Autophagy receptor ZmNBR1 promotes the autophagic degradation of ZmBRI1a and enhances drought tolerance in maize

Drought stress is a crucial environmental factor that limits plant growth,development,and *** of misfolded proteins can help alleviate the damage caused in plants experiencing ***,the mechanism of autophagy-mediated drought tolerance in plants remains largely ***,we cloned the gene for a maize(Zea mays)selective autophagy receptor,NEXT TO BRCA1GENE 1(ZmNBR1),and identified its role in the response to drought *** observed that drought stress increased the accumulation of *** sequencing and reverse transcriptionquantitative polymerase chain reaction showed that ZmNBR1 is markedly induced by drought ***1 overexpression enhanced drought tolerance,while its knockdown reduced drought tolerance in *** results established that ZmNBR1 mediates the increase in autophagosomes and autophagic activity under drought ***1 also affects the expression of genes related to autophagy under drought ***,we determined that BRASSINOSTEROID INSENSITIVE1A(ZmBRI1a),a brassinosteroid receptor of the BRI1-like family,interacts with *** analysis showed that ZmBRI1a negatively regulates drought tolerance in maize,and genetic analysis indicated that ZmNBR1 acts upstream of ZmBRI1a in regulating drought ***,ZmNBR1facilitates the autophagic degradation of ZmBRI1a under drought *** together,our results reveal that ZmNBR1 regulates the expression of autophagy-related genes,thereby increasing autophagic activity and promoting the autophagic degradation of ZmBRI1a under drought stress,thus enhancing drought tolerance in *** findings provide new insights into the autophagy degradation of brassinosteroid signaling components by the autophagy receptor NBR1 under drought stress.

Elevated CO_(2) increases soil redox potential by promoting root radial oxygen loss in paddy field

Soil redox potential(Eh)plays an important role in the biogeochemical cycling of soil *** its effect soil process and nutrients'availability under elevated atmospheric CO_(2) concentration and warming has seldom been ***,in this study,a field experiment was used to elucidate the effect of elevated CO_(2) concentration and warming on soil Eh,redox-sensitive elements and root radial oxygen loss(ROL).We hypothesized elevated CO_(2) and warming could alter soil Eh by promoting or inhibiting *** found that soil Eh in the rhizosphere was significantly higher than that of *** CO_(2) enhanced soil Eh by 11.5%,which corresponded to a significant decrease in soil Fe^(2+)and Mn^(2+)*** elevated CO_(2),the concentration of Fe^(2+)and Mn^(2+)decreased by 14.7%and 13.7%,*** also found that elevated CO_(2) altered rice root aerenchyma structure and promoted rice root *** elevated CO_(2),rice root ROL increased by 79.5%and 112.2%for Yangdao 6 and Changyou 5,*** had no effect on soil Eh and rice root *** warming increased the concentration of Mn^(2+)and SO_(4)^(2-)by 4.9%and 19.3%,*** was a significant interaction between elevated CO_(2) and warming on Fe^(2+)and Mn^(2+).Under elevated CO_(2),warming had no effect on the concentration of Fe^(2+)but decreased Mn^(2+)concentration *** study demonstrated that elevated atmospheric CO_(2) in the future could increase soil Eh by promoting rice root ROL,which will alter some soil nutrients'availability,such as Fe^(2+)and Mn^(2+).

Spatiotemporal evolution and assembly processes of ammonia-oxidizing prokaryotic communities in 1 000-year-old coastal reclaimed soils

Coastal marshes are transitional areas between terrestrial and aquatic *** are sensitive to climate change and anthropogenic *** recent decades,the reclamation of coastal marshes has greatly increased,and its effects on microbial communities in coastal marshes have been studied with great *** of these studies have explained the short-term spatiotemporal variation in soil microbial community ***,the impact of reclamation on the community composition and assembly processes of functional microbes(e.g.,ammonia-oxidizing prokaryotes)is often *** this study,using quantitative polymerase chain reaction and the Ion S5™XL sequencing platform,we investigated the spatiotemporal dynamics,assembly processes,and diversity patterns of ammonia-oxidizing prokaryotes in 1000-year-old reclaimed coastal salt *** taxonomic and phylogenetic diversity and composition of ammonia oxidizers showed apparent spatiotemporal variations with soil *** null modelling-based analysis showed that across all sites,the archaeal ammonia-oxidizing community was assembled by a deterministic process(84.71%),and deterministic processes were also dominant(55.2%)for ammonia-oxidizing bacterial communities except for communities at 60 years of *** assembly process and nitrification activity in reclaimed soils were positively *** abundance of the amoA gene and changes in ammonia-oxidizing archaeal and bacterial diversities significantly affected the nitrification activity in reclaimed *** findings suggest that long-term coastal salt marsh reclamation affects nitrification by modulating the activities of ammonia-oxidizing microorganisms and regulating their community structures and assembly *** results provide a better understanding of the effects of long-term land reclamation on soil nitrogen-cycling microbial communities.

Cross-resistance and genetics of field-evolved resistance to chlorfenapyr in Plutella xylostella

Chlorfenapyr is a broad-spectrum halogenated pyrrole insecticide with a unique mode of *** to the misuse and overuse of this chemical,resistance has been reported in several arthropods,including Plutella xylostella,which is one of the most destructive insect pests afflicting crucifers worldwide.A better understanding of the cross-resistance and genetics of field-evolved chlorfenapyr resistance could effectively guide resistance management ***,the chlorfenapyr resistance of a fieldderived population of P xylostella was introgressed into the susceptible IPP-S strain using a selection-assisted multigenerational backcrossing *** constructed nearisogenic strain,TH-BCsF2,shared 98.4%genetic background with the recurrent parent IPP-S *** TH-BCsF2 strain showed 275-fold resistance to chlorfenapyr,but no significant cross-resistance to spinosad,abamectin,chlorpyrifos,β-cypermethrin,indoxacarb,chlorantraniliprole,or broflanilide(no more than 4.2-fold).Genetic analysis revealed that resistance was autosomal,incompletely dominant,and conferred by 1 major gene or a few tightly linked *** synergism of metabolic inhibitors(PBO,DEM,and DEF)to chlorfenapyr was very weak(<1.7-fold),and the metabolic enzyme activities in the TH-BCsF2 strain were not significantly elevated compared with the IPP-S *** results enhances our understanding of the genetic traits of chlorfenapyr resistance,and provides essential information for improving resistance management strategies.

OsPDR20 is an ABCG metal transporter regulating cadmium accumulation in rice

Cadmium(Cd)is a non-essential toxic heavy metal,seriously posing high environmental risks to human *** genetic resources relevant to functional genes is important for understanding the metal absorption and accumulation in crops and bioremediation of Cd-polluted *** study investigated a functionally uncharacterized ATP binding cassette transporter G family(ABCG)gene encoding a Pleiotropic Drug Resistance 20(PDR20)type metal transporter which is localized to the plasma membrane of ***20 was transcriptionally expressed in almost all tissues and organs in lifespan and was strongly induced in roots and shoots of young rice under Cd *** expression of OsPDR20 in a yeast mutant ycf1 sensitive to Cd conferred cellular tolerance with less Cd *** of OsPDR20 by RNA interference(RNAi)moderately attenuated root/shoot elongation and biomass,with reduced chlorophylls in rice grown under hydroponic medium with 2 and 10μmol/L Cd,but led to more Cd accumulation.A field trial of rice grown in a realistic Cd-contaminated soil(0.40 mg/kg)showed that RNAi plants growth and development were also compromised compared to wild-type(WT),with smaller panicles and lower spikelet fertility but little effect on yield of ***,OsPDR20 suppression resulted in unexpectedly higher levels of Cd accumulation in rice straw including lower leaves and culm and *** results suggest that OsPDR20 is actively involved in Cd accumulation and homeostasis in rice *** increased Cd accumulation in the RNAi plants has the potential application in phytoremediation of Cd-polluted wetland soils.

Light-emitting diode(LED)irradiation can affect postharvest vegetable and fruit shelf life and nutritional *** this study,11 kinds of glucosinolates(GLS)and 6 types of GLS breakdown products were identified in cabbage *** investigate the effects of LED irradiation on quality and GLS metabolism,postharvest cabbage was irradiated with 25μmol/(m2·s)green LED(GL)light at room temperature for 12 *** dark condition and natural light were used as *** irradiation inhibited the respiration rate and the accumulation of malondialdehyde,and maintained higher cabbage sensory score,chlorophyll content,and antioxidant enzyme activities,thus extending its postharvest shelf ***,GL irradiation promoted GLS profile accumulation,including glucoraphanin,glucoiberin,gluconapin,gluconasturtiin,glucobrassicin,and sinigrin,which could have resulted from upregulated expression of the transcription factors MYB28 and MYB51,and the key genes involved in GLS biosynthesis(CYP83A1,CYP83B1,ST5b,and UGT74B1).Furthermore,GLS degradation was promoted by this treatment during the late stage of shelf life,as higher myrosinase activity and relative MYR and ESM expression levels were found in treated cabbage leaves,resulting in higher isothiocyanate(ITC)and indole-3-carbinol contents after day ***,GL irradiation was beneficial for cabbage preservation.

Straw return increases crop production by improving soil organic carbon sequestration and soil aggregation in a long-term wheat-cotton cropping system

Straw return is a promising strategy for managing soil organic carbon(SOC)and improving yield ***,the optimal straw return strategy for sustainable crop production in the wheat(Triticum aestivum L.)-cotton(Gossypium hirsutum L.)cropping system remains *** objective of this study was to quantify the long-term(10 years)impact of carbon(C)input on SOC sequestration,soil aggregation and crop yields in a wheat-cotton cropping system in the Yangtze River Valley,*** treatments were arranged with a single-factor randomized design as follows:no straw return(Control),return of wheat straw only(Wt),return of cotton straw only(Ct),return of 50%wheat and 50%cotton straw(Wh-Ch)and return of 100%wheat and 100%cotton straw(Wt-Ct).In comparison to the Control,the SOC content increased by 8.4 to 20.2%under straw return.A significant linear positive correlation between SOC sequestration and C input(1.42-7.19 Mg ha^(−1)yr^(−1))(P2 and 1-2 mm at the 0-20 cm soil depth were also significantly elevated under straw return,with the greatest increase of the aggregate stability in the Wt-Ct treatment(28.1%).The average wheat yields increased by 12.4-36.0%and cotton yields increased by 29.4-73.7%,and significantly linear positive correlations were also detected between C input and the yields of wheat and *** average sustainable yield index(SYI)reached a maximum value of 0.69 when the C input was 7.08 Mg ha^(−1)yr^(−1),which was close to the maximum value(SYI of 0.69,C input of 7.19 Mg ha^(−1)yr^(-1))in the Wt-Ct ***,the return of both wheat and cotton straw was the best strategy for improving SOC sequestration,soil aggregation,yields and their sustainability in the wheat-cotton rotation system.

Lacticaseibacillus rhamnosus Fmb14 ameliorates hyperuricemia-induced hepatocyte pyroptosis via NLRP3 inflammasome cascade inhibition

Enhanced organic degradation and microbial community cooperation by inoculating Bacillus licheniformis in low temperature composting

Microbial activity and interaction are the important driving factors in the start-up phase of food waste composting at low *** aim of this study was to explore the effect of inoculating Bacillus licheniformis on the degradation of organic components and the potential microbe-driven mechanism from the aspects of organic matter degradation,enzyme activity,microbial community interaction,and microbial metabolic *** results showed that after inoculating ***,temperature increased to 47.8℃ on day2,and the degradation of readily degraded carbohydrates(RDC)increased by 31.2%,and the bioheat production increased by 16.5%.There was an obvious enhancement of extracellular enzymes activities after inoculation,especially amylase activity,which increased by 7.68 times on day *** inoculated *** colonized in composting as key genus in the start-up *** network analysis and Mantel test indicated that inoculation drove the cooperation between microbial network modules who were responsible for various organic components(RDC,lipid,protein,and lignocellulose)degradation in the start-up *** function prediction suggested that carbohydrate metabolisms including starch and sucrose metabolism,glycolysis/gluconeogenesis,pyruvate metabolism,etc.,were improved by increasing the abundance of related functional genes after *** conclusion,inoculating *** accelerated organic degradation by driving the cooperation between microbial network modules and enhancing microbial metabolism in the start-up phase of composting.

Optimization of fertilizer performances in long-term garlic cropping soils

Continuous cropping is a common pattern of modern agriculture that takes regional advantages for crop yield *** the progress of mono-cropping continuously supported by intensive fertilizer inputs,such a cropping pattern often undergoes serious problems with low fertilizer use efficiencies and unsustainable crop *** this study,we dealt with a>25-year continuous garlic cropping system as an example for a problem-solving *** garlic cropping soils underwent problems characterized by loss of soil organic matter,dramatic retention of NH_(4)^(+)-N,and excess accumulation of phosphate and potash *** hydroponic simulations,we revealed that the presence of NH_(4)^(+)-N inhibited the root uptake of NO_(3)^(-)-N and K by 68% and 88%,*** the traditionally emphasized importance of K,we observed the negative effect of high K on the growth of garlic *** field experiments demonstrated that P and K applications can be reduced by 60% and 50%,respectively,without loss of *** thus developed a high-performance fertilization strategy by integrating a recomposed NPK fertilizer formulation to reduce unnecessary P and K inputs,a supplementary application of long-lasting C of woody peat to compensate for the soil C loss,and a foliar K approach to strengthen the stomatal function improvement with *** strategy allowed a 15% increase of garlic yield and a seasonal soil C profit of ca.1.8 Mg ha^(-1)even at ca.30% lower fertilizer *** study would be helpful in managing garlic fertilization and developing compound fertilizers,with broader significance for other long-term cropping soils.
