TRAIL and Celastrol Combinational Treatment Suppresses Proliferation, Migration, and Invasion of Human Glioblastoma Cells via Targeting Wnt/β-catenin Signaling Pathway

Objective To investigate the mechanistic basis for the anti-proliferation and anti-invasion effect of tumor necrosis factor-related apoptosis-induced ligand(TRAIL)and celastrol combination treatment(TCCT)in glioblastoma *** Cell counting kit-8 was used to detect the effects of different concentrations of celastrol(0-16µmol/L)and TRAIL(0-500 ng/mL)on the cell viability of glioblastoma cells.U87 cells were randomly divided into 4 groups,namely control,TRAIL(TRAIL 100 ng/mL),Cel(celastrol 0.5µmol/L)and TCCT(TRAIL 100 ng/mL+celastrol 0.5µmol/L).Cell proliferation,migration,and invasion were detected by colony formation,wound healing,and Transwell assays,*** reverse transcription polymerase chain reaction and Western blotting were performed to assess the levels of epithelial-mesenchymal transition(EMT)markers(zona occludens,N-cadherin,vimentin,zinc finger E-box-binding homeobox,Slug,and β-catenin).Wnt pathway was activated by lithium chloride(LiCl,20 mol/L)and the mechanism for action of TCCT was *** Celastrol and TRAIL synergistically inhibited the proliferation,migration,invasion,and EMT of U87 cells(P<0.01).TCCT up-regulated the expression of GSK-3β and down-regulated the expression of β-catenin and its associated proteins(P<0.05 or P<0.01),including c-Myc,Cyclin-D1,and matrix metalloproteinase(MMP)-*** addition,LiCl,an activator of the Wnt signaling pathway,restored the inhibitory effects of TCCT on the expression of β-catenin and its downstream genes,as well as the migration and invasion of glioblastoma cells(P<0.05 or P<0.01).Conclusions Celastrol and TRAIL can synergistically suppress glioblastoma cell migration,invasion,and EMT,potentially through inhibition of Wnt/β-catenin *** underlies a novel mechanism of action for TCCT as an effective therapy for glioblastoma.

The issue of sensitivity attenuation in high-pressure region has been a persistent concern for pressure-sensitive electronic *** order to tackle such trade-off between sensitivity and linear range,herein,a hybrid piezoresistive-supercapacitive(HRSC)strategy is proposed via introducing a piezoresistive porous aerogel layer between the charge collecting electrodes and iontronic films of the pressure ***,the HRSC-induced impedance regulation and supercapacitive behavior contribute to significant mitigation in sensitivity attenuation,achieving high sensitivity across wide linear range(44.58 kPa^(−1)from 0 to 3 kPa and 23.6 kPa^(−1)from 3 to 12 kPa).The HRSC pressure sensor exhibits a low detection limit of 1 Pa,fast responsiveness(~130 ms),and excellent cycling stability,allowing to detect tiny pressure of air flow,finger bending,and human ***,the HRSC sensor exhibits exceptional perception capabilities for proximity and temperature,broadening its application scenarios in prosthetic perception and electronic *** proposed HRSC strategy may boost the ongoing research on structural design of high-performance and multimodal electronic sensors.

Heat stress induced hepatocyte apoptosis in largemouth bass Micropterus salmoides via IRE1α/TRAF2/ASK1/JNK pathway

Heat stress(HS)has been shown to adversely affect fish livers and can lead to extensive *** investigate the relationship between endoplasmic reticulum(ER)stress and HS-induced apoptosis in fish livers,we isolated and cultured primary hepatocytes of largemouth bass,Micropterus salmoides by trypsin method,then established an in-vitro model of liver cells under HS(35℃).The contents of lactic dehydrogenase(LDH)and hydrogen peroxide(H2O2)were determined to evaluate the effects of HS on hepatocyte injury and oxidative ***-qPCR was performed to discover the key genes in unfolded protein response(UPR)pathways involved at different HS *** inhibitor 4-PBA and IRE1αinhibitor 4μ8C were used to further investigate the effects of HS on IRE1αapoptosis pathway in *** show that HS led to significant increases in the release of LDH,the content of H2O2,and the expressions of oxidative protein folding genes(ero1αand pdi)under HS,suggesting severe hepatocyte injury and oxidative stress happened in heat-stressed largemouth bass *** continuous activation of IRE1αpathway genes(grp78,grp94,atf6,perk,eif2a,atf4,chop,ire1α,traf2,ask1,jnk1,and jnk2)indicated that HS led significantly to ER *** particular,the mRNA expression levels of ER stress-related genes(grp78,grp94,atf6,perk,ire1α,chop,jnk1,and jnk2)in the high temperature(HT)+4-PBA group and the HT+4μ8C group were significantly down-regulated under *** 4μ8C treatment,the expression levels of apoptosis-related genes(caspase-2,caspase-3,caspase-6,caspase-7,caspase-8,caspase-9,and caspase-10)and LDH content were significantly decreased,whereas the cell survival rate was significantly increased when given 4-PBA or 4μ8C *** findings demonstrate that HS could induce liver apoptosis of largemouth bass through the IRE1αpathway,which may act as a key switch mediating liver apoptosis of largemouth bass under HS.

Differential expression of miRNAs and mRNAs in the livers of largemouth bass Micropterus salmoides under heat stress

Global warming threatens freshwater ecosystems and compromises fish *** elucidate the role of miRNAs in the livers of heat stressed largemouth bass,juvenile fish was subject to heat stress under 37°*** mRNA-seq and miRNA-seq were conducted on the liver tissues under control and heat stress *** gene expression analysis and enrichment analysis were performed on mRNA and miRNA expression profiles.A total of 406 differentially expressed genes(DEGs)were discovered,of which 212 were up-regulated and 194 were *** of the DEGs were significantly implicated in the regulation of“protein processing in endoplasmic reticulum”,“proteasome”,“steroid biosynthesis”,and“ornithine decarboxylase inhibitor activity”*** addition,47 differentially expressed miRNAs(DEMs)were identified in largemouth bass livers under heat stress,including 21 up-regulated and 25 down-regulated miRNAs.A negatively regulated miRNA-mRNA network including 12 miRNAs and 19 mRNAs was constructed with DEMs involved in“protein degradation”,“calcium ion regulation”,“cell apoptosis”,and“lipid metabolism”.Moreover,this study indicated novel-miR-144 activated the IRE1 signaling pathway by targeting txndc5 to induce liver apoptosis in largemouth bass under heat *** study revealed the involvement of miRNA regulation in largemouth bass in response to heat stress.

In this study,we successfully synthesized Er_(2)TiO_(5)@Ag nanocomposites(NCPs) using the ultrasonicmediated sol-gel technique to create a multifunctional material with enhanced photocatalytic and antibacterial *** visible light photocatalytic activities of Er_(2)TiO_(5) nanoparticles(NPs) and Er_(2)TiO_(5)@Ag NCPs were systematically evaluated under various conditions,including different concentrations of Basic Blue 41(BB 41) dye and *** results reveal a remarkable improvement in the photocatalytic degradation efficiency of Er_(2)TiO_(5)@Ag NCPs(95%) compared to Er_(2)TiO_(5) NPs(80%).Furthermore,the antibacterial efficacy of Er_(2)TiO_(5) NPs and Er2TiO_5@Ag NCPs were extensively examined against Gram-positive and Gram-negative ***,Er_(2)TiO_(5)@Ag NCPs exhibit significantly higher minimum bactericidal concentration(MBC) values compared to Er_(2)TiO_(5) *** antibacterial effect of Er_(2)TiO_(5)@Ag NCPs is particularly pronounced against *** and Pseudomonas aeruginosa,while demonstrating moderate effects on Escherichia coli and Enterococcus *** assess the biocompatibility of the synthesized materials,we investigated their internalization by MCF-7 ***,both Er_(2)TiO_(5) NPs and Er_(2)TiO_(5)@Ag NCPs are found to be effectively internalized by the cells,suggesting their potential application in biomedical ***,our study unveils the exceptional potential of Er_(2)TiO_(5)@Ag NCPs as a dual-action solution,simultaneously possessing enhanced photocatalytic efficiency and potent antibacterial *** multifunctional nanocomposite not only outperforms Er_(2)TiO_(5) and Ag but also paves the way for innovative applications in sustainable environmental remediation and advanced biomedical technologies,promising a brighter and cleaner future.

Effect of prisms on visual acuity,contrast sensitivity and nystagmus in patients with albinism

AIM:To investigate the effect of using base-out prisms on nystagmus,visual acuity and contrast sensitivity in patients with ***:In this interventional study,patients with albinism who had nystagmus were enrolled.A comprehensive eye exam was conducted,which included refraction,assessment of far and near vision acuity,and contrast sensitivity *** check for the nystagmus,a videonystagmography was *** tests were carried out in three modes:without any correction,with optical correction,and with correction using base-out prisms in three different powers,including 4,6,and 8 prism ***:Totally 23 patients with average age of 28.65±12.13 were *** was found that the use of optical correction and optical correction with prisms resulted in a statistically significant improvement in both far(at least:P<0.006)and near visual acuity(at least:P<0.001 except for prism 8;P<0.02).In addition,contrast sensitivity significantly improved at all low and medium frequencies except for correction with prism 8 in frequency 1.5(at least:P<0.01 except for prism 4,frequency 6;P=0.04).no significant improvement was observed in the evaluation of nystagmus ***:Optical correction with a prism can improve visual acuity and some spatial frequencies,but failed to improve nystagmus parameters.

The Myc gene is the essential oncogene in triple-negative breast cancer(TNBC).This study investigates the synergistic effects of combining Myc decoy oligodeoxynucleotides-encapsulated niosomes-selenium hybrid nanocarriers with X-irradiation exposure on the MDA-MB-468 cell *** and scramble ODNs for Myc transcription factor were designed and synthesized based on promoter sequences of the Bcl2 *** nanocarriers were synthesized by loading Myc ODNs and selenium into chitosan(Chi-Se-DEC),which was then encapsulated in niosome-nanocarriers(NISM@Chi-Se-DEC).FT-IR,DLS,FESEM,and hemolysis tests were applied to confirm its characterization and physicochemical ***,cellular uptake,cellular toxicity,apoptosis,cell cycle,and scratch repair assays were performed to evaluate its anticancer effects on cancer *** anticancer assessments were repeated under X-ray irradiation conditions(fractionated 2Gy).Physicochemical characteristics of niosomes containing SeNPs and ODNs showed that it is synthesized *** revealed that the anticancer effect of NISM@Chi-Se-DEC can be significantly improved in combination with X-ray irradiation *** can be concluded that NISM@Chi-Se-DEC nanocarriers have the potential as a therapeutic agent for cancer treatment,particularly in combination with radiation therapy and in-vivo experiments are necessary to confirm the efficacy of this nano-drug.
