Novel blood-based microRNA biomarker panel for early diagnosis of pancreatic cancer

AIM:To develop a panel of blood-based diagnostic biomarkers consisting of circulating microRNAs for the detection of pancreatic cancer at an early ***:Blood-based circulating microRNAs were profiled by high throughput screening using microarray analysis,comparing differential expression between early stage pancreatic cancer patients(n = 8) and healthy controls(n = 11).A panel of candidate microRNAs was generated based on the microarray signature profiling,including unsupervised clustering and statistical analysis of differential expression levels,and findings from the published *** selected candidate microRNAs were then confirmed using TaqMan real-time quantitative reverse transcription polymerase chain reaction(RT-qPCR) to further narrow down to a three-microRNA diagnostic *** three-microRNA diagnostic panel was validated with independent experimental proce-dures and instrumentation of RT-qPCR at an independent venue with a new cohort of cancer patients(n = 11),healthy controls(n = 11),and a group of high risk controls(n = 11).Receiver operating characteristic curve analysis was performed to assess the diagnostic capability of the three-microRNA ***:In the initial high throughput screening,1220 known human microRNAs were screened for differential expression in pancreatic cancer patients versus controls.A subset of 42 microRNAs was then generated based on this data analysis and current published *** microRNAs were selected from the list of 42 targets for confirmation study,and three-microRNAs,miR-642b,miR-885-5p,and miR-22,were confirmed to show consistent expression between microarray and *** three microRNAs were then validated and evaluated as a diagnostic panel with a new cohort of patients and controls and found to yield high sensitivity(91%) and specificity(91%) with an area under the curve of 0.97(P < 0.001).Compared to the CA19-9 marker at 73%,the three-microRNA panel has higher sensitivity although

Background: Among medical technologies that use ionizing radiation, CT is currently the radio diagnostic technic that can deliver the highest radiation to the Patient compared with other conventional procedures. In developing countries, the uses and risks of CT have not been well characterized. Objective: To estimate the lifetime attributable risk (LAR) incidence and mortality for cancer for each procedure for adult’s patients who had Computed Tomography examinations in 10 imaging centers in the city of Douala-Cameroon so as to provide a reference data. Materials and Methods: We conducted a cross-sectional study describing radiation dose associated with the 8 most common types of diagnostic CT studies performed on 1287 consecutive adult patients at 10 Douala radiology department. We estimated lifetime attributable risks of cancer by study type from these measured doses. Estimation of LAR for cancer incidence and mortality was based on the effective dose, patient’s sex and age at exposure using the BIER VII preferred models. Results: Mean effective dose from CT scans examinations varied from: 0.30 and 8.81 mSv. The highest doses were observed for lumbar spine CT (8.81 mSv), followed by abdomen-pelvis procedure (6.46 mSv), chest-abdomen-pelvic CT (6.61 mSv), chest CT (3.90 mSv), cervical Spine CT (3.05 mSv), head CT (1.7 mSv) and lower for sinus CT (0.30 mSv). The LAR values of all cancer from patients’ CT scans obtained vary from 67.13 excess per 100,000 (about 1 in 1489) and 0.45 excess per 100,000 (about 1 in 222,222). All cancer risk was high for lumbar spine CT in women 20 years old (67.13 excess deaths in 100,000 scans) followed by chest-abdomen-pelvic CT (50.36 excess deaths in 100,000 scans) and abdomen-pelvic CT (49.22 excess deaths in 100,000 scans) for the same age group. Rapid and accurate detection of glucose is of great significance for diabetic *** sensitive glucose sensors promise to achieve noninvasive detection technology,enabling more convenient and efficient means for large-scale screening and long-term dynamic monitoring of diabetes *** this work,we demonstrate a sensitive glucose electrochemical biosensor through the synergetic labelling strategy utilizing PbS colloidal quantum dots(CQDs)and Au nanospheres(AuNSs).The PbS CQDs/AuNSs/glucose oxidase(GOx)mixture could be stably immobilized on the carbon electrode surface via the onestep dip-coating *** electrochemical biosensor employing PbS CQDs/AuNSs/GOx-modified electrode integrates the functions of specific molecule recognition,signal transduction as well as signal *** sensor is capable of transducing the glucose enzyme-catalyzed reaction into significant current signals,exhibiting a good linear response in the glucose concentration range of 0.1μM-10 mM with the limit of detection being 1.432 nM.

Immune checkpoint inhibitors(ICIs)therapy targeting programmed cell death ligand 1(PD-L1)and programmed death protein 1(PD-1)had exhibited significant clinical benefits for cancer treatment such as triple negative breast cancer(TNBC).However,the relatively low anti-tumor immune response rate and ICIs drug resistance highlight the necessity of developing ICIs combination therapy strategies to improve the anti-tumor effect of ***,the immunomodulator epigallocatechin gallate palmitate(PEGCG)and the immunoadjuvant metformin(MET)self-assembled into tumor-targeted micelles via hydrogen bond and electrostatic interaction,which encapsulated the therapeutic agents doxorubicin(DOX)-loaded PEGCG-MET micelles(PMD)and combined with ICIs(anti-PD-1 antibody)as therapeutic strategy to reduce the endogenous expression of PD-L1 and improve the tumor immunosuppressive *** results presented that PMD integrated chemotherapy and immunotherapy to enhance antitumor efficacy in vitro and in vivo,compared with DOX or anti-PD-1 antibody for the therapy of *** micelles might be a potential candidate,which could remedy the shortcomings of antibody-based ICIs and provide synergistic effect to enhance the antitumor effects of ICIs in tumor therapy.

The application of extracellular vesicles,particularly exosomes(EXs),is rapidly expanding in the field of medicine,owing to their remarkable properties as natural carriers of biological *** study investigates utilization of exosomes derived from stromal cells of tumor adjacent normal tissues(NAF-EXs)for personalized medicine,which can be derived at the time of diagnosis by endoscopic ***,we show that exosomes(EXs)derived from NAFs demonstrate differential bio-physical characteristics,efficient cellular internalization,drug loading efficiency,pancreatic tumor targeting and delivery of ***-derived EXs(NAF-EXs)were used for loading ormeloxifene(ORM),a potent anti-cancer and desmoplasia inhibitor as a model *** found that ORM maintains normal fibroblast cell phenotype and renders them incompatible to be triggered for a CAF-like phenotype,which may be due to regulation of Ca^(2+) influx in fibroblast ***-EXs-ORM effectively blocked oncogenic signaling pathways involved in desmoplasia and epithelial mesenchymal transition(EMT)and repressed tumor growth in xenograft mouse *** conclusion,our data suggests preferential tropism of NAF-EXs for PDAC tumors,thus imply feasibility of developing a novel personalized medicine for PDAC patients using autologous NAF-EXs for improved therapeutic outcome of anti-cancer ***,it provides the opportunity of utilizing this biological scaffold for effective therapeutics in combination with standard therapeutic regimen.

Understanding the ultrafast carrier dynamics and the mechanism of two-dimensional(2D)transition metal dichalcogenides(TMDs)is key to their applications in the field of optoelectronic *** this work,a single pulse pump probe method is introduced to detect the layer-dependent ultrafast carrier dynamics of monolayer and few-layer WS_(2) excited by a femtosecond *** show that the ultrafast carrier dynamics of the layered WS_(2) films can be divided into three stages:the fast photoexcitation phase with the characteristic time of 2–4 ps,the fast decay phase with the characteristic time of 4–20 ps,and the slow decay phase lasting several hundred ***,the layer dependency of the characteristic time of each stage has been observed,and the corresponding mechanism of free carrier dynamics has been *** has been observed as well that the monolayer WS_(2) exhibits a unique rising time of carriers after *** proposed method can be expected to be an effective approach for studying the dynamics of the photoexcited carriers in 2D *** results provide a comprehensive understanding of the photoexcited carrier dynamics of layered WS_(2),which is essential for its application in optoelectronics and photovoltaic devices.

Perovskite solar cells(PSCs)have attracted widespread attention because of their remarkable efficiency,low cost,and ease of ***,the operational stability of the PSCs still suffers from the corrosion of metal electrodes induced by metal-halide ***,we propose a feasible strategy for improving the stability of inverted PSCs by using magnetron-sputtered Mo rear *** with a bismuth(Bi)buffer layer prepared by thermal evaporation,the damage from the magnetron sputtering process toward the underlayers can be effectively ***,based on the Bi+Mo bilayer electrode,the inverted PSCs with a NiMgLiO hole transport layer exhibited a power conversion efficiency(PCE)of 18.82%.In addition to their excellent stability at high temperatures,compact Mo films can inhibit the decomposition of Perovskite(Pvk)films in the *** PSCs with the Bi/Mo bilayer electrode maintained 97.9%of its initial efficiency,showing better stability than PSCs with the traditional Ag electrode after aging for 3000 h under continuous light illumination.
