Analysis of 12 variants in the development of gastric and colorectal cancers

AIM To evaluate the relation between 12 polymorphisms and the development of gastric cancer(GC) and colorectal cancer(CRC).MeTHODS In this study,we included 125 individuals with GC diagnosis,66 individuals with CRC diagnosis and 475 cancer-free individuals. All participants resided in the north region of Brazil and authorized the use of their samples. The 12 polymorphisms(in CASP8,CYP2 e1,CYP19 A1,IL1 A,IL4,MDM2,nFKB1,PAR1,TP53,TYMS,UGT1 A1 and XRCC1 genes) were genotyped in a single PCR for each individual,followed by fragment analysis. To avoid misinterpretation due to population substructure,we applied a previously developed set of 61 ancestryinformative markers that can also be genotyped by multiplex PCR. The statistical analyses were performed in Structure v.2.3.4,R environment and SPSS *** After statistical analyses with the control of confounding factors,such as genetic ancestry,three markers(rs79071878 in IL4,rs3730485 in MDM2 and rs28362491 in nFKB1) were positively associated with the development of GC. One of these markers(rs28362491) and the marker in the UGT1 A1 gene(rs8175347) were positively associated with the development of CRC. Therefore,we investigated whether the joint presence of the deleterious alleles of each marker could affect the development of cancer and we obtained positive results in all analyses. Carriers of the combination of alleles RP1 + deL(rs79071878 and rs28361491,respectively) are at 10-times greater risk of developing GC than carriers of other combinations. Similarly,carriers of the combination of deL + RARe(rs283628 and rs8175347) are at about 12-times greater risk of developing CRC than carriers of other *** These findings are important for the comprehension of gastric and CRC development,particularly in highly admixed populations,such as the Brazilian population.

We introduce the concept of truncated states obtained via iterative processes （TSI） and study its statistical features, making an analogy with dynamical systems theory （DST）. As a specific example, we have studied TSI for the doubling and the logistic functions, which are standard functions in studying chaos. TSI for both the doubling and logistic functions exhibit certain similar patterns when their statistical features are compared from the point of view of DST.

Genetic diversity and structure of Drimys brasiliensis in southern Brazil:insights for conservation

Population genetics studies are widely recognized for generating useful knowledge for biodiversity *** date,however,little is known about the levels and distribution of genetic diversity of Drymis brasiliensis(Miers LC),a tree species from the Atlantic ***,in this study,we investigated how genetic diversity is distributed within and among populations of *** from southern Brazil using allozyme markers to genotype reproductive trees(8 populations)and seedlings(3 populations).Furthermore,in two populations,we established two permanent plots(5.1 and 1 ha)to analyze fine-scale genetic structure(FSGS).Studied populations presented low levels of genetic diversity(reproductive=0.085;seedlings=0.054)and high fixation indexes(reproductive=0.396;seedlings=0.231).Genetic divergence among populations was equal to 0.05,which is significant,signaling that few populations can conserve large portions of the species total genetic *** was only detected for one population,when reproductive individuals were separated by less than40 *** genetic diversity combined with high fixation indexes clearly signal a risk of losing ***,conservation efforts should be aimed at enhancing gene flow within the studied populations.

Forensic data management and database systems in forensic investigations for cases of missing and unidentified persons in Brazil

Forensic investigations,especially those related to missing persons and unidentified remains,produce different types of data that must be managed and *** data collected and produced are extensive and originate from various sources:the police,non-governmental organizations(nGOs),medical examiner offices,specialised forensic teams,family members,and *** examples of information include,but are not limited to,the investigative background information,excavation data of burial sites,antemortem data on missing persons,and postmortem data on the remains of unidentified *** complex data must be stored in a secured place,analysed,compared,shared,and then reported to the investigative actors and the public,especially the families of missing persons,who should be kept informed of the ***,a data management system with the capability of performing the tasks relevant to the goals of the investigation and the identification of an individual,while respecting the deceased and their families,is critical for standardising *** management is crucial to assure the quality of investigative processes,and it must be recognised as a holistic integrated *** aim of this article is to discuss some of the most important components of an effective forensic data management *** discussion is enriched by examples,challenges,and lessons learned from the erratic development and launching of databases for missing and unidentified persons in *** main objective of this article is to bring attention to the urgent need for an effective and integrated system in Brazil.

Alzheimer's disease is a severe, highly disabling neurodegenerative disease, clinically characterized by a progressive decline in cognitive functions, and is the most common form of dementia in the elderly. For decades, the search for disease-modifying therapies has focused on the two main Alzheimer's disease histopathological hallmarks, seeking to prevent, mitigate, or clear the formation of extracellular aggregates of β-amyloid peptide and intracellular neurofibrillary tangles of tau protein, although without clinical success. Mesenchymal stem cell-based therapy has emerged as a promising alternative for the treatment of Alzheimer's disease, especially because it also targets other crucial players in the pathogenesis of the disease, such as neuroinflammation, synaptic dysfunction/loss, oxidative stress, and impaired neurogenesis. Herein, we review current knowledge of the therapeutic potential of mesenchymal stem cells and their extracellular vesicles for Alzheimer's disease, discussing the most recent findings in both preclinical and clinical trials as well as how advanced technologies have helped to overcome some limitations and contributed to stimulate the development of more effective treatments.

Identification of IL11RA and MeLK amplification in gastric cancer by comprehensive genomic profiling of gastric cancer cell lines

AIM To identify common copy number alterations on gastric cancer cell *** Four gastric cancer cell lines(ACP02, ACP03, AGP01 and PG100) underwent chromosomal comparative genome hybridization and array comparative genome hybridization. We also confirmed the results by fluorescence in situ hybridization analysis using the bacterial artificial chromosome clone and quantitative real time PCR *** The amplification of 9p13.3 was detected in all cell lines by both methodologies. An increase in the copy number of 9p13.3 was also confirmed by fluorescence in situ hybridization analysis. Moreover, the interleukin 11 receptor alpha(IL11RA) and maternal embryonic leucine zipper kinase(MeLK) genes, which are present in the 9p13.3 amplicon, revealed gains of the MeLK gene in all the cell lines studied. Additionally, a gain in the copy number of IL11 RA and MeLK was observed in 19.1%(13/68) and 55.9%(38/68) of primary gastric adenocarcinoma samples, respectively. COnCLUSIOn The characterization of a small gain region at 9p13.3 in gastric cancer cell lines and primary gastric adenocarcinoma samples has revealed MeLK as a candidate target gene that is possibly related to the development of gastric cancer.

Alterations in epigenetic control of gene expression play an important role in many diseases, including gastric cancer. Many studies have identified a large number of upregulated oncogenic mi RnAs and downregulated tumour-suppressor mi RnAs in this type of cancer. In this review, we provide an overview of the role of mi RnAs, pointing to their potential to be useful as diagnostic and/or prognostic biomarkers in gastric cancer. Moreover, we discuss the influence of polymorphisms and epigenetic modifications on mi RnA activity.

Flow regimes in wet conical spouted beds using glass bead mixtures

Using a high-viscosity newtonian fluid, glycerol, an experimental investigation was carried out to evaluate the stable spouting regime in conical spouted beds using four particle mixtures： a reference （monoparticles）, a binary mixture, two ternary mixtures with flat and Gaussian distributions respectively. The mixtures were selected for particle diameters （dp） ranging from 1.09 to 4.98 mm and particle diameter ratios （dpL/dps） ranging from 1.98 to 4.0. experimental data show that pressure fluctuation signals of the bed, as indicated by changes in their standard deviations, provide suitable information to identify the range of operational conditions for stable spouting. However, the analysis of skewness of curves of pressure fluctuation as a function of air velocity appears not sufficient to identify a particular flow regime. For glycerol in the spouting regime, the standard deviation is noted to increase with increasing glycerol concentration due to the growth of interparticle forces. The implications of these research findings on the drying of suspensions in conical spouted beds using glass bead mixtures are also discussed.

Reference genes for quantitative RT-PCR data in gastric tissues and cell lines

AIM:To evaluate the suitability of reference genes in gastric tissue samples and cell ***:The suitability of genes ACTB,B2M,GAPDH,RPL29,and 18S rRnA was assessed in21 matched pairs of neoplastic and adjacent nonneoplastic gastric tissues from patients with gastric adenocarcinoma,27 normal gastric tissues from patients without cancer,and 4 cell lines using reverse transcription quantitative real-time polymerase chain reaction(RT-qPCR).The ranking of the best single and combination of reference genes was determined by normFinder,genorm,BestKeeper,and *** addition,Genex software was used to determine the optimal number of reference *** validate the results,the mRnA expression of a target gene,DnMT1,was quantified using the different reference gene combinations suggested by the various software packages for ***:ACTB was the best reference gene for all gastric tissues,cell lines and all gastric tissues plus cell ***+B2M or ACTB+B2M was the best combination of reference genes for all the gastric *** the other hand,ACTB+B2M was the best combination for all the cell lines tested and was also the best combination for analyses involving all the gastric tissues plus cell *** to the Genex software,2 or 3 genes were the optimal number of references genes for all the gastric *** relative quantification of DnMT1 showed similar patterns when normalized by each combination of reference *** level of expression of DnMT1 in neoplastic,adjacent non-neoplastic and normal gastric tissues did not differ when these samples were normalized using GAPDH+B2M(P=0.32),ACTB+B2M(P=0.61),or GAPDH+B2M+ACTB(P=0.44).COnCLUSIOn:GAPDH+B2M or ACTB+B2M is the best combination of reference gene for all the gastric tissues,and ACTB+B2M is the best combination for the cell lines tested.
