Background Travoprost has been widely used for the treatment of patients with open-angle glaucoma （OAG） or ocular hypertension （OH）. The aim of this study was to evaluate the intraocular pressure （lOP） lowering efficacy of travoprost 0.004% monotherapy in patients previously treated with other topical hypotensive medications, and in previously untreated patients. Methods This open-label, 12-week study in 1651 adult patients with ocular hypertension or open-angle glaucoma who were untreated or required a change in therapy （due to either inadequate efficacy or safety issues） as judged by the investigator was conducted at 6 sites in China. Previously treated patients were instructed to discontinue their prior medications at the first visit. All the patients were dosed with travoprost 0.004% once-daily at 8 p.m. in both eyes for 12 weeks. Efficacy and safety evaluations were conducted at week 4 and 12. lOP measurements were performed at the same time of day at the follow-up visits. Results For patients transitioned to travoprost, mean lOP reductions from baseline in untreated and treated patients with different prior medications at week 12 were： latanoprost, （4.3±4.6） mmHg; β-blocker, （6.3±4.0） mmHg; α-agonist, （7.5±4.3） mmHg; topical carbonic anhydrase inhibitors, （8.0±4.9） mmHg. All mean lOP changes from baseline were statistically significant （P 〈0.001）. No treatment-related serious adverse events were reported in this study. Conclusions In patients treated with other hypotensive medications or untreated, the lOP reduction with travoprost was significant. The results of this study demonstrated the potential benefit of using travoprost as a replacement therapy in order to ensure adequate lOP control. Travoprost administered once daily was safe and well tolerated in patients with glaucoma or ocular hypertension.

Cultivated peanut (Arachis hypogaea) is an allotetraploid crop planted in Asia, Africa, and America for edible oil and protein. To explore the origins and consequences of tetraploidy, we sequenced the allotetraploid A. hypogaea genome and compared it with the related diploid Arachis duranensis and Arachis ipaensis genomes. We annotated 39 888 A-subgenome genes and 41 526 B-subgenome genes in allotetraploid peanut. The A. hypogaea subgenomes have evolved asymmetrically, with the B subgenome resembling the ancestral state and the A subgenome undergoing more gene disruption, loss, conversion, and transposable element proliferation, and having reduced gene expression during seed development despite lacking genome-wide expression dominance. Genomic and transcriptomic analyses identified more than 2 500 oil metabolism-related genes and revealed that most of them show altered expression early in seed development while their expression ceases during desiccation, presenting a comprehensive map of peanut lipid biosynthesis. The availability of these genomic resources will facilitate a better understanding of the complex genome architecture, agronomically and economically important genes, and genetic improvement of peanut.

全球SARS-CoV-2病毒的不同变异株导致已接种疫苗的人群中突破性感染的比例上升,迫切需要研发更为有效和广谱的COVID-19疫苗.本研究报告了一项mRNA疫苗RQ3013的临床前研究结果,旨在提供对SARS-CoV-2各种变异株(VOCs)的广谱保护.RQ3013疫苗包含经过假尿苷修饰的mRNA,制备成脂质纳米颗粒的形式.该mRNA编码了SARS-CoV-2的刺突(S)蛋白嵌合体,包含与免疫逃逸有关的重要突变位点.我们在多种动物模型中对RQ3013的免疫原性、保护效果和安全性进行了评估.RQ3013在小鼠、仓鼠和非人灵长类动物(NHP)中引发了显著的免疫反应.它能够诱导高滴度的抗体产生,具备对野生型、B.1.1.7、B.1.351、B.1.617.2和奥密克戎系列变异株的广泛中和能力.在小鼠和NHP中,两剂RQ3013疫苗能够有效保护上呼吸道和下呼吸道免受SARS-CoV-2及其变异株的感染.此外,在NHP中对RQ3013的安全性进行了评估,未观察到不良反应.这些结果为在临床试验中评估RQ3013提供了有力的支持,表明它可能成为对抗COVID-19及其变种的广泛保护的有希望的候选疫苗.

Prevalence of depressive and anxiety disorders in Chinese gastroenterological outpatients

AIM:To investigate the prevalence and physicians' detection rate of depressive and anxiety disorders in gastrointestinal(GI)outpatients across China. METHODS:A hospital-based cross-sectional survey was conducted in the GI outpatient departments of 13 general hospitals.A total of 1995 GI outpatients were recruited and screened with the Hospital Anxiety and Depression Scale(HADS).The physicians of the GI departments performed routine clinical diagnosis and management without knowing the HADS score *** with HADS scores≥8 were subsequently interviewed by psychiatrists using the Mini International Neuropsy-chiatric Interview(MINI)to make further diagnoses. RESULTS:There were 1059 patients with HADS score ≥8 and 674(63.64%)of them undertook the MINI interview by *** on the criteria of Diagnostic and Statistical Manual of Mental Disorders (4th edition),the adjusted current prevalence for depressive disorders,anxiety disorders,and comorbidity of both disorders in the GI outpatients was 14.39%, 9.42%and 4.66%,*** of depressive disorders with suicidal problems[suicide attempt or suicide-related ideation prior or current;module C (suicide)of MINI score≥1]was 5.84%in women and 1.64%in *** GI physicians'detection rate of depressive and anxiety disorders accounted for 4.14%. CONCLUSION:While the prevalence of depressive and anxiety disorders is high in Chinese GI outpatients, the detection rate of depressive and anxiety disorders by physicians is low.

Background:Erythropoietin is a glycoprotein that mainly regulates *** patients with chronic renal failure with anemia,darbepoetin alfa can stimulate erythropoiesis,correct anemia,and maintain hemoglobin *** study was designed to demonstrate the efficacy and safety of darbepoetin alfa injections as being not inferior to epoetin alfa injections(Recombinant Human Erythropoietin injection,rHuEPO)when maintaining hemoglobin(Hb)levels within the target range(10.0-12.0 g/dL)for the treatment of renal ***:Ninety-five patients were enrolled in this study from April 15,2013 to April 10,2014 at 25 *** this study,patients(n=95)aged 18-70 years were randomized into a once per week intravenous darbepoetin alfa group(n=56)and a twice or three times per week intravenous epoetin alfa group(n=39)for 28 weeks,who had anemia with hemoglobin levels between 6 g/dL and 10 g/dL due to chronic kidney disease(CKD)and were undergoing hemodialysis or hemofiltration with ESA-naive(erythropoiesis stimulating agent-naive).The primary efficacy profile was the mean Hb level(the non-inferiority margin was-1.0 g/dL,week 21-28);the secondary efficacy profiles were the Hb increase rate(week 0-4),the target Hb achievement cumulative rate and time,the change trends of the Hb levels,and the target Hb maintenance *** events(AEs)were observed and compared,and the efficacy and safety were analyzed between the two treatment ***,the frequencies of dose adjustments between the darbepoetin alfa and epoetin alfa groups were compared during the treatment ***?software version 9.2 was used to perform all statistical *** statistics were used for all efficacy,safety,and demographic variable analyses,including for the primary efficacy ***:The mean Hb level was 11.3 g/dL in the darbepoetin alfa group and 10.7 g/dL in the epoetin alfa group,respectively;the difference of the lower limits of the 95%confidence intervals(C

China’s coastal wetlands provide breeding,migration stopover,and wintering habitats for about 230 waterbird species,which is more than a quarter of all waterbirds in the ***-scale and high intensity human activities have resulted in serious loss and degradation of coastal wetlands over the past half century,causing population declines in many *** a literature review and expert surveys,this article reviews conservation measures taken in recent decades to protect waterbirds in China’s coastal wetlands and provides recommendations for future conservation action from three aspects:policy and administration,habitat conservation and management,and multiparty *** the past decades,many conservation legislation,regulations and action plans at the national level and more site-specific measures and interventions have been implemented,with notable improvement in the effectiveness in policy making and multi-stakeholder ***,some threats to waterbirds have been mitigated and many key sites for waterbirds have been designated as strictly protected nature ***,some critical issues still remain,mostly related to habitat conservation and management,such as coastal wetland restoration,control of invasive Spartina alterniflora,control of environmental pollution,and improvement of artificial habitat *** highlight that protecting natural tidal wetlands and improving habitat quality are critical for the conservation of coastal waterbirds,especially those highly dependent on the intertidal *** has demonstrated strong commitment to ecological conservation and restoration for the future,in terms of both funding and policies for biodiversity and wetland *** is important that this commitment to conserve coastal waterbirds is supported continuously by science-and evidence-based decisions and actions.

Overheating of the engine, the transmission and the hydraulic device is a problem when the construction machinery works on plateau. To solve this problem, we proposed an electro-controlled hydraulic driving fan cooling system (ECHDFCS). The system was applied to a 50-wheel loader. We carried out the coolant temperature simulation using fluid modeling software FLOWMASTER, followed by laboratory experiments and road tests. The results show that ECHDFCS can adjust the cooling capability of the system automatically based on machine heat dissipation requirements. The coolant temperature is consequently remained within an appropriate range. The simulation results are consistent with the experiment results when the experiment is performed on the plain, but are different from the road tests in some investigated parameters on the plateau.
