Objective:To investigate the efficiency ofβ-galactosidase gene transfer into rat kidney with ultrasound-mediated microbubble destruction via different injection ***:A total of 25 Wistar rats were randomly divided into 5 *** groups received a mixture of optison microbubbles(0.2 mL) and lacz plasmids(25μg) injection via renal artery,tail vein,anterior tibial muscle and renal parenchyma,*** control group received a mixture of PBS (xx mL) and lacz plasmids(25μg) via renal *** days after the gene transfer,ultrasound with fixed frequency and power(1 MHz,xxW) was delivered to the kidneys for 3 *** efficiency of the gene transfer and expression was evaluated on the basis ofβ-galactosidase *** side effects of this method were evaluated by immunohistological method. Results:β-galactosidase expression could be observed only in tubules but not in glomeruli and interstitial *** efficiency of renal artery group was higher than that of tail vein,anterior tibial muscle and renal parenchyma group(P<0.05).Immunohistochemical analysis revealed co-expression ofβ-galactosidase with a roximal tubule marker,megalin,which suggested that ultrasound enhanced gene transfer into the proximal tubular epithelial ***β-galactosidase expression was observed in the extrarenal *** were no evident pathological and biochemical changes after gene ***:Ultrasound-mediated microbubble destruction can transfer gene into kidney via renal artery,tail vein,anterior tibial muscle and renal *** with renal artery,administrating microbubbles via tail vein and anterior tibial muscle are more convenient and less vulnerarious.

This study was to optimize the exercise preconditioning(EP)intensity in protecting from exhaustive exercise-induced cardiac injury(EECI).A total of 98 male Sprague-Dawley rats were divided into 7 groups(n=14):the control group(C),the exhaustive exercise group(EE)and the EP+EE groups,which include the V10(53.0%VO_(2max)),V15(58.4%VO_(2max)),V20(67.0%VO_(2max)),V26(74.0%VO_(2max))and V30(80.0%VO_(2max))*** the C group,the other groups were subjected to treadmill *** serum contents of N terminal pro B type natriuretic peptide(NT-proBNP)and cardiac troponin I(cTn-I)were detected by the enzyme-linked immunosorbent assay method,ECG was recorded,heart function was detected by pressure volume catheter and the activities of mitochondrial electron transfer pathway(ET pathway)complexesⅠ,ⅡandⅣwere measured by high-resolution respiration *** to the EE group,the EP groups have shown decrease of NT-proBNP and cTn-I,improvement of mitochondrial respiratory function and car-diac *** to other EP groups,the V26 group has shown significant decrease of myocardial enzymes and improvement of mitochondrial *** correlation analysis showed the EP effect was proportional to EP intensity in the range of 53.0%VO_(2max)-74.0%VO_(2max).High intensity and long duration of exhaustive exercise caused cardiac injury and EP could decrease serum level of NT-proBNP and cTn-I,improve electrical derangement and the left ventricular function,and raise the activities of ET pathway complexesⅠ,ⅡandⅣ.The protection of EP on EECI was improved as the EP intensity was increased from 53.0%VO_(2max)to 74.0%VO_(2max)and when EP intensity was 74.0%VO_(2max),the effect was the most obvious among all the setting EP groups.

This study aimed to compare clinical features between membranous nephropathy (MN) and nonmembranous nephropathy (non-MN), to explore the clinically difTerential diagnosis of these two types, and to establish a diagnostic model of MN. After renal biopsy was obtained, 798 patients were divided into two groups based on their examination results: primary MN group (n = 248) and non-MN group (n = 550). Their data were statistically analyzed. Logistic regression analysis indicated that anti-PLA2R antibodies, IgG, and Cr were independently correlated with MN, and these three parameters were then used to establish the MN diagnostic model. A receiver operating characteristic (ROC) curve confirmed that our diagnostic model could distinguish between patients with and without MN, and their corresponding sensitivity, specificity, and AUC were 79.9%, 89.4%, and 0.917, respectively. The cutoff value for this combination in MN diagnosis was 0.34. The established diagnostic model that combined multiple factors shows a potential for broad clinical applications in differentiating primary MN from other kidney diseases and provides reliable evidence supporting the feasibility of noninvasive diagnosis of kidney diseases.

Influence of HbA1c on short-term blood pressure variability in type 2 diabetic patients with diabetic nephropathy

The aim of this study was to understand the characteristics of blood pressure （BP） variability in subjects with diabetic nephropathy （DN）, and identify the probable predictors affecting BP variability. Fifty-one chronic kidney disease （CKD）-hypertensive patients without diabetes （NDN group） and sixty type 2 diabetic patients with overt DN （DN group） were enrolled in this study. The values of short-term BP variability were obtained from 24 h ambulatory BP monitoring （ABPM）. Variance analysis or nonparametric analysis revealed that 24-h systolic BP variability and night- time systolic BP variability of the DN group were significantly higher than those of the NDN group [（12.23±3.66） vs. （10.74±3.83） mmHg, P〈0.05; （11.23±4.82） vs. （9.48±3.69） mmHg, P〈0.05]. Then the patients of the DN group were divided into two groups according to glycated hemoglobin （HbAlc） level： group A （HbA1c〈7%） and group B （HbA1c〉7%）, and the t-test showed that patients in group B had larger 24-h diastolic, daytime diastolic, and nighttime systolic/diastolic BP variability compared with group A. In the DN group, partial correlation analysis revealed that HbAlc exhibited a strong association with 24-h diastolic, daytime diastolic, nighttime systolic and diastolic BP vari- ability （P〈0.001, P〈0.001, P〈0.05, and P〈0.001, respectively）. Taken together, larger short-term BP variability was detected in hypertensive type 2 diabetic patients with overt nephropathy and renal insufficiency. It may imply that the optimal BP variability level could benefit from a better glycaemic control.

The expression, activity and clinical implication of heme oxygenase-1 (HO-1) in the chronic renal insufficiency (CRI) rat kidney and its mechanism were investigated The 5/6 nephrectomized rats were assigned to sham operation group, CRI group and Hemin group At the 8th week after second operation, blood pressure, urinary protein, serum creatinine(Scr) and BUN were measured Renal pathologic changes were observed The activity of HO and contents of erythropoietin (EPO) in serum and renal tissue were determined Immunohistochemistry was used to detect the expression and distribution of HO-1 in the CRI rat kidney As compared with CRI group, the urinary protein, blood pressure, Scr and BUN in Hemin group were reduced significantly ( Pgroup as compared with normal rats HO-1 distributed mainly in tubular epithelial cells The EPO contents in Hemin group were significantly higher than in CRI group Through up-regulating the EPO level in serum and renal tissues, HO-1 retards the progression of CRI

To investigate the effects of mycophenolate mofetil (MMF) on the process of renal interstitial fibrosis,unilateral ureteral obstruction (UUO) model was established in rats Twenty Sprague-Dawley rats underwent UUO and received vehicle ( n =10) or MMF (20 mg kg -1 d -1 , by daily gastric gavage, n= 10) during a period of 5 days following surgery, and the additional 10 rats were served as sham-operated group The rats were killed 5 days after surgery Immunohistochemistry was performed on renal tissue for proliferating cell nuclear antigen (PCNA), α-smooth muscle actin (α-SMA) and type Ⅰ and Ⅲ collagen (colⅠ, colⅢ) Histological studies were also done by MASSON staining Five days after surgery, proliferating cells in tubules, interstitium as well as interstitial myofibroblast (MyoF) infiltration and interstitial colⅠ, colⅢ deposition were all significantly reduced by MMF treatment MMF also alleviated the histological changes of UUO rats These results suggested that the reduction of interstitial MyoF infiltration may be an important event by which MMF prevents renal injury caused by UUO and MMF could be used to limit the progression of renal fibrosis
