AIM： To characterize the role of flgK and its protein product in Hpylori colonization. METHODS： The PCR cloning method identified the flgK gene. An isogenic flgK mutant was constructed by gene replacement and confirmed by Southern blot analysis and PCR analysis. The recombinant FlgK protein （r-FlgK） was purified. Electron microscopy （EM） was applied to demonstrate the flagella of H pylori. An in vitro motility test was assessed in semisolid medium. The densities of H pylori colonization with either the wild-type strain or its flgK mutant were compared among BALB/c mice with or without pre-immunization with r-FlgK. The serological responses to r-FlgK were analyzed for 70 clinical patients with different densities of H pylori colonization. RESULTS： From a duodenal ulcer strain, the flgK gene was cloned and it contained 1821 bp, with a 95.7% identity to the published sequences. No flagella were observed under EM for the mutant strain, which had a loss of motility. Hpylori density was lower in the BALB/c mice inoculated by the mutant or with pre-immunization with r-FlgK compared to unimmunized mice or mice inoculated by the wild-type strain （P 〈 0.05）. In the H pylori-infected patients, the serological responses to r-FlgK were uniformly low in titer.CONCLUSION： FlgK encoded by flgK is important for flagella formation and H pylori motility. Deficiency in FlgK or an enhanced serological response to r-FlgK can interfere with Hpylori colonization. FlgK of Hpylori could be a novel target for vaccination.

BACKGROUND An increased risk of insulin resistance(IR)has been identified in rheumatoid arthritis(RA),a chronic inflammatory disorder with elevated levels of pathogenic *** targeting proinflammatory cytokines can control the disease and improve insulin sensitivity in *** Janus kinase(JAK)signaling can regulate cytokine receptors and participate in RA pathogenesis,it remains to be elucidated whether there is a reduction of IR in such patients under JAK inhibitor(JAKi)*** To study the effect of JAKi treatment on the reduction of IR in RA patients with active *** A retrospective study was carried out from April 1,2017 to March 31,2021 in a population of non-diabetic patients with active RA who were undergoing tofacitinib(TOF)therapy with 5 mg twice-daily immediate-release *** Fifty-six RA patients,aged 30 years to 75 years(mean±SD:52.3±11.1)with disease activity score 28 values ranging from 4.54 to 7.37(5.82±0.74),were classified into high-IR(>2.0)and low-IR(≤2.0)groups based on their baseline homeostatic model assessment(HOMA)-IR *** had no previous exposure to JAKi,and received TOF therapy for no less than 6 *** 30 patients who were naïve to biologics,after a 24-week therapeutic period,the high-IR group showed reduced HOMA-IR levels (3.331 ± 1.036 vs 2.292 ± 0.707, P < 0.001). In another 26patients who were exposed to tumor necrosis factor-α or interleukin-6 blockers, the high-IR group,despite having achieved a decrease but with lower magnitude than in naïve patients, showedreduced HOMA-IR levels (2.924 ± 0.790 vs 2.545 ± 1.080, P = 0.018).CONCLUSIONIn this retrospective study, reduced IR was achieved in non-diabetic active RA patients following24 wk of TOF therapy.

The purpose of this study is to analyze variability in rainfall threshold for debris flow(critical rainfall for debris flow triggering) after the ML 7.3 Chi-Chi earthquake in central Taiwan in 1999. Two study sites with different geological conditions were surveyed in the earthquake area. Streambed surveys were conducted to continuously monitor debris flows between 1999 and 2006. During the 7-year study period,every debris flow event was identified,and the streambed characterized. Results show that the rainfall threshold for debris flow was remarkably lower just after the Chi-Chi Earthquake,but gradually recovered. To date,this rainfall threshold is still lower than the original level prior to the earthquake. This variability in rainfall threshold is closely related to the amount of sediment material in the initiation area of debris flow,which increased rapidly due to landslides resulting from the earthquake. With the increase in sediment material,the rainfall threshold was lowered severely during the first year following the Chi-Chi earthquake. However,heavy rainfalls mobilized the sediment material,causing debris flows and transporting sediment downstream. With the decrease in sediment material,the rainfall threshold recovered gradually over time. Furthermore,debris flows occurred only in the subbasins that had sufficient sediment material to cause significant movement. Hence,these results confirm that the sediment material in the initiation area of debris flow is a crucial component of the rainfall threshold for debris flow.

In addition toβ-cell failure with inadequate insulin secretion,the crucial mechanism leading to establishment of diabetes mellitus(DM)is the resistance of target cells to insulin,*** resistance(IR),indicating a requirement of beyond-normal insulin concentrations to maintain euglycemic status and an ineffective strength of transduction signaling from the receptor,downstream to the substrates of insulin *** is a common feature of most metabolic disorders,particularly type II DM as well as some cases of type I DM.A variety of human inammatory disorders with increased levels of proinflammatory cytokines,including tumor necrosis factor(TNF)-α,interleukin(IL)-6 and IL-1β,have been reported to be associated with an increased risk of *** arthritis conditions,including rheumatoid arthritis(RA),psoriatic arthritis(PsA)and ankylosing spondylitis(AS),with the involvement of proinflammatory cytokines as their central pathogenesis,have been demonstrated to be associated with IR,especially during the active disease *** is an increasing trend towards using biologic agents and small molecule-targeted drugs to treat such *** this review,we focus on the effects of anti-TNF-α-and non-TNF-α-targeted therapies on IR in patients with RA,PsA and ***-TNF-αtherapy,IL-1 blockade,IL-6 antagonist,Janus kinase inhibitor and phosphodiesterase type 4 blocker can reduce IR and improve diabetic hyper-glycemia in autoimmune-mediated arthritis.

Using partial factor productivity and data envelopment analysis approaches, this study measures and compares the operational efficiency of the 10 dominant mobile operators in Brazil, Russia, India and China （BRIC） between 2002 and 2006. The results of data envelopment analysis indicate that the two leading Brazilian mobile operators, Vivo and TIM, are fully efficient throughout the entire period of study, however, Indian mobile operators are the least efficient among BRIC operators. Interestingly, the findings of this study verify that full operational efficiency can be achieved by operators with large revenues, such as China Unicorn, and by other, y with medium and small revenues, such as Hvo, TIM and Oi. Partial faetor productivity demonstrates that 3 of the 4 Brazilian mobile operators, Vivo, TIM and Oi, have remarkable productivity ratios, and that the state-owned operator, China Unicom, has the highest revenue per capital expenditure among BRIC operators. In contrast, Indian mobile operators ＇productivity ratios are generally low compared to other BRIC mobile carriers.

In this paper, we propose a knowledge discovery method based on the fuzzy set theory to help elders with plant cultivation. Initially, the fuzzy sets are constructed by using the feature selection and statistical interval estimation. The min-max inference and the center of gravity defuzzification method are then used to output a candidate pattern set. Finally, a pattern discovery is adopted to obtain the patterns from the candidate set for the cultivation suggestions by considering the frequency weight and user＇s experience. In order to demonstrate the performance of our method in planting systems, we conduct a clicks-and-mortar cultivation platform, namely Eden Garden, for the elderly lifestyles of health and sustainability（LOHAS）. The experimental results show that the accuracy rate of our knowledge discovery method can reach up to 85%. Moreover, the results of the LOHAS index scale table present that the happiness of the elders is increasing while the elders are using our proposed method.
