Antitumor efficacy and potential mechanism of FAP-targeted radioligand therapy combined with immune checkpoint blockade

Radiotherapy combined with immune checkpoint blockade holds great promise for synergistic antitumor *** radionuclide therapy delivers radiation directly to tumor ***1004 is a fibroblast activation protein(FAP)-targeting radiopharmaceutical,conjugated with the albumin binder Evans Blue,which has demonstrated enhanced tumor uptake and retention in previous preclinical and clinical ***,we demonstrate that^(68)Ga/^(177)Lu-labeled LNC1004 exhibits increased uptake and prolonged retention in MC38/NIH3T3-FAP and CT26/NIH3T3-FAP tumor *** therapy with^(177)Lu-LNC1004 induced a transient upregulation of PD-L1 expression in tumor *** combination of^(177)Lu-LNC1004 and anti-PD-L1 immunotherapy led to complete eradication of all tumors in MC38/NIH3T3-FAP tumor-bearing mice,with mice showing 100%tumor rejection upon ***,single-cell RNA sequencing(scRNA-seq),and TCR sequencing revealed that combination therapy reprogrammed the tumor microenvironment in mice to foster antitumor immunity by suppressing malignant progression and increasing cell-to-cell communication,CD8^(+)T-cell activation and expansion,M1 macrophage counts,antitumor activity of neutrophils,and T-cell receptor diversity.A preliminary clinical study demonstrated that^(177)Lu-LNC1004 was well-tolerated and effective in patients with refractory ***,scRNA-seq of peripheral blood mononuclear cells underscored the importance of addressing immune evasion through immune checkpoint blockade *** was emphasized by the observed increase in antigen processing and presentation juxtaposed with T cell *** conclusion,our data supported the efficacy of immunotherapy combined with^(177)Lu-LNC1004 for cancer patients with FAP-positive tumors.

Precipitation patterns are vital to water resource management and hydrological research,especially in the upper reaches of inland rivers in arid and semiarid ***,estimating spatiotemporal precipitation patterns at a basin scale is challenging due to limited *** this study,spatiotemporal patterns of precipitation amount,frequency,duration,and intensity at different time scales from 2014 to 2019 are estimated using the Bayesian maximum entropy method in the Tianlaochi catchment of the Heihe River watershed,northwest *** study's results show that the annual average precipitation amount was 535.9 mm from 2014 to 2019,with precipitation amount between May and September accounting for 85.9%of the annual precipitation *** daily precipitation,the average frequency rate of light precipitation is highest at 59.55%,however,the average contribution rate of moderate precipitation is highest at 50.33%.The spatial distribution of precipitation is characterized by high-value areas concentrated in the central valley and low-value areas located at the catchment's *** most important driving factors of precipitation patterns are elevation,relative humidity,and wind *** outcomes can be used to establish accurate hydrological models in the catchment and provide support for water resource management in the Heihe River watershed.

Zircon U-Pb age and Sr-Nd-Hf isotope geochemistry of I-type granites in the Jiangda-Weixi region， Eastern Tibet: Implications for the tectonic evolution of the Paleo-Tethyan orogen

Permian–Triassic granitoids are widely distributed along the Jinshajiang suture belt， eastern Tibet， and are regarded as the result of the tectonic-magmatic activity associated with the evolution of Paleo-Tethys Ocean. This paper focuses on the high-K calc-alkaline I-type Rennong （～235 Ma） and Jiaduoling （～232 Ma） granitoid plutons， eastern Tibet. They are enriched in Light rare earth Elements （LREEs） and Large Ion Lithophile Elements （LILEs）， but depleted in High Field Strength Elements （HFSEs） with moderate-weak negative Eu anomalies （0.61–0.90）. The Rennong granites are characterized by uniform zircon εHf（t） values （-7.3 to -3.5） and negative εNd（t） values （-7.6 to -5.7）， with old two-stage Nd model ages (TDM2= 1.51–1.46 Ga) and was likely formed by partial melting of the basement rocks， whereas the Jiaduoling rock samples have variable zircon εHf（t） values （-5.7 to +5.5） but negative εNd（t） values （-7.6 to -7.7） and are proposed to be formed by hybridization of mantle-derived mafic magma， Rennong felsic magma and sediments. Mafic microgranular enclaves （MMEs） in the Jiaduoling granitic rocks， have similar zircon U-Pb ages （～237 Ma） and zircon εHf（t） values （-4.4 to +6.3） to the host rocks， indicating that zircons in the MMEs were actually xenocrysts that formed at an early stage in the granitic magma chamber. Finally， we proposed that the break-off of the Jinshajiang oceanic slab beneath Changdu–Simao Block in a post-collisional setting， resulted in the upwelling of asthenosphere in the Late Triassic， and then， generated the wide-spread intermediate-felsic magmatism along the Jinshajing belt， including the Rennong and Jiaduoling plutons.

Nitrate is a crucial environmental pollutant,and its risk on ecosystem keeps *** conversion of nitrate to ammonia can simultaneously achieve the commercialization of environmental hazards and recovery of valuable ammonia,which is green and sustainable for the ***,due to the thermodynamic and kinetic energy barriers,photocatalytic nitrate reduction usually involves a higher selectivity of the formation of nitrogen that largely limits the ammonia synthesis *** this work,we reported a green and facile synthesis of novel metallic ruthenium particle modified graphitic carbon nitride *** with bulk graphitic carbon nitride,the optimal sample had 2.93-fold photocatalytic nitrate reduction to ammonia activity(2.627 mg/h/gcat),and the NH3 selectivity increased from 50.77%to 77.9%.According to the experimental and calculated results,the enhanced photocatalytic performance is attributed to the stronger light absorption,nitrate adsorption,and lower energy barrier for the generation of *** work may provide a facile way to prepare metal modified photocatalysts to achieve highly efficient nitrate reduction to ammonia.

Ash-and-Slag Wastes of Energy Sector as Raw Material for Production of rare-earth Metals and Alumina

The results to develop a complex technology of co-extraction of germanium and gallium from the ash-and-slag wastes of coal combustion in Ukraine were presented. Based on the study of phase changes occurring in initial raw materials as a result of its processing, it was proposed to carry out a preliminary enrichment of the raw materials in order to produce secondary sublimates. Their further processing involves a combination of leaching soluble material with distillation of germanium in the form of tetrachloride. The resulting acidic solutions are trended to the gallium extraction. Optimum conditions of carrying out of processes are *** chemical and phase composition of the resulting dump products and solutions have been defined. On the basis of it ways of their processing for the purpose of reception of alumina and building materials are recommended.

Although population genetic theory is largely based on the premise that loci under study are selectively neutral, it has been acknowledged that the study of DNA sequence data under the influence of selection can be useful. In some circumstances, these loci show increased population differentiation and gene diversity. Highly polymorphic loci may be especially useful when studying populations having low levels of diversity overall, such as is often the case with threatened or newly established inva- sive populations. Using common starlings Sturnus vulgaris sampled from invasive Australian populations, we investigated se- quence data of the dopamine receptor D4 gene （DRD4）, a locus suspected to be under selection for novelty-seeking behaviour in a range of taxa including humans and passerine birds. We hypothesised that such behaviour may be advantageous when species encounter novel environments, such as during invasion. In addition to analyses to detect the presence of selection, we also esti- mated population differentiation and gene diversity using DRD4 data and compared these estimates to those from microsatellite and mitochondrial DNA sequence data, using the same individuals. We found little evidence for selection on DRD4 in starlings. However, we did find elevated levels of within-population gene diversity when compared to microsatellites and mitochondrial DNA sequence, as well as a greater degree of population differentiation. We suggest that sequence data from putatively non- neutral loci are a useful addition to studies of invasive populations, where low genetic variability is expected

Sensitivity to structure and composition is very challenging to establish in nanocatalysis due to inadequate definition of structures that are very close in *** synthesized a pair of atomically precise copper clusters that are very close in composition,[Cu_(20)H_(9)(Tf-dpf)_(10)]·BF4(Cu_(20)H_(9))and[Cu_(20)H_(8)(Tf-dpf)_(10)]·(BF_(4))_(2)(Cu_(20)H_(8)),by using a pyridyl-functionalized flexible amidinate ligand,N,N′-di(5-trifluoromethyl-2-pyridyl)*** one-hydride difference in their composition leads to significant variation in geometric and electronic structures and,consequently,distinctly different optical and catalytic ***_(20)H_(8)exhibits 25 times higher catalytic activity than Cu_(20)H_(9)(96.7%vs 3.7%in yield)in the selective hydrogenation of anα,β-unsaturated aldehyde(cinnamaldehyde)to saturated aldehyde(3-phenylpropanal).Electrospray ionization mass spectrometry combined with density functional theory calculations reveal that the greater ease of dissociation of one Tf-dpf ligand compared to Cu_(20)H_(8)is the key to its higher *** work demonstrates a clear case of structure and composition sensitivity in nanocatalysis and that one hydride,out of∼330 atoms in the nanoclusters,can make a huge difference in the catalytic *** insights will be useful in the design and synthesis of atomically precise nanocatalysts.
