Insights into the structural biology of G-protein coupled receptors impacts drug design for central nervous system neurodegenerative processes

In the last few years, there have been important new insights into the structural biology of G-protein coupled receptors. It is now known that allosteric binding sites are involved in the affinity and selec- tivity of ligands for G-protein coupled receptors, and that signaling by these receptors involves both G-protein dependent and independent pathways. the present review outlines the physiological and pharmacological implications of this perspective for the design of new drugs to treat disorders of the central nervous system. Specifically, new possibilities are explored in relation to allosteric and or- thosteric binding sites on dopamine receptors for the treatment of Parkinson＇s disease, and on muscarinic receptors for Alzheimer＇s disease. Future research can seek to identify ligands that can bind to more than one site on the same receptor, or simultaneously bind to two receptors and form a dimer. For example, the design of bivalent drugs that can reach homo/hetero-dimers of D2 dopa- mine receptor holds promise as a relevant therapeutic strategy for Parkinson＇s disease. Regarding the treatment of Alzheimer＇s disease, the design of dualsteric ligands for mono-oligomeric mus- carinic receptors could increase therapeutic effectiveness by generating potent compounds that could activate more than one signaling pathway.

Quantum information entropies of the eigenstates for the Pschl-teller-like potential

Shannon entropy for lower position and momentum eigenstates of Ptschl-teller-like potential is evaluated. Based on the entropy densities demonstrated graphically, we note that the wave through of the position information entropy density p （x） moves right when the potential parameter V1 increases and its amplitude decreases. However, its wave through moves left with the increase in the potential parameter 丨V2丨. Concerning the momentum information entropy density p（p）, we observe that its amplitude increases with increasing potential parameter V1, but its amplitude decreases with increasing丨V2丨. the Bialynicki-Birula-Mycielski （BBM） inequality has also been tested for a number of states. Moreover, there exist eigenstates that exhibit squeezing in the momentum information entropy. Finally, we note that position information entropy increases with V1, but decreases with 丨V2丨, However, the variation of momentum information entropy is contrary to that of the position information entropy.

A comparative study on emergent pollutants photo-assisted degradation using ruthenium modified titanate nanotubes and nanowires as catalysts

Degradation of duloxetine: Identification of transformation products by UHPLC-ESI(+)-HRMS/MS,in silico toxicity and wastewater analysis

Duloxetine(DUL), an antidepressant drug, has been detected in surface water and wastewater effluents, however, there is little information on the formation of its transformation products(tPs). In this work, hydrolysis, photodegradation(UV irradiation) and chlorination experiments were performed on spiked distillated water, under controlled experimental conditions to simulate abiotic processes that can occur in the environment and wastewater treatment plants(WWtPs). Eleven tPs, nine from reaction with UV light and two from chlorine contact, were formed and detected by ultra-high performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry, and nine of them had their chemical structures elucidated upon analyses of their fragmentation patterns in MS/MS spectra. the formation and degradation of the tPs were observed. the parent compound was completely degraded after30 min in photodegradation and after 24 hr in chlorination. Almost all tPs were completely degraded in the experiments. the ecotoxicity and mutagenicity of the tPs were predicted based on several in silico models and it was found that a few of these products presented more ecotoxicity than DUL itself and six tPs showed positive mutagenicity. Finally, wastewater samples were analyzed and DUL and one tP, possibly formed by chlorination process, were detected in the effluent, which showed that WWtP not only did not remove DUL, but also formed a tP.

Soil contamination by toxic trace metal elements,like barium(Ba),may stimulate various undesirable changes in the metabolic activity of *** plant responses are fast and with,direct or indirect,generation of reactive oxygen species(ROS).to cope with the stress imposed by the ROS production,plants developed a dual cellular system composed of enzymatic and non-enzymatic players that convert ROS,and their by-products,into stable nontoxic *** assess the Ba stress response of two Brassicaceae species(Brassica juncea,a glycophyte,and Cakile maritime,a halophyte),plants were exposure to different Ba concentrations(0,100,200,300 and 500µM).the plants response was evaluated through their morphology and development,the determination of plant leaves antioxidant enzymatic activities and by the production of plants secondary *** indicated that the two Brassicaceae species have the ability to survive in an environment containing Ba(even at 500µM).the biomass production of *** was slightly affected whereas an increase in biomass *** was *** stress imposed by Ba activated the antioxidant defense system in the two species,noticed by the changes in the leaves activity of catalase(CAt),ascorbate peroxidase(APX)and guaicol peroxidase(GPX),and of the secondary metabolites,through the production of total phenols and *** enzymatic response was not similar within the two plant species:CAt and APX seem to have a more important role against the oxidative stress in *** while in *** is ***,total phenols and flavonoids production was more significant in the plants aerial part than in the roots,of the both *** the two Brassicaceae species response was different,in both plants catalytic and non-catalytic transformation of ROS occurs,and both were able to overcome the Ba toxicity and prevent the cell damage.

Numerical Analysis of Stress Concentration in Non-uniformly Corroded Small-Scale Specimens

A numerical evaluation of stress concentrations of corroded plate surfaces of small-scale corroded steel specimens is compared with the experimentally estimated *** specimens were cut from a steel box girder,which was initially corroded in real seawater *** surface of all corroded specimens was analysed applying photogrammetry techniques,and a statistical description of an idealised corroded surface of each specimen was *** lives of specimens are determined from the fatigue *** on experimentally obtained fatigue lives,the stress concentration factors are calculated concerning the ideally smooth *** correlation between the statistical parameters of the corroded specimen surfaces and the estimated stress concentration factors is *** corroded surfaces,converted in graphical format,are then used for the finite element modelling in ABAQUS software,and stress concentration factors are estimated from the finite element results.A convergence study is performed to determine the appropriate finite element mesh *** between experimentally obtained and numerically estimated stress concentration factors is performed as well as correlation analysis between actual and finite element predicted crack locations.

the visions of Industry 4.0 and 5.0 have reinforced the industrial *** have also made artificial intelligence incorporated as a major *** machine faults has become a solid foundation for automatically recognizing machine failure,and thus timely maintenance can ensure safe *** learning is a promising solution that can enhance the machine fault diagnosis model by borrowing pre-trained knowledge from the source model and applying it to the target model,which typically involves two *** response to the availability of multiple datasets,this paper proposes using selective and adaptive incremental transfer learning(SA-ItL),which fuses three algorithms,namely,the hybrid selective algorithm,the transferability enhancement algorithm,and the incremental transfer learning *** is a selective algorithm that enables selecting and ordering appropriate datasets for transfer learning and selecting useful knowledge to avoid negative *** algorithm also adaptively adjusts the portion of training data to balance the learning rate and training *** proposed algorithm is evaluated and analyzed using ten benchmark *** with other algorithms from existing works,SA-ItL improves the accuracy of all *** studies present the accuracy enhancements of the SA-ItL,including the hybrid selective algorithm(1.22%-3.82%),transferability enhancement algorithm(1.91%-4.15%),and incremental transfer learning algorithm(0.605%-2.68%).these also show the benefits of enhancing the target model with heterogeneous image datasets that widen the range of domain selection between source and target domains.

Abstract A few important integrals involving the product of two universal associated Legendre polynomials Pl＇m＇, （x）,Pk＇n＇（x）and x2a（1-x2）-p-1,xb（1± x）-p-1and xc（1-x2）-p-1（1 ± x）axe evaluated using the operator form of taylor＇s theorem and an integral over a single universal associated Legendre polynomial. these integrals are more general since the quantum numbers are unequal, i.e.l＇ ≠ k＇ and m＇≠ n＇ .their selection rules are a/so given. We also verify the correctness of those integral formulas numerically.
