Chilling Injury, physicochemical Properties, and Antioxidant Enzyme Activities of Red Pitahaya (Hylocereus polyrhizus) Fruits under Cold Storage Stress

Low-temperature storage is extensively used to optimize the postharvest life of various fresh ***,red pitahaya(Hylocereus polyrhizus)fruits are sensitive to chilling injury(CI),which leads to the limitation of low-temperature *** this study,red pitahaya fruits were stored at 2,4,6,8,and 10℃,respectively,for 27 days to determine the appropriate storage *** the storage of red pitahaya fruits,storage at 8℃ was more effective in suppressing decay and maintaining quality than other low ***(2,4,and 6℃)storage decreased weight loss(WL)and maintained higher content of titratable acidity(TA),soluble sugars(SS),and total phenolics(TP)but different degrees of CI were *** CI was observed at 8℃ and 10℃.Red pitahay as stored at 8 and 10℃ were associated with better color evaluation,lower electrolyte leakage(EL),respiration rate,and lipoxygenase(LOX)activity,and higher fruit firmness,superoxide dismutase(SOD)activity,and catalase(CAT)***,higher storage temperature(10℃)resulted in higher metabolic activity leading to lower quality and antioxidant capacities compared with 8℃.Therefore,our results demonstrated that red pitahaya stored at 8℃ exhibited a protective effect on fruit quality and resisted CI development during storage.

Background and Objective:LTB4 has been shown to be involved in rheumatoid arthritis(RA)*** effect of Dioscin(Dio)on the LTB4 pathway of RA have not been reported *** study aimed at further exploring whether Dioscin’s effects on TNF-αinduced collagen-induced arthritis(CIA)rat fibroblast-like synoviocytes(FLS)connected with the LTB4 and its receptor ***&Methods:In this experiment,control group,TNF-αgroup,and different concentrations of Dioscin groups were *** viability was evaluated using MTT *** levels of LTB4 in the samples of above groups were measured using *** mRNA expression levels of LTA4H,BLT1,and BLT2 were detected by quantitative real time PCR,while the expression level of LTA4H proteins were detected using western *** distribution of LTA4H was assessed by immunofluorescence ***:the LTB4 level of TNF-αgroup in sample supernatant was higher than both control group and Dioscin groups with decreased LTB4 levels(p<0.05).Compared with the control group,the expression of LTA4H was significantly increased in TNF-αgroup(p<0.05),whereas LTA4H expressions were significantly decreased in all Dioscin groups when compared to TNF-αgroup(p<0.05).The mRNA expressions of BLT1 and BLT2 were markedly higher in TNF-αgroup than those in control group while Dioscin treatment significantly inhibited the increased expressions of BLT1 and BLT2 induced by TNF-α(p<0.05).Conclusions:These results firstly demonstrate that the protective effect of Dioscin on TNF-αinduced FLS may involve in its reducing LTB4 production by down-regulating LTA4H expression,and may inhibit its downstream pathway by decreasing LTB4 receptors *** findings suggest that dioscin produces a potential therapeutic effects for RA via its influencing LTA4H/LTB4/BLT pathway.

Gel-Based Chemical Cross-Linking Analysis of 20S Proteasome Subunit-Subunit Interactions in Breast Cancer

The ubiquitin-proteasome system plays a pivotal role in breast tumorigenesis by controlling transcription factors, thus promoting cell cycle growth, and degradation of tumor suppressor proteins. However, breast cancer patients have failed to benefit from proteasome inhibitor treatment partially due to proteasome heterogeneity, which is poorly understood in malignant breast neoplasm. Chemical crosslinking is an increasingly important tool for mapping protein three-dimensional structures and proteinprotein interactions. In the present study, two cross-linkers, bis（sulfosuccinimidyl） suberate（BS3） and its water-insoluble analog disuccinimidyl suberate（DSS）, were used to map the subunit-subunit interactions in 20 S proteasome core particle（CP） from MDA-MB-231 cells. Different types of gel electrophoresis technologies were used. In combination with chemical cross-linking and mass spectrometry, we applied these gel electrophoresis technologies to the study of the noncovalent interactions among 20 S proteasome subunits. Firstly, the CP subunit isoforms were profiled. Subsequently, using native/SDSPAGE, it was observed that 0.5 mmol/L BS^3 was a relatively optimal cross-linking concentration for CP subunit-subunit interaction study. 2-DE analysis of the cross-linked CP revealed that α1 might preinteract with α2, and α3 might pre-interact with α4. Moreover, there were different subtypes of α1α2 and α3α4 due to proteasome heterogeneity. There was no significant difference in cross-linking pattern for CP subunits between BS3 and DSS. Taken together, the gel-based characterization in combination with chemical cross-linking could serve as a tool for the study of subunit interactions within a multi-subunit protein complex. The heterogeneity of 20 S proteasome subunit observed in breast cancer cells may provide some key information for proteasome inhibition strategy.

Sequencing of Mixed Model Assembly Lines Based on Improved Shuffled Frog Leaping Algorithm

Shuffled frog leaping algorithm( SFLA) was used to solve multi-objective sequencing problem of mixed model assembly line( MMAL). Local convergence can be avoided and optimal solution can be obtained to a certain extent. However,the multi-objective sequencing problem of MMAL is an non-deterministic polynomial hard( NP-hard) problem and the shortcomings are slow convergence rate and low precision. To solve the shortcomings for optimization objectives of minimizing total utility time and keeping average consumption rate of parts, a chaos differential evolution SFLA( CDESFLA) is proposed in this study. Because SFLA is easy to fall into local optimum,the evolution operator of differential evolution algorithms is introduced in SFLA as a local search strategy,and differential mutation operator is introduced in chaotic sequence to prevent premature convergence. The examples show that the proposed CDESFLA is better for convergence accuracy than SFLA,genetic algorithm( GA) and particle swarm optimization( PSO)

Characterization of grass carp FosB, Fosl2, JunD transcription factors in response to GCRV infection

Activating protein-1(AP-1)composed of Jun and Fos family proteins,which are frequently activated by pathogens to induce cellular inflammation and immune *** this study,by using RNA sequencing(RNA-seq)technology,we found that FosB,Fosl2,and JunD expression was co-induced 12 and 20 h in grass carp(Ctenopharyngodon idella)kidney cell line(CIK cells)after grass carp reovirus(GCRV)***-length cDNA of the three genes were cloned and phylogenetic analysis indicated that the corresponding proteins shared high homology with those in higher *** tissue distribution analysis of the three genes showed they were ubiquitously expressed in various tissues of healthy grass *** infection assay,FosB,Fosl2,and JunD were up-regulated and co-expressed 8 h after GCRV infection in CIK *** interaction and co-expression network analysis revealed that FosB,Fosl2,and JunD showed similar functions and were co-expressed in mammals and *** together,these results indicated that the co-expression of the AP-1 family proteins in grass carp uniformly responds to GCRV *** findings provided insights into the further study of AP-1 transcription factors function during grass carp reovirus infection.

This paper presents an approach to train junior Windows software programmers for making software more appealing to students. This approach proposes to separate the software development technologies into three layers, which are the language layer, the application layer and the project layer. For each layer, based on the idea of PBL, this paper gives 20,12 and 1 projects to make the course interesting and easy to understand. Each student must finish a project named as LMS at the end of the course which contains many main technologies of Windows programming. According to the academic results and student surveys, most of the students can finish this course successfully and make a progress in software programming.

Molecular and FISH analysis of 45S rDNA on BAC molecule of Saccharina japonica

IGS is abundant in polymorphism,which is widely used in the analysis of intraspecific genetic diversity and phylogenetic relationships among geographical *** this study,the 45S rDNA repeat unit of Saccharina japonica was obtained for the first time by BAC clone *** total length of the 45S rDNA repeat unit of *** was 8995 bp,including 5420 bp of 18S-5.8S-25S rDNA,and 3575 bp of IGS(Intergenic Spacer),with the GC content of 51.4%.The IGS was composed of a 465 bp of 3′-outer transcribed spacer(ETS),an 874 bp 5′-ETS,and a 2236 bp non-transcribed spacer(NTS),with the GC content of 50.1%.Fiber-FISH(fiber-fluorescence in situ hybridization)analysis of the distribution of 45S rDNA repeat units on the bacterial artificial chromosome illustrated that each fiber had at least five continuously moniliform hybridization signal *** study provided a new candidate molecular marker for detecting intraspecific polymorphisms of *** addition,the successful fiber-FISH analysis of the 45S rDNA on BAC molecule would contribute to the construction of the physical map and map-based cloning of this kelp.

Some Characteristics of Neurons in the Reticular Nucleus of the Thalamus (Preliminary Data)

Our general understanding of the function of neurons is that dendrites receive information that is relayed to the axon, where action potentials are initiated and propagated to eventually trigger neurotransmitter release at synaptic terminals. Although for a number of neuron types in the mammalian brain, many neuron types do not follow this classical polarity pattern. In fact, dendrites may be the site of action potentials initiation and propagation. It should be noted that convincing evidence has been obtained for the existence of dendritic action potentials in hippocampal and neocortical neurons. With regard to the dendrite potentials of thalamic neurons in general and specifically the reticular nucleus of the thalamus, it has not yet been reported. The results of this study demonstrate, for the first time, that generation of spike potentials of different amplitudes was observed in the activity of the thalamic reticular nucleus neurons. The generation of one action potential does not interfere with the generation of another, and a spike potential of smaller amplitude can occur at the ascending or descending phase of the spike potential of large amplitude. It can be argued that the spike potentials of lower amplitudes arising in the thalamic reticular nucleus neuron are of dendrite origin. Given both the strategic position and the functional purpose of the TRN, it can be assumed that the neurons of this structure must each time be discharged with spike potentials in order to carry out their modulating effect on other areas of the nervous system of the brain without leakage.

vCourse: A Course Platform Based on Virtualization Technology

This paper presents a platform named vCourse based on the virtualization technology. vCourse supports multiple operation systems and multiple courses online. Teachers can create a course template and deploy it as many virtual machines. Students can do their exercises on those virtual machines and interact with others and the teacher. The features, structure and key mechanisms of vCourse are discussed in this paper. Two experiments are designed to evaluate the performance of vCourse. According to the test results, vCourse can run the typical software smoothly and is not very sensitive on the virtual machine numbers.
