When the NifA-mediated activation of Kleb-siella pneumontae nifU promoter is recreated in Escherichia coli, it has been observed that CRP-cAMP has an inhibitory effect on the nifU promoter. sequence analysis indicates that there is a strong CRP-binding site located upstream of the nifU promoter, overlapping completely with a previously identified Nif A-binding site. In vitro gel retardation analysis indicates that this putative CRP-binding site has similar affinity for CRP, when compared with that at the lac promoter, suggesting that CRP could effectively compete with NifA for such a binding site under physiological conditions. When this putative CRP-binding site on nifU was mutated, in vitro gel retardation analysis indicates that CRP can no longer bind to the mutant promoter. However, when constitutively expressed NifA is used as the activator, CRP-cAMP-mediated inhibitory effect on this mutant nifU promoter has no significant difference when compared with that obtained from its wild-type promoter.

to maintain genomic integrity and avoid diseases, the DNA-damage response(DDR) not only detects and repairs DNA lesions, but also contributes to the resistance to DNA-damaging *** the DDR plays a significant role in drug discovery using the principle of synthetic lethality. the incomplete current knowledge of the DDR encouraged us to develop new strategies to identify and study its components and pathways. Polycarcin V, belonging to the C-aryl glycoside natural products, is a light-activatable DNA-intercalating agent that causes DNA damage by forming a covalent [2+2]cycloadduct with thymine residue under 365–450 nm of light irradiation in a DNA-sequence-independent manner. taking advantage of the light-activatable feature and temporal control of DDR, we designed and synthesized polycarcin V-based bifunctional chemical probes, including one that cross-links DNA to DNA-binding protein to explore the DDR induced by polycarcin V and uncover novel DNA–protein interactions. Utilizing this chemical probe and activity-based protein profiling-stable isotope labeling with amino acids in cell culture, we identified 311 DNA-binding protein candidates, including known DDR factors and additional proteins that may be of interest in discovering new biology. We validated our approach by showing that our probe could specifically cross-link proteins involved in nucleotide excision repair(NER) that repair bulky DNA adducts. Our studies showed that the [2+2] cycloadduct formed by polycarcin V could indeed be repaired by NER in vivo. As a DNA-damaging agent, polycarcin V or its drug-like derivative plus blue light showed promising properties for psoriasis treatment, suggesting that it may itself hold promise for clinic applications.

As a sister work of[Phys. Rev. D 79 （2009） 094004], we incorporate the color-screening effect due to light quark pair creation into the heavy quark-antiquark potential, and investigate the effects of screened potential on the spectrum of bottomonimn. We calculate the masses, electromagnetic decays, and E1 transitions of bottomonium states. We find that the fine splittings between χbJ （J = 0, 1, 2） states are in good agreement with experimental data, and the E1 transition rates of γ（2s） → γχbJ（1P） and γ（3s） →γχbJ（2P）（J = 0, 1, 2） all agree with data within experimental errors. In particular, the mass of γ（6s） is lowered down to match that of the γ（11020）, which is smaller than the predictions of the linear potential models by more than 100 MeV. Comparison between charmonium and bottomonium in some related problems is also discussed.

Modeling gene regulatory networks （GRNs） is an important topic in systems biology. Although there has been much work focusing on various specific systems, the generic behavior of GRNs with continuous variables is still elusive. In particular, it is not clear typically how attractors partition among the three types of orbits： steady state, periodic and chaotic, and how the dynamical properties change with network＇s topological characteristics. In this work, we first investigated these questions in random GRNs with different network sizes, connectivity, fraction of inhibitory links and transcription regulation rules. then we searched for the core motifs that govern the dynamic behavior of large GRNs. We show that the stability of a random GRN is typically governed by a few embedding motifs of small sizes, and therefore can in general be understood in the context of these short motifs. Our results provide insights for the study and design of genetic networks.

the poor performance and limited availability of sexuality education programs in rural areas of western China increase the danger of younger cohorts being exposed to sexually transmitted diseases(stDs)and engaging in risky sexual behaviors.A traditionally conservative culture,low levels of economic development and the relative lack of faculty resources all contribute to the weakness of sexuality education *** study uses a sample of 164 boys and 195 girls who were senior high school students aged 15-19 in rural western China in *** study aims to explore sexuality education,and sexual and reproductive behaviors,and to identify associations between sexuality education and sexual knowledge and attitudes among adolescents in rural western *** results reveal a positive association between courses about reproductive health and lectures about stDs,and sexual knowledge;and a positive association between lectures about stDs and sexual *** study also found that access to consulting services provided by governmental organization in the field of family planning and reproductive health helped rural adolescents enhance their sexual *** such services available appears to offer a feasible method to develop a comprehensive sexuality education system in western *** study concludes that an improved school-based sexuality education program strictly implemented as a part of the standard school curriculum is an effective way to increase sexual knowledge and promote de-stigmatized sexual attitudes among adolescents in rural western China.

Background Hydrosalpinx is the most common lesion of the distal part of the *** is well known to have deleterious effect on *** two options are doable in case of hydrosalpinx:radical treatment(salpingectomy)or conservative treatment(salpingoneostomy).Results this work demonstrates that in experienced hands and with an acute selection,salpingoneostomy gives results comparable to those obtained after *** cuff salpingoneostomy technique is much better than racket form salpingoneostomy and should be preferred whenever it is *** Conservative treatment should be discussed in infertile patients having hydrosalpinx after a careful selection.

Oxygen reduction reaction(ORR)plays an important role in dictating the performance of various electrochemical energy *** platinum nanoparticles have served as the catalysts of choice towards ORR,minimizing the cost of the catalysts by diminishing the platinum nanoparticle size has become a critical route to advancing the technological ***,first-principle calculations show that carbon-supported Pt9 clusters represent the threshold domain size,and the ORR activity can be significantly improved by doping of adjacent cobalt *** is confirmed experimentally,where platinum and cobalt are dispersed in nitrogen-doped carbon nanowires in varied forms,single atoms,few-atom clusters,and nanoparticles,depending on the initial *** sample consisting primarily of Pt_(2~7)clusters doped with atomic Co species exhibits the best mass activity among the series,with a current density of 4:16Amg^(-1)_(Pt)at+0.85V *** that is almost 50 times higher than that of commercial Pt/C.
