Virulence factors of Enterococcus strains isolated from patients with inflammatory bowel disease

AIM: To determine the features of Enterococcus that contribute to the development and maintenance of the inflammatory process in patients with inflammatory bowel disease (IBD). METHODS: Multiplex polymerase chain reaction (PCR) was applied to assess the presence of genes that encode virulence factors [surface aggregating protein (asa1), gelatinase (gelE), cytolysin (cylA), extracellular surface protein (esp) and hyaluronidase (hyl)] in the genomic DNA of 28 strains of Enterococcus isolated from the intestinal tissues of children with IBD (n =16) and of children without IBD (controls; n = 12). Additionally, strains with confirmed presence of the gelE gene were tested by PCR for the presence of quorum sensing genes (fsrA, fsrB, fsrC) that control the gelatinase production. Gelatinase activity was tested on agar plates containing 1.6% gelatin. We also analysed the ability of Enterococcus strains to release and decompose hydrogen peroxide (using Analytical Merckoquant peroxide test strips) and tested their ability to adhere to Caco-2 human gut epithelium cells and form biofilms in vitro. RESULTS: A comparison of the genomes of Enterococcus strains isolated from the inflamed mucosa of patients with IBD with those of the control group showed statistically significant differences in the frequency of theasa1 gene and thegelE gene. Furthermore, the cumulative occurrence of different virulence genes in the genome of a single strain ofEnterococcus isolated from the IBD patient group is greater than in a strain from the control group, although no significant difference was found. Statistically significant differences in the decomposition of hydrogen peroxide and adherence to the Caco-2 epithelial cell line between the strains from the patient group and control group were demonstrated. Libration-Generated Average Convection in a Rotating Flat Layer with Horizontal Axis

Experimental Study of Thermal Convection and Heat Transfer in Rotating Horizontal Annulus

A genuine technological issue–the thermal convection of liquid in a rotating cavity–is investigated *** experiments are conducted within a horizontal annulus with isothermal boundaries. The inner boundaryof the annulus has a higher temperature, thus exerting a stabilising influence on the system. It is shown that whenthe layer rotation velocity diminishes, two-dimensional azimuthally periodic convective rolls, rotating togetherwith the cavity, emerge in a threshold manner. The development of convection is accompanied by a significantintensification of heat transfer through the layer. It is shown that the averaged thermal convection excitation inthe form of a system of two-dimensional rolls occurs against the background of oscillations of a non-isothermalfluid in the cavity reference frame caused by the gravity field. The excitation threshold and the structure ofconvective rolls are consistent with the results of the earlier theoretical studies by the authors performed usingthe equations of “vibrational” convection obtained by the averaging method. Furthermore, the experiments haverevealed a new type of averaged flow in the form of a spatially periodic system of toroidal vortices. It is shown thata steady streaming, excited by the inertial oscillations of the fluid, is responsible for the generation of the toroidalvortices. These flows develop in a non-threshold manner and are most clearly manifested in a case of resonantexcitation of one of the inertial modes.

Diet and nutritional factors in inflammatory bowel diseases

Inflammatory bowel disease(IBD) development is affected by complex interactions between environmental factors, changes in intestinal flora, various predisposing genetic properties and changes in the immune system. Dietary factors seem to play an underestimated role in the etiopathogenesis and course of the disease. However, research about food and IBD is conflicting. An excessive consumption of sugar, animal fat and linoleic acid is considered a risk factor for IBD development, whereas a high fiber diet and citrus fruit consumption may play a protective role. Also, appropriate nutrition in particular periods of the disease may facilitate achieving or prolonging remissions and most of all, improve the quality of life for patients. During disease exacerbation, a low fiber diet is recommended for most patients. In the remission time, an excessive consumption of alcohol and sulfur products may have a negative effect on the disease course. Attempts are also made at employing diets composed in detail in order to supplement IBD therapy. A diet with a modified carbohydrate composition, a semi-vegetarian diet and a diet low in fermentable oligosaccharides, disaccharides, monosaccharides and polyols are under investigation. Due to chronic inflammation as well as side effects of chronically used medications, patients with IBD are also at increased risk of nutritional factor deficiencies, including iron, calcium, vitamin D, vitamin B12, folic acid, zinc, magnesium and vitamin A. It should also be remembered that there is no single common diet suitable for all IBD patients; each of them is unique and dietary recommendations must be individually developed for each patient, depending on the course of the disease, past surgical procedures and type of pharmacotherapy.

Oscillatory Dynamics of a Spherical Solid in a Liquid in an Axisymmetric Variable Cross Section Channel

The dynamics of a solid spherical body in an oscillating liquid flow in a vertical axisymmetric channel of variable cross section is experimentally *** is shown that the oscillating liquid leads to the generation of intense averaged flows in each of the channel *** intensity and direction of these flows depend on the dimensionless oscillating *** the region of studied frequencies,the dynamics of the considered body is examined when the primary vortices emerging in the flow occupy the whole region in each *** a fixed frequency,an increase in the oscillation amplitude leads to a phase-inclusion holding effect,i.e.,the body occupies a quasi-stationary position in one of the cells of the vertical channel,while oscillating around its average *** is also shown that the oscillating motion of a liquid column generates an averaged force acting on the body,the magnitude of which depends on the properties of the body and its position in the *** quasi-stationary position is determined by the relative density and size of the body,as well as the dimensionless *** behavior of the body as a function of the amplitude and frequency of fluid oscillation and relative size is discussed in *** findings may be used in the future to control the position of a phase inclusion and/or to strengthen mass transfer effects in a channel of variable cross section by means of fluid oscillations.

Modeling of time dependent subsidence for coal and ore deposits

Coal and ore underground mining generates subsidence and deformation of the land surface. Those defor- mations may cause damage to buildings and infrastructures. The environmental impact of subsidence will not be accepted in the future by the society in many countries. Especially acceptance of the ground deformations decreases every year there, where the mining regions are densely urbanized, the The only solution is to limit the subsidence or its impact on the infrastructure. The first is not rentable for the mining industry, the second depends on the precise subsidence prediction and good preventing management involved in the mining areas. The precision of the subsidence prediction depends strictly on the mathematical model of the deformation phenomenon and on the uncertainty of the input data. The subsidence prediction in the geological conditions of the raw materials used to be made on the basis of numerical modeling or the stochastic models. A modified solution of the stochastic model by Knothe will be presented in the paper. The author focuses on the precise description of the deposit shape and on the time dependent displacements of the rock mass. A two parameters＇ time function has been introduced in the algorithm.

Key changes across valley floors in mountain areas affected by different levels of human impact: a case study of the Ochotnica river catchment in the Polish Carpathians

The goal of this paper was to present knowledge on changes in the morphodynamic structure of the Ochotnica River(Polish Carpathians)and transformations within its active river zone since the end of the 19th *** study used a set of archival and contemporary cartographic materials and the results of three-fold field mapping of the morphodynamic structure of the *** and indirect human interference with the natural environment of the catchment and the fluvial system was taken into account in the *** of changes in the morphodynamic structure of the Ochotnica and its active river zone since the end of the 19th century indicated significant changes in the development trend of the *** contemporary structure of the Ochotnica is more complex than in the *** has been a fragmentation of the riverbed into sections with different morphodynamic *** proportion of erosive sections has increased,which explains the reduced in the width of the active river zone of the *** average width of the active river zone of the river between 1861 and 2022 has changed from 80 m to 18 *** impact has been identified as the main reason for the changes in the morphodynamic structure of the riverbed and its active river ***,only the unregulated sect ion is characterised by free lateral migration of the riverbed and the widest active river zone(~28 m).
