Cytochrome P450 enzymes mediated by DNA methylation is involved in deoxynivalenol-induced hepatoxicity in piglets

Deoxynivalenol(DON)is an inevitable contaminant in animal feed and can lead to liver damage,then decreasing appetite and causing growth retardation in *** many molecular mechanisms are related to hepatoxicity caused by DON,few studies have been done on cytochrome P450(CYP450)enzymes and DNA *** explore the role of CYP450 enzymes and DNA methylation in DONinduced liver injury,male piglets were fed a control diet,or diet containing 1.0 or 3.0 mg/kg DON for 4 *** significantly raised the activity of aspartate aminotransferase(AST),alanine aminotransferase(ALT),and glutamyl transpeptidase(GGT)(P<0.01),leading to liver *** vivo study found that DON exposure increased the expression of CYP450 enzymes(such as CYP1A1,CYP2E1,CYP3A29)(P<0.05),and disturbed the expression of nicotinamide N-methyltransferase(NNMT),galanin-like peptide(GALP)and insulin-like growth factor 1(IGF-1)(P<0.05),in which DNA methylation affected the expression of these *** vitro study(human normal hepatocytes L02)further proved that DON elevated the expression of CYP1A1,CYP2E1 and CYP3A4(P<0.05),and inhibited cell growth in a dose-dependent manner,resulting in cell *** importantly,knockdown of CYP1A1 or CYP2E1 could alleviate DON-induced growth inhibition by promoting IGF-1 *** together,increased CYP450 enzymes expression was one of the mechanisms of hepatoxicity and growth inhibition induced by DON,suggesting that the decrease of CYP450 enzymes can antagonize the hepatoxicity in animals,which provides some value for animal feed safety.

Transient Stability Preventive Control Based on Graph Convolution Neural Network and Transfer Deep Reinforcement Learning

This study proposes a new transient-stability preventive control (TSPC) method based on graph convolutional neural networks (GCNN) and transfer deep reinforcement learning (DRL) to address non-convergence problems of traditional optimization algorithms and slow training speed of artificial intelligence algorithms for TSPC. First, a transient stability assessor (TSA) with GCNN is developed to assess current-power-flow state. Sensitivities of the transient-stability index relative to the generators are approximately calculated using TSA;generators with significant influence able to narrow action space are identified. Subsequently, the Markov decision-making process of TSPC is derived by introducing the process of TSPC. A DRL for TSPC is constructed by adding entropy to twin delayed deep deterministic policy gradient (TD3). Knowledge learned by TSA is transferred to DRL based on transfer learning, which improves learning efficiency. Finally, case studies based on the IEEE 39-bus system and an actual power grid prove the effectiveness of the proposed method. Comparisons performed with reference algorithms in literature demonstrate the proposed method has better performance in both control effect and speed.

High-quality Gossypium hirsutum and Gossypium barbadense genome assemblies reveal the landscape and evolution of centromeres

Centromere positioning and organization are crucial for genome evolution;however,research on centro-mere biology is largely influenced by the quality of available genome ***,we combined Oxford Nanopore and Pacific Biosciences technologies to de novo assemble two high-quality reference genomes for Gossypium hirsutum(TM-1)and Gossypium barbadense(3-79).Compared with previously published reference genomes,our assemblies show substantial improvements,with the contig N50 improved by 4.6-fold and 5.6-fold,respectively,and thus represent the most complete cotton genomes to *** high-quality reference genomes enable us to characterize 14 and 5 complete centromeric regions for *** and ***,*** data revealed that the centromeres of allotetraploid cotton are occupied by members of the centromeric repeat for maize(CRM)and Tekay long terminal repeat families,and the CRM family reshapes the centromere structure of the At subgenome after *** two intertwined families have driven the convergent evolution of centromeres between the two subgenomes,ensuring centromere function and genome *** addition,the reposi-tioning and high sequence divergence of centromeres between *** and *** have contributed to speciation and centromere *** study sheds light on centromere evolution in a sig-nificant crop and provides an alternative approach for exploring the evolution of polyploid plants.

Nonlinear optical oscillation in on-chip erbium-doped lithium niobate microring resonators

Erbium-doped lithium niobate(LN) on insulator active devices, such as lasers and amplifiers, have received increasing attention. The nonlinear optical oscillation in them at high power destabilizes the output of signals and cannot be ignored. In this study, we reported the experimental observation and theoretical analysis of the nonlinear optical oscillation in erbiumdoped lithium niobate-on-insulator(LNOI) microring resonators while scanning the pump wavelength. Under the same pump power, the number of oscillation cycles decreases when the wavelength scanning rate increases from 10.6 to 33.9 nm/μs. A theoretical model based on the competition between the thermo-optic nonlinearity and the photorefractive effect was introduced to interpret the oscillation in transmission. A series of parameters were obtained from the comparison between the theoretical and experimental results; some of them, the relaxation rates of the thermal and the electric field, are significantly different from those of undoped LNOI microcavities. This work provides a valuable reference for future applications of active LNOI devices.

Engineering infectious foot-and-mouth disease virus in vivo from a full-length genomic cDNA clone of the A/AKT/58 strain

Two full-length genomic cDNA clones, pTA/FMDV and pCA/FMDV, were constructed that contained three point-mutants [A174G and A308G (not present in pTA/FMDV); T1029G] in the genome compared with the wild type A/AKT/58 strain of foot-and-mouth disease virus. These two viruses were rescued by co-transfection of pCA/FMDV with pCT7RNAP, which can express T7 RNA polymerase in BHK-21 cell-lines, or by transfection of the in vitro transcribed RNA. Their biological properties were analyzed for their antigenicity, virulence in suckling-mice (LD50) and growth kinetics in BHK-21 cells. The in vivo rescued viruses showed high pathogenicity for 3-day-old unweaned mice (LD50=10?7.5). However, the in vitro transcribed RNA derived from pTA/FMDV had lower pathogenicity for suckling-mice (LD50=10?6), and the in vivo transcribed RNA recovered from pCA/FMDV co-transfected with pCT7RNAP showed no significant differences from the wild type virus. These data showed that recovery of the infectious foot-and-mouth disease virus directly from the use of in vivo techniques was better than from in vitro methods. Furthermore, the reverse genetic procedure technique was simplified to a faster one-step procedure based on co-transfection with pCT7RNAP. These results suggest that in vivo RNA tran- scripts may be more valuable for engineering recombinant foot-and-mouth disease virus than in vitro RNA transcripts, and may contribute to further understanding of the biological properties, such as replication, maturation and quasispecies, of the foot-and-mouth disease virus.

Simulations of the Tropical Intraseasonal Oscillation by the Atmospheric General Circulation Model of the Beijing Climate Center

Reflectivity Measurement of Weak Fiber Bragg Grating (FBG)

One method based on the end-face reflection as a reference was used to measure weak grating reflectivity of 0.01%-1%. For measuring ultra-weak grating reflectivity, the grating group with an identical wavelength was used. By using anhydrous alcohol and deionized water as the reflecting medium, the single- pulse FBG reflectivity of a highly doped-Ge photosensitive fiber was calculated to be about 0.747% and 0.739%, respectively; and the single-pulse FBG reflectivity of ordinary SMF fiber was about 0.016% and 0.015%, respectively. The consistent results by the two different reflecting media showed that the end-face reflection method on the grating group was feasible.
