To evaluate the effect of anti-HER-2 × anti-CD3 bi-specific antibody（BsAb） on the growth of HER-2/neu-expressing human gastric carcino.a in vitro and in vivo, an MTT assay was carried out to test the inhibitive rates of herceptin, anti-CD3 and BsAb antibodies on SGC-7901 gastric carcino.a cells. Immuno.ytochemistry methods were used to test the HER-2 level of SGC-7901. Nude mice models were employed to test the effect of HER-2 CD3 BsAh combined with effector ceils（ peripheral blood lymphatic cells of healthy human beings） on the growth of tumors in animals. Compared with that of the untreated control group, the tumor cell growth rates in vitro and in vivo will both be significantly inhibited when treated with effector cells combined with anti-CD3 McAb, herceptin or HER2 CD3 BsAb （p 〈0. 05）, and the growth inhibition is the most remarkable in the group treated with HER2 CD3 BsAb combined with effector cells. The growth of tumor xeno.rafts will also be significantly inhibited in the group treated with HER2 CD3 BsAb combined with effector cells when compared with that in the group treated with anti-CD3 McAb or the group treated with herceptin combined with effector cells（p 〈0. 05）. We can conclude that HER-2/neu is possibly a useful target for immuno.herapy of gastric carcino.a, and anti-HER2 × anti-CD3 BsAb has evident anti-tumor efficacy both, in vitro and in vivo.

Two new different Cu(Ⅱ) MOFs with the same 1,3,5-tris(1-imidazolyl) benzene(tib) ligand {[Cu(tib)2]·(H2O)2·Br2}n(1) and {[Cu2(tib)·Br·Cl]·2Br}n(2) were obtained by one pot synthesized of tib with CuBr in the presences of HCl and water. X-ray single crystal diffraction analyses indicate that both complexes 1 and 2 have two dimensional frameworks containing different building blocks. Each Cu(Ⅱ) atom in complex 1 is coordinated by four N atoms from different tib ligands. However, there are two different cryptographic Cu(Ⅱ) atoms in complex 2, one is four coordinated by two bromine atoms and two N atoms from different tib ligands, the other is six coordinated by two chloride atoms and four N atoms from different tib ligands. The thermal gravimetric analysis of complexes 1 and 2 are depicted in the paper.

A new fast learning algorithm was presented to solve the large-scale support vector machine ( SVM ) training problem of aero-engine fault *** relative boundary vectors ( RBVs ) instead of all the original training samples were used for the training of the binary SVM fault *** pruning strategy decreased the number of final training sample significantly and can keep classification accuracy almost *** , the training time was shortened to 1 / 20compared with basic SVM *** , owing to the reduction of support vector number , the classification time was also *** sample aliasing existed , the aliasing sample points which were no. of the same class were eliminated before the relative boundary vectors were *** , the samples near the relative boundary vectors were selected for SVM training in order to prevent the loss of some key sample points resulted from *** can improve classification accuracy effectively.A simulation example to classify 5classes of combination fault of aero-engine gas path components was finished and the total fault classification accuracy reached 96.1%.Simulation results show that this fast learning algorithm is effective , reliable and easy to be implemented for engineering application.

Viruses are very widespread in nature, and they cause severe diseases and yield losses in potato production. The transfer of the dsRNA-producing gene could confer a high level virus resistance by specific targeting of cognate viral RNA. In this study, we constructed a marker-free expression vector of a chimeric gene derived from the coat protein sequence of Potato virus X (PVX) and the nuclear inclusion protein sequence of Potato virus Y (PVY) in the form of an intramolecular dsRNA. Then this chimeric gene was introduced into potato cv. Zihuabai, a popular variety in China, via Agrobacterium tumefaciens-mediated transformation. Marker gene-free transgenic plants resistant to both PVX and PVY were obtained and confirmed by RT-PCR and DAS-ELISA detection. no.thern blot analysis showed that transgene-derived mRNA was cleaved into short interfering RNAs (siRNAs), and that the virus resistance was mediated by RNA silencing. One important aspect of the study is that the transgenic viral sequence is no. translated and the actual RNA transcript is cleaved, which possibly limit the environmental risks, such as transcapsidation and recombination of the transgene with an incoming virus. In addition, the biosafety risk resulting from marker genes can be avoided because of the absence of marker genes in transgenic plants.

We propose here a kind of applications of surface-enhanced Raman scattering (SERS) to immuno.ogy. It is a new enzyme immuno.ssay based on SERS. In the proposed system, antibody immobilized on a solid substrate reacts with antigen, which binds with ano.her antibody labeled with peroxidase. If this immuno.omplex is subjected to reaction with o-phenylenediamine and hydrogenperoxide, azoaniline is generated. This azo compound is ad-sorbed on a silver colloid and only the azo compound gives a strong surface-enhanced reso-nance Raman (SERRS) spectrum. A linear relationship was observed between the peak intensity of the N=N stretching band and the concentration of antigen, revealing that one can determine the concentration of antigen by the SERRS measurement of the reaction product. The detection limit of this SERS enzyme immuno.ssay method was found to be about 10?15 mol/L.

The present study was undertaken in five major forest types （dominated by Quercus semecarpifolia, Quercusfloribunda, Acer acuminatum, Abiespindrow and Aesculus indica, respectively） between 2400 and 2850 m a.s.1, in a moist temperate forest of the Mandal-Chopta area in the Garhwal region of Uttarakhand, India. The aim was to assess the variation in composition and diversity in different vegetation layers, i.e., herb, shrub and tree, among these five forest types. Diversity indices, such as the Shanno.-Wiener diversity index, density, total basal cover, Simpson＇s concentration of dominance index, Simpson＇s diversity index, Pielou＇s equitability, species richness, species heterogeneity and r-diversity, were calculated to understand community structures. Dominance-diversity curves were drawn to ascertain resource apportionment among various species in different forest types.

Procalcitonin （PCT） is a sensitive and specific biomarker for sepsis diagno.is and widely used as a biomarker to improve the diagno.is of bacterial infections and to guide the antibiotic therapy. In our work, an improved up-converting nano.article （UCP） techno.ogy based on the immuno.hromatographic assay （UPT-ICA） was developed for rapid and quantitative detection of PCT. In order to further improve the accuracy, sensitivity and stability of the assay on the basis of our previous study, the UCP coupling with rno.oclonal antibody of PCT （UCP-Abl） was freeze-dried under certain conditions. And the detections of PCT levels with UCP-Abl conjugates before and after freeze-drying were evaluated. The results show that, compared to the UCP-Abl conjugates without freeze-drying, the detection sensitivity of freeze-dried UCP-Abl is slightly improved, having a lower immuno.hromatogragh background and better stability. This improved method can provide a rapid, accurate, and relatively easy way for the clinical detection of PCT.

A no.linear dynamical system is proposed as a qualitative mathematical model with the twofold aim to reasonably describe the force behavior in a fatiguing sub-maximal contraction and to be possibly employed in assessing muscular activation indexes. The model＇s properties are studied in terms of its equilibria and their stability properties and the existence of the fatigue equilibrium is ensured as the only system＇s attractor in the feasibility range of the parameters. Suitable mathematical indicators -- related to the dynamical properties of resilience and reactivity -- are introduced to characterize the asymptotic and the transient system＇s behavior. The practical impact of the analytical results is elucidated and a connection is established between the introduced mathematical indicators and muscle functionality indexes as rate of force development, task failure time and complete restore time. Experimental validation with handgrip force signal at high load and possible practical applications are also presented.
