Inhibitory potentials of phenolic-rich extracts from Bridelia ferruginea on two key carbohydrate-metabolizing enzymes and Fe^(2＋)-induced pancreatic oxidative stress

Objective： The current study was designed to evaluate the various antioxidant potentials and inhibitory effects of phenolic-rich leaf extracts of Bridelia ferruginea（BF） on the in vitro activities of some key enzymes involved in the metabolism of ***： In this study, BF leaf free and bound phenolic-rich extracts were used. We quantified total phenolic and flavonoid contents, and evaluated several antioxidant activities using assays for ferric reducing antioxidant power, total antioxidant activity（phosphomolybdenum reducing ability）, 1,1-diphenyl-2-picrylhydrazyl and thiobarbituric acid reactive species. Also, extracts were tested for their ability to inhibit a-amylase and α-glucosidase ***： The total phenolic and total flavonoid contents in the free phenolic extract of BF were significantly greater than in the bound phenolic extract. Also, all the antioxidant activities considered were significantly greater in the free phenolic extract than in the bound phenolic extract. In the same vein, the free phenolic-rich extract had a significantly higher percentage inhibition against α-glucosidase activity（IC50= 28.5 mg/mL） than the bound phenolic extract（IC50= 340.0 mg/mL）. On the contrary, the free phenolic extract（IC50= 210.0 mg/mL） had significantly lower inhibition against α-amylase than the bound phenolic-rich extract（IC50= 190.0 mg/mL）.Conclusion： The phenolic-rich extracts of BF leaves showed antioxidant potentials and inhibited two key carbohydrate-metabolizing enzymes in vitro.

Objective: To examine the influence of Glyphaea (G.) brevis twig extract on the mitochondrial dehydrogenase activity, integrity of the tight junctions between adjacent cells, mitochondria, apoptosis, nucleus and expression of inhibin-β, stem cell factor, and androgen binding protein in TM4 Sertoli cells. Methods: TM4 cell line was used in this study as it exhibited properties similar to the Sertoli cells. TM4 Sertoli cells were exposed to G. brevis twig extract (0.1, 1.0, 10.0, 100.0, or 1000.0μg/mL) for 24, 48 and 72 h. Parameters studied included cell viability [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide assay], mitochondrial membrane potential (tetra methyl rhodamine ethyl ester dye), transepithelial electrical resistance, apoptosis (Annexin V Alexa Fluor?488/propidium iodide assay) and mRNA expression (quantitative reverse transcription polymerase chain reaction). Results: G. brevis twig extract had no cytotoxic impact on cell viability, thus, considerably increasing the activity of mitochondrial dehydrogenase enzyme after 24 and 72 h exposure. Transepithelial electrical resistance values revealed substantial (P<0.05) rise in treated groups, especially after 72 h of treatment. Moreover, there was a significant decrease in mitochondrial depolarization of TM4 Sertoli cells exposed to G. brevis twig extract when compared to controls. In addition, G. brevis twig extract significantly reduced necrosis and apoptosis of TM4 Sertoli cells when compared to control. Nevertheless, fluorescence microscopy revealed that the nuclei were egg-shaped and marked uniformly with consistent cell shape at the middle of the TM4 Sertoli cells. Significant stimulatory effects were observed on mRNA levels of inhibin-β, androgen binding protein and stem cell factor. Conclusions: G. brevis twig extract may increase the secretory roles of TM4 Sertoli cells, cells proliferation, as well as cell-cell tight junction integrity. Thus, G. brevis twig may enhance spermatogenesis

Aphrodisiac activity of aqueous extract of Anthonotha macrophylla *** in female Wistar rats

OBJECTIVE： This study investigated the aphrodisiac activity of aqueous extract of Anthonotha macrophylla leaves （AEAML） in female Wistar rats. METHODS： Thirty female rats were assigned into five groups of six animals each, such that rats in groups A, B, C, D and E received orally 1 mL of distilled water only, 5.7 mg/kg body weight （BW） of Exus Ginseng （a polyherbal formulation）, 25, 50 and 100 mg/kg BW of AEAML, respectively. After 1 h of treatment, female sexual behaviour parameters were monitored for 30 min. Luteinizing hormone （LH）, follicle stimulating hormone （FSH）, estradiol （E） and testosterone were also determined using standard methods. RESULTS： Six secondary metabolites were detected in AEAML with alkaloids （13.00 mg/L） being the highest. AEAML at 25 and 50 mg/kg BW significantly （P〈0.05） increased the darting frequency, hopping frequency, lordosis frequency, anogenital grooming, genital grooming and licking behaviour, whereas the darting latency, hopping latency and lordosis latency decreased （P〈0.05） in a manner comparable with Exus Ginseng. The 25 and 50 mg/kg BW of AEAML increased （P〈0.05） the serum concentrations of FSH, LH and E, like Exus Ginseng, whereas testosterone content was not altered （P〉0.05）. The 100 mg/kg BW of AEAML did not alter （P〉0.05） the sexual behaviour parameters, serum FSH, LH and E contents, whereas testosterone content increased significantly. The ovarian histology of the animals treated with Exus Ginseng, AEAML at 25 and 50 mg/kg BW revealed developing and ruptured follicles with numerous corpora lutea in the stroma, whereas the 100 mg/kg BW of AEAML produced fewer follicles and corpora lutea. CONCLUSION： The enhanced proceptive, receptive and orientation components of the female sexual behaviour by the 25 and 50 mg/kg BW of AEAML confers sexual invigorating potential on the plant. This study thus justifies the folkloric claim of the plant as a female aphrodisiac.

Parquetina nigrescens leaves： chemical profile and influence on the physical and biochemical indices of sexual activity of male Wistar rats

OBJECTIVE： The leaves of Parquetina nigrescens have been claimed in folk medicine to be useful for managing sexual dysfunction, but there is inadequate scientific evidence for this claim. This investigation was conducted to assess the effects of aqueous leaf extract of Parquetina nigrescens （AEPN） in rats induced with sexual dysfunction. METHODS： Male rats were allocated into various groups after being induced into sexual dysfunction with paroxetine hydrochloride. The groups were treated with distilled water, PowMaxM （reference drug）, and the AEPN at 20, 40 and 80 mg/kg body weight （BW） respectively for 7 d. Frequencies of mount （MF）, intromission （IF）, ejaculation （EF）, as well as latencies of mount （ML）, intromission （IL） and post - ejaculatory interval （PEI） were progressively monitored with receptive female rats on days 1, 3 and 7. The precentage of mounted, intromitted, and ejaculated rats, intromission ratio （IR）, copulatory efficiency （CE） and intercopulatory interval （ICI） were also computed. Standard methods were employed to determine the levels of serum luteinizing and follicle stimulating hormones （LH and FSH）, testosterone, nitric oxide （NO） and the activity of phosphodiesterase V （PDE5）. RESULTS： The plant contained alkaloids, saponins, flavonoids, cardiac glycosides, steroids, tannins, phlobatannins, cardenolides, phenolics, anthraquinones and triterpenes with alkaloids （2.32 mg/g） occurring in greatest quantity while flavonoids, anthraquinones and cardenolides （0.01 mg/g, each） were the least. All the 20 standard amino acids were detected in the plant leaf, with the lowest concentration being 0.30 mg/g for methionine and the highest being 2.12 mg/g for cysteine. Furthermore, P. nigrescens leaves contained Na, K, Fe, Mg, Zn, Ca, Cu, Mn, P, Pb, Cd, Ni, Cr, and Co, with Fe present in the highest and Co the lowest concentrations. Abortifacient Potential of Aqueous Extract of Senna alata Leaves in Rats

Objective To study the abortifacient activity of aqueous extract of Senna alata leaves in female Wistar *** Pregnant rats weighing 143.65 ± 13.11 g were completely randomized into 5 groups（A-E）.Rats in group A（negative control） were orally administered,once daily with 0.5 ml of distilled water on days 10-18 post-coitum while those in groups B,C,D,and E were treated exactly like the negative control except they received the same volume containing 2.85 mg/kg body weight of mifepristone（group B,reference drug）,250,500 and 1 000 mg/kg body weight of the extract respectively（groups C-E）.Results Phytochemical screening of the extract showed positive results for saponins（1.22%）,flavonoids（1.06%）,cardiac glycosides（0.20%）,cardenolides and dienolides（0.18%）,phenolics（0.44%） and alkaloids（0.52%）.Respiratory distress,salivation,diarrhoea,changes in the appearance of hair as well as maternal mortality were not observed at any time during the exposure period except in the mifepristone-treated animals where there was episode of diarrhoea and *** extract significantly reduced（P〈0.05） the number of life foetus,weight and survival ratio of the foetus,numbers of implantations and Corpora lutea,implantation index,progesterone,prolactin,estradiol,follicle stimulating and luteinizing hormones whereas the number of dead foetus,number and percentage of rats that aborted,percentage vaginal opening,resorption index,pre-and post-implantation losses increased *** addition,mifepristone-treated animals produced resorption index that compared well with the distilled water *** was also no dead or life foetus and serum progesterone concentration was increased in the animals treated with *** cases of abortion were accompanied with vaginal ***,the final weight of the rats increased significantly,the feed and water intake were not significantly altered in all the treatment *** weight of the uterus,uterine-body weight ratio,le

Antifertility effects of 60-day oral gavage of ethanol extract of Spondias mombin leaves in guinea pigs: A biochemical, reproductive and histological study

Objective:To investigate the impact of ethanol extract of Spondias(S.)mombin leaves on the biochemical parameters of testicular function,hormonal profile,sperm profile and reproductive outcomes of guinea ***:Sexually matured male[(1000.40±8.12)g]and female[(810.00±7.09)g]guinea pigs were *** testicular function study,a total of 24 male guinea pigs were randomised into 4 groups of 6 guinea pigs *** A(control)was orally administered 1 mL of physiological saline,once daily for 60 days while groups B,C and D were treated like the control group except that they were orally administered 100,250 and 500 mg/kg body weight of ethanol extract of *** *** the fertility study,the same animal groupings and treatments in the testicular function study were *** male guinea pigs were paired with the females(1:1)and afterwards examined for pregnancy ***:The ethanol extract of *** leaves contained saponins,alkaloids,flavonoids,tannins,steroids,phenolics,phlobatannins,cardiac glycosides,cardenolides and dienolides with saponins(4.80 mg/mL)occurring the most whilst cardenolides and dienolides(0.08 mg/mL)were the *** ethanol extract of *** leaves significantly and dose dependently reduced the activities of alkaline phosphatase,glutamate dehydrogenase,3-hydroxy-3-methylglutaryl coenzyme A reductase,malic enzyme,17-β-hydroxysteroid dehydrogenase,lactate dehydrogenase,catalase and superoxide dismutase as well as levels of testosterone,glycogen,total protein and ascorbic acid in the testes when compared with the control group(P<0.05).All the doses of ethanol extract of *** leaves also reduced the levels of sorbitol dehydrogenase,3-β-hydroxysteroid dehydrogenase,glucose-6-phosphate dehydrogenase and sialic acid whereas the levels of testicular acid phosphatase,gamma glutamyl transferase and cholesterol increased dose dependently(P<0.05).The serum luteinising hormone,testosterone and estradiol were reduced after the admin

Reactive oxygen species detoxification potentials of Sorghum bicolor polyphenolic extract was investigated in the liver of N-nitrosodiethylaminetreated *** rats,weighing(135±5.5)g were completely randomized into 7 groups(A–G)of five rats *** in C,D,E and F were administered orally once daily at 24-h interval for 7 d with 500,125,250 and 500 mg/kg body weight of polyphenolic extract of ***,*** G was given 100 mg/kg body weight of vitamin *** the sixth day,groups B,D,E,F and G were administered with 100 mg/kg body weight N-nitrosodiethylamine(NDEA).Group A,which served as the control was treated like the test groups except,that the animals received distilled water *** oxygen species detoxifying enzymes(superoxide dismutase,catalase,glutathione peroxidase,glutathione reductase and glucose 6-phosphate dehydrogenase)activities were significantly(P<0.05)induced by *** inductions significantly(P<0.05)attenuated the NDEA-mediated decrease in reactive oxygen species detoxifying enzymes and compared favourably with vitamin ***-mediated elevation in the concentrations of oxidative stress biomarkers;malondialdehyde,conjugated dienes,lipid hydroperoxides,protein carbonyl and percentage DNA fragmentation were significantly(P<0.05)lowered by *** polyphenolic ***,the results obtained from this study revealed that the polyphenolic extract of *** grains enhanced the detoxification of reactive oxygen species in NDEA-treated *** polyphenols also prevented the peroxidation of lipid,oxidation of proteins as well as fragmentation of DNA component in the liver of rats and hence gave the evidence of possible prophylactic potentials of *** grains.©2013 Beijing Academy of Food *** and hosting by Elsevier *** rights reserved.

Toxicity profile of ethanolic extract of Azadirachta indica stem bark in male Wistar rats

Objective:To investigate the loxic implications of ethanolic stem bark extract of Azadirachta indica(A-indica)at 50,100,200 and 300 mg/kg body weight in Wistar ***:Fifty male rats of Wistar strains were randomly grouped into five(A-F.)of ten animals *** in Group A(control)were orally administered 1 mL of distilled water on daily basis for 21 days while those in Groups B-E received same volume of the extract corresponding to 50,100,200 and 300mg/kg body ***:The extract did not significantly(P>0.05)alter the levels of albumin,total protein,red blood cells and factors relating to it whereas the white blood cell,platelets,serum triacylglycerol and high-density lipoprotein cholesterol decreased significantly(Pbiochemical parameters of toxicity have consequential effects on the normal functioning of the organs of the ***,the ethanolic extract of *** stem bark at the doses of 50,100,200 and 300 mg/kg body weight may not be completely safe as an oral remedy and should be taken with caution if absolutely necessary.
