The present study aims to evaluate the environmental effect on fungal community composition associated with biodeterioration occurring in stones （soapstone） at two distinct locations in Minas Gerais State, Brazil： Congonhas city and Sanctuary of Cara^a. Four collections of fungal communities over one year were obtained from both research sites from the soapstone block surfaces exposed for over two decades. The molecular diversity profile of the fungal community at the two localities was obtained by DGGE （Denaturing Gradient Gel Electrophoresis）, and the genomes of the most representative population were sequenced. DGGE showed the formation of two clusters with filamentous fungal communities. Sequencing of the most representative bands revealed the presence of fungi associated with the biodeterioration of soapstone. In addition, many of the identified species were associated with photobionts that could generate lichens, indicating that environmental characteristics affect the occurrence of filamentous fungi, which leads to biodeterioration of stones. Authors＇ study focused on an environmental variation of an extreme habitat for fungi associated with soapstone in the state of Minas Gerais, Brazil and identified the presence of interesting rock-inhabiting fungal communities including species related to lichens, which can accelerate the deterioration of stones by the production of organic acids.

Immunogenicity and virus-like particle formation of rotavirus capsid proteins produced in transgenic plants

The human pathogen, group A rotavirus, is the most prevalent cause of acute infantile and pediatric gastroenteritis worldwide, especially in developing countries. There is an urgent demand for safer, more effective and cheaper vaccines against rotavirus infection. Plant-derived antigens may provide an exclusive way to produce economical subunit vaccines. Virus-like particles, constituting viral capsid proteins without viral nucleic acids, are considered a far safer candidate compared with live attenuated viral vaccines. In this study, the rotavirus capsid proteins VP2, VP6 and VP7 were co-expressed in transgenic tobacco plants, and their expression levels, formation of rotavirus-like particles (RV VLPs) and immunogenicity were extensively studied. Quantitative real-time RT-PCR and Western blot analysis revealed that the expression level of vp6 was the highest while vp7 was expressed at the lowest levels. The RV VLPs were purified from transgenic tobacco plants and analyzed by electron microscopy and Western blot. Results indicated that the plant-derived VP2, VP6 and VP7 proteins self-assembled into 2/6 or 2/6/7 RV VLPs with a diameter of 60-80 nm. When orally delivered into mice with cholera toxin as an adjuvant, the total soluble protein extracted from transgenic tobacco plants induced rotavirus-specific antibodies comparable with those of attenuated rotavirus vaccines, while VP 2/6/7 induced higher serum IgG and fecal IgA titers compared with VP 2/6.

The complete mitochondrial DNA sequence contains useful information for phylogenetic analyses of metazoa. In this study, the complete mitochondrial DNA sequence of sea cucumber Stichopus horrens （Holothuroidea： Sdchopodidae： Stichopus） is presented. The complete sequence was determined using normal and long PCRs. The mitochondrial genome of Stichopus horrens is a circular molecule 16257 bps long, composed of 13 protein-coding genes, two ribosomal RNA genes and 22 transfer RNA genes. Most of these genes are coded on the heavy strand except for one protein-coding gene （had6） and five tRNA genes （tRNAser（UcN）, tRNAGtn, tRNAAla, tRNA val, tRNAASp） which are coded on the light strand. The composition of the heavy strand is 30.8% A, 23.7% C, 16.2% G, and 29.3% T bases （AT skew=0.025; GC skew=-0.188）. A non-coding region of 675 bp was identified as a putative control region because of its location and AT richness. The intergenic spacers range from 1 to 50 bp in size, totaling 227 bp. A total of 25 overlapping nucleotides, ranging from 1 to 10 bp in size, exist among 11 genes. All 13 protein-coding genes are initiated with an ATG. The TAA codon is used as the stop codon in all the protein coding genes ex- cept nad3 and nad4 that use TAG as their termination codon. The most frequently used amino acids are Leu （16.29%）, Ser （10.34%） and Phe （8.37%）. All of the tRNA genes have the potential to fold into typical cloverleaf secondary structures. We also compared the order of the genes in the mitochondrial DNA from the five holothurians that are now available and found a novel gene arrangement in the mitochondrial DNA of Stichopus horrens.

Population growth has consequences for intense use of aquatic ecosystems. Samples were taken, between 2013 and 2014, in the reservoirs ＂Algodoeiro＂ and ＂Gloria＂ in Sergipe, Brazil. TSI （Trophic State Index） and WQI （Water Quality Index） were used. It was determined the parameters such as conductivity, dissolved oxygen, total nitrogen, total phosphorus and chlorophyll. TSI has been applied, in ＂Algodoeiro＂, which was classified as mesotrophic. With the increase of rainfall in 2014, this reservoir changed its condition to eutrophic. ＂Gloria＂ reservoir was already classified in super-eutrophication. Regarding the WQI, the reservoirs were classified in Class IV, indicating a critically degraded condition.

In order to understand the effect of different land use on soil physics and nutrients properties of the debris flow bottomland, a case study at Daqing gully （in Xiaojiang Basin, Yunnan） was conducted in 2004. Soil samples were taken at depth of 0-10,10-20,20-40,40-60,60-80 cm under three land use patterns crop bottomland （CL）, forest bottomland （FL）, and barren bottomland（BL）. The results showed that the developing bottomland to CL promoted soil toaccumulate total phosphorus （TP） and available phosphorus （AP）, pH value transferred from neutral to alkalescency, and organic matter decreased significantly. Furthermore, the contents of total nitrogen （TN） and available nitrogen （AN） in CL were lower than that of FL and BL because the growth of crops consumed more nutrients in soil. The results also showed that the contents of TP, AP and available potassium （AK） in soil were positively correlated with soil particle.

The role of the intraseasonal oscillation (ISO) on the seasonal and interannual variations of the Indian summer monsoon is investigated based on the analysis of observational data. It is shown that the ISO significantly contributes to the establishment of low-level westerlies during the monsoon onset and developing periods. The effect of the ISO on the annual cycle of the monsoon is through nonlinear eddy momentum transport. On the interannual timescale, the Indian summer monsoon rainfall exhibits a significant out-of-phase relationship with the ISO intensity over the Indian monsoon region. In strong ISO years it appears the weak monsoon when there is an abnormal high over the India subcontinent in the lower troposphere. In weak ISO years there exists an abnormal low and the strong monsoon ap-pears.

Chlorhexidine (CHX) is regarded as one of the most successful antiplaque agents in controlling the formation of dental biofilm. Nevertheless, molecular mechanisms of their effects in Streptococcus mutans are largely unknown. In this work, the effects of sub-lethal and lethal concentrations of chlorhexidine (CHX) on planktonic or biofilm-organized Streptococcus mutans cells were investigated in dose- and time-dependent manner. The Minimum Inhibitory Concentration (MIC) and the Minimum Bactericidal Concentration (MBC) for planktonic cells and biofilm conditions were determined by standard methods. Quantitative PCR (qPCR) was used to quantify the relative levels of glucosyltransferase B (gtfB), gtfC and gtfD transcription of S. mutans in the presence of CHX. The CHX activity in the initial biofilm structure and morphological alterations in planktonic cells were examined by Scanning Electron Microscopy (SEM). The results indicate that CHX increased expression of gtfC and gtfD in planktonic S. mutans cells and CHX reduced the expression of gtfB, gtfC, and gtfD in biofilms. High concentrations of CHX resulted in several wilted S. mutans planktonic cells with spilled intracellular material, while decreased cells’ chain length and matrix was found when the initial biofilm was exposed to increasing concentrations of CHX. CHX’s effects against bacteria depend on the type of growth organization and the concentration and time of exposure to the drug. At sub-lethal concentrations, CHX affects the expression of glucosyltransferases, which may have anticariogenic effect.
