Shankhpushpi is a reputed drug from an Indian system of medicine for treating mental disorders and enhancing memory. Two herbs, namely Convolvulus prostratus Forssk. and Evolvulus alsinoides(L.) L., are commonly known as Shankhpushpi. Ambiguous vernacular identity can affect the scientific validity of the Shankpushpi-based herbal drug therapy. In the present investigation, a novel and sensitive multiplex PCR method based on polymorphism in the internal transcribed spacer(ITS) region was developed to establish the molecular identity of C. prostratus and E. alsinoides. DNA was isolated and the ITS region was amplified, sequenced and assembled. Sequences were aligned to identify variable nucleotides in order to develop plant-specific primers. Primers were validated in singleplex reactions and eventually a multiplex assay was developed. This assay was tested for sensitivity and validated by amplifying DNA isolated from the simulated blended powdered plant material. Primers developed for C. prostratus resulted into a 200 bp amplicon and 596 bp for E. alsinoides. The assay was found to be sensitive enough for amplification of low quantities of DNA. The method can detect 10% of the mixing of plants with each other in blended material. This PCR assay can be used for rapid botanical identification of Shankhpushpi plant materials and will improve evidence-based herbal drug therapy.

Aim: To observe the alterations in the biochemical and biophysical changes in the sperm membrane during sperm maturation in male rats treated with the water extract of the fruit pericarp of S. mukorossi. Methods: Adult male Sprague-Dawley rats were gavaged the aqueous extract of the fruit pericarp of S. mukorossi at a dose of 50 mg/kg/d for 45 days. On day 46, the sperm parameters were observed in different sections of the epididymis and the sperm superoxide dismutase and the lipid peroxidation was determined and compared with the controls. The testis and epididymis were routinely prepared for histological examination under the light microscope. Results: No significant differences in the sperm number and morphology were observed between the control and treated groups. However, a significant inhibition (Pbnormal distribution of the superoxide dismutase activity, being minimum in the caput and maximum in the corpus, which was just opposite to that of the controls. Conclusion: The study provides a unique observation where the plant extract alters the sperm membrane physiology without change the testicular and epididymal morphology.

Plants show phenotypic changes when challenged with herb.v.rous insects. The mechanisms underlying these changes include the activation of transcriptional responses, which are dependent on the attacking insect. Most transcriptomic studies on crucifer-insect interactions have focused on the model plant Arabidopsis thaliana, a species that faces low herb.v.re pressure in nature. Here, we study the transcriptional responses of plants from a wild black mustard （brassica nigra） population to herb.v.res of different feeding guilds using an A. thaliana-based whole-genome microarray that has previously been shown to be suitable for transcriptomic analyses in brassica. Transcriptional responses of b. nigra after infestation with either Pieris rapae caterpillars or brevicoryne brassicae aphids are analyzed and compared. Additionally, the insect-induced expression changes of some individual genes are analyzed through quantitative real-time polymerase chain reaction. The results show that feeding by both insect species results in the accumulation of transcripts encoding proteins involved in the detoxification of reactive oxygen species, defensive proteins and glucosinolates and this is correlated with experimental evidence in the literature on such biochemical effects. Although genes encoding proteins involved in similar processes are regulated by both insects, there was little overlap in the induction or repression of individual genes. Furthermore, P. rapae and b. brassicae seem to affect different phytohormone signaling pathways. In conclusion, our results indicate that b. nigra activates several defense-related genes in response to P rapae or b. brassicae feeding, but that the response is dependent on the attacking insect species.

Poly(glycolic acid)is a biocompatible as well as biocomposable polymer with superior mechanical and barrier properties and,consequently,has found important applications in both medical and packaging ***,the high hydrolysis rate in a high humidity environment restricts its *** this work,a solid-state drawing process after melt extrusion is applied in order to produce fibrous PGA with enhanced mechanical properties and a much better resistance towards *** crystal structure of PGA gradually transformed from spherulites into oriented fibrous crystals in the stretching direction upon solid-state ***,both the length of microfibril and the size of lamellae increased initially with the drawing ratio(DR),while the chain-folded lamellae transformed into extended-chain fibrils at high(er)*** oriented structures lead to an overall improvement of the mechanical properties of PGA,e.g.,the tensile strength increased from 62.0±1.4 MPa to 910±54 MPa and the elongation at break increased from around 7%to 50%.Meanwhile,the heat capacity of totally mobile amorphous PGA(∆C_(p)^(0)=0.64 J·g^(−1)·℃^(−1))was reported for the first time,which was used to analyze the content of mobile amorphous fraction(XMAF)and rigid amorphous fraction(XRAF).both the oriented chain-folded lamellae crystals and the tightly arranged RAF are beneficial to prevent water molecules from penetrating the matrix,thus improving the resistance towards *** a consequence,the fibrous PGA with a DR of 5 showed a tensile strength retention rate of 17.3%higher in comparison with the undrawn sample after 7-days accelerated ***,this work provides a feasible method to improve the mechanical and resistance towards hydrolysis performance of PGA,which may broaden its application and prolong the shelf-life of PGA products.

Optical tweezers have elucidated numerous biological processes, particularly by enabling the precise manipulation and measurement of tension. One question concerns the biological relevance of these experiments and the generalizability of these experiments to wider biological systems. Here, we categorize the applicability of the information garnered from optical tweezers in two distinct categories: the direct relevance of tension in biological systems, and what experiments under tension can tell us about biological systems, while these systems do not reach the same tension as the experiment, still, these artificial experimental systems reveal insights into the operations of biological machines and life processes.
