Abortifacient Potential of Aqueous Extract of Senna alata Leaves in Rats

Objective To study the abortifacient activity of aqueous extract of Senna alata leaves in female Wistar *** Pregnant rats weighing 143.65 ± 13.11 g were completely randomized into 5 groups（A-E）.Rats in group A（negative control） were orally administered,once daily with 0.5 ml of distilled water on days 10-18 post-coitum while those in groups B,C,D,and E were treated exactly like the negative control except they received the same volume containing 2.85 mg/kg body weight of mifepristone（group B,reference drug）,250,500 and 1 000 mg/kg body weight of the extract respectively（groups C-E）.Results Phytochemical screening of the extract showed positive results for saponins（1.22%）,flavonoids（1.06%）,cardiac glycosides（0.20%）,cardenolides and dienolides（0.18%）,phenolics（0.44%） and alkaloids（0.52%）.Respiratory distress,salivation,diarrhoea,changes in the appearance of hair as well as maternal mortality were not observed at any time during the exposure period except in the mifepristone-treated animals where there was episode of diarrhoea and *** extract significantly reduced（P〈0.05） the number of life foetus,weight and survival ratio of the foetus,numbers of implantations and Corpora lutea,implantation index,progesterone,prolactin,estradiol,follicle stimulating and luteinizing hormones whereas the number of dead foetus,number and percentage of rats that aborted,percentage vaginal opening,resorption index,pre-and post-implantation losses increased *** addition,mifepristone-treated animals produced resorption index that compared well with the distilled water *** was also no dead or life foetus and serum progesterone concentration was increased in the animals treated with *** cases of abortion were accompanied with vaginal ***,the final weight of the rats increased significantly,the feed and water intake were not significantly altered in all the treatment *** weight of the uterus,uterine-body weight ratio,le

Sulfonylureas are widely used oral anti-diabetic ***,its long-term usage effects on patients’lifespan remain controversial,with no reports of influence on animal ***,the anti-aging effects of chlorpropamide along with glimepiride,glibenclamide,and tolbutamide were studied with special emphasis on the interaction of chlorpropamide with mitochondrial ATP-sensitive K+(mito K-ATP)channels and mitochondrial complex *** delayed aging in Caenorhabditis elegans,human lung fibroblast MRC-5 cells and reduced doxorubicin-induced senescence in both MRC-5 cells and *** addition,the mitochondrial membrane potential and ATP levels were significantly increased in chlorpropamide-treated worms,which is consistent with the function of its reported targets,mito K-ATP *** levels of mitochondrial reactive oxygen species(mt ROS)were observed in chlorpropamide-treated ***,the lifespan extension by chlorpropamide required complex II and increased mt ROS levels,indicating that chlorpropamide acts on complex II directly or indirectly via mito K-ATP to increase the production of mt ROS as a pro-longevity *** study provides mechanistic insight into the anti-aging effects of sulfonylureas in ***.

Evaluation of joint toxicity of heavy metals and herbicide mixtures in soils to earthworms(Eisenia fetida)

It is widely acknowledged that a simplified and robust approach to evaluating thecombined effects of chemical mixtures is critical for ecological risk assessment(ERA)of contaminated *** earthworm(Eisenia fetida)was used as a model to study the combined effects of polymetallic contamination and the herbicide siduron in field soil using a microcosm *** responses of multiple biomarkers,including the activities of catalase(CAT),superoxide dismutase(SOD),glutathione reductase(GR)and acetylcholine esterase(AChE),the concentrations of glycogen,soluble protein(SP),malonaldehyde(MDA),and metallothionein(mt),and the neutral red uptake test(NRU),were *** analysis,Principal Component Analysis(PCA)and Spearman’s Rank Correlations analysis(BVSTEP)revealed that the activities of AChE and CAT and the NRU content were the prognostic biomarkers capturing the minimum data set of all the *** Cd(tissue Cd)in earthworms was closely related to the health status of worms under combined contamination of heavy metals and *** integrated effect(E mix)calculated based on the activities of AChE and CAT and NRU content using the stress index method had significantly linear regression with internal Cd(p<0.01).E mix(10),E mix(20),and E mix(50)were then calculated,at 1.27,1.63 and 2.71 mg/kg dry weight,*** could be concluded that a bioassaybased approach incorporating multivariate analysis and internal dose was pragmatic and applicable to evaluating combined effects of chemical mixtures in soils under the guidance of the top-down evaluation concept of combined toxicity.

How to accurately simulate the distribution of forest species based upon their biological attributes has been a traditional biogeographical *** gap models are very useful tools for examining the dynamics of forest succession and revealing the species structure of *** the present study,the GFSM(Gongga Forest Succession Model) was developed and applied to simulate the distribution,composition and succession process of forests in 100 m elevation *** results indicate that the simulated results of the tree species,quantities of the different types of trees,tree age and differences in DBH(diameter at breast height) composition were in line with the actual situation from 1400 to 3700 MASL(meters above sea level) on the eastern slope of ***,the dominant species in the simulated results were the same as those in the surveyed ***,the GFSM model can best simulate the features of forest dynamics and structure in the natural conditions of *** work provides a new approach to studying the structure and distribution characteristics of mountain ecosystems in varied ***,the results of this study suggest that the biogeochemistry mechanism model should be combined with the forestsuccession model to facilitate the ecological model in simulating the physical and chemical processes involved.

BackgroundIn the normal life cycle of the parasite(Echinococcus multilocularis)that causes alveolar echinococcosis,domestic and wild carnivores act as definitive hosts,and rodents act as intermediate *** presented study contributes to the research on the distribution and transmission pattern of *** in China having identified sheep as an unusual intermediate host taking part in the domestic transmission of alveolar echinococcosis in Gansu Province,*** 2020 to 2021,nine whitish different cyst-like were collected from the liver of sheep in Gansu Province for examination.A near complete mitochondrial(mt)genome and selected nuclear genes were amplified from the cyst-like lesion for *** confirm the status of the specimen,comparative analysis with reference sequences,phylogenetic analysis,and network analysis were *** isolates displayed≥98.87%similarity to *** NADH dehydrogenase sub-unit 1(nad1)(894 bp)reference sequences deposited in ***,amplification of the nad4 and nad2 genes also confirmed all nine samples as *** with>99.30%***,three nuclear genes,pepck(1545 bp),elp-exons VII and VIII(566 bp),and elp-exon IX(256 bp),were successfully amplified and sequenced for one of the isolates with 98.42%similarity,confirming the isolates were correctly identified as *** analysis also correctly placed the isolates with other *** a result of the discovery of *** in an unusual intermediate host,which is considered to have the highest zoonotic potential,the result clearly demonstrated the necessity for expanded surveillance in the area.

目的本研究旨在利用动态增强MR成像技术(DCE-MR)检测血液病患者骨髓强化特征变化,研究其与骨髓肿瘤细胞浸润状态的相关性。方法25例血液病经DCE-MR成像及髂嵴穿刺活检,测定骨髓灌注的最大强化率(PER),最大强化斜率值(Slopemax),峰值时间(TTP),平均时间(mt),以及骨髓活检分析肿瘤分数(TF)。结果肿瘤浸润组较缓解期组PER高(临床缓解期为0.252±0.156,肿瘤组为0.592±0.433),统计学上有显著性差异(P0.05),mt值两组差异不明显(P>0.05)。结论动态增强MR成像能够监测血液病骨髓肿瘤细胞浸润状态,为恶性血液病治疗随访提供了一种新的无创性监测手段。

Matrix metalloproteinases contribute to kidney fibrosis in chronic kidney diseases

Matrix metalloproteinases(MMPs) are members of the neutral proteinase family. They were previously thought to be anti-fibrotic because of their ability to degrade and remodel of extracellular matrix. However, recent studies have shown that MMPs are implicated in initiation and progression of kidney fibrosis through tubular cell epithelial–mesenchymal transition(Emt) as well as activation of resident fibroblasts, endothelial-mesenchymal transition(Endo mt) and pericyte-myofibroblast transdifferentiation. Interstitial macrophage infiltration has also been shown to correlate with the severity of kidney fibrosis in various chronic kidney diseases. MMPs secreted by macrophages, especially MMP-9, hasbeen shown by us to be profibrotic by induction of tubular cells Emt. Emt is mainly induced by transforming growth factor-β(TGF-β). However, MMP-9 was found by us and others to be up-regulated by TGF-β1 in kidney tubular epithelial cells and secreted by activated macrophages, resulting in Emt and ultimately kidney fibrosis. Therefore, MMP-9 may serve as a potential therapeutic target to prevent kidney fibrosis in chronic kidney disease. This review, by a particular focus on Emt, seeks to provide a comprehensive understanding of MMPs, especially MMP-9, in kidney fibrosis.
