Application of wastewater-based epidemiological monitoring of COVID-19 for disease surveillance in the city

Wastewater-based surveillance serves as a supplementary approach to clinical surveillance of COVID-19 during the *** study aimed to track the prevalence of the disease and the viral genetic variability through wastewater-based surveillance in the post-epidemic *** January to December 2023,samples were collected from the influent lines of two wastewater treatment plants(WWTPs),concentrated using PEG8000,and subjected to detection of the target genes ORF 1ab and N of SARS-CoV-2 via reverse transcriptional quantitative PCR(RT-qPCR).For next-generation sequencing(NGS),high-quality samples from both wastewater and clinical patients were *** analysis were performed using R software to evaluate the correlation between the SARSCoV-2 RNA concentrations in wastewater and positive rate of reported cases,indicating a positive *** diversity patterns of SARS-CoV-2 in wastewater resembled those in the patient source based on Principal Coordinates Analysis(PCoA)with three clusters for different *** rise of RNA concentration in wastewater indicates the growth of cases and the emergence of new variants,serving as an early warning of potential viral mutations,disease outbreaks even possible ***,the genomic surveillance of wastewater could help identify new variants that may not be captured through population monitoring,especially when sample sizes are ***,surveillance of SARS-CoV-2 in municipal wastewater has emerged as a reliable,earlywarning monitoring system for COVID-19 in the post-epidemic era.

Tripartite partnership, compared with the bilateral model, is envisioned as an opportunity to foster stronger and more trusting partnerships [1]. However, China has historically been characterized as principally reliant on bilateral engagement in global health [2]. The China-Tanzania Cooperation Project on Malaria control was conducted by National Institute of Parasitic diseases at China CDC and Chinese center for Tropical diseases Research, Ifakara Health Institute in Tanzania and funding agencies from 2015 to 2022, with an aim to explore the applicability of Chinese experience on malaria control to reduce malaria disease burden in the local context of Tanzania. In the tripartite project funded by the former UK Department for International Development (DFID) and later the Bill & Melinda Gates Foundation (BMGF), China's partners were expected to share the extensively parallel practices achieved from the control and elimination of malaria for over 70 years, UK's partners were to deliver resources and design global health strategies, while Tanzanian local staff collaborated with triangular partners to ensure the project implementation [3]. In the project, the trilateral cooperation has transparent management mechanism to ensure the concrete management and implementation of the project [4] (Additional file 1: Figure S1). This Brief is to provide recommendations for China's engagement in global health by distilling the experience from the innovative tripartite partnership of the project.

Real-world study(RWS)gained prominence starting with a significant investigation on ramipril’s impact on hypertension in 1993(1).A key advancement for the Food and Drug Administration(FDA)occurred with the enactment of the 21st Century Cures Act in 2016,emphasizing RWS(2).The FDA’s Real-world Evidence Program establishes real-world data(RWD)as“data relating to patient health status and/or the delivery of health care routinely collected from a variety of sources”,while real-world evidence(RWE)is“clinical evidence about the usage and potential benefits or risks of a medical product derived from analysis of RWD”(3).The proliferation of big data,electronic medical records(EMR),electronic health records(EHR),and medical claims data offers vast information resources facilitating *** access to RWD allows for worldwide monitoring of the impacts of various public health initiatives like vaccination *** randomized clinical trials(RCTs)traditionally gauge vaccine efficacy and shortterm safety,RWS can assess vaccine performance and safety across larger,more diverse populations and are adept at identifying rare events not easily discernible in RCTs due to their infrequency.

Coexistence of multiple coronaviruses in several bat colonies in an abandoned mineshaft

Since the 2002–2003 severe acute respiratory syndrome(SARS) outbreak prompted a search for the natural reservoir of the SARS coronavirus, numerous alpha- and betacoronaviruses have been discovered in bats around the world. Bats are likely the natural reservoir of alpha- and betacoronaviruses, and due to the rich diversity and global distribution of bats, the number of bat coronaviruses will likely increase. We conducted a surveillance of coronaviruses in bats in an abandoned mineshaft in mojiang County, Yunnan Province, China, from 2012–2013. Six bat species were frequently detected in the cave: Rhinolophus sinicus, Rhinolophus affinis, Hipposideros pomona, Miniopterus schreibersii, Miniopterus fuliginosus, and Miniopterus fuscus. By sequencing PCR products of the coronavirus RNA-dependent RNA polymerase gene(Rd Rp), we found a high frequency of infection by a diverse group of coronaviruses in different bat species in the mineshaft. Sequenced partial Rd Rp fragments had 80%–99% nucleic acid sequence identity with well-characterized Alphacoronavirus species, including Bt CoV HKU2, Bt CoV HKU8, and Bt CoV1,and unassigned species Bt CoV HKU7 and Bt CoV HKU10. Additionally, the surveillance identified two unclassified betacoronaviruses, one new strain of SARS-like coronavirus, and one potentially new betacoronavirus species. Furthermore, coronavirus co-infection was detected in all six bat species, a phenomenon that fosters recombination and promotes the emergence of novel virus strains. Our findings highlight the importance of bats as natural reservoirs of coronaviruses and the potentially zoonotic source of viral pathogens.
