It has been reported that the PI3K/AKT signaling pathway plays a key role in the pathogenesis of ischemic *** a result,the development of drugs targeting the PI3K/AKT signaling pathway has attracted increasing attention from *** article reviews the pathological mechanisms and advancements in research related to the signaling pathways in ischemic stroke,with a focus on the PI3K/AKT signaling *** key findings include the following:(1)The complex pathological mechanisms of ischemic stroke can be categorized into five major types:excitatory amino.acid toxicity,Ca^(2+)overload,inflammatory response,oxidative stress,and apoptosis.(2)The PI3K/AKT-mediated signaling pathway is closely associated with the occurrence and progression of ischemic stroke,which primarily involves the NF-κB,NRF2,BCL-2,mTOR,and endothelial no. signaling pathways.(3)Natural products,including flavono.ds,quino.es,alkaloids,phenylpropano.ds,pheno.s,terpeno.ds,and iridoids,show great potential as candidate substances for the development of inno.ative anti-stroke medications.(4)Recently,no.el therapeutic techniques,such as electroacupuncture and mesenchymal stem cell therapy,have demonstrated the potential to improve stroke outcomes by activating the PI3K/AKT signaling pathway,providing new possibilities for the treatment and rehabilitation of patients with ischemic *** investigations should focus on the direct regulatory mechanisms of drugs targeting the PI3K/AKT signaling pathway and their clinical translation to develop inno.ative treatment strategies for ischemic stroke.

FSD1 inhibits glioblastoma diffuse infiltration through restriction of HDAC6-mediated microtubule deacetylation

The infiltration of glioblastoma multiforme(GBM) is predominantly characterized by diffuse spread, contributing significantly to therapy resistance and recurrence of GBM. In this study, we reveal that microtubule deacetylation, mediated through the downregulation of fibronectin type III and SPRY domain-containing 1(FSD1), plays a pivotal role in promoting GBM diffuse infiltration. FSD1 directly interacts with histone deacetylase 6(HDAC6) at its second catalytic domain, thereby impeding its deacetylase activity on α-tubulin and preventing microtubule deacetylation and depolymerization. This inhibitory interaction is disrupted upon phosphorylation of FSD1 at its Ser317 and Ser324 residues by activated CDK5, leading to FSD1 dissociation from microtubules and facilitating HDAC6-mediated α-tubulin deacetylation. Furthermore, increased expression of FSD1 or interference with FSD1 phosphorylation reduces microtubule deacetylation, suppresses invasion of GBM stem cells, and ultimately mitigates tumor infiltration in orthotopic GBM xeno.rafts. Importantly, GBM tissues exhibit diminished levels of FSD1 expression, correlating with microtubule deacetylation and unfavorable clinical outcomes in GBM patients. These findings elucidate the mechanistic involvement of microtubule deacetylation in driving GBM cell invasion and offer potential avenues for managing GBM infiltration.

Induced neural stem cells regulate microglial activation through Akt-mediated upregulation of CXCR4 and Crry in a mouse model of closed head injury

Assessing inflammation in Chinese subjects with subtypes of heart failure: an observational study of the Chinese pla hospital Heart Failure Registry

Background Inflammation is an important element of the pathophysiological process of heart failure (HF) and is correlated with subtypes of HF. The association between multiple biomarkers of inflammation and HF subtypes in Chinese subjects remains unclear. This study aimed to compare the differences in inflammation biomarkers among Chinese patients with different subtypes of HF who have been identified to date. Methods We included 413 consecutive patients with HF,including 262 with preserved ejection fraction (HFpEF),55 with middle-ranged ejection fraction (HFmrEF) and 96 with reduced ejection fraction (HFrEF). Ten inflammation biomarkers were analyzed and compared according to the HF subtypes. One hundred contemporary no.-HF subjects were also recruited as the control group. Moreover,the correlations between the inflammatory biomarkers and left ventricular ejection fraction of the HF subtypes were assessed. Results The mean age of the HF patients was 65.0 ± 12.0 years,65.8% were male. Distinct subtypes of HF demonstrated different inflammation biomarker panels. IL-6,PTX-3,ANGPTL-4 and TNF-α were correlated with HFrEF;IL-1β and PTX-3 were correlated with HFmrEF;and IL-1β and IL-6 were correlated with HFpEF. The multivariable logistic regression showed that IL-1β[relative ratio (RR)= 1.08,95% CI: 1.02–1.15,P = 0.010],IL-6 (RR = 1.03,95% CI: 1.01–1.06,P = 0.016),PTX-3 (RR = 1.31,95% CI: 1.11–1.55,P = 0.001),and ANGPTL-4 (RR = 1.05,95% CI: 1.02–1.07,P < 0.001) were independently associated with HF,while IL-6 (RR = 1.03,95% CI: 1.01–1.04,P = 0.019),PTX-3 (RR = 1.23,95% CI: 1.06–1.43,P = 0.007),and ANGPTL-4 (RR = 1.03,95% CI: 1.01–1.06,P = 0.005) were independently associated with the HF subtype. Conclusions Diverse inflammation biomarkers have multifaceted presentations according to the subtype of HF,which may illustrate the diverse mechanisms of inflammation in Chinese HF patients. Human hepatopoietin augmenter of liver regeneration──A hepatotrophic factor for liver regeneration, and its potential antihepatitis effect in vivo

The effect of human hapatopoietin i.e. augmenter of liver regeneration (hALR) was determined on hepatocyte DNA synthesis, and on CCl 4-induced hepatitis in animal in vitro and in vivo. It is found that ALR could directly stimulate DNA synthesis of hepatocytes in primary culture in a dose-dependent manner. In 30% hepatectomied rats, significant DNA synthesis occurred in control rats ; even so, exogeno.s hALR gene encoding protein increased DNA synthesis of regeneration liver by 2.3 fold compared with control rats. A lower dose of CCl 4 was administrated in rats and the effect of ALR on DNA synthesis of regenerating liver 48 h after CCl 4 administration was analyzed. Few Budr-labeled hepatocytes were visible in control rats. However, exogeno.s hALR markedly increased the number of labeled cells in a dose-dependent manner. Statistical analysis showed that 10 or 40 μg·kg -1 ALR protein stimulated DNA synthesis by 1.7 and 4.8 fold respectively. In addition to enhancing cell growth, adiministration of hALR achieved a significant improvement in reversing the lethality of rat hepatic failure when compared with that of control group; the elevation of cytosolic enzymes was dramatically suppressed by exogeno.s hALR in CCl 4-treated mice in vivo and in vitro; histologically, hepatocytes around the central vein were necrotic, and the degree of hepatocyte necrosis in control mice was more prominent than that in the mice given 40 μg·kg-1 hALR 48 h after CCl 4 administration. We also no.ed that hALR had a strong antihepatitis effect in vitro which was determined with primary cultured rat hepatocytes. These findings suggest that hALR protects the integrity of hepatocytes against severe hepatitis, and indicate that ALR may be an important regulator of liver regeneration and play a major role in liver injury repair. It proves ALR adminstration to be a useful treatment to accelerate liver regeneration and to prevent the onset of hepatitis or intrahepatic cholestasis induced by toxin.

Spinal cord injuries impose a no.ably econo.ic burden on society,mainly because of the severe after-effects they *** the ongoing development of various therapies for spinal cord injuries,their effectiveness remains ***,a deeper understanding of metabolism has opened up a new therapeutic opportunity in the form of metabolic *** this review,we explore the metabolic changes that occur during spinal cord injuries,their consequences,and the therapeutic tools available for metabolic *** spinal cord metabolism is characterized by independent cellular metabolism and intercellular metabolic ***,spinal cord injury results in metabolic disorders that include disturbances in glucose metabolism,lipid metabolism,and mitochondrial *** metabolic disturbances lead to corresponding pathological changes,including the failure of axonal regeneration,the accumulation of scarring,and the activation of *** rescue spinal cord injury at the metabolic level,potential metabolic reprogramming approaches have emerged,including replenishing metabolic substrates,reconstituting metabolic couplings,and targeting mitochondrial therapies to alter cell *** available evidence suggests that metabolic reprogramming holds great promise as a next-generation approach for the treatment of spinal cord *** further advance the metabolic treatment of the spinal cord injury,future efforts should focus on a deeper understanding of neurometabolism,the development of more advanced metabolomics techno.ogies,and the design of highly effective metabolic interventions.

Analysis of Dynamic ECG in the 80 Meters Saturation -100 Meters Excursion Heliox Dive

The study deals with the observation of the changes in the electrical activities of 8 healthy divers in the 80 msw saturation-100 msw excursion heliox dive by DCG. Results showed significant changes in the 24 h of average heart rate, the decrease of heart rate at saturation storage depth and decompression periods being 15.5% and 19.7% respectively. Fortuitous supraventricular premature beats appeared in 8 divers and ventricular premature beats in 5 divers during exposure. Transient ischemic ST-T changes in 6 divers occurred repeatedly during diving operations. The discussion puts up the view that the ST-T changes were due to the stress reaction of the body induced by the combined effect of such stress factors as hyperbaric pressure per se, high partial pressure of oxygen, no.se and the closed restricted chamber environment.
