Comparative effects of plant growth regulators on leaf and stem explants of Labisia pumila var. alata

Objective: Labisia pumila var. alata, commonly known as 'Kacip Fatimah' or 'Selusuh Fatimah' in Southeast Asia, is traditionally used by members of the Malay community because of its post-partum medicinal properties. Its various pharmaceutical applications cause an excessive harvesting and lead to serious shortage in natural habitat. Thus, this in vitro propagation study investigated the effects of different plant growth regulators (PGRs) on in vitro leaf and stem explants of L. pumila. Methods: The capabilities of callus, shoot, and root formation were evaluated by culturing both explants on Murashige and Skoog (MS) medium supplemented with various PGRs at the concentrations of 0, 1, 3, 5, and 7 mg/L. Results: Medium supplemented with 3 mg/L indole-3-butyric acid (IBA) showed the optimal callogenesis from both leaf and stem explants with (72.34±19.55)% and (70.40±14.14)% efficacy, respectively. IBA was also found to be the most efficient PGR for root induction. A total of (50.00±7.07)% and (77.78±16.47)% of root formation were obtained from the in vitro stem and leaf explants after being cultured for (26.5±5.0) and (30.0±8.5) d in the medium supplemented with 1 and 3 mg/L of IBA, respectively. Shoot formation was only observed in stem explant, with the maximum percentage of formation ((100.00±0.00)%) that was obtained in 1 mg/L zeatin after (11.0±2.8) d of culture. Conclusions: Callus, roots, and shoots can be induced from in vitro leaf and stem explants of L. pumila through the manipulation of types and concentrations of PGRs.

Brown spot formation with necrosis and sloughing off pellicle and sunken surface lesions under visibly healthy shells is one of the quality problems of Iranian *** symptomatic kernels were investigated for isolation of the causative *** quality attributes of infected and non-infected hazelnuts were *** results showed that the total phenol content and antioxidant activity of the infected hazelnut samples were higher than non-infected *** infection enhanced the soluble protein content and affected the soluble protein profiles,either in the pattern or in the intensity of the protein *** color parameters decreased,which led to darker infected hazelnut *** infection also caused the alteration of the fatty acids composition of hazelnuts,which was a remarkable increment in the relative amounts of saturated fatty acids and a decrement in the unsaturated fatty *** optical microscopic and Scanning Electron Microscope(SEM)micrograph observation of the defective hazelnut kernel indicated the kernel parenchyma tissue had been damaged,and the microbial culture from many necrotic points revealed that this contamination due to the presence of yeast-like fungus,white to slightly creamy,round colonies with raised centers and peripheral mycelial *** pathogenicity of a yeast-like fungus(isolate of NAS 201)was proved by fulfilling Koch’s postulates,and the morphological and molecular identification(internal transcribed spacer region as DNA barcoding)indicated the presence of the fungal agent Eremothecium *** our knowledge,this is the first report of the occurrence of yeast spots in hazelnut in Iran.

The impact of epigenetic modifications like DNA methylation on plant phenotypes has expanded the possibilities for crop *** methylation plays a part in the regulation of both the chromatin structure and gene expression,and the enzyme involved,DNA methyltransferase,executes the methylation process within the plant *** regulating crucial biological pathways,epigenetic changes actively contribute to the creation of the ***,epigenome editing may assist in overcoming some of the drawbacks of genome editing,which can have minor off-target consequences and merely facilitate the loss of a gene’s *** drawbacks include gene knockout,which can have such off-target *** review provides examples of several molecular characteristics of DNA methylation,as well as some plant physiological processes that are impacted by these epigenetic changes in the *** also discuss how DNA alterations might be used to improve crops and meet the demands of sustainable and environmentally-friendly farming.

Temperature-Induced Stress on Growth and Expression of Hsp in Freshwater alga Scenedesmus quadricauda

Objective To investigate the impact of various levels of sublethal temperature （26℃, 31 ℃, 33℃, 36℃, and 39℃） on growth and heat shock protein （hsp） expression in freshwater green alga Scenedesmus quadricauda. Methods Impact of selected levels of temperature on growth rate （based on optical density）, population count, chlorophyll-a and biomass of the alga was evaluated in artificial growth medium for 19 days. To determine the induction of hsp in the alga, it was exposed to selected temperature levels for 3 h and further kept for 6 h at culturing condition at 26℃. Induction of hsp was confirmed by immuno-detection followed by SDS-polyacrylamide gel electrophoresis. Results The selected growth parameters such as growth rate, population count, chlorophyll-a and biomass were reduced significantly （P〈0.001） at 39℃. However, hsp 70 expression was observed only at 39℃. Conclusion Temperature up to 36℃ may be considered as the limit of safe exposure for thermal, stress for the alga Scenedesmus quadricauda.

Enhancement of filtration efficiency by electrical charges on nebulized particles

There have been few investigations of effects of electrical charge, carried by lab-generated particles, on filtration efficiency testing. Here, we measured the elementary charge on particles and the fraction of particles carrying that charge with a combined electrometer, differential mobility analyzer, and scanning mobility particle sizer. A typical solid NaCI aerosol and liquid diethylhexyl sebacate （DEHS） aerosol were generated with Collison and Laskin nebulizers, respectively. Our experimental results showed that NaCI aerosols carried more charge after aerosol generation. The average net elementary charge per particle was approximately 0.07. The NaCI aerosol was overall positively charged but contained a mixture of neutral and charged particles. Individual particles could carry at most four elementary charges. According to constant theorem, we speculated that original NaC1 aerosol contained 17% neutral, 45% positive-, and 38% negative-charged particles in the diameter range from 30 to 300nm. A Kr-85 neutralizer was used to decrease the charge on the NaCI particles. Our results indicated that the DEHS aerosol was electrically neutral. The effects of electric charge on particle collection by electret and electroneutral high efficiency particulate air （HEPA） filters were analyzed. Theoretical calculations suggested that charges on original NaCI aerosol particles enhanced the filtration efficiency of HEPA filters,

Re-estimation of absolute gamma ray intensities of ^(56)Mn using k_0-standardization

The thermal neutron capture gamma ray facility at Pakistan Research Reactor (PARR-1) is being used forthe re-estimation of various properties like capture cross-sections, resonance integral, absolute gamma intensities, *** different isotopes. The data for gamma ray transitions from the capture of thermal neutrons by 55Mn are not ingood agreement specifically below 2 MeV. So there is a need to re-estimate its intensities with better accuracy. Analyticalgrade MnCl2 powder and high purity Mn metal pieces were used in this study. Standard 152Eu and 60Co radioactivesources as well as thermal neutron capture γ-rays in chlorine were chosen for efficiency calibration. The k0standardization technique was applied for these measurements to eliminate systematic errors in efficiencies. Chlorinealso acted as a comparator in k0- factor calculations. The results have been tabulated for the main gamma rays from56Mn in the low as well as in the medium energy regions. The absolute intensities are in good agreement with most ofthe reported values.

Deposition of Diamond-Like Carbon Films with Femtosecond KrF Laser Pulses

Hydrogen-free diamond-like carbon (DLC) films with the optical band gaps of 2.0-2.7 e V and the microhardness of 40-55 GPa are deposited by femtosecond KrF laser pulses. The film microstructure of nanometer spheroid is observed. The filrn microhardness is improved with the increase of ion kinetic energy of plasma plume from 300 to 600 eV. The characteristics of DLC film deposition with fs pulses is discussed.

Phase analysis of AlFe2B2 by synchrotron X-ray diffraction,magnetic and M?ssbauer studies

The structural and magnetic properties of essentially phase pure AlFe2B2prepared by arc melting were compared with a sample containing impurities. Analysis was carried out by means of synchrotron X-ray diffraction, magnetic measurements and M?ssbauer spectroscopy. The analysis of synchrotron X-ray diffraction data confirmed the orthorhombic structure of space group Cmmm for AlFe2B2with Al13Fe4as main impurity phase. The magnetic measurements revealed an unsaturated ferromagnetic state in AlFe2B2with a transition temperature（Tc） of 286 K. Isothermal magnetization measurment at 5 K gave a saturation magnetization of 0.7 μB per Fe atom while Arrott plots establish the second order nature of ferromagnetic transition. The thermal evolution of M?ssbauer spectra confirmed the ferromagnetic nature of this material revealing an hyperfine field of 73 kOe and an isomer shift of 0.46 mm/s at 100 K. The M?ssbauer spectra of a phase pure sample was compared with the one containing impurity phases. It is suggested that the low temperature paramagnetic contribution in M?ssbauer spectra of phase pure material may have its origin in some intrinsic phenomena arising from defects or inhomogeneities in crystal structure.
