molecular testing sensitivity,which allows for early diagnosis of the 2019 coronavirus disease(cOVId-19),could be affected by sample quality,storage,and transportation timeframe to the laboratory,along with bias related to the pre-analytic *** present study reports the selection and decontamination of nasopharyngeal samples during cOVId-19 management at the institut pasteur côte d’*** objective of this work was to organize sample reception management and report a complete picture of sample selection and decontamination in the context of diagnosis activity *** administrative note creating the selection and decontamination unit of nasopharyngeal samples initiated activities in May *** required human resources and necessary materials were identified and put in *** activity consisted of receiving,sorting,decontaminating,and sending nasopharyngeal samples to different diagnostic *** were recorded *** a six-month period of activities,from a total amount of 11,401 containers received and decontaminated,174,085 samples were selected.A proportion of 92.0%of these specimens met the diagnostic standards,while 7.0%that were found acceptable showed minor ***,a rate of 1.0%of samples with major abnormalities could not be used for cOVId-19 testing and,therefore,were ***,the non-conformity rate was reduced by 2.4%after the first term *** and decontamination of nasopharyngeal samples are crucial steps in biosafety optimization for the technical staff and quality improvement of sample care.

Since its outbreak in december 2019 in Wuhan Province (china), the coronavirus (cOVId-19) disease quickly spread around the world in such a way that most response plans were outdated. There was an urgent need to change and adapt response strategies as the virus globally spread. Entire firms and economies were brought to a standstill in order to reduce the virus’ capacity to spread and to limit some of the short-term impacts in order to save time and find out solutions to come back to a more or less normal way of life. Thus, most of the countries that closed their air, sea and land borders had to reopen them progressively, with travel restrictions submitted to rigid controls. In côte d’ivoire, as in all other countries, air travellers leaving the territory were required to provide a certificate for a negative cOVId-19 test, valid for 24 to 72 hours depending on the country of destination. However, the national system implemented could not provide a result before 48 hours. The objective of this work was to develop an alternative strategy to the system for air travellers who were in a hurry and those who had a computer bug in obtaining their result. A total of 38,444 air travellers benefited from this strategy implemented by the institut pasteur de côte d’ivoire during these two years.

Background: Some national and international strategies for the detection and prevention of emerging infectious diseases have been established across sectors. The capacity to carry out these tasks varies from country to country, and that remains largely undervalued. The pasteur institute of c?te d’ivoire has created and implemented the capacity of national reference centers to fight against emerging and other infectious diseases. Objective: Show on the one hand the strategies used to develop the National Reference centers and the IPcI and on the other hand the results obtained by performing these strategies. Method: datas collection by documentary analysis (published scientific articles and grey literature) was done on Google Scholar, PUBMEd and institutional reference documents. The documentary research was carried out to have a better understanding of strategies used to create and develop the NRcs in microbiology of communicable diseases. Results: Seven integrated strategies were launched: 1-training and workforce development;2-investigation of epidemics or public health events;3-strengthening laboratory epidemiological research;4-strengthening surveillance systems;5-improving communication with partners and stakeholders;6-building national and international collaborations;and 7-strengthening technical and technological platforms. In two decades, the number of researchers has risen from 10 in 2004 to ninety (90) in 2021, with 12 senior researchers and 32 junior researchers. A number of health service staff had attended a qualifying training course, 27 investigations into outbreaks and other public health events had been carried out, 18 short-term research projects had been launched, major surveillance programs and epidemiological research efforts on vector-borne, food-borne and nosocomial infections had begun, and several scientific manuscripts had been published or were edited in the writting press. conclusion: The Ivorian experience shows that, with concerted eff

Achievements, challenges and prospects for the development of broadly protective multivalent vaccines and therapeutic antibodies against hand, foot and mouth disease

Hand, foot and mouth disease(HFMd) is a significant health concern in the Asia–Pacific regions for infants and young children in recent years. However, no vaccines or therapeutics are available at present. The causative agents for HFMd include human enterovirus 71(EV71), coxsackievirus A16(cVA16) and some other viruses. Recently, tremendous progress has been made in the development of monovalent and bivalent vaccines against HFMd. A few neutralizing monoclonal antibodies against EV71 or cVA16 have been identified and characterized. Here, we reviewed some achievements for the development of broadly protective vaccines and neutralizing antibodies against HFMd, and discussed challenges and prospects toward broadly protective multivalent vaccines and therapeutic antibodies against HFMd.

cryptosporidium spp. infection is one of the causes of diarrhea in people living with HIV/AIdS. The objective of this study is to compare the sensitivity of microscopy and molecular biology to determine the prevalence of cryptosporidium spp. in Patients Living With HIV (PLWH). This is a descriptive cross-sectional study conducted in three care centers for people living with HIV/AIdS in Abidjan. It took place from November 2018 to March 2020. Sociodemographic data were obtained via a questionnaire. Stool and blood samples were collected and analyzed for microscopy and Nested PcR detection of cryptosporidium spp. Blood samples were analyzed for cd4+ count. A total of 363 stool samples were collected from the three sites. Individuals aged 40 - 50 years (36.52%) were most likely to participate in the study. HIV Type 1 accounted for 86.22% of the study population. The samples collected consisted of 47.65% diarrheal stool. Microscopic examination of the stool yielded a prevalence of 3.86% for cryptosporidium spp. while the prevalence was 3.96% with molecular identification. No statistically significant difference was observed between these two prevalences (χ2 = 0.26;p = 0.609). cd4+ count was the factor associated with cryptosporidium spp. infection for both microscopy (OR = 0.887, p = 0.001) and PcR (OR = 0.896, p = 0.001). This study demonstrated that Nested PcR improves the detection of cryptosporidium spp. in patient diagnosis.

The effect of host plants on genotype variability in fitness and honeydew composition of Aphis fabae

Aphid species can be polyphagous, feeding on multiple host plants across genera. As host plant species can have large variation in their phloem composition, this can affect aphid fitness and honeydew composition. Previous research showed significant intraspecific genotype variation in the composition of the honeydew carbohydrates of the black bean aphid Aphis fabae, with the ant attractant trisaccharide melezitose showing especially large variation across different genotypes. In this study, we test if variation in melezitose and carbohydrate composition of aphid honeydew could be linked to the adap- tation of specific aphid genotypes to particular host plants. To this end, 4 high and 5 low melezitose secreting genotypes of the black bean aphid Aphisfabae were reared on 4 com- mon host plants： broad bean, goosefoot, beet, and poppy. The carbohydrate composition, and in particular melezitose secretion, showed important aphid genotype and host plant in- teractions, with some genotypes being high melezitose secreting on 1 host plant but not on another. However, the interaction effects were not paralleled in the fitness measurements, even though there were significant differences in the average fitness across the different host plants. On the whole, this study demonstrates that aphid honeydew composition is influenced by complex herbivore-plant interactions. We discuss the relevance of these findings in the context of ant-aphid mutualisms and adaptive specialization in aphids.

HIV-induced membraneless organelles orchestrate post-nuclear entry steps

HIV integration occurs in chromatin sites that favor the release of high levels of viral progeny;alternatively, the virus is alsoable to discreetly coexist with the host. The viral infection perturbs the cellular environment inducing the remodelling of thenuclear landscape. Indeed, HIV-1 triggers the nuclear clustering of the host factor cPSF6, but the underlying mechanism ispoorly understood. Our data indicate that HIV usurps a recently discovered biological phenomenon, called liquid–liquid phaseseparation, to hĳack the host cell. We observed cPSF6 clusters as part of HIV-induced membraneless organelles (HIV-1 MLOs) inmacrophages, one of the main HIV target cell types. We describe that HIV-1 MLOs follow phase-separation rules and representfunctional biomolecular condensates. We highlight HIV-1 MLOs as hubs of nuclear reverse transcription, while the double-strandedviral dNA, once formed, rapidly migrates outside these structures. Transcription-competent proviruses localize outside but nearHIV-1 MLOs in LEdGF-abundant regions, known to be active chromatin sites. Therefore, HIV-1 MLOs orchestrate viral events priorto the integration step and create a favorable environment for the viral replication. This study uncovers single functional host–viralcomplexes in their nuclear landscape, which is markedly restructured by HIV-1.

Although IL-12 plays a critical role in priming Th1 and cytotoxic T lymphocyte(cTL) responses, Toll-like receptor(TLR) signaling only induces low amounts of IL-12 in dendritic cells and macrophages, implying the existence of stringent regulatory mechanisms. In this study, we sought to uncover the mechanisms underlying TLR-induced IL-12 expression and the Th1 response. By systemic screening, we identified a number of protein kinases involved in the regulation of TLRinduced IL-12 expression. In particular, PI3 K, ERK, and m TOR play critical roles in the TLR-induced Th1 response by regulating IL-12 and IL-10 production in innate immune cells. Moreover, we identified c-fos as a key molecule that mediates m TOR-regulated IL-12 and IL-10 expression in TLR signaling. Mechanistically, m TOR plays a crucial role in c-fos expression, thereby modulating NFκB binding to promoters of IL-12 and IL-10. By controlling the expression of a special innate gene program, m TOR can specifically regulate the TLR-induced T cell response in vivo. Furthermore, blockade of m TOR by rapamycin efficiently boosted TLR-induced antigen-specific T and B cell responses to HBV and HcV vaccines. Taken together, these results reveal a novel mechanism through which m TOR regulates TLR-induced IL-12 and IL-10 production, contributing new insights for strategies to improve vaccine efficacy.

Hepatitis c virus vaccine development: old challenges and new opportunities

Hepatitis c virus(HcV), an enveloped positive-sense single-stranded RNA virus, can cause chronic and end-stage liver diseases. Approximately 185 million people worldwide are infected with HcV. Tremendous progress has been achieved in the therapeutics of chronic hepatitis c thanks to the development of direct-acting antiviral agents(dAAs), but the worldwide use of these highly efective dAAs is limited due to their high treatment cost. In addition, drug-resistance mutations remain a potential problem as dAAs are becoming a standard therapy for chronic hepatitis c. Unfortunately, no vaccine is available for preventing new HcV infection. herefore, HcV still imposes a big threat to human public health, and the worldwide eradication of HcV is critically dependent on an efective HcV vaccine. In this review, we summarize recent progresses on HcV vaccine development and present our views on the rationale and strategy to develop an efective HcV vaccine.
