Serum metabolic effects of corn oligopeptides with 7-day supplementation on early post-surgery primary liver cancer patients: a double-blind randomized controlled trial

Background:Liver cancer as the main leading cancer has caused heavy burdens *** prognosis of liver cancer is closely related with postoperative nutrition *** oligopeptides(COPs)are protein hydrolysates produced by enzymatic treatments,which have shown potential bioactivities,such as inhibiting angiotensin I-converting enzyme,resisting lipid peroxidation and ***,the correlation between COPs and liver cancer patients is still unknown and the potential mechanism of COPs on liver cancer is unclear as *** aim of this study was to assess effects of 7-day intervention of COPs after surgery on liver function and serum metabolic profiles of liver cancer ***:Patients were assigned into COPs intervention group(n=50)and control group(n=91)for 7 *** were scheduled at 1st day and 7th day after liver resection surgery respectively,mainly including anthropometric,biochemical indexes and liquid chromatography-mass spectrometry(LC/MS)***:Seven-day supplementation of COPs on early post-surgery liver cancer patients down-regulated levels of alanine aminotransferase,aspartate aminotransferase,total bilirubin,direct bilirubin and up-regulated prothrombin time activity and prealbumin ***/MS analysis revealed metabolic signatures including regulation of 16 metabolites,which was closely related with two metabolic pathways(nicotinate and nicotinamide metabolism,fatty acid metabolism).Conclusions:COPs supplementation has displayed the potentials on alleviating the injury of liver function and it may be due to regulation of fatty acid metabolism,nicotinate and nicotinamide metabolism,lipid peroxidation and anti-inflammatory *** researches are warranted in future to confirm the exact mechanisms.

During the coronavirus disease 2019(COVID-19)pandemic,the Japanese government implemented several infection control measures,such as states of emergency(SoE)and school closures,and encouraged universal masking and hand *** activity restrictions and improved hygiene were found to be associated with declines in the incidence of infuenza during the 2019–2020 winter season[1].Prior studies have found that the incidence of seven pediatric infections,such as pharyngoconjunctivitis and varicella,decreased during and after the school closures in Japan[2].As activity restrictions continue to be lifted,there is growing concern that pediatric infections may suddenly return given that the pool of susceptible children increased during the pandemic[3].In this study,we use national infectious disease sentinel data from January 2012 to October 2021 to assess changes in the reported number of pediatric infections in Japan during the COVID-19 *** hypothesize that the reporting of many infectious diseases decreased during the early stages of the pandemic but ultimately returned to normal when activity restrictions were lifted.

GB7 acetate is a galbulimima alkaloid obtained from Galbulimima ***,information regarding its structure,biological activities,and related mechanisms is not entirely available.A series of spectroscopic analyses,structural degradation,interconversion,and crystallography were performed to identify the structure of GB7 *** MTT assay was applied to measure cell proliferation on human colorectal cancer HCT 116 *** expressions of the related proteins were measured by Western *** electron microscopy(TEM),acridine orange(AO)and monodansylcadaverine(MDC)staining were used to detect the presence of autophagic vesicles and autolysosomes.A transwell assay was performed to demonstrate metastatic *** consumption rate(OCR)and extracellular acidification rate(ECAR)assays were performed to determine the mitochondrial oxidative phosphorylation(OXPHOS)and glycolysis activity of HCT 116 *** data showed that GB7 acetate suppressed the proliferation and colony-forming ability of HCT 116 *** with GB7 acetate significantly induced the formation of autophagic vesicles and ***7 acetate upregulated the expressions of LC3 and Thr172 phosphorylated adenosine 5'-monophosphate(AMP)-activated protein kinase a(p-AMPKa),which are key elements of *** addition,GB7 acetate suppressed the metastatic capabilities of HCT 116 ***,the production of matrix metallo-proteinase-2(MMP-2)and MMP-9 was reduced,whereas the expression of E-cadherin(E-cad)was ***,GB7 acetate significantly reduced mitochondrial OXPHOS and *** conclusion,the structure of the novel Galbulimima alkaloid GB7 acetate was ***7 acetate was shown to have anti-proliferative,pro-autophagic,anti-metastatic,and anti-metabolite capabilities in HCT 116 *** study might provide new insights into cancer treatment efficacy and cancer chemoprevention.

Genome-wide and Interaction Linkage Scan for Nonsyndromic Cleft Lip with or without Cleft Palate in Two Multiplex Families in Shenyang,China

Objectives To identify the loci involved in nonsyndromic cleft lip with or without cleft palate （NSCL/P） in Northern Chinese people in Shenyang by using genomewide and interaction linkage *** Two multiplex families in Shenyang from North China were ascertained through probands with NSCL/*** of every member was drawn for DNA extraction and *** were available for 382 autosomal short tandem repeat （STR） markers from the ABI Prism Linkage Mapping Set version *** between markers and NSCL/P was assessed by 2-point parametric LOD scores,multipoint heterogeneity parametric LOD scores （HLODs）,and multipoint nonparametric linkage score （NPL）.Results The initial scan suggested linkage on Chromosomes 1,2,and *** subsequent fine mapping,1q32-q42 showed a maximum multipoint LOD score of 1.9（empirical P=0.013） and an NPL score of 2.35 （empirical P=0.053）.For 2p24-p25,the multipoint NPL increased to 2.94 （empirical P=0.007）.2-locus interaction analysis obtained a maximum NPL score of 3.73 （P=0.00078） and a maximum LOD score of 3 for Chromosome 1 （at 221 cM） and Chromosome 2 （at 29 cM）.Conclusion Both parametric and nonparametric linkage scores greatly increased over the initial linkage scores on 1q32-q42,suggesting a susceptibility locus in this *** linkage gave a strong evidence for a candidate region on chromosome *** superiority of 2-locus linkage scores compared to single-locus scores gave additional evidence for linkage on 1q32-q42 and 2p24-p25,and suggested that certain genes in the two regions may contribute to NCSL/P risks with interaction.

With the convergence of high-performance computing(HPC),big data and artificial intelligence(AI),the HPC community is pushing for"triple use"systems to expedite scientific ***,supporting these converged applications on HPC systems presents formidable challenges in terms of storage and data management due to the explosive growth of scientific data and the fundamental differences in I/O characteristics among HPC,big data and AI *** this paper,we discuss the driving force behind the converging trend,highlight three data management challenges,and summarize our efforts in addressing these data management challenges on a typical HPC system at the parallel file system,data management middleware,and user application *** HPC systems are approaching the border of exascale computing,this paper sheds light on how to enable application-driven data management as a preliminary step toward the deep convergence of exascale computing ecosystems,big data,and AI.

Enhancing the Crystallization Performance of Poly(L-lactide) by Intramolecular Hybridizing with Tunable Self-assembly-type Oxalamide Segments

In this work, hydroxyl-terminated oxalamide compounds N^(1),N^(2)-bis(2-hydroxyethyl)oxalamide(OXA1) and N^(1),N^(1)′-(ethane-1,2-diyl)bis(N^(2)-(2-hydroxyethyl)oxalamide(OXA2) were synthesized to initiate the ring-opening polymerization of L-lactide for preparation of oxalamide-hybridized poly(L-lactide)(PLA_(OXA)), i.e., PLA_(OXA1) and PLA_(OXA2). The crystallization properties of PLA were improved by the self-assembly of the oxalamide segments in PLA_(OXA) which served as the initial heterogeneous nuclei. The crystal growth kinetics was studied by HoffmanLauritzen theory and it revealed that the nucleation energy barrier of PLA_(OXA1) and PLA_(OXA2) was lower than that of PLA. Consequently, PLA_(OXA) could crystallize much faster than PLA, accompanied with a decrease in spherulite size and half-life crystallization time by 74.8% and 86.5%(T=125 ℃), respectively. In addition, the final crystallinity of PLA_(OXA1) and PLA_(OXA2) was 6 and 8 times higher, respectively, in comparison with that of neat PLA under a controlled cooling rate of 10 ℃/min. The results demonstrate that the hybridization of oxalamide segments in PLA backbone will serve as the self-heteronucleation for promoting the crystallization rate. The higher the content of oxalamide segments(PLA_(OXA2) compared with PLA_(OXA1)) is, the stronger the promotion effect will be. Therefore, this study may provide a universal approach by hybridizing macromolecular structure to facilitate the crystallization of semi-crystalline polymer materials.

Establishment of probabilistic model for Salmonella Enteritidis growth and inactivation under acid and osmotic pressure

The growth and survival characteristic of Salmonella Enteritidis under acidic and osmotic conditions were ***,a probabilistic model based on the theory of cell division and mortality was established to predict the growth or inactivation of *** experimental results demonstrated that the growth curves of planktonic and detached cells showed a significant difference(p<0.05)under four conditions,including pH5.0+0.0%NaCl,pH7.0+4.0%NaCl,pH6.0+4.0%NaCl,and pH5.0+4.0%*** the established primary and secondary models could describe the growth of *** well by estimating four mathematics evaluation indexes,including determination coefficient(R2),root mean square error(RMSE),accuracy factor(Af)and bias factor(Bf).Moreover,sequential treatment of 15%NaCl stress followed by pH 4.5 stress was the best condition to inactivate *** in 10 h at 25◦*** probabilistic model with Logistical or Weibullian form could also predict the inactivation of *** well,thus realize the unification of predictive model to some extent or generalization of inactivation ***,the primary 4-parameter probabilistic model or generalized inactivation model had slightly higher applicability and reliability to describe the growth or inactivation of *** than Baranyi model or exponential inactivation model within the experimental range in this study.

Tissue distribution of Lycium barbarum polysaccharides in rat tissue by fluorescein isothiocyanate labeling

To date, in vivo investigations of polysaccharide’s pharmacokinetics are significantly restricted by the difficulty in their detection. This study was conducted to establish the quantitative determination of Lycium barbarum polysaccharides(LBPs) based on fluorescein isothiocyanate(FITC) pre-labeling and to investigate their tissue distribution in rat. We obtained the calibration curves linear over the range of 0.0–25 μg/m L in rat tissue samples with correlation coefficients greater than 0.99. The inter-day and intra-day precisions(RSD, %) were within 15%, and the relative recovery ranged 95.2%–102.4%, with RSD range 1.48%–9.58%, indicating that this experiment was suitable for the determination of LBPs. The fluorescence intensity was measured after 24 h storage at room temperature, 3 times of freeze-cycle and cryopreservation at –20 ℃ for 15 day, these results indicated that the stability of the samples was good. LBP-FITC was mainly absorbed by the small intestine and stomach, and mainly excreted in the urine through the kidney;this distinct difference in the tissue distribution of LBPs could be attributed to the size of these LBPs in relation to the pore sizes of the vascular beds in the kidney and liver. Results showed in this study enable us to comprehensively understand the biological effects of LBPs following its oral ingestion.

Simulating optimal vaccination times during cholera outbreaks

The use of cholera vaccines has been increasingly recognized as an effective control measure in cholera endemic countries. However, guidelines for using vaccination during cholera outbreaks are still to be established, and it remains an open question as to how and when the vaccines should be deployed to best control ongoing cholera outbreaks. Here we formulate a new optimal control model to assess the value of cholera vaccines in epidemic settings and cost-effective optimal times to deploy a vaccine. Our results suggest that as long as the vaccine prices are sufficiently low, vaccination should always start from or immediately after the onset of a cholera outbreak.
