Vulvectomy: Indications and Results in the General Surgery department of the Ignace Deen chu Hospital in Conakry

Introduction: The aim of this study was to report the indications and evaluate the results of vulvectomy in the general surgery department of Ignace Deen Hospital, chu Conakry. Materials and Methods: This was a retrospective descriptive study, conducted over a period of five (05) years from January 1, 2018 to December 31, 2022, in the general surgery department of Ignace Deen Hospital, chu Conakry. We included in our study all patient records in whom vulvectomy was performed. Results: We recorded 15 cases of vulvectomy out of 453 perineal surgeries, i.e. 3.31%, with a mean age of 43.56 years and extremes of 35 and 69 years. Vulvar cancer was the most common diagnosis (46.67%), followed by Buschke-Lowenstein (33.33%) and anal canal cancer extending to the vulva (20%). Six patients had undergone biopsy (40%). Vulvectomy with lymph node dissection was performed in only 9 patients (60%), and all surgical specimens were sent to anatomical pathology (100%). Conclusion: Vulvectomy is a surgical technique most often indicated for the treatment of vulvar cancer.

Prion-Like Activity of Pathological Tau Species Derived from Alzheimer's Brain

Background:Neurofibrillary tangles(NFTs),the insoluble and fibrillar intraneuronal accumulations of hyperphosphorylated tau protein,are a defining histopathological hallmark of the Alzheimer’s disease(AD) brain. NFTs initiate in subcortical regions,transentorhinal area,and entorhinal cortex,then appear in the hippocampal formation and some parts of the neocortex,followed by most of the neocortex. The distribution of tau pathology correlates with the progression of AD. Injection of misfolded tau aggregates derived from AD and transgenic mouse brains or generated in vitro induces tau pathology not only at the injection sites,but also in the anatomically connected brain regions in a time-dependent manner,introducing the concept of "prion-like propagation of tau pathology". The prion-like activity of the pathological tau species and the role of tau hyperphosphorylation in seeding and propagation of tau pathology are currently under debate. Methods:To determine the prion-like activity of pathological tau species in AD brain,we isolated different pathological tau species,soluble tau(S-tau),heat-stable soluble tau(HS-tau),oligomeric tau(O-tau),sarkosyl insoluble tau 1(SI1-tau),and sarkosyl insoluble tau 2(SI2-tau),from AD brains,analyzed their truncation and phosphorylation by Western blots and determined their prion-like activity by capturing normal tau in vitro and seeding tau aggregation in cultured cells,and inducing tau pathology in vivo. Results:We found that these tau species isolated from AD brain showed differences in truncation,site-specific phosphorylation,and SDSand reducing agent-resistant high molecular weight aggregation. S-tau,AD O-tau and SI1-tau,but not HS-tau and SI2-tau,captured tau151-391 from cell lysates and induced tau aggregation in cultured cells. AD O-tau showed the highest prion-like activity. Dephosphorylation of AD O-tau ameliorated its prion-like activity to capture tau in vitro,to seed tau aggregation in cultured cells and to induce tau path

HLCDG1, which locates in chromosome 5q33, is a novel gene cloned recently. The HLCDG1 expression was significantly down regulated in the primary lung carcinoma. It was previously studied that HLCDG1 acted like a tumor suppressor gene. In this paper, proteomics studies were performed to analyze the proteomic expression patterns in the HLCDG1-transfected human lung carcinoma cell line (A549-HLCDG1) and in the control vector-transfecred human lung carcinoma cell line (A549-vector). Employing two dimensional gel eleetrophoresis (2DE), the global pattern of protein expressions in A549-HLCDG1 human lung adenocarcinoma cell line expressing stably HL-CDG1 gene were compared with those of control A549-vector cell line to generate a differential protein expression catalog. Forty-two differentially expressed proteins were screened. Thirteen differential proteins were identified by matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS), which were 6 upregulated (MSH5, MOD, MDH precursor, ETFβ, Prxd Ⅵ and JM23) and 7 downregulated (PLC-δ1, hnRNPA2,hnRNPB1, TIM, TCTP, nm23H-1 and PrxdⅤ) proteins in A549-HLCDG1 cells compared to control A549-vector cells. The above identified proteins were involved in energy metabolism, transcription regulation, antioxidation,cell cycle, metastasis, DNA methylation and mismatch repair. Therefore, these differential expression proteins by HLCDG1 transfection may play some important roles for investigation of the biochemical basis of growth suppression of HLCDG1 gene in lung carcinoma cells A549. Further understanding of this data base may provide valuable resources for the developing novel diagnostic markers and therapeutic targets of lung cancer.
