Herein lie the crosstalk and regulation between AQP1 and Emmprin in SMMC7221 cells by means of siRNA technology and deglycosylation method. Firstly, HAQP1, rather than hAQP3, was selectively upregulated in SMMC7221 cells by FBS, flollowed by the upregulated expression of Emmprin. Emmprin gene silencing caused a remarkable change in the expression ofAQP1 gene, just like its downstream gene, MMP9, meanwhile the water permeability and cell migration were also descended prominently. Furthermore, when treated with tunicamycin, Emmprin was deglycosylated, which made the expression of AQP 1 significantly declined, followed by remarkably decreased cell membrane water permeability and cell migration. Taken together, all the data indicates the expression level and the modification of Emmprin by glycosylation are the key factors in regulating the expression of AQP1.

Objective: To investigate the correlation between E670 G polymorphism of proprotein convertase subtilisin/kexin type 9(PCSK9) gene and coronary heart disease(CHD), and contrastively study the regional differences of E670 G polymorphism of PCSK9 gene between patients with CHD among the Han population in Hainan and three provinces in the northeast of China(TPNC), providing scientific basis for prevention and treatment of patients with CHD in different regions. Methods: A total of 233 cases of patients with CHD were selected from the Han population in Hainan and TPNC as the experimental group(118 cases from Hainan, 115 cases from TPNC), and 239 cases with non-CHD were selected among the Han population also in the two regions as control group(125 cases from Hainan, 114 cases from TPNC). The triglyceride(TG), total cholesterol(TC), high density lipoprotein cholesterol and low density lipoprotein cholesterol(LDL-C) levels of plasma were tested and PCR-RFLP method was used to test the E670 G polymorphism of PCSK9 gene. The statistical software package SPSS 21.0 was used for the statistical analysis and P<0.05 was considered as statistically significant. Results: The levels of systolic pressure, diastolic blood pressure, fasting blood sugar, TC, TG, and LDL-C of patients in CHD group were significantly higher than those in non-CHD group, while the high density lipoprotein cholesterol level was lower than that in non-CHD group(P<0.05). In CHD group, the frequencies of AG, GG genotypes of PCSK9 gene and G allele were higher than those in non-CHD group(P<0.05), and in CHD group, the frequencies of AG, GG genotypes and G allele of patients both in Hainan and TPNC were higher than those in control group(P<0.05). Among the patients with CHD, the frequencies of GG genotype and G allele of patients in Hainan were lower than those in TPNC(P<0.05), and in CHD group, the levels of TG, TC and LDL-C of GG genotype were higher than those of AA genotype(P<0.05). Summary： Human interleukin-15 （hIL-15） is an important cytokine to activate endothelial cells and can be regulated by many other cytokines. The aim of this study is to examine the ability of interferon-γ （IFN-γ）, and tumor necrosis factor-or （TNF-α to induce the production of human interleukin-15 （hIL-15） and IL-15 receptor （IL-15Rα by human umbilical vein endothelial cells （HUVECs）. The data are summarized as follows： 1. Northern blot revealed that IL-15 mRNA was up-regulated by IFN-γ and TNF-α 2. Intracellular IL-15 protein was visualized by fluorescence microscopy, whereas the expression of IL-15 on the surface of HUVECs was detected by fluorescence activated cell sorting （FACS）, and no detectable IL-15 in the medium was verified by ELISA. 3. IL-15Rα was detected on the surface of HUVECs by FACS after IFN-α and TNF-α stimulation, whereas Western blotting revealed that the elevated expression on surface IL-15Rα was not due to the increased protein expression. The conclusion demonstrated from our results is that IFN-α and TNF-α play an important role in regulating the expression of IL-15 and IL-15Rα on the surface of HUVECs.

Background：Since 2010,two versions of National Guidelines aimed at promoting the management of ST-segment elevation myocardial infarction （STEMI） have been formulated by the Chinese Society of ***,little is known about the changes in clinical characteristics,management,and in-hospital outcomes in rural ***：In the present multicenter,cross-sectional study,participants were enrolled from rural hospitals located in Liaoning province in northeast China,during two different periods （from June 2009 to June 2010 and from January 2015 to December 2015）.Data collection was conducted using a standardized *** total,607 and 637 STEMI patients were recruited in the 2010 and 2015 cohorts,***：STEMI patients in rural hospitals were older in the second group （63 years vs.65 years,P =0.039）.We found increases in the prevalence of hypertension,prior percutaneous coronary intervention （PCI）,and prior *** the past 5 years,the cost during hospitalization almost *** proportion of STEMI patients who underwent emergency reperfusion had significantly increased from 42.34% to 54.47% （P 〈 0.0001）.Concurrently,the proportion of primary PCI increased from 3.62% to 10.52% （P 〈 0.0001）.The past 5 years have also seen marked increases in the use of guideline-recommended drugs and clinical ***,in-hospital mortality and major adverse cardiac events did not significantly change over time （13.01% vs.10.20%,P =0.121;13.34% vs.13.66%,P =0.872）.Conclusions：Despite the great progress that has been made in guideline-recommended therapies,in-hospital outcomes among rural STEMI patients have not significantly ***,there is still substantial room for improvement in the quality of care.

Background : Hypertension and dyslipidemia are considered reversible risk factors for cardiovascular disease. The purpose of this study was to explore the impact of traditional and nontraditional blood lipid profiles on the risk of left ventricular hypertrophy(LVH) and to explore the superposition effect of dyslipidemia combined with *** : Data on 9134 participants(53.5 ±10.3 years old) from the northeast China Rural Cardiovascular Health Study(NCRCHS) were statistically analyzed. The blood lipid profile was measured by total cholesterol(TC), low-density lipoprotein cholesterol(LDL-C), high-density lipoprotein cholesterol(HDL-C), total glyceride(TG), and calculated nontraditional blood lipid indices including non-HDL-C, atherosclerosis index(AI), TC/HDL-C, and residual cholesterol(RC).Results : After the adjustment of age and gender, the odds ratios(ORs) of LVH in patients with hypertension, high LDL-C, high non-HDL-C, high AI, and high TC/HDL-C were 3.97(3.31– 4.76), 1.27(1.02– 1.59), 1.21(1.04– 1.39), 1.33(1.15– 1.53), and 1.42(1.22– 1.65), respectively. After full adjustment of potential confounding factors, high AI and TC/HDL-C were associated with LVH rather than traditional blood lipid *** combination of hypertension and nontraditional dyslipidemia(defined by high AI and TC/HDL-C) was associated with the highest risk of LVH, especially in participants under 45 years of age. The risk was more significant in men, 5.09-fold and 6.24-fold,respectively, compared with 3.66-fold and 4.01-fold in *** : People with dyslipidemia defined by nontraditional blood lipid indices(high AI and high TC/HDL-C) and hypertension were more likely to develop LVH.

The launch of the World Journal of Nephrology （WJN） has distinct purposes. Its main purpose is to incorporate the fields of diabetes, hypertension, urology, cardiol-ogy and neurology, which are related to kidney disease, and to make all of our readers, contributors and allied health professionals feel at home with the WJN. The WJN aims to rapidly report new theories, methods and techniques for prevention, diagnosis, treatment, reha-bilitation and nursing skill in the feld of nephrology. The WJN will cover diagnostic imaging, disorders of kidney development, renal obstruction, atrophy and regenera-tion, kidney tumors, renal pharmacology and therapy, renal replacement therapies including transplantation, and Chinese herbal medicine. The WJN issues will in-clude an editorial, frontier, invited review of articles and commentaries in addition to original articles submitted. The WJN will solicit articles from investigators in areas of diabetes and hypertension, and high priority will be given to those articles with an emphasis on the pre-vention of dialysis. Final decision for publication will be based on the merit of the article, language and lucidity.

Blood conservation pediatric cardiac surgery in all ages and complexity levels

AIM To demonstrate the feasibility of blood conservation methods and practice across all ages and risk categories in congenital cardiac *** We retrospectively analyzed a collected database of 356 patients who underwent cardiac surgery using cardiopulmonary bypass(CPB) from 2010-2015. The patients were grouped into blood conservation(n = 138) and nonconservation(n = 218) groups and sub-grouped based on their ages and procedural complexity scores. RESULTS There were no statistical differences in gender,weight,pre-operative and pre-CPB hematocrit levels in both groups. Despite equivalent hematocrit levels during and after CPB for both groups,there was significantly less operative homologous blood utilized in blood conservation group across all ages and complexity levels. CONCLUSION Blood conservation surgery can be performed in con-genital patients needing cardiac surgery in all age groups and complexity categories. The above findings in addition to attendant risks and side effects of blood transfusion and the rising cost of safer blood products justify blood conservation in congenital cardiac surgery.
