Scutellarin(SCU)is a herbal flavonoid glucuronide with multiple pharmacological activities,including antioxidant,anti-inflammation,vascular relaxation,anti-platelet,and myocardial ***,the effect of SCU on complete Freund’s adjuvant(CFA)-induced rheumatoid arthritis(RA)had not been *** this study,we investigated the beneficial effects of SCU in the CFA-induced RA mice model and the anti-arthritic activity was evaluated by paw ***-linked immunosorbent assay(ELISA)was carried out to evaluate the plasma levels of immunoglobulin(Ig)G,IgE,tumor necrosis factor(TNF)-α,interleukin(IL)-1β,IL-6,receptor activator of nuclear factor-κB ligand(RANKL),and osteoprotegerin(OPG).Histological slides were prepared from the harvested paws of mice to determine the pathological changes in the *** proportions of T helper type 1(Th1)and T helper type 2(Th2)cells of CD4+T lymphocyte subsets were analyzed by flow *** expression of Kelch-like ECHassociated protein 1(Keap1),nuclear factor erythroid 2-related factor 2(Nrf2),and heme oxygenase-1(HO-1)was analyzed using real-time quantitative PCR(RT-qPCR)and western blotting *** present study demonstrated that SCU prevented CFA-induced RA,and inhibited the expression of inflammation factors,IgG,IgE,TNF-α,IL-1β,and *** SCU also reduced the RANKL level,it increased OPG expression in RA *** Th1/Th2 ratio was significantly lower in mice treated with ***,HO-1 expression was reduced while the expression of Keap1 and Nrf2 was elevated following SCU *** provide preliminary evidence to employ SCU in arthritis treatment which might be related to the regulation of Th1/Th2 balance and the Keap1/Nrf2/HO-1 pathway.

An alginate-degrading bacterium, identified as Ochrobactrum sp. on the basis of 16S rDNA gene sequencing, was isolated from brown algal samples collected from the waters in close vicinity to the Dongtou isles of the East China Sea. The strain, designated WZUH09-1, is able to depolymerize alginates with higher enzyme activity than that of others reported so far.

Carbapenem-resistant Enterobacteriaceae(CRE)have spread worldwide as a global threat and CRE infection is associated with a significant ***,data on epidemiology and treatment of CRE infection in children are comparatively lacking.[1]Therefore,we retrospectively conducted a matched case-control study to summarize the epidemiological characteristics,risk factors,treatment,and outcomes of nosocomial CRE infections in a children patient population,and also to identify the antimicrobial resistance and resistance genotyping of CRE isolates.

Low back pain,mainly caused by intervertebral disc degeneration(IVDD),is a common health problem;however,current surgical treatments are less than ***,it is essential to develop novel non-invasive surgical methods for IVDD ***,we describe a therapeutic strategy to inhibit IVDD by injecting hydrogels modified with the extracellular matrix of costal cartilage(ECM-Gels)that are loaded with cartilage endplate stem cells(CESCs).After loaded with CESCs overexpressing Sphk2(Lenti-Sphk2-CESCs)and injected near the cartilage endplate(CEP)of rats in vivo,ECM-Gels produced Sphk2-engineered exosomes(Lenti-Sphk2-Exos).These exosomes penetrated the annulus fibrosus(AF)and transported Sphk2 into the nucleus pulposus cells(NPCs).Sphk2 activated the phosphatidylinositol 3-kinase(PI3K)/p-AKT pathway as well as the intracellular autophagy of NPCs,ultimately ameliorating *** study provides a novel and efficient non-invasive combinational strategy for IVDD treatment using injectable ECM-Gels loaded with CESCs that express Sphk2 with sustained release of functional exosomes.

Safety and clinical efficacy of linezolid in children:a systematic review and meta-analysis

Background We aimed to evaluate the tolerability and efficacy of linezolid in children for treating suspected and diagnosed Gram-positive bacterial *** A systematic literature search was conducted up to April 23,2021,using linezolid and its synonyms as search *** reviewers independently identified and extracted relevant randomized controlled trials and prospective cohort *** extracted studies were included in a single-rate meta-analysis of adverse events and clinical outcomes using random-effects *** A total of 1082 articles were identified,and nine studies involving 758 children were included in the *** overall proportion of adverse events was 8.91%[95%confidence interval(CI)=1.64%–36.52%],with diarrhea(2.24%),vomiting(2.05%),and rash(1.72%)being the most *** incidences of thrombocytopenia and anemia were 0.68%and 0.16%,*** specific adverse events,including rash and gastrointestinal events,were more frequent in the oral administration *** terms of efficacy,the overall proportion of clinical improvement was 88.80%(95%CI=81.31%–93.52%).Children with a history of specific bacteriological diagnosis or concomitant antibiotic therapy had a 1.13-fold higher clinical improvement than children without such *** proportion of microbial eradication was 92.68%(95%CI=84.66%–96.68%).The proportion of all-cause mortality was 0.16%(95%CI=0.00%–7.75%).Conclusions Linezolid was well-tolerated in pediatric patients and was associated with a low frequency of adverse events,such as anemia,thrombocytopenia,and ***,linezolid was effective in children with diagnosed and suspected Gram-positive infections.

Oral nano-antioxidants improve sleep by restoring intestinal barrier integrity and preventing systemic inflammation

Sleep deprivation(SD) is a severe public health threat that can cause systemic inflammation and nerve damage. Few effective and side-effect-free drugs are available to address SD. However, the bidirectional communications between the brain and gut provide new strategies for anti-SD therapeutics. Here we explored oral delivery of fullerene nano-antioxidants(FNAO) in the SD model to improve sleep by regulating abnormal intestinal barrier and systemic inflammation via the brain-gut axis. SD caused excessive reactive oxygen species(ROS) production and hyperactive inflammatory responses in the intestines of zebrafish and mouse models, leading to disturbed sleep patterns and reduced brain nerve activity. Of note,based on the property of the conjugated π bond of the C60structure to absorb unpaired electrons, oral FNAO efficiently reduced the excessive ROS in the intestines, maintained redox homeostasis and intestinal barrier integrity, and ameliorated intestinal and systemic inflammation, resulting in superior sleep improvement. Our findings suggest that maintaining intestinal homeostasis may be a promising avenue for SD-related nerve injury therapy.

Three new compounds, including one chloro-furoeremophilanoid （1）, one eremophiladiolide （2）, a rare dimer of nor- furoeremophilanoid, ligulatrovine A （3）, and a known furoeremophilanolide （4）, were isolated from Ligularia atroviolacea. The structures of compounds 1-4 were elucidated by spectroscopic methods including 1D and 2D NMR experiments as well as X- ray diffraction study.

BACKGROUND Shigella flexneri(***)is a major pathogen causing acute intestinal infection,but the systematic oxidative damage incurred during the course of infection has not been *** To investigate the incurred systemic RNA oxidative damage and the diagnostic value of RNA oxidative metabolites during ***-induced intestinal *** In this study,a Sprague-Dawley rat model of acute intestinal infection was established by oral gavage with *** *** changes in white blood cells(WBCs)and cytokine levels in blood and the inflammatory response in the colon were *** also detected the RNA and DNA oxidation in urine and *** *** infection induced an increase in WBCs,C-reactive protein,interleukin(IL)-6,IL-10,IL-1β,IL-4,IL-17a,IL-10,and tumor necrosis factorα(TNF-α)in *** note,a significant increase in urinary 8-oxo-7,8-dihydroguanosine(8-oxo-Gsn),an important marker of total RNA oxidation,was detected after intestinal infection(P=0.03).The urinary 8-oxo-Gsn level returned to the baseline level after recovery from *** addition,the results of a correlation analysis showed that urinary 8-oxo-Gsn was positively correlated with the WBC count and the cytokines IL-6,TNF-α,IL-10,IL-1β,and IL-17α.Further detection of the oxidation in different tissues showed that *** infection induced RNA oxidative damage in the colon,ileum,liver,spleen,and *** Acute infection induced by *** causes increased RNA oxidative damage in various tissues(liver,spleen,and brain)and an increase of 8-oxo-Gsn,a urinary *** 8-oxo-Gsn may be useful as a biomarker for evaluating the severity and prognosis of infection.
