Trend Analysis and Intervention Effect Starting Point Detection of COVID-19 Epidemics Using Recalibrated Time Series Models—Worldwide,2020

Objective:This study aimed to identify a model for short-term coronavirus disease 2019(COVID-19)trend prediction and intervention evaluation.Methods:We compared the autoregressive integrated moving average(ARIMA)model and Holt exponential smoothing(Holt)model on predicting the number of cumulative COVID-19 cases in China.Based on the mean absolute percentage error(MAPE)value,the optimal model was selected and further tested using data from the United States,Italy and Republic of Korea.The intervention effect starting time points and abnormal trend changes were detected by observing the pattern of differences between the predicted and real trends.Results:The recalibrated ARIMA model with a 5-day prediction time span has the best model performance with MAPEs ranged between 2%and 5%.The intervention effects started to show on February 7 in the mainland of China,March 5 in Republic of Korea and April 27 in Italy,but have not been detected in the US as of May 19.Temporary abnormal trends were detected in Korea and Italy,but the overall epidemic trends were stable since the effect starting points.Conclusion:The recalibrated ARIMA model can detect the intervention effects starting points and abnormal trend changes;thus to provide valuable information support for epidemic trend analysis and intervention evaluation.

Association between Serum Uric Acid and the Early Marker of Kidney Function Decline among Chinese Middle-Aged and Older Population:Evidence from the China health and Retirement Longitudinal Study

Objective To evaluate the association between serum uric acid(SUA)and kidney function decline.Methods data was obtained from the China health and Retirement Longitudinal Study on the Chinese middle-aged and older population for analysis.The kidney function decline was defined as an annual estimated glomerular filtration rate(e GFR)decrease by>3 mL/min per 1.73 m^(2).Multivariable logistic regression was applied to determine the association between SUA and kidney function decline.The shape of the association was investigated by restricted cubic splines.Results A total of 7,346 participants were included,of which 1,004 individuals(13.67%)developed kidney function decline during the follow-up of 4 years.A significant dose-response relation was recorded between SUA and the kidney function decline(OR 1.14,95%CI 1.03-1.27),as the risk of kidney function decline increased by 14%per 1 mg/d L increase in SUA.In the subgroup analyses,such a relation was only recorded among women(OR 1.22,95%CI 1.03-1.45),those aged5 mg/dL was associated with a significantly higher risk of kidney function decline.Conclusion The SUA level was associated with kidney function decline.An elevation of SUA should therefore be addressed to prevent possible kidney impairment and dysfunction.

EpiMix:A novel method to estimate effective reproduction number

Transmission potential of a pathogen,often quantified by the time-varying reproduction number R t,provides the current pace of infection for a disease and indicates whether an emerging epidemic is under control.In this study,we proposed a novel method,EpiMix,for R t estimation,wherein we incorporated the impacts of exogenous factors and random effects under a Bayesian regression framework.Using Integrated Nested Laplace Approx-imation,EpiMix is able to efficiently generate reliable,deterministic R t estimates.In the simulations and case studies performed,we further demonstrated the method's robust-ness in low-incidence scenarios,together with other merits,including its flexibility in selecting variables and tolerance of varying reporting rates.All these make EpiMix a potentially useful tool for real-time R t estimation provided that the serial interval distri-bution,time series of case counts and external influencing factors are available.

On March 11,2020,the World health Organization(WHO)formally declared coronavirus disease 2019(COVID-19)a worldwide pandemic as the virus spreads rapidly with new cases and deaths rising exponentially in many countries.As of March 12,there were 125,048 confirmed cases and 4,613 deaths,and the numbers are still surging affecting 118 countries(1).Now we know that regional efforts to contain individual outbreaks have failed.The next phase of epidemic control is mitigation,and China has implemented multiple effective measurements such as mandatory citywide lockdowns to isolate and block the spread since January 2020.Recent data has shown evidence of controlling the epidemic(2).

Summary What is already known about this topic?Reducing population mobility and increasing the vaccination rate for severe acute respiratory syndrome coronavirus 2 can decrease the transmission of coronavirus disease 2019(COVID-19).What is added by this report?In order to reduce the incidence of COVID-19 to the levels of influenza after restoring normal mobility,the efficacy against infection needs to be increased to 40%and the efficacy against symptomatic disease needs to be increased to 90%.The efficacy against infection has a more important impact compared to efficacy against symptomatic disease or death on the transmission of COVID-19 at the population level.What are the implications for public health practice?The population should continue maintaining nonpharmaceutical interventions and minimize international movement to prevent transmission of COVID-19.Furthermore,developing new vaccines or promoting booster vaccinations should be considered to increase efficacy.

SNX14 deficiency-induced defective axonal mitochondrial transport in Purkinje cells underlies cerebellar ataxia and can be reversed by valproate

Loss-of-function mutations in sorting nexin 14(SNX14) cause autosomal recessive spinocerebellar ataxia20, which is a form of early-onset cerebellar ataxia that lacks molecular mechanisms and mouse models. We generated Snx14-deficient mouse models and observed severe motor deficits and cell-autonomous Purkinje cell degeneration. SNX14 deficiency disrupted microtubule organization and mitochondrial transport in axons by destabilizing the microtubule-severing enzyme spastin, which is implicated in dominant hereditary spastic paraplegia with cerebellar ataxia, and compromised axonal integrity and mitochondrial function. Axonal transport disruption and mitochondrial dysfunction further led to degeneration of highenergy-demanding Purkinje cells, which resulted in the pathogenesis of cerebellar ataxia. The antiepileptic drug valproate ameliorated motor deficits and cerebellar degeneration in Snx14-deficient mice via the restoration of mitochondrial transport and function in Purkinje cells. Our study revealed an unprecedented role for SNX14-dependent axonal transport in cerebellar ataxia, demonstrated the convergence of SNX14 and spastin in mitochondrial dysfunction, and suggested valproate as a potential therapeutic agent.

We present angle-uniform parallel coordinates,a data-independent technique that deforms the image plane of parallel coordinates so that the angles of linear relationships between two variables are linearly mapped along the horizontal axis of the parallel coordinates plot.Despite being a common method for visualizing multidimensional data,parallel coordinates are ineffective for revealing positive correlations since the associated parallel coordinates points of such structures may be located at infinity in the image plane and the asymmetric encoding of negative and positive correlations may lead to unreliable estimations.To address this issue,we introduce a transformation that bounds all points horizontally using an angleuniform mapping and shrinks them vertically in a structure-preserving fashion;polygonal lines become smooth curves and a symmetric representation of data correlations is achieved.We further propose a combined subsampling and density visualization approach to reduce visual clutter caused by overdrawing.Our method enables accurate visual pattern interpretation of data correlations,and its data-independent nature makes it applicable to all multidimensional datasets.The usefulness of our method is demonstrated using examples of synthetic and real-world datasets.

Neural regeneration is a great clinical challenge in both the central nervous system(CNS)and the peripheral nervous system(PNS).Degeneration in the CNS caused by disease(such as Alzheimer's or Parkinson's diseases)or trauma(such as traumatic brain injury and spinal cord injury)often results in permanent paralysis and loss of sensation.The spontaneous neural regeneration in the CNS is generally unsuccessful,while the PNS has an intrinsic regenerative ability.However,PNS regeneration occurs over relatively short distances.Damage to the PNS results in the motor and cognitive impairment in many cases.Biomaterials in the form of scaffolds or nanoparticles for neuroprotection and neuroregeneration have attracted much attention.Nanoparticles can be fabricated using various lioids.

Suppression of laser-induced choroidal neovascularization by intravitreal injection of tristetraprolin

AIM: To examine the effect of intravitreal adenoviral vector-mediated tristetraprolin(Ad-TTP) on VEGF m RNA expression in a rat model of laser-induced choroidal neovascularization.METHODS: Ad-TTP was prepared using a commercial kit. Retinal laser-induced photocoagulation(10 spots per eye) was performed on rats in this experimental choroidal neovascularization(CNV) model. Rats were divided into four groups: control(single intravitreal injection of balanced salt solution, n =10), laser-induced CNV(photocoagulation only, n =20), laser-induced CNV plus Ad-TTP injection(photocoagulation plus a single intravitreal Ad-TTP injection, n =20) and Ad-TTP injection only(n =10). Changes in choroidal morphology were evaluated in ten rats in the laser only and the laser plus Ad-TTP groups. Two weeks after laser injury, the size of CNV was calculated by perfusion with high-molecular-weight fluorescein isothiocyanate(FITC)-dextran. VEGF m RNA expression in retina-choroid tissue from ten rats in each group was measured by reverse transcription polymerase chain reaction(RT-PCR). RESULTS: Two weeks after treatment, the area of laser-induced CNV was reduced by approximately 60% in the rats given the Ad-TTP injection compared with that in the laser-only group. There was a tendency toward decreased VEGF m RNA expression in the Ad-TTP injection groups.CONCLUSION: A single intravitreal injection of Ad-TTP significantly suppressed CNV size in this experimental laser-induced CNV model. Ad-TTP injection also decreased VEGF m RNA expression compared with that inthe laser-induced CNV group. The present study is meaningful as the first study to investigate the effect of tristetraprolin delivered via intravitreal injection.

Human pluripotent stem cells:Towards therapeutic development for the treatment of lifestyle diseases

There are two types of human pluripotent stem cells: Embryonic stem cells(ESCs) and induced pluripotent stem cells(iPSCs),both of which launched themselves on clinical trials after having taken measures to overcome problems: Blocking rejections by immunosuppressants regarding ESCs and minimizing the risk of tumorigenicity by depleting exogenous gene components regarding iP SCs.It is generally assumed that clinical applications of human pluripotent stem cells should be limited to those cases where there are no alternative measures for treatments because of the risk in transplanting those cells to living bodies.Regarding lifestyle diseases,we have already several therapeutic options,and thus,development of human pluripotent stem cell-based therapeutics tends to be avoided.Nevertheless,human pluripotent stem cells can contribute to the development of new therapeutics in this field.As we will show,there is a case where only a short-term presence of human pluripotent stem-derived cells can exert long-term therapeutic effects even after they are rejected.In those cases,immunologically rejections of ESC-or allogenic iP SC-derived cells may produce beneficial outcomes by nullifying the risk of tumorigenesis without deterioration of therapeutic effects.Another utility of human pluripotent stem cells is the provision of an innovative tool for drug discovery that are otherwise unavailable.For example,clinical specimens of human classical brown adipocytes(BAs),which has been attracting a great deal of attention as a new target of drug discovery for the treatment of metabolic disorders,are unobtainable from living individuals due to scarcity,fragility and ethical problems.However,BA can easily be produced from human pluripotent stem cells.In this review,we will contemplate potential contribution of human pluripotent stem cells to therapeutic development for lifestyle diseases.
