Chemoselective labeling-based spermatozoa glycan imaging reveals abnormal glycosylation in oligoasthenotspermia

Spermatogenesis, maturation, capacitation and fertilization are precisely regulated by glycosylation. However, the relationship between altered glycosylation patterns and the onset and development of reproductive disorders is unclear, mainly limited by the lack of in situ imaging techniques for spermatozoa glycosylation. We developed an efficient and highly specific spermatozoa glycan imaging technique based on the robust chemoselective labeling of sialic acid(Sia) and N-acetyl-D-galactosamine(Gal/GalNAc). We further proposed a “tandem glycan chemoselective labeling” strategy to achieve simultaneous imaging of two types of glycans on spermatozoa. We applied the developed method to the spermatozoa from oligozoospermic patients and diabetic mice and found that these spermatozoa showed higher levels of Sia and Gal/Gal NAc expression than the normal groups. Moreover, spermatozoa from diabetic mice showed a severe decrease in number, viability, and forward motility, suggesting that in vivo glucose metabolism disorders may lead to an elevated level of spermatozoa glycosylation and have a correlation with the development of oligoasthenotspermia. Our work provides a research tool to reveal the relationship between glycosylation modification and spermatozoa quality, and a promising clue for the development of glycan-based reproductive markers.

The COVID-19 pandemic has caused severe global disasters,highlighting the importance of understanding the details and trends of epidemic transmission in order to introduce efficient intervention measures.While the widely used deterministic compartmental models have qualitatively presented continuous “analytical” insight and captured some transmission features,their treatment usually lacks spatiotemporal variation.Here,we propose a stochastic individual dynamical(SID)model to mimic the random and heterogeneous nature of epidemic propagation.The SID model provides a unifying framework for representing the spatiotemporal variations of epidemic development by tracking the movements of each individual.Using this model,we reproduce the infection curves for COVID-19 cases in different areas globally and find the local dynamics and heterogeneity at the individual level that affect the disease outbreak.The macroscopic trend of virus spreading is clearly illustrated from the microscopic perspective,enabling a quantitative assessment of different interventions.Seemingly,this model is also applicable to studying stochastic processes at the “meter scale”,e.g.,human society’s collective dynamics.

Jurassic integrative stratigraphy,biotas,and paleogeographical evolution of the Qinghai-Tibetan Plateau and its surrounding areas

The Qinghai-Tibetan Plateau experienced a unique geological evolution during the Jurassic,driven by the termination of the Palaeotethys and the reduction of the Neotethys.The Indian Plate separated from the northern margin of Gondwana and drifted northward from the Southern Hemisphere.Given that the timing of strata serves as the basis for reconstructing geological history,the present work aimed to develop a new multiple stratigraphic and chronologic framework for the Jurassic strata of the Qinghai-Tibetan Plateau region via a synthesis of the material on lithostratigraphy,palaeontology,iso-radiometric dating,magnetostratigraphy,and other techniques with an emphasis on recent progress and findings.The new framework included the Jurassic System from the four major subdivisions of the plateau:the Baryan Har,Qiangtang,Lhasa-Gandise,and Southern Xizang(Himalaya).Ultimately,a more complete,refined biostratigraphic sequence was proposed,comprising the most common fossils in the plateau and those that are stratigraphically significant for the Jurassic stratigraphy,including ammonites,bivalves,brachiopods,foraminifera,radiolarians,and dinoflagellate cysts for the marine strata,and pollen and spores,and charophytes for the terrestrial sediments.This biostratigraphic framework was correlated with the Jurassic international standard zonation of the Geological Time Scale 2020 via standard or representative species or genera of ammonites.Based on this framework,we constructed a lateral correlation of the Jurassic strata between different basins of the plateau.The palaeontologic correlation in the present work shows that the Lhasa-Gandise Block had a closer relationship with the Qiangtang Block than with the Southern Xizang Himalaya during the Jurassic Period.Meanwhile,the Lhasa-Gandise Block and Qiangtang Block shared similar marine fauna features of the north marginal East Tethys.This contrasts the opinion suggesting that the Yarlung Zangbo Tethys was a small back-arc basin.A combination of stratigraphical,palaeontological,a

Mott Gap Filling by Doping Electrons through Depositing One Sub-Monolayer Thin Film of Rb on Ca_(2)CuO_(2)Cl_(2)

Understanding the doping evolution from a Mott insulator to a superconductor probably holds the key to resolve the mystery of unconventional superconductivity in copper oxides. To elucidate the evolution of the electronic state starting from the Mott insulator, we dose the surface of the parent phase Ca_(2)CuO_(2)Cl_(2) by depositing Rb atoms, which are supposed to donate electrons to the CuO_(2) planes underneath. We successfully achieved the Rb sub-monolayer thin films in forming the square lattice. The scanning tunneling microscopy or spectroscopy measurements on the surface show that the Fermi energy is pinned within the Mott gap but close to the edge of the charge transfer band. In addition, an in-gap state appears at the bottom of the upper Hubbard band(UHB), and the Mott gap will be significantly diminished. Combined with the Cl defect and the Rb adatom/cluster results, the electron doping is likely to increase the spectra weight of the UHB for the double occupancy. Our results provide information to understand the electron doping to the parent compound of cuprates.

Moyamoya disease(MMD)is a chronic occlusive cerebrovascular disease with the development of a network of abnormal vessels.Immune inflammation is associated with the occurrence and development of MMD.However,the mechanisms underlying the formation of the abnormal vascular network remain unclear.Twenty-eight patients with MMD,26 ischemic stroke patients,and 26 unrelated healthy volunteers were enrolled in this study The data showed that the levels of granulocyte-macrophage colony-stimulating factor(GM-CSF)were higher in MMD patients than in healthy controls(P<0.01),and GM-CSF was mainly from Th1 and Th17 cells in MMD.We found that increased GM-CSF drove monocytes to secrete a series of cytokines associated with angiogenesis,inflammation,and chemotaxis.In summary,our findings demonstrate for the first time the important involvement of GM-CSF in MMD and that GM-CSF is an important factor in the formation of abnormal vascular networks in MMD.

IL-17 induces NSCLC cell migration and invasion by elevating MMP19 gene transcription and expression through the interaction of p300-dependent STAT3-K631 acetylation and its Y705-phosphorylation

The cancer cell metastasis is a major death reason for patients with non-small cell lung cancer(NSCLC).Although researchers have disclosed that interleukin 17(IL-17)can increase matrix metalloproteinases(MMPs)induction causing NSCLC cell metastasis,the underlying mechanism remains unclear.In the study,we found that IL-17 receptor A(IL-17RA),p300,p-STAT3,Ack-STAT3,and MMP19 were up-regulated both in NSCLC tissues and NSCLC cells stimulated with IL-17.p300,STAT3 and MMP19 overexpression or knockdown could raise or reduce IL-17-induced p-STAT3,Ack-STAT3 and MMP19 level as well as the cell migration and invasion.Mechanism investigation revealed that STAT3 and p300 bound to the same region(−544 to−389 nt)of MMP19 promoter,and p300 could acetylate STAT3-K631 elevating STAT3 transcriptional activity,p-STAT3 or MMP19 expression and the cell mobility exposed to IL-17.Meanwhile,p300-mediated STAT3-K631 acetylation and its Y705-phosphorylation could interact,synergistically facilitating MMP19 gene transcription and enhancing cell migration and invasion.Besides,the animal experiments exhibited that the nude mice inoculated with NSCLC cells by silencing p300,STAT3 or MMP19 gene plus IL-17 treatment,the nodule number,and MMP19,Ack-STAT3,or p-STAT3 production in the lung metastatic nodules were all alleviated.Collectively,these outcomes uncover that IL-17-triggered NSCLC metastasis involves up-regulating MMP19 expression via the interaction of STAT3-K631 acetylation by p300 and its Y705-phosphorylation,which provides a new mechanistic insight and potential strategy for NSCLC metastasis and therapy.

A sustainable process to 100%bio-based nylons integrated chemical and biological conversion of lignocellulose

Considerable progress has been made in recent years to the development of sustainable polymers from bio-based feedstocks.In this study,100%bio-based nylons were prepared via an integrated chemical and biological process from lignocellulose.These novel nylons were obtained by the melt polymerization of 3-propyladipic acid derived from lignin and 1,5-pentenediamine/1,4-butanediamine derived from carbohydrate sugar.Central to the concept is a three-step noble metal free catalytic chemical funnelling sequence(Raney Ni mediated reductive catalytic fractionation-reductive funnelling-oxidative funnelling),which allowed for obtaining a single component 3-propyladipic acid from lignin with high efficiency.The structural and thermodynamic properties of the obtained nylons have been systematically investigated,and thus obtained transparent bio-based nylons exhibited higher Mw(>32,000)and excellent thermal stability(Td5%>265℃).Considering their moderate Tg and good melt strength,these transparent bio-based nylons could serve as promising functional additives or temperature-responsive materials.

Effects of dense planting patterns on photosynthetic traits of different vertical layers and yield of wheat under different nitrogen rates

A two-year field experiment was conducted to measure the effects of densification methods on photosynthesis and yield of densely planted wheat.Inter-plant and inter-row distances were used to define ratefixed pattern(RR)and row-fixed pattern(RS)density treatments.Meanwhile,four nitrogen(N)rates(0,144,192,and 240 kg N ha-1,termed N0,N144,N192,and N240)were applied with three densities(225,292.5,and 360×10^(4)plants ha^(-1),termed D225,D292.5,and D360).The wheat canopy was clipped into three equal vertical layers(top,middle,and bottom layers),and their chlorophyll density(Ch D)and photosynthetically active radiation interception(FIPAR)were measured.Results showed that the response of Ch D and FIPAR to N rate,density,and pattern varied with different layers.N rate,density,and pattern had significant interaction effects on Ch D.The maximum values of whole-canopy Ch D in the two seasons appeared in N240 combined with D292.5 and D360 under RR,respectively.Across two growing seasons,FIPAR values of RR were higher than those of RS by 29.37%for the top layer and 5.68%for the middle layer,while lower than those of RS by 20.62%for the bottom layer on average.With a low N supply(N0),grain yield was not significantly affected by density for both patterns.At N240,increasing density significantly increased yield under RR,but D360 of RS significantly decreased yield by 3.72%and 9.00%versus D225 in two seasons,respectively.With an appropriate and sufficient N application,RR increased the yield of densely planted wheat more than RS.Additionally,the maximum yield in two seasons appeared in the combination of D360 with N144 or N192 rather than of D225 with N240 under both patterns,suggesting that dense planting combined with an appropriate N-reduction application is feasible to increase photosynthesis capacity and yield.

Curve Classification Based onMean-Variance Feature Weighting and Its Application

The classification of functional data has drawn much attention in recent years.The main challenge is representing infinite-dimensional functional data by finite-dimensional features while utilizing those features to achieve better classification accuracy.In this paper,we propose a mean-variance-based(MV)feature weighting method for classifying functional data or functional curves.In the feature extraction stage,each sample curve is approximated by B-splines to transfer features to the coefficients of the spline basis.After that,a feature weighting approach based on statistical principles is introduced by comprehensively considering the between-class differences and within-class variations of the coefficients.We also introduce a scaling parameter to adjust the gap between the weights of features.The new feature weighting approach can adaptively enhance noteworthy local features while mitigating the impact of confusing features.The algorithms for feature weighted K-nearest neighbor and support vector machine classifiers are both provided.Moreover,the new approach can be well integrated into existing functional data classifiers,such as the generalized functional linear model and functional linear discriminant analysis,resulting in a more accurate classification.The performance of the mean-variance-based classifiers is evaluated by simulation studies and real data.The results show that the newfeatureweighting approach significantly improves the classification accuracy for complex functional data.

Downregulation of Serum PTEN Expression in Mercury-Exposed Population and PI3K/AKT Pathway-Induced Inflammation

Objective This study investigated the impact of occupational mercury(Hg) exposure on human gene transcription and expression, and its potential biological mechanisms.Methods Differentially expressed genes related to Hg exposure were identified and validated using gene expression microarray analysis and extended validation. Hg-exposed cell models and PTEN lowexpression models were established in vitro using 293T cells. PTEN gene expression was assessed using qRT-PCR, and Western blotting was used to measure PTEN, AKT, and PI3K protein levels. IL-6 expression was determined by ELISA.Results Combined findings from gene expression microarray analysis, bioinformatics, and population expansion validation indicated significant downregulation of the PTEN gene in the high-concentration Hg exposure group. In the Hg-exposed cell model(25 and 10 μmol/L), a significant decrease in PTEN expression was observed, accompanied by a significant increase in PI3K, AKT, and IL-6 expression.Similarly, a low-expression cell model demonstrated that PTEN gene knockdown led to a significant decrease in PTEN protein expression and a substantial increase in PI3K, AKT, and IL-6 levels.Conclusion This is the first study to report that Hg exposure downregulates the PTEN gene, activates the PI3K/AKT regulatory pathway, and increases the expression of inflammatory factors, ultimately resulting in kidney inflammation.
