Objective:To evaluate the efficacy of multiparametric magnetic resonance imaging(mp-MRI)using Prostate Imaging Reporting and Data System version 2.0(PI-RADSv2)definitions in detecting organ-confined prostate ***:All patients who underwent radical prostatectomy between January 1,2014 and December 30,2014 were *** underwent mp-MRI within 180 days before *** with prior pelvic irradiation or androgen deprivation therapy were *** embedded,whole-mount histopathology was centrally reviewed and correlated with imaging for tumour location,Gleason score(GS)and ***:There were 39 patients included,of which 35(90%)had mp-MRI done post-biopsy.A total of 93 cancer foci were identified on whole-mount pathology,of which mp-MRI detected 63(68%).Of those detected by mp-MRI,14 were PI-RADS 3(n=6 for GS 6,n=8 for GS 7,no GS≥8)and 49 were PI-RADS 4e5(nZ7 for GS 6,nZ33 for GS 7,and nZ9 for GS≥8).There were 30(32%)cancer foci missed by mp-MRI(n=15 for GS 6,n=13 for GS 7 and n=2 for GS≥8).A lesion classified as PI-RADS 4e5 predicted a higher grade cancer on pathology as compared to PI-RADS 3(for GS 7 lesions,odds ratio[OR]=3.53,95%CI:0.93e13.45,p=0.064).The mp-MRI size detection limit was 20 mm2 and 100 mm2 for 50%and 75%probability of cancer,*** associating with radiological and pathologic stage,the weighted Kappa value was 0.69(p<0.0001).The sensitivity and positive predictive values for this study were 68%(95%CI:57%e77%)and 78%(95%CI:67%e86%),***:In this predominantly post-biopsy cohort,mp-MRI using PI-RADSv2 reporting has a reasonably high diagnostic accuracy in detecting clinically significant prostate cancer.

Background: Nanotechnology symbolizes a broad discipline with enormous potential in cancer treatment bridging one of the bottlenecks of traditional approaches in cancer therapy which is an inability to deliver adequate quantities of anti-cancer drugs to the tumor area. Studies on nanoparticles indicate their importance in cancer angiogenesis and metastasis. Aim: The present study assessed anti-angiogenesis and anti-metastatic effects of biogenic silver nanoparticles (AgNPs) synthesized from neem plant (Azadirachta indica). Methods: Chicken chorioallantoic membrane (CAM) and two-dimensional (2D) wound healing assays were used to study anti-angiogenic and anti-metastatic effects of the AgNPs respectively. Twenty-four fertilized eggs were divided into four groups: two biogenic AgNPs treatments at 100 μg/ml and 200 μg/ml;negative control (1% DMSO) and positive control (cyclophosphamide). On day 8 of incubation, filter discs impregnated with different concentration levels of the treatments were placed on the CAM. On day 12 of incubation, the CAMs were imaged using a stereomicroscope, scaled using ImageJ, and different morphometric and spatial parameters computed using AngioTool software. Vessel area, vessel percent area, total number of junctions, total vessel length, average vessel length, mean lacunarity, and junction density were measured. The crown-rump length (CRL) and fetal weight were also recorded on day 16 of incubation. In order to determine relative gene expression profiles of iNOS and VEGF, total RNA was extracted from the CAM, and qRT-PCR was performed with β-Actin as a reference gene. For the 2D wound healing assay, DU145 human prostate cells were grown in Dulbecco’s Modified Eagle’s Medium supplemented with 10% Fetal Bovine Serum. Improving caregiving competence, stress coping, and mental well-being in informal dementia carers

AIM: To study the effectiveness of Reitman centre "Coaching, Advocacy, Respite, Education, Relationship, and Simulation"(CARERS) program, which uses problem-solving techniques and simulation to train informal dementia carers. METHODS: Seventy-three carers for family members with dementia were included in the pilot study. Preand post-intervention data were collected from carers using validated measures of depression, mastery, role captivity and overload, caregiving competence and burden, and coping styles. To assess program effec-tiveness, mean differences for these measures were calculated. One-way ANOVA was used to determine if change in scores is dependent on the respective baseline scores. Clinical effects for measures were expressed as Cohen's D values. RESULTS: Data from 73 carers were analyzed. The majority of these participants were female(79.5%). A total of 69.9% were spouses and 30.1% were children of the care recipient. Participants had an overall mean age of 68.34 ± 12.01 years. About 31.5% of participating carers had a past history of psychiatric illness(e.g., depression), and 34.2% sustained strained relationships with their respective care recipients. Results from carers demonstrated improvement in carers' selfperception of competence(1.26 ± 1.92, P < 0.0001), and significant reduction in emotion-focused coping(measured by the Coping Inventory of Stressful Situations,-2.37 ± 6.73, P < 0.01), Geriatric Depression scale(-0.67 ± 2.63, P < 0.05) and Pearlin's overload scale(-0.55 ± 2.07, P < 0.05), upon completion of the Program. Secondly, it was found that carers with more compromised baseline scores benefited most from the intervention, as they experienced statistically significant improvement in the following constructs: competence, stress-coping style(less emotion-oriented), sense of mastery, burden, overload. Overprescription of opioid analgesia is common following ambulatory Otolaryngology—Head and Neck surgery procedures: A multicenter study

Background: The rise in the use of prescription opioids for postoperative analgesia within surgery has mirrored an increased trend of opioid-related morbidity within Canada and the United States. This study prospectively studied daily pain levels and medication requirements postoperatively in patients undergoing elective Otolaryngology—Head and Neck surgery ***: Patients were asked to prospectively document their pain level and medication use daily for 7 days postoperatively. A final survey was used to quantify unused medication left at home and clarify each patient's disposal plan. We included patients undergoing elective outpatient or short stay surgeries from three tertiary care centers in Toronto, Ontario from September 2016 to September 2017. Previous opioids users or patients suffering from chronic pain were ***: A final cohort of 56 eligible adult patients were included in the study. The most common procedures were thyroidectomy (n = 19), endoscopic sinus surgery (n= 10), tympanoplasty/ossiculoplasty (n= 7), and cochlear implant (n = 5). Most patients received a prescription for acetaminophen/codeine (n = 29, 51.8%) or acetaminophen/oxycodone (n = 22, 39.3%) and used on average 29% of their initial prescription. Patients most commonly opted to keep their unused narcotics at home (n = 23, 41%). A total of 710 tablets of narcotics were overprescribed in our study population, 351 of which were kept in patients'' home for future ***: There is a clear tendency to overestimate postoperative pain resulting in significant overprescription of opioids among Otolaryngologists.

The Effect of Catena Position on Greenhouse Gas Emissions from Dambo Located Termite (<i>Odontotermes transvaalensis</i>) Mounds from Central Zimbabwe

Methane (CH4), carbon dioxide (CO2) and nitrous oxide (N2)O) are greenhouse gases (GHGs) which cause global warming. Natural sources of GHGs include wetlands and termites. Previous studies have quantified GHG emissions from upland termites and no study has reported GHG emissions from seasonal wetlands (dambo) located termite mounds. The objective of this study was to evaluate the effect of dambo catena position on termite mound distribution and GHG emissions. It was hypothesized that mound density and GHG emissions from Odontotermes transvaalensis mounds, vary with catena position. The evaluated catena positions were margin, mid-slope, lower slope and bottom. Mound density was significantly lower in the bottom when compared to the other catena positions. The mean GHG fluxes were 88 μg m2 hr-1, 0.78 mg m2 hr-1 and 1361 mg m2 hr-1 for N2) O, CH4 and CO2 respectively. Fluxes varied with catena position and were 0.48, 0.72, 1.35 and 0.79 mg m-2 hr-1 for CH4 , and 1173.7, 1440.7, 1798.7 and 922.8 mg m-2 hr-1 for CO2 in the margin, mid-slope, lower slope and the bottom catena position respectively. For N2) O, there were no significant differences between catena positions. It was concluded that dambo located Odontotermes transvaalensis termite mounds are an important source of GHGs, and emissions varied with catena position for CO2 and CH4.

Linking gold mineralization to regional-scale drivers of mineral systems using in situ U-Pb geochronology and pyrite LA-ICP-MS element mapping

Proterozoic orogens commonly host a range of hydrothermal ores that form in diverse tectonic settings at different times. However, the link between mineralization and the regional-scale tectonothermal evolution of orogens is usually not well understood, especially in areas subject to multiple hydrothermal ***-scale drivers for mineral systems vary between the different classes of hydrothermal ore, but all involve an energy source and a fluid pathway to focus mineralizing fluids into the upper crust. The mount Olympus gold deposit in the Proterozoic Capricorn Orogen of Western Australia, was regarded as an orogenic gold deposit that formed at ca. 1738 Ma during the assembly of Proterozoic Australia. However,the trace element chemistry of the pyrite crystals closely resembles those of the Carlin deposits of Nevada,with rims that display solid solution gold accompanied by elevated As, Cu, Sb, Hg, and Tl, surrounding gold-poor cores. New SHRIMP UeP b dating of xenotime intergrown with auriferous pyrite and ore-stage alteration minerals provided a weighted mean^(207) Pb*/^(206) Pb* date of 1769 ± 5 Ma, interpreted as the age of gold mineralization. This was followed by two discrete episodes of hydrothermal alteration at 1727 ± 7 Ma and 1673 ± 8 Ma. The three ages are linked to multiple reactivation of the crustal-scale Nanjilgardy Fault during repeated episodes of intracratonic reworking. The regional-scale drivers for Carlin-like gold mineralization at mount Olympus are related to a change in tectonic regime during the final stages of the intracratonic 1820 -1770 Ma Capricorn Orogeny. Our results suggest that substantial sized Carlin-like gold deposits can form in an intracratonic setting during regional-scale crustal reworking.

Miombo woodlands near human settlements are under significant pressures from human activities,with negative consequences on their structure and *** studies are limited,we assessed the structure and species composition of a portion of miombo woodland along an increasing disturbance gradient from a national park,through a buffer zone to communal lands in northeast *** concentric plots of 2 and 11 m radii were established in each area to record woody species composition,diameter,height,basal area,density and volume as well as evidence of *** of site,growth stage and their interaction on vegetation diversity and structural parameters were tested using a general linear model(GLM).Principal component analysis(PCA) tested the association between species and site and ANOVA the differences in the level of disturbance across *** diversity did not differ between sites except for evenness,which increased with *** and richness were greater in seedlings and saplings than mature trees across sites,*** and mature tree diameters differed significantly between *** and density of mature trees declined with increasing disturbance while seedling densities peaked at intermediate disturbance levels(buffer zone).Tree harvesting,was more evident in the buffer zone and in the communal area relative to the national *** contrast,fire frequency was greater in the national park and in the buffer zone relative to the communal *** results of this study identify a true miombo woodland dominated by Brachystegia boehmii with a stable population,as illustrated by an inverse-J shape in diameter class distribution on all sites,and that these woodlands are generally resilient to disturbances,maintaining similar species composition and structure at various levels of ***,continued monitoring of disturbance levels and miombo woodland response is recommended to ensure sustainable utilisation of

The SHC1 adaptor protein directly binds to all members of the HER/ERBB receptor tyrosine kinase family and forms a critical link between the receptors and downstream effectors1,2. Since SHC1 only binds to receptors in their activated states, it is well positioned to monitor the translation of signals from the receptors to downstream targets which is of critical importance in ERBB-driven cancers. We affinity purified SHC1 followed by parallel reaction monitoring(PRM)-based quantification to measure changes in SHC1 complex formation in 19 cancer cell lines with various ERBB levels1. We targeted 35 SHC interactors for quantitation. Our data showed that the cell lines varied significantly in the type and number of proteins(from 6 to 32 proteins) that were recruited to SHC1. We further captured temporal profiles of the SHC1 complexes before and during EGF stimulation in four ERBB2/HER2 positive breast cancer cell lines(SKBR3, BT-474, HCC-1954, HCC-1419) with different sensitivity towards trastzumab3, a clinical inhibitor for ERBB2/HER2. We detected significantly stronger association of promitogenic components of SHC1 complexes at basal level in drug-sensitive cells over drug-resistant cells. This pattern is not observed for components involved in negatively regulation ERBB signaling. Moreover, the duration, but not the maximum level, of the association for pro-mitogenic components is apparently longer in drug-sensitive cells, suggesting defects in signal down regulation in these cells. Not all of these observations could be predicted from either m RNA or protein expression levels. Taking together, our analysis of dynamic SHC1 complex formation supports SHC1 as a function-base probe for monitoring ERBB activation status and a potential diagnostic tool for drug sensitivity prediction.
