Genetic and environmental control of fruit maturation,dry matter and firmness in apple(Malus×domestica Borkh.)

For any given genotype,the environment in which an apple is grown can influence the properties of the fruit *** there has been extensive research on the mechanism of the genetic control of fruit quality traits,less effort has been made to investigate the way that these genetic mechanisms interact with the *** address this issue,we employed a large‘Royal Gala’×‘Braeburn’population of 572 seedlings replicated over sites in three climatically diverse apple-growing regions in new *** for traits including fruit maturation timing,firmness and dry matter content was performed at each of these three sites for a single growing season(2011),and at two sites(Motueka and Hawke’s Bay)for two seasons(2009 and 2010).The phenotype data collected over 2 years at two sites enabled the detection of 190 quantitative trait loci(QTL)that controlled these traits regardless of year or growing location,as well as some chromosomal loci that influenced the traits in a single given environment or *** those loci that were environmentally stable over three sites,there was an interdependency of fruit maturation date,dry matter content and storage potential within this population,with two regions on Linkage Groups(LGs)10 and 16 strongly *** these loci were used in a marker-assisted selection programme to select for progeny bearing firmer fruit,this would have the unintentional consequence of selecting,high dry matter content,later maturing *** addition,a further 113 new QTLs with a smaller effect were identified,some of which were exhibited only in a single growing environment,demonstrating the underlying complexity of control of traits determining fruit quality,in addition to the need for being aware of environmental effects when developing new apple varieties.

Soil particle size range correction for improved calibration relationship between the laser-diffraction method and sieve-pipette method

Particle size fraction(clay, silt, and sand) is an important characteristic that influences several soil functions. The laser-diffraction method(LDM) provides a fast and cost-effective measurement of particle size distribution, but the results usually differ from those obtained by the traditional sieve-pipette method(SPM). This difference can persist even when calibration is applied between the two methods. This partly relates to the different size ranges of particles measured by the two methods as a result of different operational principles, i.e., particle sedimentation according to Stokes’ Law vs. Mie theory for laser beam scattering. The objective of this study was to identify particle size ranges of LDM equivalent to those measured by SPM and evaluate whether new calibration models based on size range correction can be used to improve LDM-estimated particle size fractions, using 51 soil samples with various texture collected from five soil orders in new zealand. Particle size distribution was determined using both LDM and SPM. Compared with SPM, original data from LDM underestimated the clay fraction(new calibration models(based on the corrected size ranges of these two methods) improved the estimation of clay and silt contents by LDM. Compared with soil-specific models(i.e., different models were developed for different soils), a universal model may be more parsimonious for estimating particle size fractions if the samples to be assessed represent multiple soil orders.

The apple dwarfing rootstock‘Malling9’(‘M9’)has been used worldwide both to reduce scion vigour and as a genetic source for breeding new *** of‘M9’segregate for rootstock-induced dwarfing of the scion,indicating that this trait is controlled by one or more genetic factors.A quantitative trait locus(QTL)analysis of a rootstock population derived from the cross between‘M9’×‘Robusta5’(non-dwarfing)and grafted with‘Braeburn’scions identified a major QTL(Dw1)on linkage group(LG)5,which exhibits a significant influence on dwarfing of the scion.A smaller-effect QTL affecting dwarfing(Dw2)was identified on LG11,and four minor-effect QTLs were found on LG6,LG9,LG10 and *** analysis indicates that the combination of Dw1 and Dw2 has the strongest influence on rootstock-induced dwarfing,and that Dw1 has a stronger effect than *** markers linked to Dw1 and Dw2 were screened over 41 rootstock accessions that confer a range of effects on scion *** majority of the dwarfing and semi-dwarfing rootstock accessions screened carried marker alleles linked to Dw1 and *** suggests that most apple dwarfing rootstocks have been derived from the same genetic source.

Characterization of the proteases in the midgut of the xylophagous larvae of Oemona hirta （Coleoptera： Cerambycidae）

The protein digestive capability of the larvae of the longhorn beetle （Oemona hirta, Coleoptera： Cerambycidae, Fabricius, 1775） was investigated. This species feeds only on wood where there is a high proportion of vascular tissue. The pH of the midgut, the major digestive organ, was alkaline and protein hydrolysis was maximal at alkaline pH. Use of specific synthetic peptide substrates showed that the major protease activities were the endopeptidases, trypsin and chymotrypsin-like activity, and the exopeptidase, leucine aminopeptidase and the pH curves corresponded to that with protein substrate. Studies using a range ofserine protease inhibitors as well as specific inhibitors ofmetalloproteases, cysteine proteases and aspartate proteases confirmed a serine protease-based digestive system similar to earlier reports of sapwood-feeding Cerambycids. Control of these insect pests using protease inhibitors is discussed.

Does Particulate Organic Matter Fraction Meet the Criteria for a Model Soil Organic Matter Pool?

There is a well-recognized need for improved fractionation methods to partition soil organic matter into functional pools. Physical separation based on particle size is widely used, yielding particulate organic matter(POM, i.e., free or "uncomplexed" organic matter> 50 μm) as the most labile fraction. To evaluate whether POM meets criteria for an ideal model pool, we examined whether it is:1) unique, i.e., found only in the > 50 μm fraction and 2) homogeneous, rather than a composite of different subfractions. Following ultrasonic dispersion, sand(> 50 μm) along with coarse(20–50 μm) and fine(5–20 μm) silt fractions were isolated from a silt loam soil under long-term pasture at Lincoln, new zealand. The sand and silt fractions contained 20% and 21% of total soil C, *** adopted a sequential density separation procedure using sodium polytungstate with density increasing step-wise from 1.7 to 2.4 g cm^(-3) to recover organic matter(light fractions) from the sand and silt fractions. Almost all(ca. 90%) the organic matter in the sand fraction and a large proportion(ca. 60%–70%) in the silt fractions was recovered by sequential density separation. The results suggested that POM is a composite of organo-mineral complexes with varying proportions of organic and mineral materials. Part of the organic matter associated with the silt fractions shared features in common with POM. In a laboratory bio-assay, biodegradability of POM varied depending on land use(pasture > arable cropping). We concluded that POM is neither homogeneous nor unique.

Apple dwarfing rootstocks cause earlier shoot termination and reduced root and shoot *** identify physiological factors responsible for rootstock-induced growth restriction,we compared vascular-enriched gene expression between two dwarfing rootstocks(‘M27’and‘M9’)and the vigorous rootstock‘M793’using RNA sequencing and quantitative reverse transcriptase *** expressed genes common to both dwarfing rootstocks belonged to five main biological processes:(1)primary metabolism,(2)cell wall synthesis and modification,(3)secondary metabolism,(4)hormone signalling and response and(5)redox *** promoting the biosynthesis of amino acids,lipids and cell walls were downregulated in dwarfing rootstocks,whereas genes promoting the breakdown of these compounds were *** only exception to this trend was the upregulation of starch synthesis genes in dwarfing ***-structural carbohydrate analysis demonstrated that starch concentrations in‘M9’roots,stems and grafted‘Royal Gala’(‘RG’)scions were double that of equivalent tissues from‘RG’homo-grafted trees(‘RG’/‘RG’).Fructose and glucose concentrations were much lower in all three tissues of the‘RG’/‘M9’***,these data indicate that dwarfing rootstocks are in a state of sugar depletion and reduced cellular activity despite having large starch *** significant finding was the over-accumulation of flavonoids and the downregulation of auxin influx transporters MdAUX1 and MdLAX2 in dwarfing *** propose that both factors reduce polar auxin *** results of this study contribute novel information about the physiological state of dwarfing rootstocks.

Responses of 9 lepidopteran species to Bacillus thuringiensis： How useful is phylogenetic relatedness for selecting surrogate species for nontarget arthropod risk assessment？

To evaluate phylogenetic relatedness as a proxy for susceptibility to Bacillus thuringiensis （Bt） when selecting species to act as surrogates for others in prerelease test- ing, we examined the responses of 11 laboratory-reared lepidopteran colonies, comprising members of 2 families, 5 genera, and 9 species, to a commercial Bt preparation. Sur- vival, pupal mass, and timing of pupation and adult emergence of 2 noctuids （Spodoptera litura and Helicoverpa armigera） and 7 tortricids （Cnephasia jactatana, Ctenopseustis obliquana, Ctenopseustis herana, Planotortrix octo, Planotortrix notophaea, Planotor- trix excessana [2 different laboratory colonies], and Epiphyas postvittana [2 colonies]） were examined after feeding first instar larvae with artificial diet containing 5 μL/100 mL Dipel ES （Bt subsp, kurstaki）. Bt caused significant larval mortality in all species except S. litura, in which only pupation was delayed compared with untreated controls. Neither of the noctuid species tested would act as a suitable surrogate for the other in tests of Bt impacts on survival. With the exception of the 2 colonies orE. postvittana, which differed from each other not only in their responses to Bt but also in their development times when not treated with Bt, species within each tortricid genus had similar responses to Bt and thus could act as surrogates for each other. Members of different genera within this family could represent each other only if relatively coarse measurement endpoints （e.g., toxic or not） were considered adequate for assessing risks to nontarget species in the field.

Genotyping-by-sequencing of pear (Pyrus spp.) accessions unravels novel patterns of genetic diversity and selection footprints

Understanding of genetic diversity and marker-trait relationships in pears(Pyrus spp.)forms an important part of gene conservation and cultivar *** of Asian and European pear species,and interspecific hybrids were planted in a common garden ***-by-sequencing(GBS)was used to genotype 214 accessions,which were also phenotyped for fruit quality traits.A combination of selection scans and association analyses were used to identify signatures of *** of genetic diversity,population structure and introgression were also *** 15000 high-quality SNP markers were identified from the GBS data,of which 25%and 11%harboured private alleles for European and Asian species,*** clustering analysis suggested negligible gene flow,resulting in highly significant population differentiation(Fst=0.45)between Asian and European *** hybrids displayed an average of 55%and 45%introgression from their Asian and European ancestors,***(firmness,acidity,shape and so on)variation between accessions was significantly associated with genetic *** frequencies at large-effect SNP loci were significantly different between genetic groups,suggesting footprints of directional *** scan analyses identified over 20 outlier SNP loci with substantial statistical support,likely to be subject to directional selection or closely linked to loci under selection.
