Owing to the widespread distribution of mosquitoes capable of transmitting Zika virus, lack of clinical vaccines and treatments, and poor immunity of populations to new infectious diseases, Zika virus has become a global public health concern. Recent studies have found that Zika virus can continuously infect human brain microvascular endothelial *** cells are the primary components of the blood–brain barrier of the cerebral cortex, and further infection of brain tissue may cause severe damage such as encephalitis and fetal pituitary disease. The present study found that a biologically active base, piperlongumine(PL), inhibited Zika virus replication in human brain microvascular endothelial cells, Vero cells, and human umbilical vein endothelial cells. PL also significantly increased heme oxygenase-1(HO-1) gene expression, while silencing HO-1 expression and using the reactive oxygen species scavenger, N-acetylcysteine, attenuated the inhibitory effect of PL on Zika virus replication. These results suggest that PL induces oxidative stress in cells by increasing reactive oxygen species. This, in turn, induces an increase in HO-1 expression, thereby inhibiting Zika virus replication. These findings provide novel clues for drug research on the prevention and treatment of Zika virus.

Short term effect of choroid thickness in the horizontal meridian detected by spectral domain optical coherence tomography in myopic children after orthokeratology

AIM： To investigate choroidal thickness changes in the horizontal meridian after orthokeratology. METHODS： This is a prospective cross-sectional observed study. Subjects （n=30; 11.3±1.7y） with low-to-moderate myopia （-1.0 to -6.0 diopters）, wore orthokeratology （Ortho-K） lenses for 3mo. Before and after Ortho-K, OCT scans were made through the fovea in the horizontal meridian. Choroid thickness around the fovea was acquired by custom software. The analyzed regions along the horizontal meridian were divided into 7 equal zones. Ocular parameters were measured by Lenstar LS 900 non-contact biometry. RESULTS： Only the right eye ocular parameters were analyzed in this study. Before Ortho-K, choroidal thickness along the horizontal meridian was 273.7±31.8 μm in the temporal zone, 253.1±38.6 μm in the macula zone, and 194.8±52.2 μm in the nasal zone. After Ortho-K, the choroid was thicker in each horizontal zone （P〈0.05）. The increased thickness was greatest in the temporal zone （13.5±22.5 μm） and least in the nasal zone （8.4±14.2 μm）. The axial length （AL） increased 0.02 mm （P〉0.05）. The choroid thickness change in each horizontal zone was negatively correlated with AL （r, -0.3 to -0.4; P〈0.05） except one of the nasal zones. CONCLUSION： In myopic children, the thickness of the choroid is greatest in the temporal zone and thinnest in the nasal zone. After nightly Ortho-K for 3mo, the thickness increase along the horizontal meridian. The choroid thickness changes are negatively correlated with the change of AL.

Molecular imprinted nanoparticles(MINPs) can memorize the shape and functional group positions complementary to template, which account for the large drug loading capacity and slow drug release behavior as drug carriers. We synthesized MINPs via precipitation polymerization with vinblastine(VBL) as a model drug, and investigated the drug loading,releasing property in vitro and bio-distribution in vivo. The obtained MINPs, from 300 to 450 nm,had smooth surface and favorable dispersibility. The entrapment efficacy and drug loading capacity of VBL loaded MINPs(MINPs-VBL) were 83.25% and 8.72% respectively. In PBS(pH 7.4),MINPs-VBL showed sustained release behavior. The cumulative release percentage reached about 70% during 216 h and no burst release was observed. The releasing behavior of MINPsVBL in vitro conformed to the first-order kinetics model. MINPs-VBL and commercially available vinblastine sulfate injection(VBL injection) were injected via tail vein of SD rats respectively to investigate the bio-distribution. MINPs-VBL group showed higher concentration of VBL in tissues and serum than VBL injection group after 60 min, and the drug level in liver was the highest. MINPs-VBL exhibited liver targeting trend to some extent, which was based on the evaluation of drug targeting index(DTI) and drug selecting index(DSI).

AVP(4-8) Improves Cognitive Behaviors and Hippocampal Synaptic Plasticity in the APP/PS1 Mouse Model of Alzheimer’s Disease

Memory deficits with aging are related to the neurodegeneration in the brain, including a reduction in arginine vasopressin(AVP) in the brain of patients with Alzheimer’s disease(AD). AVP(4-8), different from its precursor AVP, plays memory enhancement roles in the CNS without peripheral side-effects. However, it is not clear whether AVP(4-8) can improve cognitive behaviors and synaptic plasticity in the APP/PS1 mouse model of AD. Here, we investigated for the first time the neuroprotective effects of AVP(4-8) on memory behaviors and in vivo long-term potentiation(LTP) in APP/PS1-AD *** results showed that:(1) APP/PS1-AD mice had lower spontaneous alternation in the Y-maze than wild-type(WT) mice, and this was significantly reversed by AVP(4-8);(2) the prolonged escape latency of APP/PS1-AD mice in the Morris water maze was significantly decreased by AVP(4-8), and the decreased swimming time in target quadrant recovered significantly after AVP(4-8) treatment;(3) in vivo hippocampal LTP induced by high-frequency stimulation had a significant deficit in the AD mice, and this was partly rescued by AVP(4-8);(4) AVP(4-8)significantly up-regulated the expression levels of postsynaptic density 95(PSD95) and nerve growth factor(NGF)in the hippocampus of AD mice. These results reveal the beneficial effects of AVP(4-8) in APP/PS1-AD mice,showing that the intranasal administration of AVP(4-8)effectively improved the working memory and long-term spatial memory of APP/PS1-AD mice, which may be associated with the elevation of PSD95 and NGF levels in the brain and the maintenance of hippocampal synaptic plasticity.

The incidence of hepatocellular carcinoma(HCC)is signifi-cantly lower in women than men,implying that estrogen receptors(ERs)may play an important role in this sex ***,considerable progress has been made in expanding our understanding of the mechanisms of ERs in *** one of the most important ERs,ERαfunctions as a tumor suppressor in the progression of HCC through various pathways,such as STAT3 signaling pathways,lipid metabolism-related signaling pathways,and non-coding ***,the function of ERαwas reduced with the changes of some molecules in the liver,which may develop further into HCC and make it difficult to achieve an effective hormone treatment ***,there are signs that individualized hormone therapy according to the activity of ERαwill overcome this *** on these observa-tions,it is particularly imperative to reassess and extend the function of ERα.In this review,we mainly elucidated molecular mechanisms associated with ERαin HCC and investigated the individualized hormone therapy based on these mechanisms,with the aim of providing new insights for HCC treatment.

CD74 is a protein whose initial role in antigen presentation was recognized two decades ago. Recent studies have revealed that it has additional functions as a receptor for macrophage migration inhibitory factor and as a receptor for an important human pathogen, Helicobacter pylori （H pylon）. The role of CD74 as a receptor is important because after binding of migration inhibitory factor or H pylori, NF-κB and Erkl/2 activation occurs, along with the induction of proinflammatory cytokine secretion. This review provides an up-to-date account of the functions of CD74 and how it might be involved in inflammation and cancer within the gastrointestinal tract.

Protective effect of inosine against cell death after photochemically induced focal cerebral ischemia was investigated in adult rats. Intraperitoneal injection of inosine was started 24 h before operation and continued till the animals were *** TUNEL method and Nissl staining were used to identify apoptosis and necrosis,***-positive cells were located in the transitional zone between the necrotic core and normal *** mean maximum area of the necrotic cores and mean number of TUNEL-positive cells 24 h after operation were reduced significantly in the inosine-treated rats when compared with those in the saline-treated control animals. We suggest that inosine can significantly reduce both necrosis and apoptosis following cerebral ischemia induced by photothrombosis in adult rats.
