Non-pulmonary allergic diseases and inflammatory bowel disease: A qualitative review

While the etiological underpinnings of inflammatory bowel disease(IBD) are highly complex, it has been not-ed that both clinical and pathophysiological similarities exist between IBD and both asthma and non-pulmonary allergic phenomena. In this review, several key points on common biomarkers, pathophysiology, clinical manifesta-tions and nutritional and probiotic interventions for both IBD and non-pulmonary allergic diseases are *** and mast cell activity show common behav-iors in both IBD and in certain allergic disorders. IgE also represents a key immunoglobulin involved in both IBD and in certain allergic pathologies, though these links require further study. Probiotics remain a critically important intervention for both IBD subtypes as well as multiple allergic phenomena. Linked clinical phenomena, especially sinonasal disease and IBD, are discussed. In addition, nutritional interventions remain an underuti-lized and promising therapy for modification of both al-lergic disorders and IBD. Recommending new mothers breastfeed their infants, and increasing the duration of breastfeeding may also help prevent both IBD and al-lergic diseases, but requires more investigation. While much remains to be discovered, it is clear that non-pulmonary allergic phenomena are connected to IBD in a myriad number of ways and that the discovery of com-mon immunological pathways may usher in an era of vastly improved treatments for patients.

Global COVID-19 Pandemic Waves: Limited Lessons Learned Worldwide over the Past Year

Major depressive disorder(MDD)is a chronic,generally episodic and debilitating disease that affects an estimated 300 million people worldwide,but its pathogenesis is poorly *** heritability estimate of MDD is 30–40%,suggesting that genetics alone do not account for most of the risk of major *** factor known to associate with MDD involves environmental stressors such as childhood adversity and recent life *** studies have emerged to show that the biological impact of environmental factors in MDD and other stress-related disorders is mediated by a variety of epigenetic *** epigenetic modification alterations contribute to abnormal neuroendocrine responses,neuroplasticity impairment,neurotransmission and neuroglia dysfunction,which are involved in the pathophysiology of ***,epigenetic marks have been associated with the diagnosis and treatment of *** evaluation of epigenetic modifications holds promise for further understanding of the heterogeneous etiology and complex phenotypes of MDD,and may identify new therapeutic ***,we review preclinical and clinical epigenetic findings,including DNA methylation,histone modification,noncoding RNA,RNA modification,and chromatin remodeling factor in *** addition,we elaborate on the contribution of these epigenetic mechanisms to the pathological trait variability in depression and discuss how such mechanisms can be exploited for therapeutic purposes.

Identifying microRNA panels specifically associated with hepatocellular carcinoma and its different etiologies

Aim:Deregulation of microRNAs(miRNAs)expression has been identified in hepatocellular carcinoma(HCC),but few results are *** objective of this study is to investigate“HCC tumor type specific”and“tumor common”miRNA ***:The authors integrate and analyze clinical,etiologic and miRNA profiles data from 9 types of solid tumors in The Cancer Genome Atlas(TCGA)and HCC data from columbia university medical center(CUMC).Results:Levels of 33 miRNAs were significant different between HCC tumor and paired non-tumor tissues(over 2-fold changes)after Bonferroni correction for multiple comparisons,and most(28 miRNAs)were down-regulated in HCC *** this panel,the authors well classified HCC tumor tissues with 4 misclassifications among 48 paired *** this panel in an additional 302 HCC tumor tissues,the authors almost perfectly distinguished tumor from non-tumor tissues with only two misclassifications(99%of HCC tissues correctly classified).Evaluating miRNA profiles in 32 independent HCC paired tissues from CUMC,the authors observed 40 miRNAs significantly deregulated in HCC with over 2-fold changes;14 overlapped with those identified in *** analyses by HCC etiology found that 4 upregulated and 8 downregulated miRNAs were significantly associated with alcohol-related *** were 7 and 4 miRNAs significantly associated with hepatitis B virus-and hepatitis C virus-related HCC,*** for the first time revealed that miR-24-1,miR-130a and miR-505 were significantly down-regulated only in HCC tumors;miR-142 and miR-455 were significantly down-regulated in HCC,but up-regulated in 5 other solid tumors;suggesting their HCC“tumor type specific”characteristics.A panel of 8 miRNAs was significant in at least 5 tumor types,including HCC,and was identified as“tumor common”***:The authors concluded that aberrant miRNA panels have HCC“tumor type specificity”and may be affected by etiologic factors.

Risk factors for hepatocellular carcinoma in cirrhosis due to nonalcoholic fatty liver disease: A multicenter, case-control study

AIM To identify risk factors associated with hepatocellular carcinoma(HCC), describe tumor characteristics and treatments pursed for a cohort of individuals with nonalcoholic steatohepatitis(NASH) cirrhosis. METHODS We conducted a retrospective case-control study of a well-characterized cohort of patients among five liver transplant centers with NASH cirrhosis with(cases) and without HCC(controls).RESULTS Ninety-four cases and 150 controls were included. Cases were significantly more likely to be male than controls(67% vs 45%, P < 0.001) and of older age(61.9 years vs 58 years, P = 0.002). In addition, cases were more likely to have had complications of end stage liver disease(83% vs 71%, P = 0.032). On multivariate analysis, the strongest association with the presence of HCC were male gender(OR 4.3, 95%CI: 1.83-10.3, P = 0.001) and age(OR = 1.082, 95%CI: 1.03-1.13, P = 0.001). Hispanic ethnicity was associated with a decreased prevalence of HCC(OR = 0.3, 95%CI: 0.09-0.994, P = 0.048). HCC was predominantly in the form of a single lesion with regional lymph node(s) and distant metastasis in only 2.6% and 6.3%, respectively. Fifty-nine point three percent of individuals with HCC underwent locoregional therapy and 61.5% underwent liver transplantation for HCC. CONCLUSION Male gender, increased age and non-Hispanic ethnicity are associated with HCC in NASH cirrhosis. NASH cirrhosis associated HCC in this cohort was characterized by early stage disease at diagnosis and treatment with locoregional therapy and transplant.

Rupture of intracranial aneurysms is the most common cause of nontraumatic subarachnoid hemorrhage,but the intricate neuroinflammatory processes which contribute to aneurysm pathophysiology are not well‑*** evidence has implicated the complement cascade in the progression of aneurysms from their formation to *** this article,we identify and review studies that have sought to determine the role of the complement system in the aneurysm *** studies were generally conducted by immunhistological analyses on aneurysm tissue collected intraoperatively,and multiple components of the complement cascade and its modulators were identified in specific regions of the aneurysm *** results of the studies suggest that the complement cascade is locally upregulated and disinhibited in the perianeurysmal environment,and that it contributes to chronic as well as acute immunological damage to the aneurysm *** the future,understanding the mechanisms at work in complement‑mediated damage is necessary to leading the development of novel therapies.

Pericyte Plasticity in the Brain

Cerebral pericytes are perivascular cells that stabilize blood vessels. Little is known about the plasticity of pericytes in the adult brain in vivo. Recently, using stateof-the-art technologies, including two-photon microscopy in combination with sophisticated Cre/loxP in vivo tracing techniques, a novel role of pericytes was revealed in vascular remodeling in the adult brain. Strikingly, after pericyte ablation, neighboring pericytes expand their processes and prevent vascular dilatation. This new knowledge provides insights into pericyte plasticity in the adult brain.
