantraniliprole （about 1/3 LC50, 12 mg L-1） 12 and 24 h, respectively. The results show that there is a strong correlation between the enzyme activity and resistance, and GST is likely the main detoxiifcation mechanism responsible for resistance to chlorantraniliprole in P. xylostella L., cytochrome P450 monooxygenase （P450） and carboxy-lesterase （CarE） are involved in to some extent.

Population Genetic Analysis of Blumeria graminis f. sp. tritici in Qinghai Province, China

To gain more precise information about molecular genetic variation for wild populations of Blumeria graminis f. sp. tritici from Qinghai Province, China, 38 single-colony isolates were purified from samples collected from Haidong District, Xining City and Hainan Tibetan Autonomous Prefecture in 2010. The virulence of 21 isolates among them was tested at seedling stage on 34 wheat cultivars（lines） carrying known powdery mildew（Pm） resistant genes. The results showed that V1 a, V3 a, V3 c, V3 e, V5 a, V6, V7, V8 and V19 had high virulence frequencies（〉75%）, indicating a wide distribution; and V1 c, V5 b, V12, V13, V16, V21, VXBD, V2＋6, V2＋Mld and V4＋8, with less distribution, appeared to be lower in frequencies（0-20%）. The Nei＇s gene diversity（H）, Shannon＇s information index（I） and the percentage of polymorphic loci（P） were 0.23, 0.35 and 67.65%, respectively, which revealed a virulent diversity. The results from single nucleotide polymorphisms（SNPs） of 38 isolates showed that three housekeeping genes were found to contain a total of 9 SNP sites. 10 haplotypes（H1-H10） were inferred from the concatenated sequences, with 1 haplotype（H1） comprising of over 55% of Qinghai population. Phylogenic analysis did not show obvious geographical subdivision between the isolates. A multilocus haplotype network presented a radial structure, with H1 in the central as an inferred ancestor. Using analysis of molecular variance（AMOVA）, we found 1.63% of the total variation was among populations and 98.37% within populations, with a low fixations index（FST=0.01634, P〈0.05）. This revealed a relatively high genetic diversity but a low genetic divergence in Qinghai population. Moreover, the molecular data on gene flow（Nm=6.32） confirmed the migration of pathogen populations among areas in Qinghai Province.

Release performance and sustained-release efficacy of emamectin benzoate-loaded polylactic acid microspheres

High-performance liquid chromatography （HPLC） was employed to determine drug release rates based on emamectin benzoate concentrations in the medium. Release kinetics equations were used to fit the drug release behavior. The effects of particle size and release medium pH on the release rate were also investigated. The indoor toxicity of emamectin benzoate-loaded polylactic acid microspheres on the diamondback moth larva （Plutella xylostella） was studied to explore drug sustained-release performance. In acidic and neutral media, the drug release behavior of the microspheres was in accord with the first-order kinetics equation. Increasing the spray dosage of emamectin benzoate-loaded polylactic acid microspheres initially resulted in an equivalent insecticidal efficacy with the conventional emamectin benzoate microemulsion. However, the drug persistence period was four-fold longer than that observed using the conventional formulation. The developed emamectin benzoate-loaded polylactic acid microspheres showed dramatic sustained-release performance. A treatment threshold of greater than 35 mg mL-1 was established for an efficient accumulated release concentration of emamectin benzoate-loaded microspheres.

American sloughgrass(Beckmannia syzigachne(Steud.) Fernald) is one of the most competitive and malignant weeds in rice-wheat rotation fields in China. American sloughgrass populations in the Jiangsu Province of China became less sensitive to acetohydroxyacid synthase(AHAS) inhibitors after repeated application for many years in these areas. Two suspected resistant American sloughgrass populations(R1 and R2) collected in the field were detected the resistance to inhibitors of AHAS in whole-plant dose-response assays, compared to the susceptible(S) population. These assays indicated that R1 showed low resistance to mesosulfuron-methyl(3.32-fold), imazapic(2.84-fold) and pyroxsulam(1.55-fold), moderate resistance to flazasulfuron(4.67-fold) and pyribenzoxim(7.41-fold), and high resistance to flucarbazone(11.73-fold). However, using a combination of the cytochrome P450 inhibitor, malathion, with mesosulfuron-methyl resulted in a reduction in R1 resistance relative to mesosulfuron-methyl alone. Furthermore, R2 was highly resistant to flazasulfuron(34.90-fold), imazapic(11.30-fold), flucarbazone(49.20-fold), pyribenzoxim(12.94-fold), moderately resistant to mesosulfuron-methyl(9.77-fold) and pyroxsulam(6.26-fold), and malathion had no effect on R2 resistance to mesosulfuron-methyl. The fulllength of AHAS genes was sequenced and the AHAS enzymes were assayed in vitro in order to clarify the mechanism of resistance to AHAS inhibitors in R1 and R2 populations. The results demonstrated that R2 had a Pro-197-Ser mutation in the AHAS gene, and the sensitivity of R2 to the five AHAS inhibitors was decreased, which may result in R2 resistance to AHAS inhibitors. There was no mutation in the AHAS gene of R1, and there were no significant differences in enzyme sensitivity between susceptible(S) and resistant(R1) populations. An enhanced metabolism may be the main mechanism of R1 resistance to AHAS inhibitors.

Management of pesticide residues in China

This paper reviewed management of pesticide residues in China including laws and regulations, the supervision system, the standard system, and the quality and safety of agricultural products. The process of establishment and internationalization of standards for pesticide residues were also discussed. Results indicate that the progress of the management of pesticide residues has been steadily made in China. However, the following aspects which refer to updates to regulations, supervising efficiency, standard system, risk assessment, international cooperation and communication, should be further improved. China should draw lessons from international experience, and then establish its own management system, which focuses on pesticides controls by strictly following relevant laws and technical standards to ensure the quality and safety of agricultural products.

Assessment of Rice Cultivars in China for Field Resistance to Aphelenchoides besseyi

The effect ofAphelenchoides besseyi on 27 cultivars of rice （23japonica and 4 indica） was assessed in the field for two seasons during 2010 and 2011. The vigorous pathogenic nematodes culturing on Botrytis cinerea were used for this experiment. Inoculation was carried out at the tilling stage; the growth parameters and nematode population were recorded at the end of growth of rice plants. The results showed that the cultivars differed in their response to infection. Most of cultivars were lack of the characteristic symptom of white tip, which was seen less frequently than the other two symptoms, namely small grains and erect panicles; moreover, the expression of symptoms was probably hereditary. The infection lowered the values of all the measured biological parameters, namely length of the stem and of the panicle, the number of filled grains per panicle, and 100-grain weight, in all the cultivars. The final nematode population indicated that the threshold of economic damage had also been exceeded in 10 cultivars, and none of them was immune. Three japonica cultivars proved most vulnerable whereas Tetep, an indica type, showed a level of resistance potentially useful in controlling A. besseyi.

Chemical mutagenesis and soybean mutants potential for identification of novel genes conferring resistance to soybean cyst nematode

The resistance of soybean(Glycine max(L.) Merr.) to soybean cyst nematode(SCN, Heterodera glycines Ichinohe), which is a devastating pathogen in soybean production and causes a large quantity of annual yield loss worldwide, can shift during the long-term interaction and domestication. It is vital to identify more new resistance genetic sources for identification of novel genes underlying resistance to SCN for management of this pathogen. In the present study, first, two ethane methylsulfonate-mutagenesis soybean M2 populations of PI 437654, which shows a broad resistance to almost all of SCN races, and Zhonghuang 13, which is a soybean cultivar in China conferring strong resistance to lodging, were developed. Many types of morphological phenotypes such as four-and five-leaflet leaves were observed from these two soybean M2 populations. Second, 13 mutants were identified and confirmed to exhibit alteration of resistance to SCN race 4 through the forward genetic screening of 400 mutants of the PI 437654 M2 population, the rate of mutants with alteration of SCNinfection phenotype is 3.25%. Third, these identified mutants were further verified not to show any changes in the genomic sequences of the three known SCN-resistant genes, GmSHMT08, GmSNAP18 and GmSANP11, compared to the wildtype soybean; and all of them were still resistant to SCN race 3 similar to the wild-type soybean. Taken together, we can conclude that the 13 mutants identified in the present study carry the mutations of the new gene(s) which contribute(s) to the resistance to SCN race 4 in PI 437654 and can be potentially used as the genetic soybean sources to further identify the novel SCN-resistant gene(s).
