Altered 3D genome reorganization mediates precocious myeloid differentiation of aged hematopoietic stem cells in inflammation

Inflammation is a driving force of hematopoietic stem cells(HSCs) aging, causing irreversible exhaustion of functional HSCs. However, the underlying mechanism of HSCs erosion by inflammatory insult remains poorly understood. Here, we find that transient LPS exposure primes aged HSCs to undergo accelerated differentiation at the expense of self-renewal, leading to depletion of HSCs. Meanwhile, the central regulator nuclear factor kappa B(NF-κB) mediating functional impairment by inflammation insult induces differential transcriptional response in aged HSCs compared with young HSCs, with precocious activation of myeloid lineage genes. Altered compartmentalization and chromatin loop formation are associated with aging-related differential transcriptional response in HSCs upon lipopolysaccharide(LPS)stimulation. Mechanistically, enhancer and promoter regions of myeloid lineage genes in aged HSCs are more accessible and display more rapid and prominent CTCF occupancy upon LPS stimulation. Our study provides comprehensive resources for the three-dimensional(3D)genome structure of HSCs and sheds light into the ordered genome organization and the associated transcriptome signature underlying HSCs aging.

Methionine restriction(MR)is an effective dietary strategy to regulate energy metabolism and alleviate oxidative stress and inflammation in the body,especially in the middle-aged and elderly ***,the high methionine content of meat products makes this dietary strategy impossible to combine with protein supplementation and *** barley(HB),a low-methionine cereal,not only provides the body with protein but also has improved glucose metabolism and antioxidant and anti-inflammatory ***,this study evaluated the feasibility of HB as a source of methionine-restricted dietary protein and the potential ***-aged C57BL/6J mice were fed a control diet(CON),a high-fat diet(HFD),a whole-grain HB high-fat diet(HBHF),or a HBHF+methionine diet(HBHFmet)for 25 *** results showed that the HBHF could keep the body weight,fasting glucose,insulin,homeostasis model assessment of insulin resistance(HOMA-IR),blood lipids,inflammation,and oxidative stress of HFD mice at normal *** with the HFD groups,HBHF inhibited pancreatic cell apoptosis and improved insulin secretion while improving hepatic and skeletal muscle glucose ***,these efficacies were attenuated in HBHFmet group *** findings suggest that HBHF has an MR strategy.

Low survival rate occurs in patients who initially experience a spontaneous return of circulation after cardiac arrest（CA）. In this study, we induced asphyxial CA in adult male Sprague-Daley rats, maintained their body temperature at 37 ± 0.5°C, and then observed the survival rate during the post-resuscitation phase. We examined neuronal damage in the hippocampus using cresyl violet（CV） and Fluore-Jade B（F-J B） staining, and pro-inflammatory response using ionized calcium-binding adapter molecule 1（Iba-1）, glial fibrillary acidic protein（GFAP）, and tumor necrosis factor-alpha（TNF-α） immunohistochemistry in the hippocampus after asphyxial CA in rats under normothermia. Our results show that the survival rate decreased gradually post-CA（about 63% at 6 hours, 37% at 1 day, and 8% at 2 days post-CA）. Rats were sacrificed at these points in time post-CA, and no neuronal damage was found in the hippocampus until 1 day post-CA. However, some neurons in the stratum pyramidale of the CA region in the hippocampus were dead 2 days post-CA. Iba-1 immunoreactive microglia in the CA1 region did not change until 1 day postCA, and they were activated（enlarged cell bodies with short and thicken processes） in all layers 2 days postCA. Meanwhile, GFAP-immunoreactive astrocytes did not change significantly until 2 days post-CA. TNF-α immunoreactivity decreased significantly in neurons of the stratum pyramidale in the CA1 region 6 hours post-CA, decreased gradually until 1 day post-CA, and increased significantly again 2 days post-CA. These findings suggest that low survival rate of normothermic rats in the early period of asphyxia-induced CA is related to increased TNF-α immunoreactivity, but not to neuronal damage in the hippocampal CA1 region.

Effects of oat β-glucan–based prebiotic blended formulas on defecation and gut microbiota in chinese adults: a randomized clinical trial

Digestive health is important for overall well-being， but gut health problems are common in the chinese population. The objective of this study was to investigate the effects of oat β-glucan–based prebiotic blended formulas in humans on digestive health by assessing defecation frequen-cy， defecation symptoms， stool quality， self-reported digestive feelings， defecation satisfaction， and gut bacteria. A parallel， randomized， double-blind， controlled trial was conducted in 168 eli-gible adults randomized to three groups for 14 days: Intervention Groups A and B received two different doses of oat β-glucan–based prebiotic blended formulas and a Control Group received oat β-glucan mixed with hydroxypropylmethyl-cellulose and microcrystalline cellulose. Partici-pants completed a defecation diary and Bristol stool chart daily and digestive system question-naires weekly. Fecal samples were collected on Day 0 and Day 14 to evaluate gut bacteria using 16S rDNA analysis. Both formulas improved defecation frequency， stool quality， defecation symptoms， defecation satisfaction， and on digestive health of human， compared with baseline (P chinese adults. These formulas are a feasible nutritional strategy for digestive health.

BACKGROUND:The high morbidity,high mortality and low survival rate of cardiac arrest(CA)cause a heavy global *** aimed to analyze the changes in scientific output related to CA over the past two ***:We analyzed the scientific output related to CA from 2000 to 2020 via the Web of *** data were analyzed using CiteSpace ***:In total,28,312 articles relating to CA were identified in the Web of *** volume of scientific research output in the field of global CA research was mainly distributed in the Americas,Europe and Asia,covering a wide *** the 28,312 articles,the research content of the highly cited literature mainly focused on CA,mild hypothermia treatment,and prognosis of CA ***:Various scientific methods were applied to reveal scientific productivity,collaboration,and research hotspots in the CA research *** resuscitation(CPR),extracorporeal membrane oxygenation(ECMO),survival and target temperature management are research *** research on CA will continue to focus on its treatment and prognosis to improve the survival rate of CA patients.
