29.1. Primary spectra The cosmic radiation incident at the top of the terrestrial atmosphere includes all stable charged particles and nuclei with lifetimes of order 108 years or longer. Technically, ＂primary＂ cosmic rays are those particles accelerated at astrophysical sources and ＂secondaries＂ are those particles produced in interaction of the primaries with interstellar gas. Thus electrons, protons and helium, as well as carbon, oxygen, iron, and other nuclei synthesized in stars, are primaries. Nuclei such as lithium, beryllium, and boron （which are not abundant end-products of stellar nucleosynthesis） are secondaries. Antiprotons and positrons are also in large part secondary. Whether a small fraction of these particles may be primary is a question of current interest.

Three-dimensional models: from cell culture to Patient-Derived Organoid and its application to future liposarcoma research

Liposarcoma is one of the most common soft tissue sarcomas, however, its occurrence rate is still rare compared to other cancers. Due to its rarity, experiments are an essential approach to elucidate liposarcoma pathobiology. Conventional cell culture-based research (2D cell culture) is still playing a pivotal role, while several shortcomings have been recently under discussion. , mouse models are usually adopted for pre-clinical analyses with expectations to overcome the issues of 2D cell culture. However, they do not fully recapitulate human dedifferentiated liposarcoma (DDLPS) characteristics. Therefore, three-dimensional (3D) culture systems have been the recent research focus in the cell biology field with the expectation to overcome at the same time the disadvantages of 2D cell culture and animal models and fill in the gap between them. Given the liposarcoma rarity, we believe that 3D cell culture techniques, including 3D cell cultures/co-cultures, and Patient-Derived tumor Organoids (PDOs), represent a promising approach to facilitate liposarcoma investigation and elucidate its molecular mechanisms and effective therapy development. In this review, we first provide a general overview of 3D cell cultures compared to 2D cell cultures. We then focus on one of the recent 3D cell culture applications, Patient-Derived Organoids (PDOs), summarizing and discussing several PDO methodologies. Finally, we discuss the current and future applications of PDOs to sarcoma, particularly in the field of liposarcoma.

Co-delivery of mRNA and SPIONs through amino-ester nanomaterials

Nanoparticles have been widely explored for combined therapeutic and diagnostic applications. For example, lipid-based nanoparticles have been used to encapsulate multiple types of agents and achieve multi-functions. Herein, we enabled a co-delivery of mRNA molecules and superparamagnetic iron oxide nanoparticles （SPIONs） by using an amino-ester lipid-like nanomaterial. An orthogonal experimental design was used to identify the optimal formulation. The optimal formulation, MPA-Ab-8 LLNs, not only showed high encapsulation of both mRNA and SPIONs, but also increased the r2 relaxivity of SPIONs by more than 1.5-fold in vitro. MPA-Ab-8 LLNs effectively delivered mRNA and SPIONs into cells, and consequently induced high protein expression as well as strong MRI contrast. Consistent herewith, we observed both mRNA-mediated protein expression and an evident negative contrast enhancement of MRI signal in mice. In conclusion, amino-ester nanomaterials demonstrate great potential as delivery vehicles for theranostic applications.

Benefits of Stochastic Optimization for Scheduling Energy Storage in Wholesale Electricity Markets

We propose a two-stage stochastic model for optimizing the operation of energy storage. The model captures two important features: uncertain real-time prices when day-ahead operational commitments are made;and the price impact of charging and discharging energy storage. We demonstrate that if energy storage has full flexibility to make real-time adjustments to its day-ahead commitment and market prices do not respond to charging and discharging decisions, there is no value in using a stochastic modeling framework, i.e., the value of stochastic solution is always zero. This is because in such a case the energy storage behaves purely as a financial arbitrageur day ahead, which can be captured using a deterministic *** show also that prices responding to its operation can make it profitable for energy storage to "waste" energy, for instance by charging and discharging simultaneously, which is normally sub-optimal. We demonstrate our model and how to calibrate the price-response functions from historical data with a practical case study.

Machine Learning-Based Presymptomatic Detection of Rice Sheath Blight Using Spectral Profiles

Early detection of plant diseases,prior to symptom development,can allow for targeted and more proactive disease *** objective of this study was to evaluate the use of near-infrared(NIR)spectroscopy combined with machine learning for early detection of rice sheath blight(ShB),caused by the fungus Rhizoctonia *** collected NIR spectra from leaves of ShBsusceptible rice(Oryza sativa L.)cultivar,Lemont,growing in a growth chamber one day following inoculation with ***,and prior to the development of any disease *** vector machine(SVM)and random forest,two machine learning algorithms,were used to build and evaluate the accuracy of supervised classification-based disease predictive *** partial least squares discriminant analysis was used to confirm the *** most accurate model comparing mockinoculated and inoculated plants was SVM-based and had an overall testing accuracy of 86.1%(N=72),while when control,mock-inoculated,and inoculated plants were compared the most accurate SVM model had an overall testing accuracy of 73.3%(N=105).These results suggest that machine learning models could be developed into tools to diagnose infected but asymptomatic plants based on spectral profiles at the early stages of disease *** testing and validation in field trials are still needed,this technique holds promise for application in the field for disease diagnosis and management.

Aging gracefully: social engagement joins exercise and enrichment as a key lifestyle factor in resistance to age-related cognitive decline

Cognitive impairment is a consequence of the normal aging process that effects many species, including humans and rodent models. Decline in hippocampal memory function is especially prominent with age and often reduces quality of life. As the aging population expands, the need for interventional strategies to prevent cognitive decline has become more pressing. Fortunately, several major lifestyle factors have proven effective at combating hippocampal aging, the most well-known of which are environmental enrichment and exercise. While the evidence supporting the beneficial nature of these factors is substantial, a less well-understood factor may also contribute to healthy cognitive aging: social engagement. We review the evidence supporting the role of social engagement in preserving hippocampal function in old age. In elderly humans, high levels of social engagement correlate with better hippocampal function, yet there is a dearth of work to indicate a causative role. Existing rodent literature is also limited but has begun to provide causative evidence and establish candidate mechanisms. Summed together, while many unanswered questions remain, it is clear that social engagement is a viable lifestyle factor for preserving cognitive function in old age. Social integration across the lifespan warrants more investigation and more appreciation when designing living circumstances for the elderly.

Conservation practices are encouraged to improve soil health and sustain agronomic crop ***-3 is often used as a multi-nutrient extractant to determine soil fertility status.A study inves-tigated the impacts of the conservation practices of gypsum,cover crops,and crop rotation on 28 Mehlich-3 extractable elements,of which 11 were considered plant nutrients,from soil at three mid-western US *** was collected from 0 to 15 and 15-30 cm depths 5 years after implementing the conservation *** consisted of(1)with and without cereal rye(Secale cereale L.)winter cover,(2)continuous soybean[Glycine max(L.)Merr.]***-corn(Zea mays L.)rotation,and(3)annual gypsum application(0,1.1,and 2.2 Mg ha-1).Differences were observed by site,depth,and conservation practice depending on the element *** interactive effects were observed among *** most consistent effect was observed for crop rotation across *** only affected the site with the greatest clay content,where more Ca and S were retained,and Mg and Mn *** crop only affected elements at this high clay site,where different elements were positively or negatively *** suggest that not one practice fits all,and optimum conservation practices must be tailored for the site.

High-grade pancreatic intraepithelial lesions： prevalence and implications in pancreatic neoplasia

BACKGROUND： High-grade pancreatic intraepithelial neoplasia（Pan IN-3）, a precursor of pancreatic ductal adenocarcinoma（PDAC）, is not univ.rsally detected in resected pancreatic neoplasms. We sought to determine the prevalence and prognostic relevance of Pan IN-3 lesions in primary surgical resections of PDACs and intraductal papillary mucinous neoplasms（IPMNs）.METHODS： A retrospective review of a tertiary care center pathology database（1/2000-6/2014） was performed. Demographics, imaging, pathology, disease-recurrence, and survival data were ***： A total of 458 patients who underwent primary pancreatic resection were included. “Pan IN-3” lesions were found in 74（16.2%） patients who either had PDAC（n=67） or main duct（MD）-IPMN（n=7）. Among IPMN-MDs, Pan IN-3 lesions were exclusively found in those with pathological evidence of chronic pancreatitis. For PDACs, the median overall survival（OS） for pancreata with Pan IN-3 lesions was significantly better than those without（OS 1.12 years, interquartile range [IQR] 0.72, 2.05 years vs OS 0.86 years, IQR 0.64,1.60 years respectively; P=0.04）. Multivariate Cox regression analysis demonstrated that the presence of Pan IN-3 lesions was associated with a reduced risk of death（HR=0.43; 95% CI： 0.23-0.82; P=0.01）.CONCLUSIONS： Following primary resection of pancreatic adenocarcinoma, the lower survival observed in patients without Pan IN-3 lesions might suggest a state of complete or accelerated transformation. Further investigations are necessary to validate these findings that might impact disease prognosis and management.

Control of Directional Macromolecular Trafficking Across Specific Cellular Boundaries:A Key to Integrative Plant Biology

There is now solid evidence that cell-to-cell trafficking of certain proteins and RNAs plays a critical role in trans-cellular regulation of gene expression to coordinate cellular differentiation and development. Such trafficking also is critical for viral infection and plant defense. The mechanisms of trafficking remain poorly understood. Although some proteins may move between cells by diffusion, many proteins and RNAs move in a highly regulated fashion. Regulation is likely achieved through interactions between distinct protein or RNA motifs and cellular factors. Some motifs and factors have been identified. One of the major focuses for future studies is to identify all motifs and their cognate factors and further elucidate their roles in trafficking between specific cells. With increasing information from such studies, we should be able to develop an understanding of the mechanisms that regulate trafficking of various proteins and RNAs across all and specific cellular boundaries. On the basis of such mechanistic knowledge, we can further investigate how the trafficking machinery has evolved to regulate developmental and physiological processes in a plant, how pathogens have co-evolved to use this machinery for systemic spread in a plant, and how plants use this machinery for counterdefense.
