Review of thrust measurement techniques for micro-thr usters

Green fl uorescent protein(GFP)has been widely used for monitoring gene expression and protein localization in diverse ***,highly sensitive imaging equipment,like fl uorescence microscope,is usually required for the visualization of GFP,limitings its application to fi xed locations in samples.A reporter that can be visualized in realtime regardless the shape,size and location of the target samples will increase the fl exibility and ef fi ciency of research ***,we report the application of a GFP-like protein,called eYGFPuv,in both transient expression and stable transformation,in two herbaceous plant species(Arabidopsis and tobacco)and two woody plant species(poplar and citrus).We observed bright fl uorescence under UV light in all of the four plant species without any effects on plant growth or *** was shown to be effective for imaging transient expression in leaf and root *** a focus on in vitro transformation,we demonstrated that the transgenic events expressing 1x eYGFPuv could be easily identi fi ed visually during the callus stage and the shoot stage,enabling early and ef fi cient selection of ***,whole-plant level visualization of eYGFPuv revealed its ubiquitous stability in transgenic *** addition,our transformation experiments showed that eYGFPuv can also be used to select transgenic plants without *** work demonstrates the feasibility of utilizing 1x eYGFPuv in studies of gene expression and plant transformation in diverse plants.

The bipartite begomoviruses (Geminiviridae family), which are DNA viruses that replicate in the nucleus of infected cells, encode the nuclear shuttle protein (NSP) to facilitate the translocation of viral DNA from the nucleus to the cytoplasm via nuclear pores. This intracellular trafficking of NSP-DNA complexes is accessorized by the NSP-interacting guanosine triphosphatase (NIG) at the cytosolic side. Here, we report the nuclear redistribution of NIG by AtWWPI, a WW domain-containing protein that forms immune nuclear bodies (NBs) against begomoviruses. We demonstrated that AtWWPI relocates NIG from the cytoplasm to the nucleus where it is confined to AtWWP1-NBs, suggesting that the NIG-AtWWP1 interaction may interfere with the NIG pro-viral function associated with its cytosolic localization. Consistent with this assumpti on joss AtWWPI functi on cuased plants more susceptible to begomovirus infection, whereas overexpression of AWWP1 enhaneed plant resistance to begomovirus. Furthermore, we found that a mutant versi on of AtWWPI defective for NB formatio n was noIonger capable of interacti ng with and relocating NIG to the nucleus and lost its immune function against begomovirus. The antiviral function of AtWWP1-NBs, however, could be antagonized by viral infection that induced either the disruption or a decrease in the number of AtWWP1-NBs. Collectively, these results led us to propose that AtWWPI organizes nuclear structures into nuclear foci, which provide intrinsic immunity against begomovirus infection.

Background Our earlier studies indicate that melatonin inhibits the proliferation of prolactinoma and induces apoptosis of pituitary prolactin-secreting tumor in rats. Melatonin has also been shown to induce apoptosis and to reduce the production of ATP in breast tumor cells. This study analyzed the levels of the four mitochondrial respiratory complexes and the production of ATP and also the effects of melatonin treatment of prolactinoma. Methods In the in vivo study, mitochondria were harvested from control pituitaries or prolactinoma collected from the pituitaries of melatonin- and 17-13-estradiol （E2）-treated male rats. In the in vitro study, prolactinoma cells mitochondria were harvested. Activities of the four mitochondrial respiratory complexes were assayed using fluorometer. ATP production of prolactinoma cells was estimated using bioluminescent methods. Results Elevated levels of four mitochondrial respiratory complexes activities and ATP production were recorded in prolactinoma cells. Moreover, in both in vivo and in vitro studies, melatonin inhibited the activities of mitochondrial respiratory complexes and the production of ATP in prolactinoma cells. Conclusions There is a link between mitochondria[ function increase and tumorigenesis. Melatonin induces apoptosis of pituitary prolactin-secreting tumor of rats via the induction of mitochondrial dysfunction and inhibition of energy metabolism.

Background:Virtual global health partnership initiatives(VGHPIs)evolved rapidly during the COVID-19 pandemic to ensure partnership *** the current landscape for VGHPI use and preference is *** study aimed to increase understanding of GH partners’perspectives on ***:From 15 October to 30 November 2020,An online,international survey was conducted using snowball sampling to document pandemic-related changes in partnership activities,preferences for VGHPIs,and perceived acceptability and *** survey underwent iterative development within a diverse author group,representing academic and clinical institutions,and the non-profit *** from their professional global health networks were invited,including focal points for global health partnerships while excluding trainees and respondents from the European Economic *** stratified responses by country income classification and partnership *** used descriptive statistics to characterize responses,defining statistical significance asα=***:A total of 128 respondents described 219 partnerships.152/219(69%)partnerships were transnational,157/219(72%)were of>5 years duration,and 127/219(60%)included bidirectional site ***-income country(HIC)partners sent significantly more learners to low-to middle-income country(LMIC)partner sites(p<0.01).Participants commented on pandemic-related disruptions affecting 217/219(99%)partnerships;195/217(90%)were disruption to activities;122/217(56%)to communication;73/217(34%)to access to professional support;and 72/217(33%)to *** indicated that VGHPIs would be important to 206/219(94%)of their partnerships moving *** were overall differences in resource availability,technological capacity,and VGHPI preferences between LMIC and HIC respondents,with a statistically significant difference in VGHPI acceptability(p<0.001).There was no significant difference between groups regarding VGHP
