Modulation of immunity and inflammatory gene expression in the gut, in inflammatory diseases of the gut and in the liver by probiotics

The potential for the positive manipulation of the gut microbiome through the introduction of beneficial microbes, as also known as probiotics, is currently an active area of investigation. The FAO/WHO define probiotics as live microorganisms that confer a health benefit to the host when administered in adequate amounts. However, dead bacteria and bacterial molecular components may also exhibit probiotic properties. The results of clinical studies have demonstrated the clinical potential of probiotics in many pathologies, such as allergic diseases, diarrhea, inflammatory bowel disease and viral infection. Several mechanisms have been proposed to explain the beneficial effects of probiotics, most of which involve gene expression regulation in specific tissues, particularly the intestine and liver. Therefore, the modulation of gene expression mediated by probiotics is an important issue that warrants further investigation. In the present paper, we performed a systematic review of the probiotic-mediated modulation of gene expression that is associated with the immune system and inflammation. Between January 1990 to February 2014, PubMed was searched for articles that were published in English using the MeSH terms “probiotics' and 'gene expression' combined with “intestines', 'liver', 'enterocytes', 'antigen-presenting cells', 'dendritic cells', 'immune system', and 'inflammation'. Two hundred and five original articles matching these criteria were initially selected, although only those articles that included specific gene expression results (77) were later considered for this review and separated into three major topics: the regulation of immunity and inflammatory gene expression in the gut, in inflammatory diseases of the gut and in the liver. Particular strains of Bifidobacteria, Lactobacilli, Escherichia coli, Propionibacterium, Bacillus and Saccharomyces influence the gene expression of mucins, Toll-like receptors, caspases, nuclear factor-κ

Objective:To investigate in vitro larvicidal and antioxidant enzymes potential of the medicinal plants Ginkgo biloba（***）,Stevia rebaudiana（***） and Parthenium hysterophorous（***） against Anopheles stephensi（***） 4th instars larvae. Methods:For evaluation of larvicidal potential,the ethanolic,methanolic and dichloromethane leaves extracts of three different plants were used in dose-dependent experiments in two media, while the antioxidant enzymes activities were investigated using four different methods viz., superoxide dismutase,peroxidase,ascorbate and ***:*** has developed resistance to various synthetic insecticides,making its control increasingly *** comparative performance of ethanolic extracts（65%-90%） was found better than the methanolic extract（70%-87%） and dichloromethane extract（60%-70%）.Among the three plants extracts tested in two media,*** exhibited higher larvicidal activity with LC50（24 h） in methanolic extract than *** and *** and *** exhibited the strongest antioxidative enzymes activity and *** were less active and no significant difference was ***:These three plants exhibit larvicidal potential and can be further used for vector control alternative to synthetic insecticide due to eco-friendly and diseases control,furthermore these plant species have potent antioxidative enzyme activities, therefore,making them strong natural candidate particularly for diseases which are caused due to free radicals.

The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). With an exposure of 5.6 ton-years, the sensitivity of Panda X-4 T could reach a minimum spin-independent dark matter-nucleon cross section of 6 × 10^(-48) cm^2 at a dark matter mass of 40 Ge V/c^2.

Traditionally,fi sh carcass is headed and gutted to prepare a skin-on fi llet in surimi *** this study,tail and/or belly of grass carp carcass were further cut off in the fi lleting ***,composition,gelling properties and aroma profile of surimi as affected by filleting methods were investigated by SDS-PAGE,ELISA,SEM,magnetic resonance imaging(MRI)and electron *** tail off increased surimi yield by 14.0%,whereas cutting belly off decreased it by 11.2%.Cutting tail or belly off signifi cantly decreased fat content,but did not change protein content,ash content,cathepsin(B,H and L)contents,nor protein *** breaking force and whiteness of surimi gel signifi cantly increased after cutting tail *** belly off slightly increased whiteness of surimi *** microstructure of all the surimi gels was compact and uniform,with fractal dimensions(Df)ranging from 2.81 to *** for the macrostructure,cutting tail off apparently improved the integrity of surimi gel while cutting belly off did *** profi les of the surimi prepared under different fi lleting methods could be clearly distinguished by linear discriminant analysis(LDA).Our results indicate that cutting tail off contribute positively to surimi production.

Effect of sodium alginate-based hydrogel loaded with lutein on gut microbiota and inflammatory response in DSS-induced colitis mice

In order to effectively deliver lutein to the inflamed colon and better exert its pharmacological activity,this paper constructed a sodium alginate hydrogel-based delivery system loaded with lutein nanoparticles,evaluated the regulation on the expression and secretion of related inflammatory factors in mice with colitis,and its impact on intestinal microbial *** results showed that comparing lutein crystal and its nanoparticle,lutein hydrogel alleviated dextran sodium sulfate(DSS)-induced colitis in mice more effectively by adjusting fecal heme content,colon tissue damage,and inflammatory factor ***,lutein hydrogel increased the expression of intestinal tight junction proteins zonula occluden-1(ZO-1),claudin-1 and occludin to maintain the integrity of the intestinal-barrier,inhibited the nuclear factor-κB(NF-κB)pathway and reduced expression and secretion of inflammatory factors including tumour necrosis factor-α(TNF-α),inducible nitric oxide synthase(iNOS),NOD-like receptors 3(NLRP3)and interleukin(IL)-1β.In addition,the intestinal microbial environment of mice with colitis was improved by down-regulating the relative abundance of Desulfovibrionaceae and up-regulating the relative abundance of Erysipelotrichaceae and *** a slow-release carrier to load lutein nanoparticles,sodium alginate-based hydrogel has potential application prospect.

Mineral constituents of a prized edible mushroom(Tricholoma matsutake)and soils beneath the fruiting bodies from the production areas across China

Fruiting bodies(500 g per site) of Tricholoma matsutake and the surface layer of soils collected from 20 spatially distant areas with pristine backgrounds across China were analyzed for potassium,magnesium,calcium,zinc,iron,copper,manganese and cadmium using inductively coupled plasma optical emission *** terms of the bioconcentration and bioexclusion concept,K,Mg,Zn,and Cu were highly bioconcentrated,and their bioconcentration factor values varied between 75-615,2-107,38-603 and 7-76,respectively,across the 20 ***,Mn and Cd were moderately bioconcentrated and their bioconcentration factors(BCFs) varied between 0.6-34.0,0.4-37.0 and 0.9-7.0 ***,Ca was excluded(BCFfood and can contain a wide spectrum of both essential and hazardous mineral compounds that accumulate at elevated concentrations even if grown in pristine *** estimated intake rate of Cd in the fruiting bodies indicates cause for concern associated with this metal resulting from the daily consumption of between 200 and 400 g of fruiting bodies on a frequent basis during the mushrooming season.

Dorsomorphin induces cancer cell apoptosis and sensitizes cancer cells to HSP90 and proteasome inhibitors by reducing nuclear heat shock factor 1 levels

Objective: Heat shock factor 1(HSF1), a transcriptional regulator of heat shock proteins(HSPs), is an attractive therapeutic target for cancer. However, only a few HSF1 inhibitors have been identified so ***: The mRNA and protein levels of HSF1, HSPs, cleaved PARP, and phosphorylated HSF1 were examined by real-time PCR and Western blot. Forced expression, RNA interference, and immunofluorescence assay were used for mechanistic *** viability and apoptosis were measured by WST-8 assay and flow cytometry, respectively. Xenograft studies were performed in nude mice to evaluate the effect of dorsomorphin and an HSP90 inhibitor on tumor ***: Dorsomorphin suppressed multiple stimuli-induced and constitutive HSPs expression in cancer cells. Mechanistic studies revealed that dorsomorphin reduced heat-induced HSP expression independent of adenosine monophosphate activated protein kinase. Dorsomorphin reduced heat-stimulated HSF1 Ser320 phosphorylation and nuclear translocation, as well as resting nuclear HSF1 levels in cancer cells. Dorsomorphin induced cancer cell apoptosis by inhibiting HSF1 expression. A structure-activity study revealed that the 4-pyridyl at the 3-site of the pyrazolo [1, 5-a]pyrimidine ring is critical for the anti-HSF1 activities of dorsomorphin. Dorsomorphin sensitized cancer cells to HSP90 and proteasome inhibitors and inhibited HSP70 expression induced by these inhibitors in vitro. In tumor-bearing nude mice, dorsomorphin enhanced HSP90 inhibitor-induced cancer cell apoptosis, tumor growth inhibition, and HSP70 ***: Dorsomorphin is an HSF1 inhibitor. It induces cancer cell apoptosis, sensitizes cancer cells to both HSP90 and proteasome inhibitors, and suppresses HSP upregulation by these drugs, which may prevent the development of drug ***, dorsomorphin and its derivates may serve as potential precursors for developing drugs against cancer.

Morchella angusticeps Peck,one of the most popular edible mushrooms,has attracted great attention due to its delicious taste and healthy ***,both its biological effects and the possible mechanism of action have not yet been *** investigated the anti-proliferative activity of the phenolic extract derived from Morchella angusticeps Peck(MPE)against HepG2 human hepatocellular carcinoma *** showed that MPE at non-cytotoxicity doses significantly inhibited the proliferation of HepG2 cells in a dose-dependent manner with inhibitory rates ranging from 18 to 90%(Pnuclear antigen(PCNA).Additionally,MPE downregulated TRAF-2 and p-p53,while upregulated p-ASK1 and ***,it could be inferred that MPE might induce the anti-proliferative function to HepG2 cells through the p38/MAPK signal transduction pathway.

Polymorphisms in NF-κB,PXR,LXR,PPARγ and risk of inflammatory bowel disease

AIM:To investigate the contribution of polymorphisms in nuclear receptors to risk of inflammatory bowel disease(IBD) . METHODS:Genotypes of nuclear factor(NF) -κB(NFKB1) NFκB-94ins/del(rs28362491) ;peroxisome proliferatoractivated receptor(PPAR) -γ(PPARγ) PPARγPro12Ala(rs1801282) and C1431T(rs 3856806) ;pregnane X receptor(PXR) (NR1I2) PXR A-24381C(rs1523127) ,C8055T(2276707) ,and A7635G(rs 6785049) ;and liver X receptor(LXR) (NR1H2) LXR T-rs1405655-C and T-rs2695121-C were assessed in a Danish case-control study of 327 Crohn's disease patients,495 ulcerative colitis(UC) patients,and 779 healthy *** ratio(OR) and 95%CI were estimated by logistic regression models. RESULTS:The PXR A7635G variant,the PPARγPro12Ala and LXR T-rs2695121-C homozygous variant genotypes were associated with risk of UC(OR:1.31,95% CI:1.03-1.66,P=0.03,OR:2.30,95%CI:1.04-5.08,P=0.04,and OR:1.41,95%CI:1.00-1.98,P=0.05,respectively) compared to the corresponding homozygous wild-type *** never smokers,PXR A7635G and the LXR T-rs1405655-C and T-rs2695121-C variant genotypes were associated with risk of IBD(OR:1.41,95%CI:1.05-1.91,P=0.02,OR:1.63,95%CI:1.21-2.20,P=0.001,and OR:2.02,95%CI:1.36-2.99,P=0.0005,respectively) compared to the respective homozygous variant *** A7635G(rs6785049) variant genotype was associated with a higher risk of UC diagnosis before the age of 40 years and with a higher risk of extensive disease(OR:1.34,95%CI:1.03-1.75 and OR:2.49,95%CI:1.24-5.03,respectively) . CONCLUSION:Common PXR and LXR polymorphisms may contribute to risk of IBD,especially among never smokers.
