Oligodendroglial heterogeneity in health,disease,and recovery:deeper insights into myelin dynamics

Decades of research asserted that the oligodendroglial lineage comprises two cell types:oligodendrocyte precursor cells and ***,recent studies employing single-cell RNA sequencing techniques have uncovered novel cell states,prompting a revision of the existing *** forward,the oligodendroglial lineage should be delineated into five distinct cell states:oligodendrocyte precursor cells,committed oligodendrocyte precursor cells,newly formed oligodendrocytes,myelin-forming oligodendrocytes,and mature *** new classification system enables a deeper understanding of the oligodendroglia in both physiological and pathological *** this uniform terminology will facilitate comparison and integration of data across ***,including the consolidation of findings from various demyelinating models,is essential to better understand the pathogenesis of demyelinating ***,comparing injury models across species with varying regenerative capacities can provide insights that may lead to new therapeutic strategies to overcome remyelination ***,by standardizing terminology and synthesizing data from diverse studies across different animal models,we can enhance our understanding of myelin pathology in central nervous system disorders such as multiple sclerosis,Alzheimer's disease,and amyotrophic lateral sclerosis,all of which involve oligodendroglial and myelin dysfunction.

Molybdenum and cadmium co-induce necroptosis through Th1/Th2 imbalance-mediated endoplasmic reticulum stress in duck ovaries

Innovative approaches to genome editing in avian species

The tools available for genome engineering have significantly improved over the last 5 years, allowing scientist to make precise edits to the genome. Along with the development of these new genome editing tools has come advancements in technologies used to deliver them. In mammals genome engineering tools are typically delivered into in vitro fertilized single cell embryos which are subsequently cultured and then implanted into a recipient *** avian species this is not possible, so other methods have been developed for genome engineering in birds. The most common involves in vitro culturing of primordial germ cells(PGCs), which are cells that migrate through the embryonic circulatory system to the developing gonad and colonize the gonad, eventually differentiating into the gonadocytes which produce either sperm or ova. While in culture the PGCs can be modified to carry novel transgenes or gene edits, the population can be screened and enriched, and then transferred into a recipient embryo. The largest drawback of PGC culture is that culture methods do not transfer well across avian species, thus there are reliable culture methods for only a few species including the chicken. Two newer technologies that appear to be more easily adapted in a wider range of avian species are direct injection and sperm transfection assisted gene editing(STAGE).The direct injection method involves injecting genome engineering tools into the circulatory system of the developing embryo just prior to the developmental time point when the PGCs are migrating to the gonads. The genome engineering tools are complexed with transfection reagents, allowing for in vivo transfection of the PGCs. STAGE utilizes sperm transfection to deliver genome engineering tools directly to the newly fertilized embryo. Preliminary evidence indicates that both methodologies have the potential to be adapted for use in birds species other than the chicken, however further work is needed in this area.

Cortisol levels in skimmed milk during the first 22 weeks of lactation and response to short-term metabolic stress and lameness in dairy cows

Background： Cortisol is secreted into blood in reaction to acute stress, but also in phases of diminished feed intake and changed animal behavior. As cows do not always show clear signs of discomfort, reliable diagnostic markers could be used to provide information regarding individual cows＇ distress. The objective of this study was to establish an ether free immunoassay for the detection of cortisol and to determine values during the first 22 weeks of lactation. Furthermore, the response in milk cortisol levels was assessed during times of metabolic stress and pain associated symptoms of lameness. Methods： Milk yield and composition, blood serum glucose, NEFA and BHBA as well as milk cortisol were determined in 24 multiparous Holstein-Friesian cows over the course of the first 22 weeks of lactation. animals were further checked for signs of clinical diseases on a daily basis. Two feed restrictions over three days （FR; 70 % of precious ad libitum intake） were performed during the 4th wk and the 21st wk, respectively. An ELISA for cortisol measurement in easily accessible bovine skimmed milk was established and applied. Results： On the last day of FR in early lactation, a reduction in milk yield and changes in serum metabolites compared to respective previous values were detected. The FR in mid-lactation resulted in no changes in milk production and serum metabolites. Milk cortisol was highest during first wk of lactation and remained on comparable levels thereafter. Milk yield and composition were not influenced by FR. Lameness resulted in enhanced milk cortisol levels. Conclusion： Milk cortisol could be used as an indicator of painful symptoms such as lameness. Higher values of milk cortisol levels during first wk of lactation should be taken into account for interpretation.

Real-world treatment patterns of gastrointestinal neuroendocrine tumors: A claims database analysis

AIM To describe real-world treatment patterns of gastrointestinal neuroendocrine tumors(GI NET).METHODS In this retrospective cohort study,we used 2009-2014 data from 2 United States commercial claims databases to examine newly pharmacologically treated patients using tabular and graphical techniques. Treatments included somatostatin analogues(SSA),cytotoxic chemotherapy(CC),targeted therapy(TT),interferon(IF) and combinations. We identified patients at least 18 years of age,with ≥ 1 inpatient or ≥ 2 outpatient claims for GI NET who initiated pharmacologic treatment from 7/1/09-6/30/14. A 6 mo clean period prior to first treatment ensured patients were newly treated. Patients were followed until end of enrollment or the study end date,whichever was *** We identified 2258 newly treated GI NET patients: mean(SD) age was 55.6 years(SD = 9.7),47.2% of the patients were between 55 and 64 years,and 48.8% were female. All regions of the United States were represented. 59.6% started first-line therapy with SSA monotherapy(964 with octreotide LAR,380 with octreotide SA,and 1 with lanreotide),33.3% CC,3.6% TT,and 0.5% IF. The remainder received combinations. Mean follow up was 576 d. Overall mean first-line therapy duration was 361 d(449 d for SSA,215 for CC,267 for TT). 58.9% of patients had no pharmacological treatment beyond first line. The most common secondline was combination therapy with SSA. In graphical pattern analysis,there was no clear pattern visible after first line *** In this study,60% of patients initiated treatment with SSA alone or in combination. The relatively long time to discontinuation suggests possible sustained effectiveness and tolerability.

Genome-wide differences in DNA methylation changes in caprine ovaries between oestrous and dioestrous phases

Background: DNA methylation plays a vital role in *** genome DNA methylation changes during the oestrous phase(ES) and dioestrous phase(DS) in the ovaries of Guanzhong dairy goats were investigated using bisulphite sequencing to understand the molecular biological mechanisms of these goats' oestrous ***: We discovered distinct genome-wide DNA methylation patterns in ES and DS ovaries.A total of 26,910 differentially methylated regions were upregulated and 21,453 differentially methylated regions were downregulated in the ES samples compared with the DS samples(P-values ≤0.05 and fold change of methylation ratios ≥2).Differentially methylated region analysis showed hypomethylation in the gene body regions and hypermethylation in the joining region between upstream regions and gene *** methylation ratios of the STAR,FGF2,FGF12,BMP5 and SMAD6 genes in the ES samples were lower than those of the DS samples(P-values≤0.05 and fold change of methylation ratios ≥2).Conversely,the methylation ratios of the EGFR,TGFBR2,IGF2 BP1 and MMD2 genes increased in the ES samples compared with the DS *** addition,223 differentially methylated genes were found in the GnRH signalling pathway(KO04912),ovarian steroidogenesis pathway(KO04913),oestrogen signalling pathway(KO04915),oxytocin signalling pathway(KO04921),insulin secretion pathway(KO04911) and MAPK signalling pathway(KO04010).Conclusions: This study is the first large-scale comparison of the high-resolution DNA methylation landscapes of oestrous and dioestrous ovaries from dairy *** studies and our investigations have shown that the NR5 A2,STAR,FGF2 and BMP5 genes might have potential application value in regulating caprine oestrus.

Fermentation characteristics of Megasphaera elsdenii J6 derived from pig feces on different lactate isomers

D-Lactate-utilizing bacteria play important roles in maintaining the balance of gut lactate; however, studies on gut D-lactate-uti-lizing bacteria have been limited. This study aimed to isolate and identify D-lactate-utilizing bacteria from pig gut using the Hungate rol-tube method, and to investigate their metabolic characteristicsin vitro. Six different anaerobes were isolated from pig feces, which were identiifed as related toBacteroides fragilis, Bacteroides acidifaciens,Veilonela denticariosi, Veilonela caviae,Bacteroides uniformis, andMegasphaera elsdenibased on the 16S rRNA gene sequences. Al strains had a signiifcant ability to utilize D-lactate, which was concluded afterin vitro fermentation with 25 mmol L–1 D-lactate as the primary carbon source. Of al 6 strains,M. elsdeniJ6 showed the highest efifciency of D-lactate utilization and produced a higher ratio of butyrate in total short chain fatty acids （SCFAs）. Thus, thein vitro fermentation characteristics of this strain in D-, L-, and DL-lactate mixtures （D-lactate：L-lactate=1：1 or 1：2） were further studied. The results showed that M. elsdeni J6 preferred utilizing D-lactate, and produced more SCFA when using D-lactate as the primary carbon source. The ifndings suggest that the administration of D-lactate-utilizing bacteria such asM. elsdeni J6 may have a potential advantage in the aleviation of D-lactic acidosis in the animal gut.

Feeding of processed vegetable wastes to bulls and its potential environmental benefit

The study was conducted with the objectives to quantify year round availability of different vegetables waste(VW) in a wholesale market and to determine the inclusion level of a processed VW(VWP) in the diets of bulls. The daily VW biomass availability at Kawran bazaar, Dhaka, bangladesh was quantified by weighing the vegetable supply and their wastes by visiting 2 days in a week. Concurrently, VW of cucumber, bitter gourd, spotted gourd, brinjal, pumpkin, potato, tomato, ladies finger, and snake gourd representing 0.21, 0.18, 0.17, 0.16, 0.09, 0.07, 0.06, 0.03, and 0.02 as fresh fractions, respectively were blended, dried and stored while adding rice polish and common salt at 200 and 20 g/kg DM, respectively;it was tested in bulls as an ingredient of concentrate mixture. Four dietary groups, each of 6 bulls, with initial average live weight(LW) of 85.47 ± 17 kg, were fed fresh German grass(Echinochloa polystachya)ad libitum supplemented with 4 different concentrates containing 0, 10%, 20% and 30% VWP at the rate of1% of LW for 89 days. The availability of VW biomass of the market was 42.51 t/d and recycling of them as feed, instead of using landfills, might reduce annual methane emission by 0.43 Gg. The inclusion of VWP in the diet up to 9.7% of DM, or 0.30% of LW of bulls showed no significant effect on the DM intake,digestibility, growth performance and health status of bulls. The dietary DM intake represented 3.10%,3.09%, 3.20% and 3.14% of LW resulting in daily gain of 302, 300, 312 and 344 g, respectively. The digestibility of DM of diets was 56.9%, 62.8%, 62.8% and 63.4%, respectively. It was concluded that VWP may be included at a level of 9.7% of the diet(DM basis) or 0.30% of LW of bulls.
