BACKGROUND Colorectal polyps,which are characterized by a high recurrence rate,represent preneoplastic conditions of the *** to unclear mechanisms of pathogenesis,first-line therapies for non-hereditary recurrent colorectal polyps are limited to endoscopic *** recent studies suggest a mechanistic link between intestinal dysbiosis and polyps,the exact compositions and roles of bacteria in the mucosa around the lesions,rather than feces,remain *** To clarify the composition and diversity of bacteria in the mucosa surrounding or 10 cm distal to recurrent intestinal *** Mucosal samples were collected from four patients consistently with adenomatous polyps(Ade),seven consistently with non-Ade(Pol),ten with current Pol but previous Ade,and six healthy individuals,and bacterial patterns were evaluated by 16S rDNA *** discriminant analysis and Student’s t-tests were used to identify the genus-level bacteria differences between groups with different colorectal polyp ***’s correlation coefficients were used to evaluate the correlation between intestinal bacteria at the genus level and clinical *** The results confirmed a decreased level of probiotics and an enrichment of pathogenic bacteria in patients with all types of polyps compared to healthy *** changes were not restricted to the mucosa within 0.5 cm adjacent to the polyps,but also existed in histologically normal tissue 10 cm distal from the *** differences in bacterial diversity were observed in the mucosa from individuals with normal conditions,Pol,and *** abundance of Gram-negative bacteria,including Klebsiella,Plesiomonas,and Cronobacter,was observed in Pol group and Ade group,suggesting that resistance to antibiotics may be one risk factor for bacterium-related harmful ***,age and gender were linked to bacteria changes,indicating the potential involvement of sex hormone

Objective The activities and products of carbohydrate metabolism are involved in key processes of ***,its relationship with hepatocellular carcinoma(HCC)is *** The cancer genome atlas(TCGA)-HCC and ICGC-LIRI-JP datasets were acquired via public *** expressed genes(DEGs)between HCC and control samples in the TCGA-HCC dataset were identified and overlapped with 355 carbohydrate metabolism-related genes(CRGs)to obtain differentially expressed CRGs(DE-CRGs).Then,univariate Cox and least absolute shrinkage and selection operator(LASSO)analyses were applied to identify risk model genes,and HCC samples were divided into high/low-risk groups according to the median risk ***,gene set enrichment analysis(GSEA)was performed on the risk model *** sensitivity of the risk model to immunotherapy and chemotherapy was also *** A total of 8 risk model genes,namely,G6PD,PFKFB4,ACAT1,ALDH2,ACYP1,OGDHL,ACADS,and TKTL1,were ***,the risk score,cancer status,age,and pathologic T stage were strongly associated with the prognosis of HCC *** the stromal score and immune score had significant negative/positive correlations with the risk score,reflecting the important role of the risk model in immunotherapy ***,the stromal and immune scores had significant negative/positive correlations with risk scores,reflecting the important role of the risk model in immunotherapy ***,we found that high-/low-risk patients were more sensitive to 102 drugs,suggesting that the risk model exhibited sensitivity to chemotherapy *** results of the experiments in HCC tissue samples validated the expression of the risk model *** Through bioinformatic analysis,we constructed a carbohydrate metabolism-related risk model for HCC,contributing to the prognosis prediction and treatment of HCC patients.

Amyotrophic lateral sclerosis refers to a neurodegenerative disease involving the motor system,the cause of which remains unexplained despite several years of ***,the journey to understanding or treating amyotrophic lateral sclerosis is still a long *** to current research,amyotrophic lateral sclerosis is likely not due to a single factor but rather to a combination of mechanisms mediated by complex interactions between molecular and genetic *** progression of the disease involves multiple cellular processes and the interaction between different complex mechanisms makes it difficult to identify the causative factors of amyotrophic lateral ***,we review the most common amyotrophic lateral sclerosis-associated pathogenic genes and the pathways involved in amyotrophic lateral sclerosis,as well as summarize currently proposed potential mechanisms responsible for amyotrophic lateral sclerosis disease and their evidence for involvement in amyotrophic lateral *** addition,we discuss current emerging strategies for the treatment of amyotrophic lateral *** the emergence of these new therapies may help to further our understanding of the pathogenic mechanisms of the disease.

Enhanced Grain Refining Effect of Mg–Zr Master Alloy on Magnesium Alloys via a Synergistic Strategy Involving Heterogeneous Nucleation and Solute‑Driven Growth Restriction

Zirconium(Zr)emerges as the most effective grain refiner for magnesium(Mg)alloys incorporating ***,Zr is introduced in the form of an Mg–Zr master ***,within Mg–Zr master alloys,Zr predominantly exists in a particle form,which tends to aggregate due to attractive van der Waals *** clustered Zr is prone to settling,thereby reducing its refining impact on Mg *** this work,a combined pretreatment process for Mg–Zr master alloys was proposed,encompassing the introduction of a physical field to intervene the agglomeration of particle Zr and the employ of high-temperature dissolution and peritectic reactions to promote the solid solution of *** results demonstrate that the particle Zr within the pretreated Mg–Zr master alloy is effectively dispersed and refined,and greater solute Zr levels can be *** subsequent grain refinement ability was studied on a typical Mg–6Zn–0.6Zr(wt%)*** outcome highlights that an improvement in the grain refinement efficacy(32.4%)of Mg–Zr master alloys was obtained with a holding time of 60 *** pretreated Mg–Zr master alloy significantly augments the efficiency of grain refinement for Mg alloys through a synergistic strategy involving heterogeneous nucleation and solute-driven growth *** crucial factor in achieving effective grain refinement of Zr in Mg alloys lies in regulating the presence and morphology of Zr in the Mg–Zr master alloy,distinguishing between particle Zr and solute *** study introduces a novel method for developing more efficient Mg–Zr refiners.

The current impedance spectroscopy measurement techniques face difficulties in diagnosing solar cell faults due to issues such as cost,complexity,and ***,a novel system was developed for precise broadband impedance spectrum measurement of solar cells,which was composed of an oscilloscope,a signal generator,and a sampling *** results demonstrate concurrent accurate measurement of the impedance spectrum(50 Hz-0.1 MHz)and direct current voltametric *** analysis with Keithley 2450 data yields a global relative error of approximately 6.70%,affirming the *** excitation signals(sine,square,triangle,pulse waves),sine wave input yields the most accurate data,with a root mean square error of approximately 13.3016 and a global relative error of approximately 4.25%compared to theoretical *** reference resistance expands the half circle in the impedance *** of reference resistance to that of the solar cell enhances the accuracy by mitigating line resistance *** error is lower in high-frequency regions due to a higher signal-to-noise ratio.

The consistency of the cell has a significant impact on battery capacity,endurance,overall performance,safety,and service life ***,it is challenging to identify cells with high consistency and no loss of battery *** paper presents a cell screening algorithm that integrates genetic and numerical differentiation ***,a mathematical model for battery consistency is established,and a multi-step charging strategy is proposed to satisfy the demands of fast charging ***,the genetic algorithm simulates biological evolution to efficiently search for superior cell combinations within a short time while evaluating capacity,voltage consistency,and charge/discharge ***,through experimental validation and comparative analysis with similar algorithms,our proposed method demonstrates notable advantages in terms of both search efficiency and performance.

A Review of Type 1 and Type 2 Intraductal Papillary Neoplasms of the Bile Duct

Intraductal papillary neoplasm of the bile duct(IPNB)is a heterogeneous disease similar to intraductal papillary mucinous neoplasm of the *** lesions have been recognized as one of the three major precancerous lesions in the biliary tract since *** 2018,Japanese and Korean pathologists reached a consensus,classifying IPNBs into type l and type 2 *** are more prevalent in male patients in East Asia and are closely related to diseases such as cholelithiasis and *** a molecular genetic perspective,IPNBs exhibit early genetic variations,and different molecular pathways may be involved in the tumorigenesis of type 1 and type 2 *** histological subtypes of IPNBs include gastric,intestinal,pancreaticobiliary,or oncocytic subtypes,but type 1 IPNBs typically exhibit more regular and well-organized histological features than type 2 IPNBs and are more commonly found in the intrahepatic bile ducts with abundant *** to the rarity of these lesions and the absence of specific clinical and laboratory features,imaging is crucial for the preoperative diagnosis of IPNB,with local bile duct dilation and growth along the bile ducts being the main imaging *** resection remains the optimal treatment for IPNBs,but negative bile duct margins and the removal of lymph nodes in the hepatic hilum significantly improve the postoperative survival rates for patients with IPNBs.

The pathogenic mechanism of TAR DNA-binding protein 43(TDP-43)in amyotrophic lateral sclerosis

The onset of amyotrophic lateral sclerosis is usually characterized by focal death of both upper and/or lower motor neurons occurring in the motor cortex,basal ganglia,brainstem,and spinal cord,and commonly involves the muscles of the upper and/or lower extremities,and the muscles of the bulbar and/or respiratory ***,as the disease progresses,it affects the adjacent body regions,leading to generalized muscle weakness,occasionally along with memory,cognitive,behavioral,and language impairments;respiratory dysfunction occurs at the final stage of the *** disease has a complicated pathophysiology and currently,only riluzole,edaravone,and phenylbutyrate/taurursodiol are licensed to treat amyotrophic lateral sclerosis in many industrialized *** TAR DNA-binding protein 43 inclusions are observed in 97%of those diagnosed with amyotrophic lateral *** review provides a preliminary overview of the potential effects of TAR DNAbinding protein 43 in the pathogenesis of amyotrophic lateral sclerosis,including the abnormalities in nucleoplasmic transport,RNA function,post-translational modification,liquid-liquid phase separation,stress granules,mitochondrial dysfunction,oxidative stress,axonal transport,protein quality control system,and non-cellular autonomous functions(e.g.,glial cell functions and prion-like propagation).

Time-dependent deviation of bridge pile foundations caused by adjacent large-area surcharge loads in soft soils and its preventive measures

Time-dependent characteristics(TDCs)have been neglected in most previous studies investigating the deviation mechanisms of bridge pile foundations and evaluating the effectiveness of preventive *** this study,the stress-strain-time characteristics of soft soils were illustrated by consolidation-creep tests based on a typical engineering *** extended Koppejan model was developed and then embedded in a finite element(FE)model via a user-material subroutine(UMAT).Based on the validated FE model,the time-dependent deformation mechanism of the pile foundation was revealed,and the preventive effect of applying micropiles and stress-release holes to control the deviation was *** results show that the calculated maximum lateral displacement of the cap differs from the measured one by 6.5%,indicating that the derived extended Koppejan model reproduced the deviation process of the bridge cap-pile foundation with *** additional load acting on the pile side caused by soil lateral deformation was mainly concentrated within the soft soil layer and increased with the increase in load *** with t=3 d(where t is surcharge time),the maximum lateral additional pressure acting on Pile 2#increased by approximately 47.0%at t=224 *** bridge pile foundation deviation in deep soft soils,stress-release holes can provide better prevention compared to micropiles and are therefore recommended.
