African swine fever(ASF)is a devastating infectious disease in swine that is severely threatening the global pig *** efficacious vaccine is urgently ***,we used the Chinese ASFV HLJ/18 as a backbone and generated a series of genedeleted *** virulence,immunogenicity,safety,and protective efficacy evaluation in specific-pathogen-free pigs,commercial pigs,and pregnant sows indicated that one virus,namely HLJ/18-7GD,which has seven genes deleted,is fully attenuated in pigs,cannot convert to the virulent strain,and provides complete protection of pigs against lethal ASFV *** study shows that HLJ/-18-7GD is a safe and effective vaccine against ASFV,and as such is expected to play an important role in controlling the spread of ASFV.

African swine fever(ASF)continues to cause enormous economic loss to the global pig *** there is no safe and effective vaccine,accurate and timely diagnosis of ASF is essential to implement control *** immunofluorescence assay(IFA)is a gold standard serological method recommended by the World Organization for animal Health(WOAH).In this study,we used primary fetal kidney cells to establish a wild boar cell line(BK2258)that supported the efficient replication of ASF virus(ASFV)SD/DY-I/21 and showed visible cytopathic effect(CPE).Moreover,using BK2258,we established a sensitive and specific IFA for ASFV antibody *** standardize and evaluate the performance of this assay,we used serum samples from pigs infected with the low virulent genotype I SD/DY-I/21 and genotype II HLJ/HRB1/20,and immunized with the vaccine candidate HLJ/18-7GD,field samples,and negative serum *** IFA reacted with the ASFV-positive sera and displayed bright fluorescence *** was no non-specific green fluorescence due to cellular senescence or other cell damage-causing *** to a commercial indirect enzyme-linked immunosorbent assay(iELISA),ASFV antibodies were detected 1–4 days earlier using our *** detection limits of the IFA and iELISA for the same ASFV-antibody positive serum samples were 1:25,600 and 1:6,400,respectively,indicating that the IFA is more sensitive than *** newly established IFA was highly specific and did not cross-react with sera positive for six other important porcine pathogens(i.e.,Classical swine fever virus(CSFV),Porcine reproductive and respiratory syndrome virus(PRRSV),Porcme circovirus type 2(PCV2),Pseudorabies virus(PRV),Foot-and-Mouth disease virus type O(FMDV/O),and Porcine epidemic diarrhea virus(PEDV)).This study thus provides a sensitive,specific,and reliable detection method that is suitable for the serological diagnosis of ASF.

African swine fever(ASF) was first discovered and reported in East Africa in 1921 and it has been listed as a notifiable animal disease by the World Organization for animal Health(WOAH) due to the high morbidity and mortality(WOAH 2019). African swine fever virus(ASFV) can persist in pig farm environments, carcasses, and various pig products(Petrini et al. 2019). ASF has caused significant economic losses to the pig industry since its outbreak in China in 2018(Zhao et al. 2019).

African swine fever virus(ASFV)has been circulating in China for more than two years,and it is not clear whether the biological properties of the virus have ***,we report on our surveillance of ASFVs in seven provinces of China,from June to December,2020.A total of 22 viruses were isolated and characterized as genotype II ASFVs,with mutations,deletions,insertions,or short-fragment replacement occurring in all isolates compared with Pig/HLJ/2018(HLJ/18),the earliest isolate in *** isolates had four different types of natural mutations or deletion in the EP402R gene and displayed a nonhemadsorbing(non-HAD)*** isolates were tested for virulence in pigs;two were found to be as highly lethal as HLJ/***,two non-HAD isolates showed lower virulence but were highly transmissible;infection with 106 TCID50 dose was partially lethal and caused acute or sub-acute disease,whereas 103 TCID50 dose caused non-lethal,sub-acute or chronic disease,and persistent *** emergence of lower virulent natural mutants brings greater difficulty to the early diagnosis of ASF and creates new challenges for ASFV control.

prevention of African swine fever,a disease caused by African swine fever virus(ASFV),requires maintenance of high biosecurity standards,which principally relies on *** the perfect disinfectant against ASFV is difficult because of the lack of relevant ***,we aimed to find the most effective disinfectant and to optimise its concentration as well as contact time to confirm the viricidal effect against ASFV in *** evaluated the viricidal activity of three concentrations each of six common disinfectants against ASFV using immersion disinfection assay(IDA)and spray disinfection assay(SDA);the concentrations of these disinfectants at which complete viral inactivation occurred were almost same as the manufacturer-recommended concentrations,but the exposure times for viral inactivation are *** following disinfectants(assay:concentration,exposure time)showed complete inactivation:iodine and acid mixed solution(IDA/SDA:0.5%,10 min);compound potassium peroxymonosulfate(IDA:0.25%,30 min;SDA:0.25%,60 min);citric acid(IDA:0.25%,60 min;SDA:0.5%,60 min);sodium dichloroisocyanurate(IDA:0.125%,60 min;SDA:0.25%,60 min);and glutaral ang deciquam(IDA/SDA:0.2%,60 min);and deciquam(IDA/SDA:0.5%,60 min).However,in the presence of organic material contamination,disinfectants did not show a marked inactivation ***,disinfection procedures should be performed in two steps:thorough mechanical cleaning followed by application of *** conclusion,all the tested disinfectants can inactivate ASFV;these can be used as alternative disinfectants to enhance biosecurity.

African swine fever(ASF) is a hemorrhagic disease caused by African swine fever virus(ASFV), which belongs to the Asfarviridae family. ASF has become prevalent in Africa since it was first reported in Kenya in 1921(Rowlands et al. 2008;Costard et al. 2009). In 1957, it was introduced to Portugal in Europe, after which ASFV rapidly spread to other European countries and has already been eradicated in several countries except Sardinia(Mur et al. 2016). In 2007, ASF was introduced to Georgia and has continued its spread to other countries in Europe(Smietanka et al. 2016;Kolbasov et al. 2018;Garigliany et al. 2019;Linden et al. 2019). In China, an ASF outbreak was first reported on August 3, 2018 in a pig farm in Shenyang, Liaoning Province(Zhou et al. 2018). Since then, the dramatic spread of ASF throughout China has caused huge economic losses to the swine industry(Jiang et al. 2021).
