Background and Aims:The results of basic research implicate the vascular endothelial growth factor(VEGF)family as a potential target of hepatopulmonary syndrome(HPS).However,the negative results of anti-angiogenetic therapy in clinical studies have highlighted the need for markers for ***,we aimed to determine whether VEGF family members and their receptors can be potential biomarkers for HPS through clinical and experimental ***:Clinically,patients with chronic liver disease from two medical centers were enrolled and examined for *** were divided into HPS,intrapulmonary vascular dilation[positive contrast-enhanced echocardiography(CEE)and normal oxygenation]and CEE-negative *** information and perioperative clinical data were compared between HPS and non-HPS *** levels of VEGF family members and their receptors were *** parallel,HPS rats were established by common bile duct ***,lung and serum samples were collected for the evaluation of pathophysiologic changes,as well as the expression levels of the above ***:In HPS rats,all VEGF family members and their receptors underwent significant changes;however,only soluble VEGFR1(sFlt-1)and the sFlt-1/placental growth factor(PLGF)ratio were changed in almost the same manner as those in HPS ***,through feature selection and internal and external validation,sFlt-1 and the sFlt-1/PLGF ratio were identified as the most important variables to distinguish HPS from non-HPS ***:Our results from animal and human studies indicate that sFlt-1 and the sFlt-1/PLGF ratio in serum are potential markers for HPS.

Background The performance of intravenous tenecteplase in patients who had an acute ischaemic stroke with large/medium vessel occlusion or severe stenosis in an extended time window remains unknown. We investigated the promise of efficacy and safety of different doses of tenecteplase manufactured in China, in patients who had an acute ischaemic stroke with large/ medium vessel occlusion beyond 4.5- hour time window. Methods The CHinese Acute tissue- Based imaging selection for Lysis In Stroke- Tenecteplase was an investigator- initiated, umbrella phase IIa, open- label, blinded- endpoint, Simon’s two- stage randomised clinical trial in 13 centres across China's Mainland. Participants who had salvageable brain tissue on automated perfusion imaging and presented within 4.5-24 hours from time of last seen well were randomised to receive 0.25 mg/ kg tenecteplase or 0.32 mg/kg tenecteplase, both with a bolus infusion over 5-10 s. The primary outcome was proportion of patients with promise of efficacy and safety defined as reaching major reperfusion without symptomatic intracranial haemorrhage at 24-48 hours after thrombolysis. Assessors were blinded to treatment allocation. All participants who received tenecteplase were included in the analysis. Results A total of 86 patients who had an acute ischaemic stroke identified with anterior large/medium vessel occlusion or severe stenosis were included in this study from November 2019 to December 2021. All of the 86 patients enrolled either received 0.25 mg/kg (n=43) or 0.32 mg/kg (n=43) tenecteplase, and were available for primary outcome analysis. Fourteen out of 43 patients in the 0.25 mg/kg tenecteplase group and 10 out of 43 patients in the 0.32 mg/kg tenecteplase group reached the primary outcome, providing promise of efficacy and safety for both doses based on Simon’s two- stage design. Posttraumatic stress disorder(PTSD)is a complex mental disorder notable for traumatic experience *** current first-line treatments are linked with clinically important symptom reduction,a large proportion of patients retained to experience considerable residual symptoms,indicating pathogenic mechanism should be illustrated *** studies reported that newly formed myelin could shape neural circuit function and be implicated in fear memory ***,its role in PTSD remains to be *** this study,we adopted a restraint stress-induced PTSD mouse model and found that PTSD-related neuropsychiatric symptoms were accompanied by increased myelination in the posterior parietal cortex and ***,but not risperidone or sertraline,has a more profound rescue effect on neuropsychological behaviors and myelin *** mechanistic experiments revealed that fluoxetine could directly interfere with oligodendroglial differentiation by upregulating Wnt *** data demonstrated the correlation between PTSD and abnormal myelination,suggesting that the oligodendroglial lineage could be a target for PTSD treatment.

Patients with small-cell lung cancer (SCLC) relapse within months after completing previous therapies. This study aimed to investigate the efficacy and safety of anlotinib as third- or further-line therapy in patients with short-term relapsed SCLC from ALTER1202. Patients with short-term relapsed SCLC (disease progression within 3 months after completing ≥ two lines of chemotherapy) in the anlotinib (n = 67) and placebo (n = 34) groups were analyzed. The primary endpoint was progression-free survival (PFS). The secondary endpoints included overall survival, objective response rate (ORR), disease control rate, and safety. Anlotinib significantly improved median PFS/OS (4.0 vs. 0.7 months, P line anlotinib treatment was associated with improved survival benefit. Further studies are warranted in this regard.

In this study, we conducted a clinical analysis of lymphocyte subtypes in 268 patients with Parkinson＇s disease （PD） to assess their clinical impact as a potential marker of advanced PD in Chinese patients. The participants comprised 268 sporadic PD patients and 268 healthy controls. The numbers of natural killer （NK） cells and CD3＋, CD3＋CD4＋, CD3＋CD8＋, and CD19＋ lympho- cytes from peripheral blood were determined by immunos- taining and flow cytometric analysis and the percentages of these CD＋ T cells were calculated. The ratio of regulatory T （Treg）/helper T 17 （Thl7） lymphocytes from 64 PD patients and 46 controls was determined by flow cytometric analysis. The results showed that the percentage of NK cells was higher in advanced PD patients than in controls （22.92% ± 10.08% versus 19.76% ±10.09%, P= 0.006）, while CD3＋ T cells are decreased （62.93% ±9.27% versus 65.75% ± 9.13%, P = 0.005）. The percentage of CD19＋ B cells in male patients was lower （P = 0.021） than in female patients, whereas NK cells were increased （P 〈 0.0001）. The scores on the Unified Parkinson＇s Disease Rating Scale （UPDRS） and the Non-Motor Symptoms Scale in late-onset PD patients were significantly higher than those in earlyonset patients （P = 0.024 and P = 0.007, respectively）. The percentage of CD19＋ B cells in patients with UPDRS scores 〉24 was lower than in those with scores 〈24 （10.17% ± 4.19% versus 12.22% ± 5.39%, P = 0.009）. In addition, the Treg/Th17 ratio in female patients was higher than that in female controls （13.88 ± 6.32 versus 9.94 ±4.06, P = 0.042）. These results suggest that the percentages of NK cells, CD3＋ T cells, and CD19＋ B cells along with the Treg/Th17 ratio in peripheral blood may be used to predict the risk of PD in Chinese individuals and provide fresh avenues for novel diagnostic biomarkers and therapeutic designs.
