JrATHB-12 mediates JrMYB113 and JrMYB27 to control the anthocyanin levels in different types of red walnut

Red walnut has broad market prospects because it is richer in anthocyanins than ordinary walnut. However,the mechanism driving anthocyanin biosynthesis in red walnut is still unknown. We studied two types of red walnut, called red walnut 1(R1), with a red pericarp and seed coat, and red walnut 2(R2), with a red seed coat only. R1 mostly contained cyanidin-3-O-galactoside, while R2 contained a various amounts of cyanidin-3-Ogalactoside, cyanidin-3-O-arabinoside, and cyanidin-3-O-glucoside. The LDOX-2(LOC109007163) and LDOX-3(LOC109010746) genes, which encode leucoanthocyanidin dioxygenase/anthocyanidin synthase(LDOX/ANS),were preliminarily indicated as the crucial genes for anthocyanin biosynthesis in R1 and R2, respectively. The MYB differential genes analysis showed that MYB27 and MYB113 are specifically expressed in the red parts of R1 and R2,respectively, and they are regarded as candidate regulatory genes. Ectopic expression in Arabidopsis and transient injection in walnut showed that both MYB27 and MYB113 were located in the nucleus and promoted anthocyanin accumulation, while MYB27 promoted the expression of LDOX-2, and MYB113 promoted the expression of LDOX-3and UAGT-3. Yeast one-hybrid and electrophoretic mobility shift assays showed that MYB27 could only bind to the LDOX-2 promoter, while MYB113 could bind to the promoters of both LDOX-3 and UAGT-3. In addition, we also identified an HD-Zip transcription factor, ATHB-12, which is specifically expressed in the pericarp. After silencing the expression of ATHB-12, the R2 pericarp turned red, and MYB113 expression increased. Further experiments showed that ATHB-12 could specifically interact with MYB113 and bind to its promoter. This suggests that MYB27controls R1 coloration by regulating LDOX-2, while MYB113 controls R2 coloration by regulating LDOX-3 and UAGT-3, but ATHB-12 can specifically bind to and inhibit the MYB113 of the R2 pericarp so that it becomes unpigmented. PaPYL9 is involved in the regulation of apricot fruit ripening through ABA signaling pathway

Abscisic acid(ABA)is a major regulator of non-climacteric fruit ripening;however,the role of ABA in the ripening of climacteric fruit is not clear.Here,as a typical climacteric fruit,apricots were used to investigate the role of ABA in fruit ripening.Based on weighted gene coexpression network analysis(WGCNA)of our previous transcriptome data,we treated‘Danxing’fruit with exogenous ABA and obtained ABA receptor genes,genes related to ABA biosynthesis and signal transduction,and analyzed the response of these candidate genes to exogenous ABA during fruit ripening.Subsequently,the full length of candidate PYLs genes were cloned,and their putative function were analyzed by phylogenetic analysis and protein structure domain analysis.And then the function of one candidate gene PaPYL9 was verified by using transgenic tomato.Furthermore,the response genes in transgenic tomato were screened by transcriptome sequencing,and ultimately the related regulatory network was proposed.The results showed that the injection of exogenous 1.89 mmol·L^(-1) ABA remarkably promoted fruit coloration,and increased the color index for red grapes(CIRG)and the total soluble solids(TSS)content,but significantly decreased the firmness and titratable acid(TA)content(p<0.01).Nordihydroguaiaretic acid(NDGA),the inhibitor of ABA,appeared to have the converse role in TA,TSS,CIRG and firmness,during the ripening process.One NCED(9-cis-epoxycarotenoiddioxygenase)and five ABA receptor genes related to signal transduction were mined from the transcriptome data of apricot fruit through WGCNA.Compared with the control,the expression levels of NCED1,PYL9(PYR/PYL/RCAR),SnRK2(SUCROSE NON-FERMENTING1(SNF1)-RELATED PROTEIN KINASE 2S),and ABF2(ABRE-binding bZIP transcription)were induced dramatically by ABA treatment(p<0.01),while NDGA treatment significantly inhibited their expression.Based on gene expression and protein domain analysis,we inferred that PaPYL9 is putatively involved in apricot fruit ripening.Overexpression of PaPYL9 in Micro-TOM tomatoes resu

Novel wild edible mushroom Astraeus hygrometricus(Pers.)Morgan induces robust apoptosis on human acute lymphoblastic leukemia cells through a RONS-subsisted mitochondria-dependent pathway

Objective:To explore the therapeutic effects of the novel wild edible mushroom Astraeus hygrometricus(Pers.)Morgan(A.hygrometricus)on human acute lymphoblastic leukemia cells.Methods:Extensive screening of the antiproliferative and chemopreventive potential of different extracts from 5 wild mushrooms,A.hygrometricus,Phallus sp.,Lentinus sp.,Tricholoma sp.,and Serpula sp.was performed against a panel of 6 cancer cell lines and normal cells using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Apoptosis determination,cell cycle profiling,intracellular reactive oxygen species(ROS)and reactive nitrogen species(RNS),and mitochondrial membrane potential were analyzed by flow cytometry.The activity of caspases was measured colorimetrically,and the expression pattern of mitochondrial proteins was analyzed.Results:The methanol extract of A.hygrometricus and MOLT-4 cells were identified as the most potent extract exhibiting antiproliferative activity and most sensitive cell line,respectively.The mushroom extract induced robust selective apoptosis in MOLT-4 cells and arrested cell cycle progression at the G0/G1 stage.The extract disrupted the mitochondrial membrane potential and enhanced ROS production in MOLT-4 cells.The methanol extract induced apoptosis by downregulating the expression of Bcl-2,increasing the expression of Bax,and activating the caspase cascade.Conclusion:The novel wild edible mushroom is a potential repository of biomolecules for the development of antileukemic drugs.

Date palm(Phoenix dactylifera),one of the most ancient crops,is grown commercially in>30 countries.Using whole plastome assemblies,phylogenetic analyses revealed that cultivated date palm accessions share the same clade with Phoenix sylvestris,Phoenix pusilla and Phoenix acaulis,which are native to the Indian subcontinent,and Phoenix caespitosa that is native to the Arabian Peninsula and the deserts of Somalia.Analysis of genetic diversity and genetic relationships among date palm accessions from 13 producing countries involved 195 date palm accessions that were genotyped at 19 microsatellite loci.Extensive genetic diversity was observed,with many accessions heterozygous for most markers in this clonally propagated crop.The average number of alleles per locus(42.1),expected heterozygosity(0.8),observed heterozygosity(0.47)and fixation indices(FST=0.42)demonstrated substantial genetic diversity and population structure.Iraqi accessions were found to have the richest allelic diversity,and the most private alleles.The model-based Bayesian method indicated that these accessions could be broadly divided into two structure groups,one group with predominantly African accessions and another predominantly Asian.Some germplasm,especially from Tunisia and Iraq,deviated from this generalization.Many accessions in the STRUCTURE-derived groups were found to be genetic admixtures,with gene flow between Asian and African groups.Indian and Pakistani date palms were found to be most closely related to North African germplasm.

Citrus huanglongbing（HLB） has become a major disease and limiting factor of production in citrus areas that have become infected. The destruction to the affected citrus industries has resulted in a tremendous increase to support research that in return has resulted in significant information on both applied and basic knowledge concerning this important disease to the global citrus industry. Recent research indicates the relationship between citrus and the causal agent of HLB is shaped by multiple elements, in which host defense responses may also play an important role. This review is intended to provide an overview of the importance of HLB to a wider audience of plant biologists. Recent advances on host-pathogen interactions, population genetics and vectoring of the causal agent are discussed.

Dear Editor,Utilization of heterosis is fundamental to modern agricultural improvement and contributes significantly to food security and agricultural development(Ouyang et al.,2022).However,heterosis of F,hybrids cannot be stably inherited.Apomixis,or asexual reproduction through seeds,offers the potential for the fixation of heterosis in F,hybrids and the formation of clonal offspring,which could have considerable implications for plant breeding and agriculture(Schmidt,2020).But naturally occurring apomixis has never been found in major crops.Recently,synthetic apomixis has been successfully generated by inducing incomplete meiotic development combined with parthenogenesis development to produce cloned rice(Oryza sativa)seeds(Khanday et al.,2019;Wang et al.,2019;Vernet et al.,2022;Wei et al.,2023).

Soil-borne plant pathogens represent a serious threat that undermines commercial walnut(Juglans regia)production worldwide.Crown gall,caused by Agrobacterium tumefaciens,and Phytophthora root and crown rots,caused by various Phytophthora spp.,are among the most devastating walnut soil-borne diseases.A recognized strategy to combat soil-borne diseases is adoption of resistant rootstocks.Here,resistance to A.tumefaciens,P.cinnamomi,and P.pini is mapped in the genome of Juglans microcarpa,a North American wild relative of cultivated walnut.Half-sib J.microcarpa mother trees DJUG 31.01 and DJUG 31.09 were crossed with J.regia cv.Serr,producing 353 and 400 hybrids,respectively.clonally propagated hybrids were genotyped by sequencing to construct genetic maps for the two populations and challenged with the three pathogens.Resistance to each of the three pathogens was mapped as a major QTL on the long arm of J.microcarpa chromosome 4D and was associated with the same haplotype,designated as haplotype b,raising the possibility that the two mother trees were heterozygous for a single Mendelian gene conferring resistance to all three pathogens.The deployment of this haplotype in rootstock breeding will facilitate breeding of a walnut rootstock resistant to both crown gall and Phytophthora root and crown rots.

Breast cancer resistance protein(BCRP)is an important resistance protein that significantly impacts anticancer drug discovery,treatment,and rehabilitation.Early identification of BCRP substrates is quite a challenging task.This study aims to predict early substrate structure,which can help to optimize anticancer drug development and clinical diagnosis.For this study,a novel intelligent approach-based methodology is developed by modifying the ResNet101 model using transfer learning(TL)for automatic deep feature(DF)extraction followed by classification with linear discriminant analysis algorithm(TLRNDF-LDA).This study utilized structural fingerprints,which are exploited by DF contrary to conventional molecular descriptors.The proposed in silico model achieved an outstanding accuracy performance of 98.56%on test data compared to other state-of-the-art approaches using standard quality measures.Furthermore,the model’s efficacy is validated via a statistical analysisANOVAtest.It is demonstrated that the developedmodel can be used effectively for early prediction of the substrate structure.The pipeline of this study is flexible and can be extended for in vitro assessment efficacy of anticancer drug response,identification of BCRP functions in transport experiments,and prediction of prostate or lung cancer cell lines.

What is already known about this topic?The inclusion of meningococcal vaccines in the national Immunization Program(NIP)over several years has significantly reduced the incidence of meningococcal meningitis in China to historic lows.Worldwide,there has been a diversification of meningococcal serogroups,leading to a shift in dominant serogroups in China from serogroup A to serogroups C and B,accompanied by a rise in reports of serogroups Y and W.What is added by this report?An outbreak of serogroup Y Neisseria meningitidis(Nm)in a secondary vocational school involved a single confirmed severe case and 24 individuals with laboratory-confirmed Nm carriage.Epidemiological investigation revealed that the outbreak was localized to the classroom of the confirmed case.Prolonged close contact within a confined space was identified as a significant risk factor for Nm transmission.The genotype sequence identified was type 1655(ST-1655),which is categorized under clonal complex 23(CC-23)and bears resemblance to 8 previously confirmed cases of serogroup Y meningococcal meningitis within Guangdong Province.This suggests that serogroup Y infections continue to sporadically emerge and have become prevalent strains.What are the implications for public health practice?This outbreak underscores the critical need to enhance surveillance of meningococcal serogroups and population carrier,and advocate for vaccination with MenY-containing vaccines.

Epidemiology and genetic characterization of tet(X4)-positive Klebsiella pneumoniae and Klebsiella quasipneumoniae isolated from raw meat in Chengdu City, China

The rapid spread of mobile tigecycline resistance presents a significant public health threat,particularly with the increasing prevalence of tet(X4)-positive Enterobacterales across various species.This study aimed to inves-tigate the epidemic features and transmission dynamics of tet(X4)-positive Klebsiella pneumoniae(K.pneumo-niae)through the analysis of 206 raw meats,including pork(n=182),beef(n=16),duck(n=5),and chicken(n=3).These samples were collected from schools,markets,and restaurants in Chengdu City,China.A total of 25 isolates were obtained from 13 administrative regions.All isolates exhibited resistance to tetracycline,tigecycline,ampicillin,chloramphenicol,and florfenicol.Over half of the isolates also demon-strated resistance to streptomycin(80%),sulfamethoxazole/trimethoprim(72%),ciprofloxacin(64%),and ampicillin/sulbactam(56%).Among these strains,14 distinct sequence types(STs)were identified,revealing evidence of inter-regional clonal spread,notably among 9 K.pneumoniae ST3393.Phylogenetic analysis revealed the presence of two K.pneumoniae ST5 closely resembling hypervirulent K.pneumoniae from Jiangsu.Importantly,12 isolates were capable of transferring tigecycline resistance to Escherichia coli J53.Further plasmid analysis showed that the tet(X4)-harboring plasmids in K.pneumoniae could be classified into four types,primarily belonging to the IncFIA(HI1)/HI1A/HI1B hybrid plasmid(n=16)and IncFII plasmid(n=7),which significantly contributed to the cross-species dissemination of tet(X4).In summary,this study highlights the prevalence of MDR tet(X4)-positive K.pneumoniae in Chengdu,driven predominantly by clonal expansion and plasmid-mediated horizontal gene transfer.These findings emphasize the importance of contin-uous surveillance of tet(X4)-positive K.pneumoniae in raw meat and the implementation of effective measures to control their spread.
