Beach pea or beach cowpea(Vigna marina(Burm.)Merr.)belongs to the family *** is a close relative of cultivated Vigna species such as adzuki bean(***),cowpea(***),mung bean(***),and blackgram(***),and is distributed throughout the *** its ability to tolerate salt stress,beach pea has great potential to contribute salt-tolerance genes for developing salt-tolerant cultivars in cultivated Vigna ***,it is still underutilized in Vigna breeding programs.A draft genome sequence of beach pea was generated using a high-throughput next-generation sequencing platform,yielding 23.7 Gb of sequence from 79,929,868 filtered reads.A de novo genome assembly containing 68,731 scaffolds gave an N50 length of 10,272 bp and the assembled sequences totaled 365.6 Mb.A total of 35,448 SSRs,including 3574 compound SSRs,were identified and primer pairs for most of these SSRs were *** analysis identified 50,670 genes with mean coding sequence length 1042 *** analysis revealed highest sequence similarity with ***,followed by *** with the *** genome revealed 16,699 SNPs and 2253 InDels and comparison with the *** genome revealed 17,538 SNPs and 2300 *** our knowledge this is the first draft genome sequence of beach pea derived from an accession(ANBp-14-03)adapted locally in the Andaman and Nicobar Islands of *** draft genome sequence may facilitate the genetic enhancement in cultivated Vigna species.

Down Regulation of Survivin Gene and Up Regulation of p53 Gene expression by siRNA Induces Apoptosis in human Hepatocellular Carcinoma cell Line Hepg2

Survivin gene may be a good target for cancer gene therapy because it is over expressed in a variety of human tumors including human hepatocellular carcinoma but not in differen- tiated adult tissues. To explore the effects of the siRNA of survivin gene inducing apoptosis in human hepatocellular cancer cells, three siRNAs cpusiRNA1, cpusiRNA2 and cpusiRNA3 were designed and transferred into human hepatocellular carcinoma cell line Hepg2 (Hepg2) by lipofection. MTT test showed that the growth of Hepg2 decreased when it was transfected with 25nM, 50nM, 100nM, 150nM, 200nM, 400nM siRNA respectively after 48 hours. And the change of mRNA and protein of survivin gene and p53 gene had been detected by RT-PCR and Western blot. Cells presented an increase in apoptosis index was assayed by flow cytometry. Small interfering RNA can exert a knockdown of survivin gene expression and up regulation of p53 gene to induce apoptosis and to inhibit the growth of Hepg2.

Assessment of Variation in Morpho-Physiological Traits and Genetic Diversity in Relation to Submergence Tolerance of Five Indigenous Lowland Rice Landraces

The present study evaluated submergence responses in 88 lowland indigenous rice(Oryza sativa L.) landraces from Koraput, India, to identify submergence-tolerant rice genotypes. In pot experiments, variations in survival rate, shoot elongation, relative growth index, dry matter, chlorophyll, soluble sugar and starch contents were evaluated in two consecutive years under well-drained and completely submerged conditions. Principal component analysis showed that the first three axes contributed 96.820% of the total variation among the landraces, indicating wide variation between genotypes. Major traits such as survival rate, relative growth index, soluble sugar and starch contents appeared to be important determinants of phenotypic diversity among the landraces. Phenotypic coefficient of variance was higher than genotypic coefficient of variance for all the traits and all showed high heritability(90.38%–99.54%). Five rice landraces(Samudrabali, Basnamundi, Gadaba, Surudaka and Dokarakuji) were the most tolerant to submergence. When submerged for up to 14d, Samudrabali, Basnamundi and Godoba were notable for having greater survival rates than a standard submergence tolerant variety FR13 A, and also notable for elongating more vigorously and accumulating more biomass. These three landraces may therefore be especially useful in lowland rice growing areas that are affected by both moderate stagnant water and flash flooding. Molecular genotyping revealed that the submergence tolerance of Samudrabali, Basnamundi and Godoba is linked to the presence of one or more different Sub1 loci and it may well prove useful for breeding improved submergence tolerant rice varieties, thereby assising to improve yield stability in the rainfed lowland agro-ecosystem.

Insights on the solubilization products after combined alkaline and ultrasonic pre-treatment of sewage sludge

This work provides insights on the solubilization products after a simultaneous combination of alkaline and ultrasonic （ALK ＋ ULS） pre-treatment of sewage sludge. Soluble chemical oxygen demand （SCOD） increased from 1200 to 11,000 mg/L after such treatment. Organics with molecular weight around 5.6 kDa were solubilized because of the synergistic effect of ultrasound and alkali. Organics with molecular weight larger than 300 kDa increased from 7.8% to 60%, 16% and 42.3% after ULS, ALK and ALK ＋ ULS treatment, respectively. Excitation emission matrix fluorescence spectroscopy analysis identified soluble microbial product-like and humic acid-like matters as the main solubilization products. Sludge anaerobic biodegradability was significantly enhanced with the simultaneous application of ALK ＋ ULS pre-treatment. ALK ＋ ULS pre-treatment resulted in 37.8% biodegradability increase compared to the untreated sludge. This value was higher compared to the biodegradability increase induced by individual ALK pre-treatment （5.7%） or individual ULS pre-treatment （20.7%） under the same conditions applied.

Impact of ozone assisted ultrasonication pre-treatment on anaerobic digestibility of sewage sludge

Impact of ultrasonication（ULS） and ultrasonication-ozonation（ULS-Ozone） pre-treatment on the anaerobic digestibility of sewage sludge was investigated with semi-continuous anaerobic reactors at solid retention time（SRT） of 10 and 20 days. The control, ULS and ULS-Ozone reactors produced 256, 309 and 348 m L biogas/g CODfedand the volatile solid（VS） removals were 35.6%, 38.3% and 42.1%, respectively at SRT of 10 days. At SRT of20 days, the biogas yields reached 313, 337 and 393 m L biogas/g CODfedand the VS removal rates were 37.3%, 40.9% and 45.3% in the control, ULS and ULS-Ozone reactors, ***-Ozone pre-treatment increased the residual organic amount in the digested *** soluble residual organics were found to contain macromolecules with molecular weights（MW） larger than 500 k Da and smaller polymeric products with MW around 19.4and 7.7 k Da. These compounds were further characterized to be humic acid-like substances with fluorescent spectroscopy analysis.

Clinical Study on Autosomal Dominant Polycystic Kidney Disease among South Indians

Autosomal dominant polycystic kidney disease (ADPKD) is a commonly inherited disorder in humans, with a frequency of 1 in 100 in the general population. ADPKD is caused by mutations in PKD1 gene (85%) located on human chromosome 16p13.3;Mutations in the PKD2 gene contribute to 15% of ADPKD incidence and is located on human chromosome 4q2-23. A total of hundred ADPKD patients and age and sex matched healthy individuals were selected for the study. The study was aimed to evaluate the prevalence of ADPKD, lipid profile and level of calcium (Ca), sodium (Na), iron (Fe) and potassium (K) in patients with ADPKD and in healthy individuals. The lipid profile was analyzed using commercially available span kit and semiautoanalyzer (Erba, Chem 5X). The Ca, Na, Fe and K concentrations were estimated by flame photometry (ELICO, CL 22D) and atomic absorption spectroscopy (ELICO, SL 173) and significant changes were noted at p 0.05. The ADPKD patients were observed to have lipid abnormalities, hyponatremia, cholesterolemia, renal osteodystrophy, cardiovascular problems and anemia. Therefore, the study reveals an association between the lipid profile and Ca, Na, Fe and K levels with ADPKD among South Indian population.

Whiteflies(Bemisia tabaci)are polyphagous invasive hemipteran insects that cause serious losses of important crops by directly feeding on phloem sap and transmitting pathogenic *** insects have emerged as a major threat to global agriculture and food *** synthesized insecticides are currently the only option to control whiteflies,but the ability of whiteflies to evolve resistance against insecticides has made the management of these insects very *** host-plant resistance against whiteflies identified in some crop plants has not been exploited to a great *** engineering approaches,such as transgenics and RNA interference(RNAi),are potentially useful for the control of *** plants harboring insecticidal toxins/lectins developed via nuclear or chloroplast transformation are a promising vehicle for whitefly ***-stranded RNAs(dsRNAs)of several insect genes,delivered either through microinjection into the insect body cavity or orally via an artificial diet and transiently or stably expressed in transgenic plants,have controlled whiteflies in model plants and in some crops at the laboratory level,but not at the field *** this review,we highlight the merits and demerits of each delivery method along with strategies for sustained delivery of dsRNAs via fungal entomopathogen/endosymbiont or nontransgenic RNAi approaches,foliar sprays,root absorption or nanocarriers as well as the factors affecting efficient RNAi and their biosafety *** sequencing and transcriptome studies of whitefly species are facilitating the selection of appropriate genes for RNAi and gene-editing technology for the efficient and resilient management of whiteflies and their transmitted viruses.

Genetic Diversity in ex-situ Conserved Lens culinaris for Botanical Descriptors, Biochemical and Molecular Markers and Identification of Landraces from Indigenous Genetic Resources of Pakistan

Lentil, one of the oldest legumes was investigated for diversity based on botanical descriptors, total seed proteins, isozymes and random amplified polymorphic DNA （RAPD） markers. About one fourth of accessions were heterogeneous for botanical descriptors and a seed protein profile. The germplasm collected from the province of Baluchiatan revealed the prevalence of indigenous landraces as high diversity was observed for all of the techniques. Diversity explored through various techniques revealed validity irrespective of the sample size or geographic pattem, RAPD being the best choice for investigating both inter- and intra-acceaaion variation in lentil. Although all of the techniques were able to resolve genetic diversity in lentil, isozymea and seed proteins gave low levels of genetic diversity, suggesting that more investigation into isozymea of specific proteins is required. RAPD is the best option for determining inter- and intra-acceaaion variation, and will be required to extend germplaama and primers to continue the study of botanical descriptors.
