Antagonism of Transcription Factor MYC2 by EDS1/PAD4 Complexes Bolsters Salicylic Acid Defense in Arabidopsis Effector-Triggered Immunity

In plant immunity, pathogen-activated intracellular nucleotide binding/leucine rich repeat （NLR） receptors mobilize disease resistance pathways, but the downstream signaling mechanisms remain obscure. Enhanced disease susceptibility 1 （EDS1） controls transcriptional reprogramming in resistance triggered by Toll-lnterleukinl-Receptor domain （TIR）-family NLRs （TNLs）. Transcriptional induction of the salicylic acid （SA） hormone defense sector provides one crucial barrier against biotrophic pathogens. Here, we present genetic and molecular evidence that in Arabidopsis an EDS1 complex with its partner PAD4 inhibits MYC2, a master regulator of SA-antagonizing jasmonic acid （JA） hormone pathways. In the TNL immune response, EDSl/PAD4 interference with MYC2 boosts the SA defense sector independently of EDS1-induced SA synthesis, thereby effectively blocking actions of a potent bacterial JA mimic, coronatine （COR）. We show that antagonism of MYC2 occurs after COR has been sensed inside the nucleus but before or coincident with MYC2 binding to a target promoter, pANAC019. The stable interaction of PAD4 with MYC2 in planta is competed by EDS1-PAD4 complexes. However, suppression of MYC2-promoted genes requires EDS1 together with PAD4, pointing to an essential EDS1-PAD4 heterodimer activity in MYC2 inhibition. Taken together, these results uncover an immune receptor signaling circuit that intersects with hormone pathway crosstalk to reduce bacterial pathogen growth.

Jasmonic acid-isoleucine formation in grapevine(Vitis vinifera L.) by two enzymes with distinct transcription profiles

The plant hormone jasmonic acid （JA） is essential for stress responses and the formation of reproductive organs, but its role in fruit development and ripening is unclear. Conjugation of JA to isoleucine is a crucial step in the JA signaling pathway since only JA-Ile is recognized by the jasmonate receptor. The conjugation reaction is catalyzed by JA-amido synthetases, belonging to the family of Gretchen Hagen3 （GH3） proteins. Here, in vitro studies of two grapevine （Vitis vinifera L. cv Shiraz） GH3 enzymes, VvGH3-7 and VvGH3-9, demonstrated JA-conjugating activ- ities with an overlapping range of amino acid substrates, including isoleucine. Expression studies of the correspond- ing genes in grape berries combined with JA and JA-lle measurements suggested a primary role for JA signaling in fruit set and cell division and did not support an involvement of JA in the ripening process. In response to methyl JA （MeJA） treatment, and in wounded and unwounded （distal）leaves, VvGH3-9 transcripts accumulated, indicating a participation in the JA response. In contrast, VvGH3-7 was unresponsive to MeJA and local wounding, demonstrating a differential transcriptional regulation of VvGH3-7 and VvGH3- 9. The transient induction of VvGH3-7 in unwounded, distal leaves was suggestive of the involvement of an unknown mobile wound signal.

Identification and Characterization of Genes Responsible for Drought Tolerance in Rice Mediated by Pseudomonas fluorescens

Drought is one of the major abiotic stresses which adversely affect crop plants limiting growth and yield *** and functional characterization of drought stress-induced genes has contributed to a better understanding of how plants respond and adapt to the drought *** the present study,differential display technique was employed to study the gene expression of rice plants at the reproductive stage that were subjected to drought stress by withholding water,Pseudomonas fluorescens strain(Pf1) treated plants subjected for drought stress by withholding water and control(well-watered).Differentially expressed c DNAs of six genes(COX1,PKDP,b ZIP1,AP2-EREBP,Hsp20 and COC1) were identified,cloned and ***-time q PCR analysis showed that all the six genes were upregulated in drought-stressed plants treated with *** revealed that the remarkable influence of Pf1 colonization leads to drought tolerance at the reproductive *** results showed that high levels of gene expression in plants lacking adequate water can be remarkably influenced by Pf1 colonization,which might be a key element for induced systemic tolerance by microbes.

Aphid populations showing differential levels of virulence on Capsicum accessions

The green peach aphid,Myzus persicae,is one of the most threatening pests in pepper cultivation and growers would benefit from resistant ***,we identified two Capsicum acessions as susceptible and three as resistant to *** using an aphid population originating from the Netherlands(NL).Later on we identified an aphid population originating from a diferent gcographical region(Switserland,SW)that was virulent on all tested Capsicum *** objeetive of the current work is to describe in detail diferent aspects of the interaction between two aphid populations and two sclected Capsicum acessions(one that was susceptible[PB2013046]and one that was resistant[PB2013071]to population NL),including biochemical processes *** penetration graph(EPG)recordings showed similar feeding activities for both aphid populations on *** acession PB2013071 the aphid population sw was able to devote significantly more time to phloem ingestion than population *** also studied plant defense response and found that plants of acession PB2013046 could not induce an accumulation of reactive oxygen species and callose formation after infestation with either aphid ***,plants of PB2013071 induced a stronger defense response after infestation by population NL than after infestation by population *** on these results,population SW of *** seems to have overcome the resistance of PB2013071 that prevented feeding of aphids from NL *** potential mechanism by which SW population overcomes the resistance is discussed.

Quantifying cardinal temperatures and thermal time required for germination of Silybum marianum seed

The response of seed germination to environmental factors can be estimated by nonlinear regression. The present study was performed to compare four nonlinear regression models(segmented, beta, beta modified, and dent-like) to describe the germination rate–temperature relationships of milk thistle(Silybum marianum L.) at six constant temperatures, with the aim of identifying the cardinal temperatures and thermal times required to reach different germination percentiles. Models and statistical indices were calibrated using an iterative optimization method and their performance was compared by root mean square error(RMSE), coefficient of determination(R2) and Akaike information criterion correction(AICc). The beta model was found to be the best model for predicting the required time to reach 50% germination(D50),(R2= 0.99;RMSE = 0.004; AICc =-276.97). Based on the model outputs, the base, optimum, and maximum temperatures of seed germination were 5.19 ± 0.79, 24.01 ± 0.11, and 34.32 ± 0.36 °C,respectively. The thermal times required for 50% and 90% germination were 4.99 and7.38 degree-days, respectively.

Genetic diversity and phylogenetic relationships between and within wild Pistacia species populations and implications for its conservation

Although cultivation and utilization of Pistacia are fully exploited, the evolutionary history of the Pistacia genus and the relationships among the species and accessions is still not well understood. The aim of this study was to analyze random amplified polymorphic DNA（RAPD） in a total of 50 accessions of wild pistachio species, which included five populations Pistacia vera, Pistacia khinjuk,Pistacia atlantica, Pistacia mutica, and Pistacia *** levels of genetic diversity were detected within wild pistachio accessions, as revealed by using the unweighted pair-group method with arithmetic averaging and supported via analysis of molecular variance. The objectives of this investigation were to estimate marker indices, polymorphic information contents（PICs）, and genetic similarities（GS）for RAPD markers; assess the genetic diversity of Pistacia species, using GS estimated from RAPD fingerprints and molecular characterization; and facilitate the use of markers in inter-specific introgression and cultivar *** of the 149 polymerase chain reaction fragments that were scored, 146（97.98 %） were polymorphic. Genetic similarities ranged from 0.3 to 0.86 %, marker indices ranged from 2.98 to 17.74 %, and PICs ranged from 0.80 to0.99 %. Our results provided great molecular identification of all assayed genotypes, which have shown that there is large quantity of genetic diversity among the pistachio accessions. This finding might render striking information in breeding management strategies for genetic conservation and cultivar development.

Nucleic Acid Metabolism, Proline Concentration and Antioxidants Enzyme Activity in Canola (Brassica nupus L.) Under Salinity Stress

Seedling of five canola genotypes, Hyola 308, Hyola 401, Hyola 60, Option 50 and RGS003, were grown in Hoagland nutrient solution containing S1 = 0, S2 = 100, S3 = 200 and S4 = 300 mmol L-1 NaCl in controlled environment. Proline, antioxidant activities like catalase （CAT）, ascobrate peroxidase （APX） and guaiacol peroxidase （GPX） and some enzyme activities of nucleic acid metabolism were determined in shoot and root 20 days after induction of salinity stress. Results showed application of stress significantly affected plant growth components such as fresh （FW） and dry weight （DW） of canola genotypes. Among the genotypes, RGS003 had the highest reduction of FW and DW in S3 treatment. By increasing NaCl levels from 0 to 300 mmol L-1, the activity of two antioxidant enzymes （APX and CAT） in shoot and root increased but GPX in all of genotypes decreased. The increase in salinity stress, increased proline concentration in both root and shoot tissues of canola genotypes. Hyola 401 genotype had the maximum concentration of proline in root and shoot in S3 treatment. Along with increased salinity stress in all of the studied plants, salinity significantly increased the level of the total nucleic acid and the activity of DNase I in all of salinity treatments and at the S3 level, RGS003 had the maximum concentration of nucleolytic enzyme.

Haplotype Diversity at Sub1 Locus and Allelic Distribution Among Rice Varieties of Tide and Flood Prone Areas of South-East Asia

Single nucleotide polymorphisms and restriction digestion-based haplotype variations among 160 flood prone rice varieties were analyzed with enzymes Alu I and Cac8 I to generate polymorphisms at Sub1A and Sub1C loci （conferring submergence tolerance）, respectively. Haplotype associated with phenotype was used to study the haplotype variations at Sub1A and Sub1C loci and to determine their functional influence on submergence tolerance and stem elongation. Three patterns at Sub1A locus, Sub1A0 （null allele）, Sub1A1 （does not cut） and Sub1A2 （one SNP）, and four patterns at Sub1C locus, Sub1C1, Sub1C2, Sub1C3 and Sub1C4, were generated. Both tolerant Sub1A1 and intolerant Sub1A2 had the same length, but the difference was presence of a restriction site in the Sub1A2, but absent at the Sub1A1. Further, two types of polymorphism were detected at the Sub1C, one included major length polymorphisms （165, 170 and 175 bp） and the other was a single restriction site at different position. Eight haplotypes （different combinations of the two loci）, A1C1, A1C2, A1C4, A2C2, A2C4, A0C2, A0C3 and A0C4, were detected among 160 varieties. Haplotype A1C1 was comparatively more related to haplotypes A1C2 and A1C4, having the same Sub1A allele, and these haplotypes were found only in Bangladeshi, Sri Lankan and Indian varieties. Most tolerant varieties in A1C1 haplotype showed slow elongation, having tolerant specific Sub1A1 and Sub1C1 alleles. Further, the varieties Madabaru and Kottamali （A2C2） also showed moderate level of tolerance without Sub1A1 allele. These varieties were different with FR13A and also suspected to carry different novel tolerant genes at other loci. These materials could be used for hybridization with Sub1 varieties for pyramiding additional tolerant specific alleles into a single genotype for improving submergence tolerance in rice.

OS1 functions in the allocation of nutrients between the endosperm and embryo in maize seeds

Uncovering the genetic basis of seed develop-ment will provide useful tools for improving both crop yield and nutritional value, However, the genetic regulatory networks of maize (Zea mays) seed development remain largely unknown. The maize opaque endosperm and small germ 1 (osl) mutant has opaque endosperm and a small embryo,Here, we cloned OSi1 and show that it encodes a putative transcription factor containing an RWP-RK domain-Transcriptional analysis indicated that OSi1 expression is elevated in early endosperm development, especially in the basal endosperm transfer layer (BETL), conducting zone (CZ), and central starch endospcrm (CSE) cells. RNA sequencing (RNA-Seq) analysis of the osi mutant revealed sharp downregulation of certain genes in specific cell types, including ZmMRP-1 and Megl in BETL cells and a majority of zein-and starch-related genes in CSE cells. Using a haploid induction system, we show that wild-type endosperm could rescue the smaller size of osi embryo, which suggests that nutrients are allocated by tho wild type endosperm. Therefore, our dsta imply that the network regulated by 051 accomplishes a key step in nutrient allocation between endosperm and embryo within maize seeds. Identification of this network will help uncover the mechanisms regulating the nutritional balance between endosperm and embryo.
