Mechanisms of vacuolar phosphate efflux supporting soybean root hair growth in response to phosphate deficiency

Phosphorus is an essential macronutrient for plant growth and development. In response to phosphate(Pi) deficiency, plants rapidly produce a substitutive amount of root hairs; however, the mechanisms underlying Pi supply for root hair growth remain unclear. Here, we observed that soybean(Glycine max) plants maintain a consistent level of Pi within root hairs even under external Pi deficiency. We therefore investigated the role of vacuole-stored Pi, a major Pi reservoir in plant cells, in supporting root hair growth under Pi-deficient conditions. Our findings indicated that two vacuolar Pi efflux(VPE) transporters, Gm VPE1 and Gm VPE2, remobilize vacuolar stored Pi to sustain cytosolic Pi content in root hair *** analysis showed that double mutants of Gm VPE1 and Gm VPE2 exhibited reduced root hair growth under low Pi conditions. Moreover,Gm VPE1 and Gm VPE2 were highly expressed in root hairs, with their expression levels significantly upregulated by low Pi treatment. Further analysis revealed that Gm RSL2(ROOT HAIR DEFECTIVE6-like 2), a transcription factor involved in root hair morphogenesis, directly binds to the promoter regions of Gm VPE1 and Gm VPE2, and promotes their expressions under low Pi conditions. Additionally, mutants lacking both Gm RSL2 and its homolog Gm RSL3 exhibited impaired root hair growth under low Pi stress, which was rescued by overexpressing either Gm VPE1 or Gm *** together, our study has identified a module comprising vacuolar Pi exporters and transcription factors responsible for remobilizing vacuolar Pi to support root hair growth in response to Pi deficiency in soybean.

Lactic acid bacteria and the fermentation environment interact to form an intertwined system. Lactic acid bacteria are constantly evolving to adapt to different fermentation environments， causing changes in their physiological processes. To achieve a targeted improvement of their adaptability to various environments， a detail analysis of their evolutionary physiological processes is required. While several studies have been carried out in the past by using single-omics techniques to investigate their response to environmental stress， most researchers are now using a multi-omics approach to explore more detail in the biological regulatory networks and molecular mechanisms of lactic acid bacteria in response to environmental stress， thereby overcoming the limitations of single- omics analysis. Scope and approach: In this review， we describe the various single-omics approaches that have been used to study environmental stress in lactic acid bacteria， present the advantages of various multi-omics combined analysis approaches， and discuss the potential and practicality of applying emerging single-cell transcriptomics and single-cell metabolomics techniques to the molecular mechanism study of microbes response to environmental stress. Key ?ndings and conclusions: Multi-omics approaches enable the accurate identification of complex microbial physiological processes in different environments， allow people to comprehensively reveal the molecular mechanisms of microbes response to stress from different perspectives. Single-cell omics techniques， analyze the targeted regulation of microbial functions in a multi-dimensional space， provides a new perspective on understanding microbes responses environment stress.

In recent years,heavy ion accelerator technology has been rapidly developing worldwide and widely applied in the fields of space radiation simulation and particle ***,a very high uniformity in the irradiation area is required for the extracted ion beams,which is crucial because it directly affects the experimental precision and therapeutic ***,ultra-large-area and high-uniformity scanning are crucial requirements for spacecraft radiation effects assessment and serve as core specification for beamline terminal *** the 300 MeV proton and heavy ion accelerator complex at the Space Environment Simulation and Research Infrastructure(SESRI),proton and heavy ion beams will be accelerated and ultimately delivered to three irradiation *** order to achieve the required large irradiation area of 320 mm×320 mm,horizontal and vertical scanning magnets are used in the extraction beam ***,considering the various requirements for beam species and energies,the tracking accuracy of power supplies(PSs),the eddy current effect of scanning magnets,and the fluctuation of ion bunch structure will reduce the irradiation *** mitigate these effects,a beam uniformity optimization method based on the measured beam distribution was proposed and applied in the accelerator complex at *** the experiment,the uniformity is successfully optimized from 75%to over 90%after five iterations of adjustment to the PS *** this paper,the method and experimental results were introduced.

Herein based on literature available a hypothesis is provided about molecular basis for initial events in establishment *** hypothesis asserts that:"recognition and interaction that occur between organisms is *** are membrane receptors with or without soluble components derived from the respective organisms that bridge specific interactions".Organisms'prearranged recognition theory(OPRT)can be specifically applied to host-microbe interactions where most microbes are coated(opsonised)by soluble components(opsonins)from the host,but there are also some microbes that can bypass host opsonization expressing receptors for the host cells or secreting host’s opsonin-like *** receptors involved in organism's interactions,their specificity and repertoire depend on saccharides from glycoproteins,glycolipids,and polysaccharides(glycans)which are abundant extracellular *** in OPRT is possible to explain species-specific interactions and several other phenomena,such as hyper-infectivity,tissue tropism,differential sensitivity to disease depending on type O-blood,and tumoral cell *** lipid raft domain in cellular membrane is proposed as the main location where interactions will trigger cellular *** scientific and biotechnology applications and alternative routes to modify organism's interactions and consequences are *** is a novel hypothesis regarding the degree to which an organism's interactions are prearranged and the role of saccharides epitopes.

A Study on the Genetic Dynamics and Development Characteristics of Granitic Rock Avalanches in the Northern Qinling Mountains， China

Massive granitic rock avalanches are extensively developed in the middle section of the northern Qinling Mountains (NQM)， China. The current consensus is that their formation could have been connected with seismic events that occurred in the NQM. However， there is a lack of systematic discussion on the genetic dynamics of these rock avalanches. Hence， taking Earth system scientific research as a starting point， this paper systematically summarizes and discusses development characteristics， formation times and genetic dynamic mechanisms of granitic rock avalanches in the NQM based on geological investigations， high-precision remote sensing interpretations， geomorphological dating， geophysical exploration， and a large-scale shaking table model test. We identified 53 granitic rock avalanches in this area， with a single collapse area ranging from 0.01×106to 1.71×106m2. Their development time can be divided into six stages， namely， 107297～106289 years BP， 11870-11950 years BP， 11666～10279 years BP， 2260-2320 years BP， 1788-1925 years BP， and 1392～1661 years BP， which were closely related to multiple prehistoric or ancient earthquakes. We suggest that long-term coupling of internal and external earth dynamics was responsible for the granitic rock avalanches in the NQM; the internal dynamics were mainly related to subduction， collision and extrusion of different plates under the Qinling terrane， leading to the formation and tectonic uplift of the Qinling orogenic belt; and the external dynamics were closely associated with climate changes resulting in mountain denudation， freeze-thaw cycles and isostatic balance uplift. In this process， the formation and evolution of the Qinling orogenic belt play a geohazard-pregnant role， structural planes， including faults and joints， play a geohazard-controlled role， and earthquakes play a geohazard-induced role， which jointly results in the occurrence of large-scale granitic rock avalanches in the NQM. Dear Editor,Primary mediastinal large B-cell lymphoma (PMBCL) is a rare subtype of aggressive B cell lymphoma, accounting for 2%to 4%of total non-Hodgkin’s lymphoma(NHL) cases (Savage, 2022). Genomic rearrangements (e.g., isochromosome9p) and copy-number alterations at9p24.1 loci, which contain the genes encoding programmed death-ligand 1/2(PDL1/2) and JAK2, lead to the overexpression of these genes in PMBCL,distinguishing PMBCL from diffuse large B-cell lymphoma (DLBCL)(Green et al.,2010). Recent thymic single-cell gene profiling has advanced our understanding of the presence and functions of B cells in the thymus (Afzali et al., 2024). However,the etiology of thymic B cell-derived PMBCL and its cellular microenvironment remain unknown. In this study, from a single-cell perspective, we compared the single-cell profiles of PMBCL with those of DLBCL and normal lymph nodes (NLNs),and discovered a unique phenotype of PMBCL-associated malignant plasma cells.
