Intestinal anti-inflammatory activity of Ground Cherry(Physalis angulata L.)standardized CO2 phytopharmaceutical preparation

AIM To investigate the effects of Ground Cherry(Physalis angulata L.)standardized supercritical CO_2 extract in trinitrobenzenesulphonic acid(TNBS)model of rat intestinal *** The animals were divided into groups that received vehicle or *** extract(PACO_2)orally at the doses 25,50 and 100 mg/kg daily by 5 d before TNBS *** effects of PACO_2 were assessed by macroscopic analysis,biochemical determinations of the levels of myeloperoxidase(MPO),alkaline phosphatase(ALP),glutathione and cytokines(such as INF-γ,IL-1β,IL-6,IL-10 and TNF-α),gene expression evaluation(including Hsp70,heparanase,NF-κB,mitogenactivated protein kinases(Mapk)1,3,6 and 9,and the mucins genes Muc 1,2,3 and 4)and histopathological studies using optical,and electronic(transmission and scanning)*** PACO2 extract promoted a significant reduction in MPO and ALP activities,reducing oxidative stress and neutrophil *** effects were accompanied by significant reduction of colonic levels of IFN-γand IL-6 and down-regulation of heparanase,Hsp70,Mapk3,Mapk9,Muc1 and Muc2 genes expression when compared with TNBS-control *** addition,protective effects were also evidenced by reduced neutrophil infiltration,recovery of cell architecture and replacement of mucin by histopathological and ultrastructural *** Physalis angulata supercritical CO2 extract is an intestinal anti-inflammatory product that modulates oxidative stress,immune response and expression of inflammatory mediators,with potentially utility for treating inflammatory bowel disease.

CREB-binding proteins (CBP) as a transcriptional coactivator of GATA-2

The GATA family consists of six members, GATA 1-6. In this study, we focused on GATA-2, which is expressed predominantly in hematopoietic progenitor cells and plays the key role in keeping these cells in the undifferentiated status. CREB-binding proteins (CBP) are essential transcriptional coacti- vators for a large number of regulated dNA-binding transcription factors, including GATA-1. But there have been no reports on whether CBP is still a co-activator of GATA-2. Here, we used the immunopre- cipitation and pull-down experiments to show that the GATA-2 and CBP were physically binding to- gether, and clarified the binding sites CH1, CH3 , CH452 and CT1430 in CBP and N-finger, C-finger and N-C-finger in GATA-2. Luciferase assay results in our experiment indicated that CBP could increase GATA-2 transcriptional activity in the dose-dependent manner. GATA-1 is mainly expressed in differen- tiated hematopoietic cells, but still has overlap expression with GATA-2. CBP is a coactivator of GATA-2 and GATA-1. The investigation on the mechanism that could decide whether CBP binds to GATA-2 to keep hematopoietic cells in the progenitor status or to GATA-1 to start differentiation will be a very in- teresting and very meaningful project in the near future.

Effects of 17<i>β</i>-Estradiol on dopamine d2 Receptors in Thiamine-deficient Female Rats: Consequences on Sucrose, Alcohol, Water Intakes and Body Weight

Our previous studies showed that 17β-estradiol (E2) modulated dopamine d2 receptor in regulating body weight set-point. The aim of this study was to understand whether thiamine deficiency influenced the E2 modulation on dopamine d2 receptors, using bromocriptine mesylate (BR) and sulpiride (SUL) as selective central dopamine-d2 receptors agonist and antagonist respectively. We studied the E2-dopamine d2 receptors interferences in a 10-day thiamine-deficient female rats for which consumptions of water, sugar, alcohol and food were daily-recorded and their consequences on body weights assessed. Our results showed that the volume of water daily ingested doubled in thiamine-deficient female rats (OXT), while sugar and alcohol consumptions collapsed with decreased weight and food consumption. On the one hand, thiamine potentiated d2/BR activity (bromocriptine-activated d2 receptors) to induce sugar intake and inhibited the same d2/BR receptors to induce water intake. On the other hand, thiamine promoted d2/SUL receptors (sulpiride-inhibited d2 receptors) for enhanced alcohol intake, increased food consumption and weight gain. Taking together, thiamine modulated the actions of 17β-estradiol on both d2/BR and d2/SUL receptors activities.

Antioxidant Protection Mechanisms Reveal Significant Response in drought-Induced Oxidative Stress in Some Traditional Rice of Assam, India

drought tolerance levels and antioxidant protection mechanisms were evaluated for 21 traditional rice varieties of Assam, India, along with Sahbhagi dhan （drought tolerant） and IR64 （drought sensitive） as controls. drought was imposed in hydroponic culture with polyethylene glycol 6000 （PEG6000） that was initially standardized with different concentrations. All the rice varieties showed apparent decreases in growth characteristics under drought stress （initially at 15% for 7 d followed by 20% PEG6000 for 7 d in Yoshida medium）. On the basis of standard evaluation score （SES）, eight rice varieties showed high drought tolerance which were carried forward for further biochemical analyses. Based on different morpho- physiological parameters, SN03 （Bora）, SN04 （Prosad Bhog）, SN05 （Kola Joha）, SN06 （Helash Bora）, SN08 （Salihoi Bao）, SN12 （Kola Amona）, SN20 （Ronga Bora） and SN21 （Sok-Bonglong） were identified as promising drought tolerant varieties. The non-enzymatic antioxidants activities viz., glutathione, ascorbate and enzymatic antioxidant activities such as superoxide dismutase （SOd）, catalase （CAT）, guaiacol peroxidase （GPX）, ascorbate peroxidase （APX）, glutathione reductase （GR） in shoots and roots of all the selected varieties revealed significant level of protection mechanisms as compared with controls. Enhancement in activities of the overall antioxidant enzymes including SOd, GPX, CAT, GR and APX under drought stress reflects their role in the adaptation process under water stress.

Influence of the SNPs on the structural stability of CBS protein： Insight from molecular dynamics simulations

Cystathionine β-synthase is an essential enzyme of the trans-suifuration pathway that condenses serine with homocysteine to form cystathionine. Missense mutations in CBS are the major cause of inherited homocystinuria, and the detailed effect of disease associated amino acid substitutions on the structure and stability of human CBS is yet unknown. Here, we apply a unique approach in combining in silico tools and molecular dynamics simulation to provide structural and functional insight into the effect of SNP on the stability and activity of mutant CBS. In addition, principal component analysis and free energy landscape were used to predict the collective motions, thermodynamic stabilities and essential subspace relevant to CBS function. The obtained results indicate that C109R, E176K and d376N mutations have the diverse effect on dynamic behavior of CBS protein. We found that highly conserved d376N mutation, which is present in the active pocket, affects the protein folding mechanism. Our strategy may provide a way in near future to understand and study effects of functional nsSNPs and their role in causing homocystinuria.

Physicochemical and Microbiological Properties of Bissap (Hibiscus sabdariffa) and Ginger (Zingiber officinale (L) Rose) Juices Sold in the Western Part of Abidjan (Côte d’Ivoire)

Juices made from the calyx of Hibiscus sabdariffa (Malvaceae), commonly called Bissap, and the rhizome of Zingiber officinale (Zingiberaceae), known as Gnamankoudji, are widely consumed in Côte d'Ivoire. The artisanal preparation of these juices often makes their quality problematic. The aim of this study is to determine some physicochemical and microbiological properties of these drinks sold in the city of Abidjan. To do this, samples of Bissap and Gnamankoudji juices were collected near schools in the Mamie Adjoua (Yopougon) and Abobodoumé (Attécoubé) districts. Physico-chemical and microbiological analyses were performed on these collected samples. The results show that the Bissap and Gnamankoudji juices have an acid pH (2.47 and 3.71), a low protein content (0.78% and 2.11%) and a high water content (80.13% and 85.21%), respectively. The reducing sugar content ranged from 1.20% to 3.34%, with high total sugar (695 mg/mL and 812 mg/mL), low ash (0.70 ± 0.07;0.91 ± 0.01). On the other hand, these juices contain variable concentrations of minerals from one site to another (magnesium, calcium, phosphorus, potassium and iron). Potassium is the most abundant mineral element, followed by phosphorus and magnesium. Moreover, it is observed that the Gnamankoudji juices contain vitamin C (4.67 to 5.58 mg/100mL), contrary to the Bissap juice. The microbiological analysis indicates the presence of aerobic mesophilic germs (AMG) but a total absence of pathogenic germs in all juices. The important presence of nutrients and the total absence of pathogenic germs in these drinks justify their regular consumption by the population.

Joint effects of weather and interspecific competition on foraging behavior and survival of a mountain herbivore

Weather variations have the potential to in fluence species interactions, although effects on competitive interactions between species are poorly known. Both weather and competition can influence foraging behavior and survival of herbivores during nursing/weaning, a critical period in the herbivore life cycle. We evaluated the joint effects of weather and competition with red deer Cervus elaphus on the foraging behavior of adult female Apen nine chamois Rupicapra pyrenaica ornata in summer, and on winter survival of chamois kids. High temperature and low rainfall during the growing seas on of vegetation had negative effects on bite rate. Effects of weather were greater in forb patches, including cold-adapted, nutritious plants of key importanee to chamois, than in graminoid ones. Our results confirm previous indications of a negative effect of competition on bite rate of female chamois and on kid survival. Furthermore, harsh weather conditions and competition with deer had additive, negative roles on foraging behavior and survival of chamois. Growing temperatures are expected to influence distribution, growth, and/or nutritional quality of plants;competition would reduce pasture quality and food availability through resource depletion. Both factors would limit food/energy intake rates during summer, reducing survival of the youngest cohorts in winter. We suggest that interspecific competition can be an important additive factor to the effects of weather changes on behavior and demography.
