Aluminum exposure impairs oocyte quality via subcellular structure disruption and DNA damage-related apoptosis in mice

Aluminum(Al)can lead to an exposure of creature in varieties ways for its universality,and it could disturb normal physiological metabolism,with the damage to multisystem including *** the oocyte quality is critical for female reproduction,we inspected the toxicity of Al on mouse oocyte *** constructed in vitro exposure mouse model,and we found that 5 mmol/L Al had adverse effects on oocyte maturation by impairing organelle and *** spindle and misaligned chromosomes which might be considered to be caused by elevated levels of acetylation,as well as abnormal distribution of actin dynamics could hinder normal meiosis of *** dysfunction indicated that Al affected proteins synthesis,transport and digestion,which would further damage oocyte *** order to explore the mechanism of Al toxicity,our further investigation demonstrated that Al caused mitochondrial dysfunction and imbalance calcium homeostasis,resulting in limited energy ***,high level of reactive oxygen species,DNA damage and apoptosis caused by oxidative stress were also the manifestation of Al toxicity on *** conclusion,our study provided the evidence that Al exposure affected oocyte quality through its effects on spindle organization,actin dynamics,organelle function and the induction of DNA damage-related apoptosis with mouse model.

Methods to Reduce the Hypoglycemic Mortality of Alloxan in Diabetic Rabbit Model

Objective: To explore an intervention method to reduce the mortality of alloxan diabetes model, and to preliminarily analyze the mechanism of alloxan induced animal death. Methods: Healthy New Zealand rabbits were randomly divided into injection group, control group, experimental group and blank group. The single injection group was injected with 100 mg/kg alloxan once. The control group was given 5% glucose solution and 100 mg/kg alloxan was injected in two times. The experimental group was given 5% glucose solution orally, 100 mg/kg alloxan, 7 mL 0.9% NaCl intravenously and 5 mL 5% glucose intraperitoneally immediately, and blood glucose was continuously monitored, 10 mL 5% glucose intravenously and 10 mL 5% glucose intraperitoneally every 4 h in the hypoglycemic stage. The blank group does nothing. Liver and kidney tissues at different time periods were stained with HE and organ index was evaluated. Results: 1) A single injection of 100 mg/kg alloxan without any intervention resulted in 100% mortality. Before modeling, oral administration of 5% glucose solution, divided into two injections of 100 mg/kg alloxan, mortality reached 100%;A single injection of 100 mg/kg alloxan and continuous intervention of normal saline and glucose for 20 h can significantly reduce the mortality of alloxan induced diabetic rabbit model. 2) Liver and kidney tissues were damaged in different degrees at different time periods, and liver and kidney indexes were significantly increased after alloxan injection compared with the normal group, with statistical significance (P > 0.05). Conclusion: 1) Every 4 hours of hypoglycemia, 10 ml 5% glucose was injected intravenously 10 ml 5% glucose intraperitoneally. It can reduce the death rate of alloxan diabetic rabbit model and shorten the time of blood glucose measurement. 2) After the injection of alloxan, acute lesions of liver and kidney may occur in different degrees, or one of the causes of acute death of experimental animals.

Intraplantar Injection of Monosodium Iodoacetate Produces Hyperalgesia in Rats and the Mechanism Underlying the Effect

Objective: To observe the effect of intraplantar injection of monosodium iodoacetate (MIA) on pain perception in rats and to investigate the role of transient receptor potential vanilloid type 1 (TRPV1) in the effect. Methods: Adult male Wistar rats were used in the experiment. 1) MIA was injected subcutaneously into the right hindpaw of rats, the low, medium, and high doses of MIA were 0.11, 0.33, and 1 mg, respectively, then the changes of paw withdrawal thermal latency, paw withdrawal mechanical threshold, and dynamic weight bearing within 4 hours after MIA injection were measured. 2) Capsazepine (TRPV1 antagonist, 30 μg) was injected subcutaneously into the right hindpaw of rats at 2 hours after intraplantar injection of MIA (1 mg), then the changes of paw withdrawal thermal latency, paw withdrawal mechanical threshold, and dynamic weight bearing within 1 hour after capsazepine injection were measured. Results: 1) The paw withdrawal thermal latency, paw withdrawal mechanical threshold, and dynamic weight bearing decreased after intraplantar injection of MIA in rats and the effect lasted for at least 4 hours. 2) The MIA-induced reduction in paw withdrawal thermal latency, paw withdrawal mechanical threshold, and dynamic weight bearing were significantly alleviated after intraplantar injection of capsazepine in rats. Conclusion: Intraplantar injection of MIA can produce thermal pain, mechanical pain, and spontaneous pain for more than 4 hours, which may be due to the TRPV1 activation caused by MIA.

Hyperspectral image super resolution using deep internal and self-supervised learning

Correlation between TGFβ1 Gene Polymorphism and Asthma in Baise, Guangxi Children

Objective: This research was to study the correlation between the rs1800469, rs1800470, rs2241712, rs224171 and rs4803455 of TGFβ1 gene and asthma in Baise, Guangxi children. This research also studied the relationship between serum concentration of TGFβ1 and childhood asthma. Method: From June 2022 to December 2023, 121 children had physical examination in affiliated Hospital of Youjiang Medical University for nationalities were selected as control group and 118 children suffered from asthma in affiliated Hospital of Youjiang Medical University for nationalities during the same period were selected as asthma group. Result: There was no correlation between rs1800469, rs1800470, rs2241712, rs2241715, rs4803455 and asthma in Baise, Guangxi children. Linkage disequilibrium analysis showed that there were strong linkage disequilibrium among rs1800469, rs1800470, rs2241712, rs2241715 and rs4803455. Their haplotypes had no significant correlation with childhood asthma. The serum concentration of TGFβ1 in asthma group was lower than that in control group (p β1 had no significant relationship with the genotypes of rs1800469, rs1800470, rs2241712, rs2241715 and rs4803455.

Intranasal administration of stem cell-derived exosomes for central nervous system diseases

Exosomes, lipid bilayer-enclosed small cellular vesicles, are actively secreted by various cells and play crucial roles in intercellular communication. These nanosized vesicles transport internalized proteins, mRNA, miRNA, and other bioactive molecules. Recent findings have provided compelling evidence that exosomes derived from stem cells hold great promise as a therapeutic modality for central nervous system disorders. These exosomes exhibit multifaceted properties including antiapoptotic, anti-inflammatory, neurogenic, and vasculogenic effects. Furthermore, exosomes offer several advantages over stem cell therapy, such as high preservation capacity, low immunogenicity, the ability to traverse the blood-brain barrier, and the potential for drug encapsulation. Consequently, researchers have turned their attention to exosomes as a novel therapeutic avenue. Nonetheless, akin to the limitations of stem cell treatment, the limited accumulation of exosomes in the injured brain poses a challenge to their clinical application. To overcome this hurdle, intranasal administration has emerged as a non-invasive and efficacious route for delivering drugs to the central nervous system. By exploiting the olfactory and trigeminal nerve axons, this approach enables the direct transport of therapeutics to the brain while bypassing the blood-brain barrier. Notably, exosomes, owing to their small size, can readily access the nerve pathways using this method. As a result, intranasal administration has gained increasing recognition as an optimal therapeutic strategy for exosomebased treatments. In this comprehensive review, we aim to provide an overview of both basic and clinical research studies investigating the intranasal administration of exosomes for the treatment of central nervous system diseases. Furthermore, we elucidate the underlying therapeutic mechanisms and offer insights into the prospect of this approach.

Muscle Mass Is Associated with Depression in Adolescents and Young Adults

Background and Aims: The study aims to explore the association between body composition and depression in adolescents and young adults. Methods: A random cluster sampling method was used to select 41 age-matched indi-viduals (aged 11 - 30) as a non-depressive control group. The Department of Psychology at the Affiliated Hospital of Youjiang Medical University for Na-tionalities recruited 27 depressive patients within the same age range. Bioelec-trical impedance analysis measured body composition, while SCL-90, SAS, and SDS were employed for depression diagnosis. Logistic regression analysis was conducted to assess the relationship between body composition and de-pression scores. Results: The Depression Group exhibited higher levels of fat mass (p Conclusion: Depression is associated with age, gender, and muscle mass in adolescents and young people. Proper exercise and a healthy diet can increase muscle mass, thereby reducing fat mass and, as a result, reducing the prevalence of depression.

Aiming at the triangular fuzzy(TF)multi-attribute decision making(MADM)problem with a preference for the distribution density of attribute(DDA),a decision making method with TF number two-dimensional density(TFTD)operator is proposed based on the density operator theory for the decision maker(DM).Firstly,a simple TF vector clustering method is proposed,which considers the feature of TF number and the geometric distance of ***,the least deviation sum of squares method is used in the program model to obtain the density weight ***,two TFTD operators are defined,and the MADM method based on the TFTD operator is ***,a numerical example is given to illustrate the superiority of this method,which can not only solve the TF MADM problem with a preference for the DDA but also help the DM make an overall comparison.

Bioinformatics Analysis of the Biological Properties of Ewing Sarcoma

Purpose: Bioinformatics-based approach to screen and analyze differentially expressed genes associated with the biological characteristics of Ewing sarcoma. Means: The GSE17674 dataset was selected for analysis, obtained by data retrieval based on the GEO public database. The R language limma toolkit was used to screen DEmRNAs. After the data were normalized, the Metascape online analysis software and the R language clusterProfiler package were used to analyze the GO function and KEGG pathway enrichment of DEmRNAs lines, respectively. The string database was selected for PPI analysis, and the results were imported into Cytoscape software to derive the core modules and predicted core genes. The genes selected above were analyzed for tissue localization specificity. Results: Through the analysis of GSE17674, differentially expressed genes were screened out, and GO and KEGG analyses were performed on the differentially expressed genes. The GO functional enrichment analysis was mainly enriched in the process of muscle system, muscle contraction, myocyte development, contractile fibers, myogenic fibers, myofibers, myofibrillar segments, actin binding, structural composition of muscle, and actin filament binding. KEGG pathway analysis showed that the core pathways associated with the development of ES were the core genes for myocardial contraction, congestive cardiomyopathy, and hypertrophic cardiomyopathy. Five Hub genes were obtained based on Cytoscape prediction. Tissue localization specificity analysis of Hub genes was performed, and a total of 2 Hub genes with tissue specificity were screened;MYH6 was specifically expressed in cardiac cells and MYL1 was specifically expressed in skeletal muscle cells. Conclusions: The differential genes screened will help to understand the molecular mechanisms underlying the highly invasive and metastasis-prone biological characteristics of ES, as well as provide new ideas for clinical drug-targeted treatment of ES.

Non-Kramers doublet ground state in a quaternary cubic compound PrRu_(2)In_(2)Zn_(18) investigated by ultrasonic measurements

We performed ultrasonic measurements on a quaternary cubic compound PrRu_(2)In_(2)Zn_(18) to explore the ground state properties derived from non-Kramers Γ_(3) doublet of Pr^(3+).PrRu_(2)In_(2)Zn_(18) is a quaternary derivative of the ternary compound PrRu_(2)Zn_(20) that exhibits a structural phase transition at T_S=138 *** PrRu_(2)In_(2)Zn_(18),the Zn atoms at the 16c site in PrRu_(2)Zn_(20) are selectively replaced by In atoms.A monotonic increase was observed in the temperature dependence of elastic constants C_L=(C_(11)+2C_(12)+4C_(44))/3 and C_(T)=(C_(11)-C_(12)+C_(44))/3 in the temperature range around T_(S) to which an elastic softening was observed in(C_(11)-C_(12))/2 for PrRu_(2)Zn_(20).The disappearance of the softening indicates that the structural transition in PrRu_(2)Zn_(20) is suppressed by the substitution of Zn ions by In ones with a larger ionic ***,the C_(T) of PrRu_(2)In_(2)Zn_(18) exhibits a precursor Curie-type elastic softening toward low temperatures being responsible for the non-Kramers Γ_(3) ground *** discuss the ground state and the evolution of the elastic properties of the different single-crystal samples of PrRu_(2)In_(2)Zn_(18) grown under different conditions.
