A New ALS Based PMS Design and Its Evaluations

Westinghouse company (WEC) had developed a nuclear regulatory Commission (NRC) approved advanced logic system (ALS) platform based on field programmable gate array (FPGA) technology as the next generation 1E class platform for protection and monitoring system (PMS) development for nuclear power plants. In compliance with the requirements of typical PMS functions, a new ALS based PMS is designed by overcoming the restrictions of communication modules etc. The consistency with data communication independence and isolation, deterministic, diversity requirements etc. is analyzed. The evaluations indicate the design meets the requirements and can be applied for coming projects.

Metabolic-associated fatty liver disease(MAFLD),which is previously known as non-alcoholic fatty liver disease(NAFLD),represents a major health concern worldwide with limited ***,we provide evidence that ferroptosis,a novel form of regulated cell death characterized by iron-driven lipid peroxidation,was comprehensively activated in liver tissues from MAFLD *** canonical-GPX4(cGPX4),which is the most important negative controller of ferroptosis,is downregulated at protein but not mRNA ***,a non-canonical GPX4 transcript-variant is induced(inducible-GPX4,iGPX4)in MAFLD *** high fat-fructose/sucrose diet(HFFD)and methionine/choline-deficient diet(MCD)-induced MAFLD pathologies,including hepatocellular ballooning,steatohepatitis andﬁbrosis,were attenuated and aggravated,respectively,in cGPX4-and iGPX4-knockin ***4 and iGPX4 isoforms also displayed opposing effects on oxidative stress and ferroptosis in *** of iGPX4 by siRNA alleviated lipid stress,ferroptosis and cell ***,the triggered iGPX4 interacts with cGPX4 to facilitate the transformation of cGPX4 from enzymatic-active monomer to enzymatic-inactive oligomers upon lipid stress,and thus promotes ***-immunoprecipitation and nano LC–MS/MS analyses confirmed the interaction between iGPX4 and *** results reveal a detrimental role of non-canonical GPX4 isoform in ferroptosis,and indicate selectively targeting iGPX4 may be a promising therapeutic strategy for MAFLD.

Bisphenol A （BPA） has been shown to exert biological effects through estrogen receptor （ER）-dependent and ER-independent mechanisms. Recent studies suggest that prenatal exposure to BPA may increase the risk of childhood asthma. To investigate the underlying mechanisms in the actions of BPA, human fetal lung fibroblasts {hFLFs） were exposed to varying doses of BPA in culture for 24 hr. Effects of BPA on localization and uptake of BPA, cell viability, release of immune and developmental modulators, cellular localization and expression of ERa, ERβ and G-protein coupled estrogen receptor 30 （GPR30）, and effects of ERs antagonists on BPA-induced changes in endothelin-1 （ET-1） release were examined. BPA at 0.01-100 μmol/L caused no changes in cell viability after 24 hr of exposure, hFLFs expresses all three ERs. BPA had no effects on either cellular distribution or protein expression of ERa, however, at 100 μmol/L （or 23 μmol/L intracellular BPA） increased ERβ protein levels in the cytoplasmic fractions and GPR30 protein levels in the nuclear fractions. These paralleled with increased release of growth differentiation factor-15, decreased phosphorylation of nuclear factor kappa B p65 at serine 536, and decreased release of ET-1, interleukin-6, and interferon gamma-induced protein 10. ERs antagonists had no effects on BPA-induced decrease in ET-1 release. These data suggest that BPA at 100 μmol/L altered the release of immune and developmental modulators in hFLFs, which may negatively influence fetal lung development, maturation, and susceptibility to environmental stressors, although the role of BPA in childhood asthma remains to be confirmed in in viuo studies.

Rapid and robust induction of type I interferon(IFN-I) is a key event for the host against invaded viruses. Non-coding RNAs(ncRNAs) including microRNAs, long non-coding RNAs, and circular RNAs play important roles in the fine regulation of IFN-I production and IFNAR downstream signaling pathways. However, it is not clear whether small nuclear RNAs(snRNAs) involve in the regulation of IFN-I-dependent antiviral immunity. snRNA, a type of ncRNA between 100～300bp, is stably, widely, and highly expressed in cells, and very conserved among numerous species. SnRNAs mainly control the splicing of mRNA precursors, which is critical for the basic activities of life. However, the function of snRNAs in viral infectious diseases has not been reported. The E3 ubiquitin ligase TRIM25 mediates the K63 ubiquitination of the viral RNA sensor RIG-I, thereby facilitating the activation of the RIG-I downstream signaling pathway and the production of IFN-I. Our Results have shown that U1 snRNA RNVU1-18 is upregulated in the influenza virus-infected cells. RNVU1-18 interacts with TRIM25 to promotes the activation of RIG-I and its downstream antiviral signaling pathways. Our study has explored the role of RNVU1-18 in host antiviral immunity and clarified the molecular mechanisms, which will provide novel targets and strategies for the prevention and treatment of the infectious diseases caused by viruses including influenza viruses and SARS-CoV-2.

High resolution (4 mm) tof PET-CT (positron emission tomography-computed tomography) from Philips of model Ingenuity TF is newly installed at Institute of nuclear Medical Physics (INMP). 128 slice CT component incorporated with PET provides comparatively lower dose than the 511 keV annihilation photons associated with positron decay from PET scan. So, for designing shielding in our PET-CT facility, only 511 keV annihilation photons energy has been considered. The main objective of this paper is to show what measures have been taken to protect patients, occupational workers as well as environment from PET-CT radiation hazard through a cost effective design that satisfy the national regulatory demand. In this paper, AAPM (American Associations of Physicists in Medicine) Task group 108 analysis for PET and PET-CT shielding requirements is followed for our PET-CT facility shielding design. From theoretical calculation as shielding requirement, 1.1 cm Pb thickness or, 13 cm concrete thicknesses are found. Practically, all walls and ceiling are of 30.48 cm (1 foot) thick made of concrete with density 2.35 gcm-3 for more safety. As x-ray from CT is not taken into account for shielding analysis, Bangladesh Atomic Energy Commission (BAEC) conducted an extensive radiation survey at controlled, supervised and public area for CT. The report that is found meets the national regulatory requirements.

There have been many recent studies devoted to the consequences of stochasticity in protein circuitry. Stress conditions, including DNA damage, hypoxia, heat shock, nutrient deprivation, and oncogene activation, can result in the activation and accumulation of p53. Several experimental studies show that oscillations can be induced by DNA damage following nuclear irradiation. To explore the underlying dynamical features and the role of stochasticity, we discuss the oscillatory dynamics in the well-studied regulatory network motif. The fluctuations around the fixed point of a delayed system are Gaussian in the limit of sufficiently weak delayed feedback, and remain Gaussian along a limit cycle when viewed tangential to the trajectory. The experimental results are recapitulated in this study. We illustrate several features of the p53 activities, which are robust when the parameters change. Furthermore, the distribution in protein abundance can be characterized by its non-Gaussian nature.

Based on guides RG 1.109, RG 1.111 published by United States nuclear regulatory Commission (USNRC), our research concentrates in assessing radiation doses caused by radioactive substances released from the nuclear power plant (NPP) Ninh Thuan 1 under the scenario of normal operation using software package NRCDose72 provided by the USNRC. The database including the released radioactive nuclides, meteorology, terrain, population and agricultural production activities have been collected and processed to build the input data for the model calculation. The wind rose distribution obtained from the meteorological data in a five-year period from 2009-2013 showed that the radioactive nuclides released to environment spread in two main wind directions which are North East and South West. The X/Q (s/m3) and D/Q (s/m2) qualities which are, respectively, the ratio of activity concentration to release rate and that of deposition density of radioactive nuclides to release rate were calculated within an area of 80 km radius from the NPP site using XOQDOQ. Population doses were calculated using GASPAR. The XOQDOQ and GASPAR are two specific softwares in NRCDose72 package.
